Label free optofluidic DNA hybridization detection based on suspended core fiber whispering gallery mode resonator

Xuegang Li,Hongxin Zhang,Yiming Wang,Xue Zhou,Linh Viet Nguyen,Stephen C. Warren-Smith

An optofluidic DNA biosensor based on a suspended core fiber (SCF) has been proposed for the detection of specific DNA sequence fragments. The sensor is formed by coupling a micro/nano tapered optical fiber to the cladding of an SCF, producing a whispering gallery mode (WGM) effect. The WGM resonance is sensitive to samples within the channels of the SCF. The diameter of the SCF is reduced by etching to increase refractive index sensitivity, up to 274 nm/RIU. Biomarkers are captured on the inner surface of the microfluidic channel leading to a change in the local refractive index. The experimental results show that the sensor can quantitatively detect complementary DNA (cDNA) by immobilizing probe DNA (pDNA) on the surface of the microfluidic channel with a limit of detection of 1.5 nM. The proposed DNA biosensor provides a label-free method for DNA detection, which has great potential for multiplexed detection and realizing disease diagnostics.

Hyperboloid-Drum Microdisk Laser Biosensors for Ultrasensitive Detection of Human IgG

Zhihe Guo,Yingchun Qin,Peizong Chen,Jinliang Hu,Yi Zhou,Xuyang Zhao,Zhiran Liu,Yiyan Fei,Xiaoshun Jiang,Xiang Wu

Whispering gallery mode (WGM) microresonators have been used as optical sensors in fundamental research and practical applications. The majority of WGM sensors are passive resonators that require complex systems, thereby limiting their practicality. Active resonators enable the remote excitation and collection of WGM-modulated fluorescence spectra, without requiring complex systems, and can be used as alternatives to passive microresonators. This paper demonstrates an active microresonator, which is a microdisk laser in a hyperboloid-drum (HD) shape. The HD microdisk lasers are a combination of a rhodamine B-doped photoresist and a silica microdisk. These HD microdisk lasers can be utilized for the detection of label-free biomolecules. The biomolecule concentration can be as low as 1 ag mL −1 , whereas the theoretical detection limit of the biosensor for human IgG in phosphate buffer saline is 9 ag mL −1 (0.06 a M ). Additionally, the biosensors are able to detect biomolecules in an artificial serum, with a theoretical detection limit of 9 ag mL −1 (0.06 a M ). These results are approximately four orders of magnitude more sensitive than those for the typical active WGM biosensors. The proposed HD microdisk laser biosensors show enormous detection potential for biomarkers in protein secretions or body fluids.

MXene functionalized whispering gallery mode microbubble resonator for Pb2+ sensing

Lu Cai,Yu-xiao Liu,Hao Jing,Li-ke Li,Kai Gao,Jin-ze Zhang,Yu-jie Wang,Si-rui Huang

This study proposes a novel optical fiber sensor that integrates two-dimensional MXene materials with a microbubble whispering gallery mode (WGM) resonator for the detection of lead ions (Pb 2+ ). Utilizing the inherent negative surface charge of MXene nanosheets, a uniform functionalized film was formed on the inner surface of the microbubble cavity, enabling specific binding to Pb 2+ . Experimental results demonstrated a nonlinear response ( R 2  > 0.99) over a Pb 2+ concentration range of 0–1000 mg/L, with a detection limit of 5 mg/L, meeting the standard requirements for monitoring lead ions in industrial wastewater. The sensor also exhibits high selectivity toward Pb 2+ in the presence of interfering ions such as Zn 2+ , Ni 2+ , and Cd 2+ . Furthermore, owing to the inherent microfluidic channel of the microbubble cavity, this structure is suitable for real-time monitoring in complex aqueous environments, highlighting the great potential of the MXene-microbubble resonator platform for environmental heavy metal detection.

High quality two-photon pumped whispering-gallery-mode lasing from ultrathin CdS microflakes

Liyun Zhao,Qiuyu Shang,Yan Gao,Bao Jin,Tianyou Zhai,Qing Zhang

We report low threshold, room temperature two-photon pumped whispering-gallery-mode (WGM) lasing from CdS microflakes with thicknesses down to 58 nm on mica substrates grown by self-limited van der Waals epitaxy. The relationship between the lasing properties and microflake size, as well as the recombination origins, is explored towards chip-level microlasers.

High-Quality Hexagonal Nonlayered CdS Nanoplatelets for Low-Threshold Whispering-Gallery-Mode Lasing

Yang Mi,Bao Jin,Liyun Zhao,Jie Chen,Shuai Zhang,Jia Shi,Yangguang Zhong,Wenna Du,Jun Zhang,Qing Zhang,Tianyou Zhai,Xinfeng Liu

Low threshold micro/nanolasers have attracted extensive attention for wide applications in high-density storage and optical communication. However, constrained by quantum efficiency and crystalline quality, conventional semiconductor small-sized lasers are still subjected to a high lasing threshold. In this work, a low-threshold planar laser based on high-quality single-crystalline hexagonal CdS nanoplatelets (NPLs) using a self-limited epitaxial growth method is demonstrated. The as-grown CdS NPLs show multiple whispering-gallery-mode lasing at room temperature with a threshold of ≈0.6 µJ cm −2 , which is the lowest value among reported CdS-based lasers. Through power-dependent lasing studies at 77 K, the lasing action is demonstrated to originate from a exciton–exciton scattering process. Furthermore, the edge length- and thickness-dependent lasing threshold studies reveal that the threshold is inversely proportional to the second power of lateral edge length while partially affected by vertical thickness, and the lasing modes can be sustained in NPLs as thin as 60 nm. The lowest threshold emerges with the thickness of ≈110 nm due to stronger energy confinement in the vertical Fabry–Pérot cavity. The results not only open up a new avenue to fabricate nonlayered material-based coherent light sources, but also advocate the promise of nonlayered semiconductor materials for the development of novel optoelectronic devices.

Liquid crystal biosensing platform based on whispering gallery mode laser for Aβ42 detection

Peiyao An,Mengxiao Li,Shanshan Zhang,Di Lian,Chuqi Li,Chuan Wang,Miao Yu,Duo Wang,Lingyi Kong,Jun Ren,Yue Li,Haiyan Wang,Xin Yang,Zhenlin Wu

LC sensing platform based on WGM laser is developed for the AD-related biomarker Aβ42 detection. • The sensor sensitivity is approximately 0.003 nm/pg/mL, and the LOD is 3 pg/mL. • RNA aptamer is used as recognition probes to enhance the specificity of the biosensing platform. • The combination of RNA aptamer and Aβ42 causes RI changes, resulting in WGM spectral shifts. • The structure design provides an ideal sensing platform for detecting biomedical samples. Alzheimer's disease (AD) is an irreversible and progressively worsening neurodegenerative disorder. Amyloid beta (Aβ42) plays a crucial role in the early diagnosis of this disease. However, the concentration of Aβ42 in the blood is extremely low, which poses a challenge for Aβ42 detection. Herein, a liquid crystal (LC) biosensing platform based on whispering gallery mode (WGM) laser was developed for rapid and real-time monitoring of the AD-related biomarker Aβ42. The LC microcavities were functionally modified using cetyltrimethyl ammonium bromide (CTAB) and RNA aptamers. Based on this, the quantitative monitoring mechanism of Aβ42 was analyzed, stemming from the interaction between the negatively charged RNA aptamer and CTAB, leading to the reorientation of LC molecules. This alters the refractive index distribution within the microcavity, thereby enabling the shift of the WGM spectrum. The dynamic changes of molecular orientation in the LC microcavity were monitored by polarization optical microscopy and spectral analysis. The experimental results showed that the total shifts in the resonance spectrum were directly proportional to the concentration of Aβ42 within the range of 0–1000 pg/mL. The sensitivity of the sensor is 0.003 nm/pg/mL. The experiment further confirmed the practicability of this sensor in the mouse serum. The sensor is inexpensive, easy to manufacture and fast and it is potentially useful for AD-related biomarker screening after further validation.

Ultrafast Antisolvent Growth of Single-Crystal CsPbBr3 Microcavity for Whispering-Gallery-Mode Lasing

Li Zhang,Xinxin Li,Yimeng Song,Bingsuo Zou

In recent years, all-inorganic cesium lead bromide (CsPbBr3) perovskites have garnered considerable attention for their prospective applications in green photonics and optoelectronic devices. However, the development of efficient and economical methods to obtain high-quality micron-sized single-crystalline CsPbBr3microplatelets (MPs) has become a challenge. Here, we report the synthesis of CsPbBr3MPs on Si/SiO2substrate by optimizing the ultrafast antisolvent method (FAS). This technique is able to produce well-dispersed, uniformly sized, and morphologically regular tetragonal phase single crystals, which can give strong green emission at room temperature, with excellent stability and excitonic character. Moreover, the crystals demonstrated lasing with a whispering gallery mode with a low threshold. These results suggest that the single-crystalline CsPbBr3MPs synthesized by this method are of high optical quality, holding vast potential for future applications in photonic devices. Keywords:perovskites;fast growth;microstructures;lasing;photonics

Whispering gallery mode microtube sensor for real-time microfluidic monitoring of the coagulation process

Lu Cai,Hao Jing,Jin-ze Zhang,Hong-dan Wan,Zi-ting Lin,Kai Gao,Yu-xiao Liu,Yu-jie Wang,Yong Zhao

Thrombin time is an important coagulation indicator reflecting the function of the terminal common pathway of the coagulation cascade, and it holds significant clinical value in anticoagulation therapy monitoring and the assessment of hemorrhagic and thrombotic diseases. In this paper, an optical sensor based on whispering-gallery-mode microtube resonator is proposed for real-time and label-free monitoring of plasma thrombin time. The core element of the sensor is a silica microtube fabricated by HF etching of a hollow-core fiber. The wall thickness is optimized via simulation to maximize the evanescent field energy at the liquid core–inner wall interface. By covalently immobilizing thrombin on the inner wall of the microtube, local refractive index changes induced by fibrin deposition during the coagulation reaction can be sensitively captured by the evanescent field and converted into real-time shifts of the resonant wavelength. Experimental results show that the sensor exhibits good repeatability for plasma thrombin time measurement, with a standard deviation of 2.67 ms. The influence of anticoagulant drugs on thrombin time was further investigated. Thrombin time was prolonged as the concentration of heparin sodium increased, and a good linear relationship ( R ²=0.94) was obtained between heparin sodium concentration (in the range of 0∼0.3 U/mL) and thrombin time, indicating that the response of the coagulation process to anticoagulant drugs can be well monitored by the proposed method. The sensor features simple fabrication, low sample and reagent consumption, resistance to external environmental contamination, and ease of storage, providing a promising optical detection platform for point-of-care monitoring of the coagulation process.

Functionalised liquid crystal microfibers for hydrogen peroxide and catalase detection using whispering gallery mode

Rui Duan,Yanzeng Li,Yonggui Yuan,Lu Liu,Hanyang Li

In this work, we demonstrate, for the first time, a whispering gallery mode (WGM) lasing-based liquid crystal (LC) soft-matter microfiber biosensor for real-time and sensitive detection of hydrogen peroxide and catalase. The functionalised nematic LC 4-cyano-4ʹ-pentylbiphenyl (5CB) microfibers served as both optical microresonators and sensing actuators. The resonance spectral shift in WGMs can be used as a stable and reliable indicator of analyte concentrations. The detection limits for hydrogen peroxide and catalase were as low as ~0.26 μM and ~1 ng/mL, respectively. Direct measurement of the WGM lasing spectrum of LC soft-matter microfibers can be expected as an alternative method to detect analytes without the need for polarised image analysis in current LC sensor systems. This research provides a preliminary basis for the development of soft-matter microfiber biosensors with high sensitivity for detecting biochemical molecules.

Liquid Crystal Microcavity Biosensors for Real-Time Liver Injury Monitoring via Whispering Gallery Mode Laser

Jianwei Wang,Yeshuai Song,Xinyu Dou,Jiapeng Sun,Xinghua Yang,Yu Zhang,Zhihai Liu,Yanzeng Li,Hanyang Li

Real-time monitoring of liver injury is essential for preserving physiological health. Alanine aminotransferase (ALT) detection is widely regarded as a fundamental approach for the early diagnosis of liver injury. However, existing detection methods often suffer from complex operation, high costs, and limited sensitivity. Here, we introduce a real-time biosensor based on functionalized liquid crystal microcavities with whispering gallery mode (WGM) laser for rapid and sensitive ALT detection. The microcavity functionalized with stearic acid exhibits distinct optical responses to ALT-catalyzed enzymatic reactions across varying concentrations. The high-quality factor of the microcavity obviously enhances its biosensing performance. Simulations reveal variations in the electric field behavior of WGM lasing as the liquid crystal microcavity transitions between radial and bipolar configurations. Further analysis was conducted using experimental WGM spectra and corresponding polarized optical microscopy images. Experimental results demonstrate a strong linear correlation between ALT concentration and reaction time within the range of 0 to 240 U/l. This biosensor exhibits a sensitivity of 0.67 s/(U/l), indicating its potential as a promising approach for early liver injury assays. In addition to in vitro verification, in vivo validation using mouse serum samples further confirms its practical applicability, yielding results consistent with those tested by using commercial assay kits. This method offers a simple, cost-effective, and efficient detection of ALT, underscoring the potential of liquid crystal microcavities for biosensing in liver injury monitoring.

A 3D printed fiber-tip whispering-gallery-mode resonator for sensing in complex liquid environments

Feiyang Hou,Weian Wang,Jialve Sun,Shengfei Feng,Chunxiang Xu

WGM resonator integrated on a seven-core fiber tip via two-photon lithography. • Exhibits reusability and robustness in complex liquid conditions environments. • High-Q micro-rings enhanced optical resonance for sensitive in-situ detection. • Sensitive detection of organic matter was achieved. • Achieving a NEP of 6.6 mPa ⋅Hz −1/2 with bandwidth exceeding 200 MHz. Stable operation of whispering-gallery-mode (WGM) microcavity sensors in complex liquid environments remains challenging due to surface fouling, optical coupling instability, and environmental interference. Although fiber-integrated WGM devices enable miniaturized and in-situ sensing, their performance is often limited by fragile coupling configurations and poor long-term stability in liquids. Here, a fiber-tip WGM resonator is fabricated via two-photon lithography (TPL) by directly printing a microcavity onto the facet of a seven-core optical fiber (SCF) and subsequently encapsulating it with a low-refractive-index glue (LRIG) layer. This integrated architecture stabilizes optical coupling and suppresses resonance degradation in complex liquid environments, enabling high-contrast WGM spectra with quality factors on the order of 5.39 × 10 4 in liquid. Benefiting from enhanced light-matter interaction, structural robustness, and operational repeatability, the device exhibits sensitive responses to the non-specific adsorption of representative organic molecules, including ethanol, acetone, and isopropanol. Moreover, the same fiber-tip resonator enables ultra-sensitive ultrasound detection via acousto-optic interaction, achieving a noise-equivalent pressure (NEP) of 6.6 mPa ⋅Hz −1/2 with a full bandwidth exceeding 200 MHz. These results demonstrate a compact, robust, reusable, and scalable fiber-tip WGM device capable of optical and acoustic sensing in liquid environments, providing a versatile building block for integrated fiber-optic sensing platforms. Real-time monitoring of the enzymatic reaction of urease by using whispering gallery mode lasing

Rui Duan,Yanzeng Li,Hanyang Li,Hanyang Li,Hanyang Li,Jun Yang,Jun Yang,Jun Yang

A new strategy is reported here to monitor the enzymatic reactions in real time by using whispering gallery mode (WGM) lasing. The optical microcavity is formed via the self-assembly of an ultraviolet (UV)-treated nematic liquid crystal (LC) 4-cyano-4’-pentylbiphenyl (5CB). The single UV-treated 5CB microdroplet serves as both optical resonator and sensing reactor. The microdroplet configuration transitions induced wavelength shift in the WGM lasing spectra can be used as an indicator for the enzymatic reaction. The proposed sensor has a sub-microgram detection limit of urease (∼0.5 µg/ml), which is lower than the detection limit of currently available urease sensor based on LC materials. Our experimental results demonstrate that WGM lasing has unique advantages in the real-time monitoring of enzymatic reactions compared, for instance, with observation of the optical appearance under a polarized optical microscope.
