Iron is an essential nutrient for all organisms. Feruloyl-COA 6-hydroxylase 1 (F6′H1) plays a pivotal role in iron uptake in plant roots by catalyzing the biosynthesis of iron-mobilizing scopoletin, a secondary metabolite also possessing antimicrobial activity. However, it remains unclear whether F6′H1-mediated iron uptake affects plant resistance to foliar pathogens and how such a process might be regulated. Here, we show that enhanced expression of 2-oxoglutarate-dependent dioxygenases 2 (CmOGD2), a homolog of F6′H1 in Citron C-05 (Citrus medica L.), confers resistance to citrus canker caused by Xanthomonas citri subsp. citri (Xcc). CmOGD2-mediated pathogen resistance is achieved by promoting iron uptake and the accumulation of reactive oxygen species (ROS), which likely results in ferroptosis. Furthermore, CmOGD2 interacts with the enolase CmENO2 to destabilize CmZAT10.1, a transcriptional activator of CmOGD2, thereby forming a negative feedback loop that limits CmOGD2 expression. Notably, the Xcc effector pthA4 interferes with the CmOGD2–CmENO2 interaction, likely via a decoy mechanism, leading to CmZAT10.1 accumulation. These findings reveal complex regulatory mechanisms underlying the critical role of CmOGD2 in mediating Xcc resistance through iron- and ROS-dependent ferroptosis.

Xanthomonas citri subsp. citri (Xcc), the pathogen responsible for citrus canker, causes major economic damage worldwide. The type III secretion system (T3SS) is essential for Xcc pathogenicity. Here, 39 chiral 1,3,4-oxadiazole sulfide derivatives of mandelic acid were synthesized, with 17 compounds exhibiting significant inhibitory activity (>70% inhibition) against the T3SS of Xcc jx-6 at 40 μg/mL. Among these, the R-configuration WR4 significantly suppressed the symptoms of citrus canker in an in vivo bioassay, whereas neither the S-configuration WS4 nor the racemic mixture WRS4 showed inhibitory effects. Mechanistically, WR4 suppressed T3SS gene transcription and Hpa1 protein secretion, and counteracted pathogen-induced suppression of host immunity, without affecting other virulence factors. From an environmental safety perspective, WR4 exhibited low toxicity against nontarget organisms, including earthworms, silkworms, and zebrafish. These findings reveal the potential of the R-configuration derivatives as sustainable T3SS inhibitors.

Bacterial infections of plants are widespread and highly virulent, causing serious economic losses in agricultural production worldwide. These infections become particularly prevalent during rainy weather, characterized by high humidity and low light intensity (about 2–10% of a sunny day). Here we identified a class of photosensitizers that were effective in inhibiting plant bacterial infections under rainy weather with dim light. One of these compounds, compound 1 , demonstrated exceptional efficacy by eliminating more than 4 logs (99.99%) of either Xanthomonas perforans or Xanthomonas citri subsp. citri (Xcc) strain at a concentration of 6.25 μM under rainy weather conditions with a light exposure of 28.51 J/cm 2 . For Clavibacter michiganensis ( Cm ), compound 1 demonstrated a comparable bactericidal effect at a lower concentration of 3.13 μM. Furthermore, compound 1 was effective in controlling plant leaf infections during rainy weather. Moreover, compound 1 exhibited a potent inhibitory effect on the biofilms pertinent to tomato spot disease and citrus canker under natural illumination on rainy days. Additionally, compound 1 displayed remarkable resilience to rain-wash on plant leaves, retaining 41.20% of its initial concentration following exposure to three simulated rain events. These findings provide a new strategy for plant pathogen disease management highlight its feasibility even during rainy weather.

Citrus canker, a significant bacterial disease caused by Xanthomonas citri subsp. citri ( Xcc ), is currently managed primarily with copper-based treatments. However, the long-term use of these agents can result in pesticide residue and increased bacterial resistance. This study employed the vacuum fractional distillation (VFD) technique to fractionate the essential oil of Mentha haplocalyx (MHEO) to produce MHF3, which exhibited significant antibacterial activity against Xcc . The minimum inhibitory concentration (MIC) of MHF3 was 0.20 μL/mL, superior to that of the positive control, copper sulfate (156 μg/mL). Meanwhile, MHF3 also inhibited the growth of a diverse array of other pathogens with MIC values ranging from 0.78 to 100 μL/mL. Gas chromatography-mass spectrometry (GC–MS) analysis identified the primary antimicrobial components in MHF3 as isopulegol, menthol, pulegone, carvone, and piperitone, with respective MICs of 0.20, 0.78, 0.78, 0.20, and 0.39 μL/mL. The results of the antibacterial mechanism study revealed that MHF3 disrupts the bacterial cell membrane structure of Xcc , causing the leakage of intracellular proteins, nucleic acids, and AKP enzymes. MHF3 also induces ROS accumulation, inhibits biofilm formation, and significantly disrupts nucleotide, lysine, and glutathione metabolism, as well as the pentose phosphate pathway in Xcc . In vivo leaf assay results confirmed that MHF3 effectively controls citrus canker. This study reveals the potential application of the VFD technique for enriching the bioactive components of essential oils. It also provides scientific data for developing MHF3 as a green, efficient, and natural agent for managing citrus canker.
