Characterization of Cytochalasins from the Endophytic Xylaria sp. and Their Biological Functions

Qiang Zhang,Jian Xiao,Qing-Qing Sun,Jian-Chun Qin,Gennaro Pescitelli,Jin-Ming Gao

Bioassay-guided fractionation of the fermentation extract of Xylaria sp. XC-16, an endophyte from Toona sinensis led to the isolation of two new cytochalasans cytochalasin Z27, 1, and cytochalasin Z28, 2, along with three known compounds seco-cytochalasin E, 3, and cytochalasin Z18, 4, and cytochalasin E, 5. The structures of 1 and 2 were elucidated by spectroscopic and electronic circular dichroism methods. Compound 5 was shown to be potently cytotoxic against brine shrimp (LC50 = 2.79 μM), comparable to that of the positive agent toosendanin (LC50 = 4.03 μM), and also exhibited potential phytotoxic effects on Lactuca sativa and Raphanus sativus L. seedlings, which are higher than that of the positive control glyphosate. Additionally, the fungicidal effect of 2 against the phytopathogen Gibberella saubinetti was better than that of hymexazol. This is the first report of the three types of cytochalasins present in genus Xylaria. A structure–phytotoxicity activity relationship is also discussed.

Antifungal ergostane-type steroids from endophytic fungus Xylaria sp. VDL4

Jiao Yao,Lin-Hong Lu,Sheng-Yun Wei,Ting Wu,Wei Yang,Wei-Hua Wang,Fa-Zhong Yang,Yun-Xian Li,Ping Zhao,Guo-Lei Zhu

Three ergostane-type steroids, including one rare C-29 methylated xylarstane A ( 1 ) and one C22,23-epoxied xylarstane B ( 2 ), along with one reported compound 3 , were isolated from the endophytic fungus Xylaria sp. VDL4 harbored within the medicinal plant Vaccinium dunalianum . The structures of novel compounds were elucidated through comprehensive spectroscopic analysis, DP4 + -validated theoretical 13 C nuclear magnetic resonance (NMR) calculations, and electronic circular dichroism (ECD) calculations. The antifungal activities of the isolates against four phytopathogens were assessed in vitro. Compounds 1 and 3 exerted significant inhibition against Alternaria solani and Botrytis cinerea respectively, both with minimal inhibitory concentration (MIC) of 12.5 μg/mL, comparing with the positive control (Carbendazim and Thiabendazole, MICs = 12.5–25.0 μg/mL)

Carbaryl biodegradation by Xylaria sp. BNL1 and its metabolic pathway

Fei Li,Lin Di,Yuxin Liu,Qiuyun Xiao,Xiaoyu Zhang,Fuying Ma,Hongbo Yu

Although ascomycetes occupy a vaster niche in soil than the well-studied basidiomycetes , they have received limited attention in studies related to bioremediation. In this study, the degradation of carbaryl by Xylaria sp. was studied in different culture conditions and its possible metabolic pathway was elucidated. In liquid culture, 99% of the added carbaryl was eliminated when cytochrome P450 (CYP450) was active, which was similar to the degradation rate of Pleurotus ostreatus , a fungus with strong bioremediation ability. Mn 2+ is beneficial to the degradation of carbaryl. Compared to the 72.17% degradation rate in sterile soil, 59.0% carbaryl was eliminated in non-sterile soil, which suggested that Xylaria sp. BNL1 can resist microorganismal infection. Furthermore, the intracellular fractions containing laccase, CYP450, and carbaryl esterase efficiently degraded carbaryl. The presence of carbaryl metabolites suggested that Xylaria sp. BNL1 initiated its attack on carbaryl via carbaryl esterase to release α-naphthol, which was further degraded to 1,4-naphthoquinone and benzoic acid by CYP450 and laccase. Thus, our study highlights the potential of using Xylaria sp. for bioremediation.

Antifungal and α-Glucosidase Inhibitory Metabolites from Xylaria sp. VDL4 Endophyte of Vaccinium dunalianum

Yao Jiao,Wei Sheng-Yun,Huang Sai,He Ling-Feng,Wang Wei-Hua,Yang Wei,Luo Ming-Wang,Lu Lin-Hong,Li Lu,Zhao Ping,Zhu Guo-Lei

Phytochemical investigation of the ethyl acetate extract from Xylaria sp. VDL4, an endophytic fungus isolated from Vaccinium dunalianum Wight (Ericaceae), resulted in the isolation and structural characterization of a novel sesquiterpenoid, (3 S ,10 S ,6 E )-2,10-dihydroxy-2,6,10-trimethyldodeca-6,11-dien-3-ylacetate, named xylarnoid A ( 1 ), along with five known secondary metabolites ( 2–6 ). Their structures were elucidated through integrated spectroscopic analysis and electronic circular dichroism calculations. In vitro antifungal evaluation against phytopathogens ( Fusarium oxysporum , Botrytis cinerea , Phytophthora capsici , and Fusarium solani ) demonstrated significant inhibitory activities for selected compounds. Additionally, 2 , 3 , and 6 exhibited notable α -glucosidase inhibitory potential.

Antifungal Polyacetylenic Deoxyglycosides Isolated from Endophytic Fungus Xylaria sp. VDL4 Associated with Vaccinium dunalianum

Jiao Yao,Sai Huang,Lingfeng He,Shengyun Wei,Wei Yang,Qiangxin Zhang,Weihua Wang,Xiaoqin Yang,Sida Xie,Yunxian Li,Ping Zhao,Guolei Zhu

One novel C10polyacetylene rhamnoside, 4,6,8-decatriyne-1-O-α-L-rhamnopyranoside, named xylariside A (1), together with two novel C10polyacetylene quinovopyranosides, 4,6,8-decatriyne-1-O-α-D-quinovopyranoside, xylariside B (2), and 8E-decaene-4,6-diyne-1-O-α-D-quinovopyranoside, xylariside C (3), were obtained from the solid fermentation ofXylariasp. VDL4, an endophytic fungus isolated fromVaccinium dunalianumwight (Ericaceae). Their chemical structures were elucidated through a combination of spectroscopic techniques. The antifungal activities of these compounds were evaluated in vitro against four phytopathogenic fungi (Fusarium oxysporum,Botrytis cinerea,Phytophthora capsici, andFusarium solani). Compound2demonstrated significant antifungal activities, with minimum inhibitory concentration (MIC) values ranging from 3.91 to 7.81 μg/mL. Compound2’s effectiveness levels were similar to those of the reference drugs thiabendazole and carbendazim (each MIC = 0.98−15.62 μg/mL). Xylariside B (2) was further evaluated againstB. cinereain vivo. It exhibited remarkable efficacy in both the prevention and treatment of tomato and strawberry gray mold. Molecular docking studies confirmed the antifungal mechanism of compound2by revealing its binding interactions with key enzyme targets inB. cinerea, thereby supporting the observed in vitro and in vivo results. Additionally, compound2showed effective inhibition of α-glucosidase, with IC50values of 5.27 ± 0.0125 μg/mL.
