Facile one-pot fabrication of ZnO2 particles for the efficient Fenton-like degradation of tetracycline

Pei Chen,Fazhe Sun,Wei Wang,Fatang Tan,Xinyun Wang,Xueliang Qiao

This work presented a facile one-pot fabrication of zinc peroxide (ZnO 2 ) particles for Fenton-like degradation of tetracycline (TC). The effects of precipitants and their addition sequences on the synthesis of ZnO 2 were first investigated in detail. Phase-pure ZnO 2 could be obtained when H 2 O 2 was added before adding precipitant (NH 3 ·H 2 O/NaOH) or after NH 3 ·H 2 O precipitation. The Fenton-like degradation performance of ZnO 2 was evaluated using 100 mL of TC solution (50 mg/L). It was found that 10 mg of ZnO 2 could degrade about 90% of TC within 1 h in the presence of 3 mg of Fe 2 (SO 4 ) 3 ·xH 2 O at ambient temperature. Besides, increasing ZnO 2 dosage, Fe 2 (SO 4 ) 3 ·xH 2 O dosage and temperature were all beneficial to TC degradation, and the degradation process could be well described by the pseudo-second order kinetic model. The catalytic degradation process could be efficiently conducted in a wide pH range of 4–8, while the coexisted PO 4 3− and HCO 3 − were unfavorable for TC degradation. Furthermore, radical quenching experiments and electron spin resonance tests indicated that hydroxyl radical was the dominant reactive oxygen species generated in the degradation system.

The Mineralization and Detoxification of PFOA Using a Sn/ZnO2@Ti4O7Anode

Yang Zehong,Wang Zihao,Qiu Qiuhong,Luo Yuzhan,Wang Chenglong,Huang Yaobin,Zhuang Tao,Zheng Weijin,Chen Liang,Lan Liying,Liu Guangli,Fang Jiasheng,Li Yanliang,Lv Sihao,Wang Wenlong,Qiu Yongfu,Zhuo Qiongfang

A Sn/ZnO 2 @Ti 4 O 7 anode was prepared using a combination of hydrothermal synthesis and discharge plasma sintering methods for the electrochemical degradation of perfluorooctanoic acid (PFOA). The introduction of Sn/ZnO 2 significantly increased the electrochemical active surface area (ECSA) and the oxygen evolution potential (OEP) of Ti 4 O 7 . A 97.6% degradation rate of PFOA can be achieved using a Sn/ZnO 2 @Ti 4 O 7 anode, at a current density of 40 mA/cm 2 , with the initial PFOA concentration of 10 mg/L, at a pH value of 5.69, and within 120 min. Radical quenching experiments indicated that direct oxidation was the main mechanism for PFOA degradation, contributing 82.5%. The degradation efficiencies of PFOA remained above 88% in Dongjiang and Zhujiang rivers (DJR and ZJR) matrixes, confirming the system’s resistance to complex matrix. The total organic carbon (TOC) removal ratio was 56.8%, and the defluorination ratio was 48% after electrolysis of 6 h. After 36 cycles of degradation, the degradation rate was 95.6%. The toxicity assessments indicated that the degradation intermediates were significantly less toxic than the parent PFOA. Graphical

A pH-responsive ZnO2@ZIF-8/TCPP nanoplatform with self-supplied oxygen for hypoxia-relieved photodynamic therapy

Ye Gaobing,Liu Jing,Jin Yu,Hu Renren,Tian Tianli,Chen Peilei

Tumor hypoxia limits type-II photodynamic therapy (PDT) by restricting oxygen-dependent singlet oxygen production during irradiation. Here, we developed a pH-responsive ZnO 2 @ZIF-8/TCPP nanoplatform (PZZT) that integrates an acid-activated oxygen-generating core (ZnO 2 ), a ZIF-8 gatekeeper shell, and the photosensitizer TCPP, aiming to enhance PDT under hypoxic conditions. Under mildly acidic conditions, PZZT showed improved oxygen generation and enhanced DPBF consumption upon 660 nm irradiation, indicating elevated ¹O₂ production. In 4T1 cells, PZZT + laser induced stronger intracellular ROS signals and greater phototoxicity than free TCPP or control formulations. In 4T1 tumor-bearing mice, PZZT + laser achieved the most significant tumor growth inhibition without obvious systemic toxicity. Overall, PZZT provides a practical pH-activated self-oxygen-supplying strategy to improve PDT efficacy.

Injectable plant vesicles-biomineralized ZnO2 hydrogel for synergistic chemodynamic antibacterial, anti-inflammatory, and regenerative treatment of infected diabetic wounds

Yangkai Fu,Xi Guo,Luming Huang,Fei Xue,Chenyu Ma,Qinqin Huang,Yi Hu,Hao Ren,Zhengjie Meng

An injectable multifunctional hydrogel (ZE-OSG) was made via ginger-derived EV-biomineralized ZnO 2 for chronic wound healing. • ZE-OSG hydrogel showed self-healing, injectability, and prolonged wound-site retention with no toxicity. • ZE-OSG hydrogel showed ZnO 2 -based chemodynamic therapy (CDT) to kill MRSA killing and disrupt biofilm. • ZE-OSG hydrogel preserved the bioactivity of EV to reduce inflammation and promote angiogenesis, thereby accelerating wound healing. Chronic diabetic wounds are characterized by persistent bacterial infection, biofilm formation, and dysregulated inflammation, which impair angiogenesis and delay healing. In this work, we developed a multifunctional injectable hydrogel (ZE-OSG) that integrates ZnO 2 -based chemodynamic therapy (CDT) with the bioactivity of ginger-derived extracellular vesicles (EVs) to eradicate biofilms, resolve inflammation, and promote vascular regeneration. ZnO 2 -EV nanohybrids were synthesized via biomineralization of ZnO 2 directly onto EV, preserving EV bioactivity while enabling localized generation of hydrogen peroxide (H₂O₂) and hydroxyl radicals (·OH). This facilitates potent eradication of methicillin-resistant Staphylococcus aureus (MRSA) and disruption of mature biofilms. After integrating to Schiff base-crosslinked oxidized starch-gelatin hydrogel (OSG), the ZE-OSG hydrogel exhibited injectability, self-healing ability, and prolonged retention at the wound site. In a diabetic mouse model of MRSA-infected wounds, ZE-OSG significantly accelerated wound closure, reduced bacterial burden, suppressed inflammatory cytokines, promoted M1 to M2 macrophage polarization, and enhanced collagen deposition and neovascularization through upregulation of vascular endothelial growth factor (VEGF) and CD31 expression, without systemic toxicity. By integrating the EVs bioactivity with ZnO 2 -mediated antibacterial CDT within a single injectable hydrogel, ZE-OSG offers a clinically translatable strategy for the comprehensive management of infected chronic wounds.

Engineered-Doping Strategy for Self-Sufficient Reactive Oxygen Species Blossom to Amplify Ferroptosis/Cuproptosis Sensibilization in Hepatocellular Carcinoma Treatment

Jiahe Hu,Jiuxin Zhu,Kuan Wang,Xiuwei Yan,Lei Xu,Xiaoling Zhang,He Ding,Piaoping Yang,Shaoshan Hu,Rui Xie

Ferroptosis and cuproptosis are emerging modes of programmed cell death and have been increasingly used to eliminate tumor cells. However, converting ferroptosis/cuproptosis into effective treatments is challenging because of the inherent antioxidant and plasma membrane repair systems and inefficient copper ion delivery. Herein, an engineered doping method is developed to encapsulate ZnO 2 with Cu 2+ -doped ZIF-8 and modify the surface by transferrin (Tf). In the resulting ZnO 2 @Cu/ZIF-8-Tf nanosystem, Tf specifically binds to transferrin receptors for targeting and aggregation. In the tumor microenvironment, Cu 2+ /Fe 3+ is released from the nanosystem and reacted with glutathione (GSH) to produce Cu + /Fe 2+ . Excessive accumulation of Cu + interfered with the tricarboxylic acid cycle and induced coproptosis. Furthermore, the additional Fe 2+ caused iron overload and enhanced ferroptosis. ZnO 2 supplied hydrogen peroxide to mediate the overproduction of reactive oxygen species (ROS). Moreover, the depletion of GSH deactivated glutathione peroxidase 4 (GPX4) and inhibited the system X c − -GSH-GPX4 pathway, and the amplified ROS triggered lipid peroxidation and reprogrammed lipid metabolism, causing malfunctioning of both antioxidant and membrane repair systems. In summary, the ferroptosis/cuproptosis pathways are activated at multiple levels in the ZnO 2 @Cu/ZIF-8-Tf nanosystem, which ensures its outstanding antitumor effect.

Facile in-situ fabrication of ZnO2/CQD composites with promoted visible-light photocatalytic activities for organic degradation and bacterial inactivation

Pei Chen,Wei Wang,Ningning Dong,Junjie Zhang,Tao Yang,Fatang Tan,Songwei Tan,Xinyun Wang,Xueliang Qiao,Po Keung Wong

Herein, the novel zinc peroxide/carbon quantum dots (ZnO 2 /CQD) composites were synthesized by an in-situ composite process for visible-light photocatalytic tetracycline (TC) degradation and Escherichia coli ( E. coli ) inactivation. It was found that 0.1 g/L ZnO 2 /CQD composite could degrade over 80 % of 50 mg/L TC and inactivate around 7-log E. coli cells after 2 h and 50 min of irradiation, respectively. The as-obtained ZnO 2 /CQD composites exhibited superior visible-light photocatalytic performance to individual ZnO 2 , and superoxide radical (•O 2 − ) and singlet oxygen ( 1 O 2 ) were identified to be the main reactive oxygen species (ROSs) for TC degradation and E. coli inactivation. The introduction of CQD reduced the particle size and increased specific surface area, benefiting for adsorption. Meanwhile, it promoted visible-light absorption and accelerated the separation of photogenerated electron-hole pairs with a 3-fold increase in photocurrent, thus producing more ROSs under visible-light irradiation. Moreover, the ZnO 2 /CQD composites exhibited a high adaptability towards initial solution pH, common coexisting anions, oxygen-deficient   environment, diverse water sources and organic pollutants. This study not only provided highly adaptable visible-light-driven photocatalysts for organic degradation and bacterial inactivation, but also shed light on the multifunctional roles of CQD in boosting visible-light photocatalytic performance.

H2O2/acid self-supplying double-layer electrospun nanofibers based on ZnO2 and Fe3O4 nanoparticles for efficient catalytic therapy of wound infection

Lihui Yuwen,Pei Lu,Qi Zhang,Kaili Yang,Zhaowei Yin,Bin Liang,Lianhui Wang

Catalytic therapy based on nanozymes is promising for the treatment of bacterial infections. However, its therapeutic efficacy is usually restricted by the limited amount of hydrogen peroxide and the weak acidic environment in infected tissues. To solve these issues, we prepared polyvinyl alcohol (PVA)–polyacrylic acid (PAA)–iron oxide (Fe3O4)/polyvinyl alcohol (PVA)–zinc peroxide (ZnO2) double-layer electrospun nanofibers (PPF/PZ NFs). In this design, PVA serves as the carrier for ZnO2 nanoparticles (NPs), Fe3O4 NPs, and PAA. The double-layer structure of nanofibers can spatially separate the PAA and ZnO2 to avoid their reaction with each other during preparation and storage, while in the wet wound bed, PVA can dissolve and PAA can provide H+ ions to promote the generation of hydrogen peroxide and subsequent conversion to hydroxyl radicals for bacteria killing. In vitro experimental results demonstrated that PPF/PZ NFs can reduce the methicillin-resistant Staphylococcus aureus by 3.1 log (99.92%). Moreover, PPF/PZ NFs can efficiently treat the bacterial infection in a mouse wound model and promote wound healing with negligible toxicity to animals, indicating their potential use as “plug-and-play” antibacterial wound dressings. This work provides a novel strategy for the construction of double-layer electrospun nanofibers as catalytic wound dressings with hydrogen peroxide/acid self-supplying properties for the efficient treatment of bacterial infections.

Tailoring the synergetic effect of CoN center and sulfur heteroatomic in co-doped carbon electrocatalyst for high performance rechargeable ZnO2 batteries

Yanmei Kong,Wenjing He,Shanhu Chen,Asad Ali,Dandan Lyu,Liang Shen,Zhi Qun Tian

The trade-off between activity and durability of the oxygen redox reaction (OER and ORR) catalysts, caused by sluggish kinetics and distinct reaction requirements, seriously hinders the application of rechargeable Zn O 2 batteries. Although precious metal catalysts show high activity, they are constrained by cost. Transition-metal catalysts are promising alternatives, but their strong adsorption of oxygen intermediates leads to high reaction energy barriers. Herein, we developed a Co N,SC catalyst (with S atoms doped in the second coordination layer of Co N sites) via coordination reactions and ball milling method. The Co N,SC electrocatalyst exhibits excellent bifunctional performance, with a low potential difference of 0.63 V between the E 1/2(ORR) and the OER overpotential. More importantly, the homemade Zn O 2 battery has a high discharge peak power density of 265.82 mW · cm −2 , a specific capacity of 792.18 mAh · g Zn −1 , and excellent charge-discharge stability with highly reversible energy conversion efficiency. DFT calculations indicate doping with S can enhance the conductivity of the catalysts, adjust the state electron density of the Co N site, and make the d-band center of the Co 3d orbital move down. Therefore, the adsorption energy barriers of the Co N sites to the rate-determining step (O 2  → *OOH in ORR; *OH → *O in OER) are significantly reduced, thus promoting the oxygen redox catalysis reaction kinetics. Our findings open new opportunities for the rational design of non-noble metal catalysts to enhance ORR and OER performance and further improve Zn O 2 battery activities.

Core–Shell ZnO2@Cerium-Based Metal–Organic Framework with Low Turnover, Dual-Catalytic Activity for Biosafe Biofilm Dispersal and Immune Modulation

Renfei Wu,Tianjin Ge,Tianrong Yu,Qiaolan Shi,Rui Shi,Yijin Ren,Henk J. Busscher,Jian Liu,Henny C. van der Mei

The era of relying on antibiotics for curing bacterial infections is rapidly approaching an end, necessitating development of non-antibiotic-based infection-control strategies. Dispersal of infectious biofilms is a potential strategy but yields dispersed bacteria in blood that may cause sepsis. We report a bromide-loaded, core–shell ZnO2-nanoparticle/Ce-based metal–organic framework (ZnO2@CeMOF/Br) of which the ZnO2 core degrades at pH ≤ 6.5, leaving the MOF’s Ce node intact. ZnO2-core degradation initially generates a nonradical, relatively stable, low-oxidative hydrogen peroxide that can cleave matrix DNA causing dispersal of Staphylococcus aureus biofilms and reacts with bromide ions to form transient hypobromous acid. Hypobromous acid modulates macrophage polarization toward an M1-like phenotype to clear dispersed bacteria from blood. Subsequently the Ce3+/Ce4+ redox couple forming the Ce node acts as an electron shuttle upon oxidation/reduction to faciltate two catalytic reactions, maintaining hydrolysis of phosphodiester bonds and associated cleavage of matrix DNA as well as modulation of macrophage polarization. Neither growth of tissue cells or macrophages nor hemolysis are negatively affected by exposure to ZnO2@CeMOF/Br nanocatalysts at a ZnO2 nanoparticle over CeMOFs weight ratio ≤ 1.2, up until CeMOF concentrations less than at least 180 μg/mL. Under biosafe, low-turnover catalytic conditions, irrigation of infected wounds in diabetic mice with ZnO2@CeMOF/Br nanocatalysts (90 μg/mL) results in 100% survival, fast recovery of healthy body temperature and weight, lower numbers of CFUs in blood and wound and organ tissues, and macrophage polarization toward an M1-like phenotype, demonstrating potential of ZnO2@CeMOF/Br nanocatalysts for non-antibiotic-based infection control.

Hierarchical core–shell nanoplatforms based on sacrificial template-directed ZnO2@ZIF-8 for spatiotemporally controlled and self-augmented chemodynamic–photothermal antibacterial therapy

Yi-Fei Hui,Duan-Ping Hu,Yi Zhao,Shulin Li,Zhi-Feng Cai,Lin-Yi Li,Yuming Xie,Yong-Nian Zeng,Zu-Jin Lin,Fa-Qiang Tang

Sacrificial template strategy builds ZnO 2 @Hemin-ZIF-8 core-shell structure. • The seamless ZIF-8 shell prevents premature ZnO 2 leakage at physiological pH. • Hierarchical ZnO 2 @Hemin-ZIF-8@PB (ZHZHP) enables on-demand H 2 O 2 release. • ZHZHP enables dual-nanozyme cascade-based CDT and PTT to eradicate bacteria. • ZHZHP orchestrates anti-inflammation and angiogenesis to accelerate wound healing. Chemodynamic therapy (CDT) has emerged as a promising strategy for combating bacterial infections; however, the scarcity of endogenous H 2 O 2 within infection sites often bottlenecks therapeutic efficacy. Herein, a pH-responsive, H 2 O 2 self-supplying nanoplatform is meticulously engineered for spatiotemporally controlled chemodynamic–photothermal antibacterial therapy. This platform leverages ZnO 2 nanoparticles dual-role cores and sacrificial template to drive the in situ epitaxial growth of dense ZIF-8 shells (optionally encapsulated with Hemin), yielding the core–shell ZnO 2 @ZIF-8 (or ZnO 2 @Hemin-ZIF-8) intermediate. Stepwise surface engineering with hyaluronic acid (HA) and Prussian blue (PB) leads to the formation of a hierarchical core–shell structured ZnO 2 @Hemin-ZIF-8@HA@PB nanoplatform (ZHZHP). Under neutral physiological conditions, ZHZHP exhibits excellent stability as the interracially-bonded ZIF-8 shell effectively shields the ZnO 2 core from premature hydrolysis. Conversely, within the weakly acidic infection microenvironment, ZHZHP undergoes programmed disassembly, triggering the on-demand release of H 2 O 2 . Simultaneously, Hemin and PB act as peroxidase mimics to catalyze the conversion of liberated H 2 O 2 into highly toxic hydroxyl radicals (‧OH) via dual nanozyme cascades, while the PB component provides robust photothermal conversion under 808 nm NIR irradiation. This synergistic CDT/PTT approach achieves complete eradication of S. aureus and E. coli at an extremely low concentration of 50 μg mL −1 and demonstrates remarkable efficacy in disrupting both immature and mature biofilms. In vivo assays further confirm that ZHZHP significantly accelerates infected wound healing through potent antimicrobial and anti-inflammatory activities, alongside the promotion of angiogenesis. With high biocompatibility and negligible toxicity, ZHZHP represents a sophisticated strategy for the clinical management of refractory wound infections. Efficient clearance of periodontitis pathogens by S. gordonii membrane-coated H2O2 self-supplied nanocomposites in a “Jenga” style

Qinghua Cao,Xiang Xiao,Chengcheng Tao,Rui Shi,Rui Lv,Ruochen Guo,Xinyi Li,Baiyan Sui,Xin Liu,Jian Liu

As a key pathogen of periodontitis, P. gingivalis requires support of the initial colonizing bacterium (S. gordonii preferably) to form symbiotic biofilms on gingival tissues with enhanced antibiotic resistance. Here, we report a new strategy to treat periodontitis biofilms with S. gordonii membrane-coated H2O2 self-supplied nanocomposites (ZnO2/Fe3O4@MV NPs) in a “Jenga” style. Integration of our special MV coatings enables selectively enhanced internalization of the cargos in S. gordonii, thus inducing severe damage to the foundational bacterial layer and collapse/clearance of symbiotic biofilms consequently. This strategy allows us to clear the symbiotic biofilms of S. gordonii and P. gingivalis with active hydroxyl radicals (˙OH) derived from ZnO2–Fe3O4@MV NPs in a H2O2 self-supplied, nanocatalyst-assisted manner. This “Jenga-style” treatment provides a cutting-edge proof of concept for the removal of otherwise robust symbiotic biofilms of periodontitis where the critical pathogens are difficult to target and have antibiotic resistance.

pH-Activated Scallop-Type Nanoenzymes for Oxidative Stress Amplification and Photothermal Enhancement of Antibacterial and Antibiofilm Effect

Yuping Jiang,Jiaman Xu,Quan Lin,Junyao Song,Maokun Sheng,Jintae Lee,Jinsheng Shi,Xiaoying Kong,Yulong Tan

Ferric phosphate (FePOs) nanoenzymes can express peroxidase (POD) activity under the dual stimulation of an acidic environment and high H2O2 concentrations. In living organisms, this generates reactive oxygen species (ROS) in sites of lesion infection, and thus FePOs nanoenzymes can act as antimicrobial agents. Here, CeO2 and ZnO2 were immobilized in a scallop-type FePOs nanoenzyme material loaded with a photosensitizer, indocyanine green, to synthesize a multifunctional cascade nanoparticle system (FePOs–CeO2–ZnO2–ICG, FCZI NPs). H2O2 concentrations could be adjusted through the ZnO2 self-activation response to the slightly acidic environment in biofilms, further promoting the release of ROS from the POD-like reaction of FePOs, achieving amplification of oxidative stress, DNA and cell membrane damage, and exploiting the photodynamic/photothermal effects of indocyanine green to enhance the antibiofilm effects. CeO2 can remove redundant ROS by switching from Ce4+ to Ce3+ valence, enhancing its ability to fight chronic inflammation and oxidative stress and thus promoting the regeneration of tissues around infection. By maintaining the redox balance of normal cells, increasing ROS at the infection site, eliminating redundant ROS, and protecting normal tissues from damage, the synthesized system maximizes the elimination of biofilms and treatment at the infection site. Therefore, this work may pave the way for the application of biocompatible nanoenzymes.
