A decentralized electrochemical immunosensor was developed using a smartphone and a portable electrochemical workstation. The system employs a gold nanoflower-modified screen-printed carbon electrode (AuNFs/SPCE) for the sensitive measurement of adalimumab (ADL) in human serum via antigen‒antibody recognition. Scanning electron microscopy, X-ray photoelectron spectroscopy, and atomic force microscopy were used to investigate the AuNFs/SPCE. Compared with the bare SPCE, the AuNFs/SPCE has a 1.47-fold larger specific surface area, which enhances the capture efficiency of ADL for signal amplification. Electrode modification and antigen‒antibody assembly processes are characterized by cyclic voltammetry and electrochemical impedance spectroscopy. The immunosensor achieves excellent electrochemical properties by optimizing experimental conditions, which improves the signal-to-noise ratio and thus the analytical performance. Under the best experimental conditions, the immunosensor has a quantitative range of 0.05‒4 μg/mL in diluted human serum with a detection limit of 9.1 ng/mL, which is consistent with the effective therapeutic window of ADL in clinical settings. The prepared electrodes maintain 10-day stability, while with high reproducibility at different ADL concentrations. High recoveries in the range 94.08‒108.6% are obtained in human serum samples. Furthermore, ADL in the serum samples was measured by both ELISA and the developed sensor. Pearson and Bland‒Altman analyses revealed a consistent trend between the two methods. Therefore, this immunosensor is a promising tool for the point-of-care (POC) measurement of ADL in clinical samples, providing valuable insights for personalizing ADL dosing regimens in clinical practice. Delanzomib is a novel proteasome inhibitor initially developed for treating multiple myeloma. It was found to inhibit the expression of tumor necrosis factor alpha (TNF-α). This study aimed to investigate the ameliorating effect of delanzomib on collagen-induced arthritis (CIA) and to explore the pharmacodynamics and pharmacokinetics (PK) interactions between delanzomib and adalimumab. Rats with CIA were randomly assigned to receive the treatment with delanzomib, adalimumab, delanzomib combined with adalimumab, or placebo. Visual inspection and biochemical examinations including TNF-α, interleukin 6, and C-reactive protein were performed to assess arthritis severity during the treatment. The adalimumab concentration in rats was determined to evaluate the PK interaction between delanzomib and adalimumab. Also, the levels of neonatal Fc receptor (FcRn) and FcRn mRNA were measured to explore the role of FcRn in the PK interaction between delanzomib and adalimumab. As a result, delanzomib combined with adalimumab exhibited stronger anti-arthritis activity than a single drug because both drugs synergistically reduced TNF-α level in vivo. Delanzomib also decreased adalimumab elimination in rats by increasing the level of FcRn. The slower elimination of adalimumab in rats further prolonged the anti-TNF-α effect of adalimumab. Moreover, FcRn level was increased by delanzomib via suppressing FcRn degradation rather than promoting FcRn production. In conclusion, delanzomib combined with adalimumab may be a potential therapeutic approach for treating rheumatoid arthritis. The initial finding that the PK interaction occurred between delanzomib and adalimumab may have clinical relevance for patients who simultaneously take proteasome inhibitors and anti-TNF-α therapeutic proteins.

Uveitis is one of the most popular blind-causing eye diseases worldwide. Adalimumab (ADA) is used for the uveitis treatment through systemic or intravitreal injection at the expense of systemic side effects and increased medical risks. Although eye drops, a non-invasive topical treatment, could be a potential strategy to reduce side effects, it remains challenging to apply due to limited bioavailability mainly linked to poor retention time and permeation capacity for eye biological barriers. Here, we reported hydrogel eye drops composed of low-deacetylated chitosan and β-glycerophosphate as an ADA carrier and tested its toxicity, tolerability, intraocular permeability, and efficacy of non-invasive treatment for uveitis. It’s found the ADA-loaded hydrogel eye drops were more efficient than free ADA both in permeation rate and clinical efficacy for uveitis, Overall, this study provides a friendly non-invasive strategy to improve drug permeation rate and uveitis treatment efficacy, which may be valuable to clinically ophthalmic medication.
