A sensitive and selective colorimetric biosensor for determination of gentamicin, amikacin and tobramycin was proposed with the unmodified gold nanoparticles (GNPs) as the sensing element. Gentamicin, amikacin and tobramycin can rapidly induce the aggregation of gold nanoparticles and is accompanied by a color change from red to blue. The concentration of gentamicin, amikacin and tobramycin can be determined by using UV-Vis spectrometer. The experimental parameters were optimized with regard to pH, incubation time and the concentration of the GNPs. Under optimal experimental conditions, the linear range of the colorimetric sensor for gentamicin/amikacin/tobramycin were 2.67–33.93 ng mL -1 , 13.33–66.67 ng mL -1 and 20–180 ng mL -1 , respectively. The corresponding limit of detection (3σ) was 0.354 ng mL -1 , 0.999 ng mL -1 and 0.579 ng mL -1 , respectively. This assay was simple and used to detect aminoglycoside antibiotics in milk and medicine products.

Natural nanoscale polysaccharide and its application have attracted much attention in recent years. In this study, we report for the first time that a novel naturally occurring capsular polysaccharide (CPS-605) from Lactobacillus plantarum LCC-605, which can self-assemble into spherical nanoparticles with an average diameter of 65.7 nm. To endow CPS-605 with more functionalities, we develop amikacin-functionalized capsular polysaccharide (CPS) nanoparticles (termed CPS-AM NPs) with improved antibacterial and antibiofilm activities against both Escherichia coli and Pseudomonas aeruginosa . They also exhibit faster bactericidal activity than AM alone. The high local positive charge density of CPS-AM NPs facilitates the interaction between the NPs and bacteria, leading to extraordinary bactericidal efficiencies (99.9 % and 100 % for E. coli and P. aeruginosa, respectively, within 30 min) by damaging the cell wall. Very interestingly, CPS-AM NPs exhibit an unconventional antibacterial mechanism against P. aeruginosa , that is, they can induce plasmolysis , along with bacterial cell surface disruption, cell inclusion release and cell death. In addition, CPS-AM NPs exhibit low cytotoxicity and negligible hemolytic activity, showing excellent biocompatibility. The CPS-AM NPs provide a new strategy for the design of next-generation antimicrobial agents that can reduce the working concentration of antibiotics to fight against bacterial resistance .

Biofilm-associated infections (BAI) have posed serious threats to public health. Novel therapy based on carbon monoxide (CO) is being increasingly appreciated. However, CO therapy like inhaled gas treatment was impeded owing to its low bioavailability. Besides, the direct use of CO releasing molecules (CORM) showed low therapeutic efficacy in BAI. Therefore, it is vital to improve the efficiency of CO therapy. Herein, we proposed polymeric CO releasing micelles (pCORM) from self-assembly of amphiphilic copolymers containing CORM bearing block as hydrophobic part and acryloylmorpholine block as hydrophilic part. The catechol modified CORM were conjugated through pH cleavable boronate ester bonds and releasing CO passively under biofilm microenvironment. When combined with subminimal inhibitory concentration antibiotic amikacin , pCORM could significantly enhance its bactericidal efficiency against biofilm-encapsulated multidrug-resistant bacteria, representing a promising approach to combat BAI.

Background Avian colibacillosis is an infectious disease caused by pathogenic Escherichia coli (E. coli) and is one of the significant causes of economic losses in the global poultry industry. The overuse of antibiotics has led to the emergence of resistant strains, and the presence of antibiotic residues in chickens and eggs poses a threat to human health. Therefore, exploring safe adjuvants to enhance the efficacy of existing antibiotics is one of the critical issues in poultry farming and public health research. Naringenin (NG), a naturally occurring flavonoid, has been extensively studied as a therapeutic agent for various diseases, but its efficacy in combination with antibiotics for treating avian colibacillosis has not yet been explored. Results In this study, we established a model of avian colibacillosis and treated it with NG and amikacin (AMK) alone or in combination. Compared with monotherapy, the combination of NG and AMK significantly decreased bacterial loads in the heart, liver, spleen, lung, and kidney by 5.0–6.0 log 10 CFU/g ( p < 0.01). Histopathological analysis revealed that the combination treatment markedly alleviated pathological damage in the lung and duodenum compared to the AMK alone group. Furthermore, the combination therapy significantly exerted anti-inflammatory and antioxidant effects, as evidenced by reduced serum levels of tumor necrosis factor-alpha (TNF-α), interleukin-1 beta (IL-1β), and malondialdehyde (MDA), alongside increased superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities ( p < 0.05). 16 S rRNA sequencing demonstrated that the combination treatment significantly modulated the gut microbiota composition, decreasing the relative abundance of Proteobacteria from 44% in the infection group to 1.2% ( p < 0.01), while increasing Firmicutes and Bacteroidetes to levels comparable to healthy controls. Conclusions The research results provides evidence for the therapeutic potential of NG as an effective antibiotic synergist when combined with AMK against avian colibacillosis.

Aminoglycosides are often subtherapeutic to intracellular infections due to their high hydrophilicity. Here we used hyaluronic acid (HA) as a carbohydrate carrier of the aminoglycoside antibiotic, amikacin (AM) to deal with intracellular bacterial infections. The hyaluronan-amikacin conjugate (HA-AM) was synthesized by ‘click’ reaction between HA-propargyl amide (HAPA) and AM-azide. This conjugate with little cytotoxicity retained antibiotic effects on planktonic bacteria and showed better intracellular bactericidal activity than the antibiotic did. In addition, this conjugate was more efficient in reducing bacteria burden in an in vivo acute infection model than amikacin did. These results suggested that hyaluronic acid conjugation could reduce the dosage of antibiotic in treatment of intracellular bacterial infection, and further helping to alleviate the emergence of drug resistance in bacteria due to a long-term, high-dosage treatment.

The interaction mechanism of tigecycline with γ-globulin and hemoglobin in the absence and presence of amikacin was investigated through multipectral, molecular docking and molecular dynamics simulation. The results show that tigecycline and γ-globulin/hemoglobin forms a ground state complex without or with amikacin. The presence of amikacin slightly increases the binding constant of tigecycline to γ-globulin/hemoglobin, but all are of moderate binding affinity, at 10 4 L mol −1 . The equilibrium fraction of unbound tigecycline f u is >90 %, but the presence of amikacin reduces the free concentration of tigecycline in γ-globulin and hemoglobin. Whether amikacin is present or not, the interaction between tigecycline and γ-globulin/hemoglobin is a synergistic interaction driven by enthalpy and entropy. Non-covalent forces are primarily hydrophobic interactions, but also include electrostatic forces and hydrogen bonds. In the presence of amikacin, the effect of tigecycline on the skeleton structure of γ-globulin/hemoglobin is more significant. The effect of tigecycline and/or amikacin on the secondary structure of γ-globulin/hemoglobin is not significant, while the secondary structure changes in different systems are not the same. Molecular docking shows that γ-globulin/hemoglobin-tigecycline (first)-amikacin ternary system is the most stable. Molecular dynamics simulation explores the stability and dynamic behavior of γ-globulin/hemoglobin-tigecycline complex without or with amikacin.

A Eu-MOG@PAN nanofibrous membrane was fabricated for amikacin sensing. • Amikacin induces red-to-blue fluorescence switching in the solid-state membrane. • Ratiometric detection is achieved without an additional reference fluorophore. • The membrane shows high selectivity with a detection limit of 0.38 μM. • Amikacin in whey was successfully determined by visual and fluorescence analysis. Lanthanide metal–organic materials are attractive for optical sensing, but their application in complex aqueous samples is often hindered by single-channel emission, poor processability and insufficient signal reliability. Herein, we report a flexible mixed-matrix nanofibrous membrane fabricated by confining a europium metal–organic gel (Eu-MOG) within an electrospun polyacrylonitrile (PAN) scaffold. The membrane enables amikacin-triggered ratiometric luminescence sensing via nanoscale interfacial coordination. Upon exposure to amikacin, the amino groups of amikacin coordinate to Eu 3+ sites, perturb the Eu–carboxylate microenvironment and weaken the antenna effect, resulting in strong quenching of the Eu 3+ emission and simultaneous recovery of ligand-centred blue emission at 458 nm. This red-to-blue transformation provides a built-in ratiometric readout (I₄₅₈/I₆₁₉) without the need for an additional reference fluorophore. Control experiments confirm that the 458 nm emission originates from the organic ligand rather than from amikacin or PAN. Mechanistic studies based on fluorescence lifetimes, FT-IR, XPS, zeta-potential and spectral overlap reveal that the response is dominated by amikacin–Eu 3+ coordination and the consequent disruption of ligand-to-Eu 3+ energy transfer. The electrospun membrane offers an open, mechanically robust architecture that facilitates analyte access and solid-state operation. It exhibits high selectivity for amikacin over common inorganic ions, amino acids, and other antibiotics. The ratiometric response is linear over the low-concentration range of 1–9 μM, while the response at 10–100 μM can be described using a Logistic fitting model. The calculated limit of detection is 0.38 μM. The method was successfully applied to the determination of amikacin in whey samples with good reproducibility. This work presents a ligand-emission activation strategy for constructing lanthanide-based ratiometric nanofibrous membranes and demonstrates a portable platform for visual screening of antibiotic residues in dairy products. Objectives This study investigated the mechanisms underlying polymyxin B (PMB)-induced resistance and examined the role of PMB in combination with amikacin (AMK) in delaying the development of resistance. Methods In vitro continuous induction of three Klebsiella pneumoniae strains was performed using PMB alone or in combination with AMK, and variations in the minimum inhibitory concentration (MIC) were determined via the microdilution method. The Kp81 strain, which presented the most significant delay in resistance development, was selected for analysis of the expression of relevant genes via qRT–PCR. Homologous recombination techniques were employed to construct nlpE and cpxR gene knockout and complemented Kp81 strains, and the expression levels of these genes, as well as changes in the MIC following continuous induction, were assessed. Additionally, the biofilm-forming abilities of the strains were analyzed via crystal violet staining and confocal laser scanning microscopy. Results The results indicate that the combination of PMB and AMK can delay the development of resistance to PMB in K. pneumoniae . qRT–PCR analysis demonstrated a significant increase in the expression of phoP , nlpE , cpxR , and acrA in the resistant strain Kp81·R, whereas these genes were significantly downregulated in the resistant delay strain Kp81·DR. Notably, after 20 h of PMB treatment, the expression of nlpE was markedly elevated in the nlpE knockout strain. cpxR exhibited synchronized dynamic changes with nlpE in both the nlpE knockout and complemented strains, whereas acrA showed a similar expression pattern in the cpxR knockout and complemented strains. Additionally, the absence of nlpE or cpxR was found to delay PMB resistance. Furthermore, biofilm formation was significantly increased in both the resistant strains and those treated with PMB, while the combination of PMB with AMK inhibited biofilm formation. Conclusions The combination of PMB and AMK can delay the development of resistance in K. pneumoniae to PMB. The nlpE gene, as a key regulatory factor, can influence PMB resistance by modulating the CpxA/R two-component system and biofilm formation.

Bacterial drug resistance is becoming an increasingly serious problem, and the development of antibacterial synergists is urgently needed. Combining existing antibiotics with promising nonantibiotic agents is one strategy that has been shown to be effective at overcoming the widespread emergence of antibiotic-resistant pathogens. In this study, we investigated the antibacterial activities and mechanism of naringenin (NG) combined with amikacin (AMK) against multidrug-resistantEscherichia coli(E. coli). We first measured the fractional inhibitory concentration (FIC) of NG combined with antibiotics via the checkerboard method. The results indicated that the combination of NG and AMK had a synergistic effect onE. coliATCC 25922 andE. coliC7F3. In addition, this synergistic effect was verified by time-kill assays. Moreover, scanning electron microscopy (SEM) was used to observe cell morphology. The results showed that the cell wall ofE. coliwas destroyed. Furthermore, we assessed the leakage of alkaline phosphatase (AKP), K+, and protein. The extracellular AKP activity increased after the combinational group of 1/2MIC NG and 1/2MIC AMK, suggesting an impairment in cell wall permeability. An increase in the leakage of intracellular K+and protein indicated an increase in cell inner membrane permeability. These results revealed that NG and AMK inhibitedE. coliby damaging cell walls and membranes. In addition, PI uptake rapidly increased after treatment with NG and AMK. Confocal laser scanning microscopy (CLSM) revealed that NG caused cell wall and cell membrane damage inE. coli. In summary, our results provide a new strategy for responding to the development ofE. colidrug resistance.

An amikacin-sensitive monoclonal antibody (MAb) assay and immunochromatographic test strip were developed and applied for the detection of amikacin residues in bovine milk and chicken eggs. The immunoassay was specific to amikacin and showed no cross-reactivity with other aminoglycosides. The half maximum inhibitory concentration (IC50) of the assay was 0.65 ng/mL and the results were obtained within 90 min. Recoveries from spiked food matrices were within the range of 73.55–84.61% for bovine milk and 73.70–105.75% for whole egg. The strip test results were obtained within 10 min and showed a visual detection limit of 5.0 ng/mL for both food matrices. These results show that the MAb immunoassay and strip test developed in this study are very specific to amikacin and sufficiently sensitive for detection and routine monitoring of amikacin residues in food.

Plant essential oils possess broad-spectral antimicrobial property, but the applications are impeded by their insolubility in water, extreme volatility, and strong irritation. Nanoparticle-stabilized emulsion (Pickering emulsion) gels are colloidal systems with ability to accommodate two immiscible phases in one system. The thick adsorption nanoparticle layers and the cross-linked networks in continuous phase could provide protective barriers for antibacterial oil and achieve on-demand controlled release. An emulsion hydrogel templated from gelatin nanoparticle-stabilized emulsion is one-pot constructed by conducting a tunable cross-linking process between oxidized dextran (Odex) and amikacin in the continuous phase and concomitantly trapping tea tree essential oil (TO) droplets in the three-dimensional network. The resulted emulsion hydrogel presents tunable gelation time, adequate mechanical strength, fascinating injectability, and self-healing capability. It is pH-responsiveness and presents controlled release of amikacin and TO, exhibiting a long-term bacteriostasis of 144 h. The emulsion hydrogel facilitates the outstanding wound healing efficiency in 14 days (95.2 ± 0.8 % of wound closure), accompanied with enhanced collagen deposition and angiogenic activities. The incorporation of TO into emulsion hydrogel system reduced its irritation and improved its biosafety, showing potential application in bacteria inhibition even as implants in vivo .
