Conjugation of haloalkane dehalogenase DhaA with arabinogalactan to increase its stability

Meiqi Wang,Weili Yu,Lijuan Shen,He Zheng,Xuan Guo,Jinyi Zhong,Tao Hu

Haloalkane dehalogenase DhaA can catalyze the hydrolytic cleavage of carbonhalogen bonds, along with production of the corresponding alcohol, a proton and a halide. However, DhaA suffers from poor environmental tolerance, such as sensitivity to high temperature, low pH and hypersaline. Arabinogalactan (AG) is a hydrophilic polysaccharide with highly branched long chains. DhaA was conjugated with AG to improve the environmental stability of DhaA in the present study. Each DhaA was averagely conjugated with 4∼5 AG molecules. Conjugation of AG essentially maintained the enzymatic activity of DhaA (91.4 %) without apparent structural alteration. The hydration layer formed by AG could reduce the solvent accessible area of DhaA and slow the protonation process, thereby improving the pH and high salt stability of DhaA. In particular, the remaining activities of the conjugate (AG-DhaA) were 35.3 % after treatment at pH4.0 for 1 h, and 80.8 % in 1 M NaCl after treatment for 16 h. As compared with DhaA, AG-DhaA showed slightly different kinetic parameters (K M of 1.90 μmol/L and k cat of 2.60 s -1).

A novel Stauntonia leucantha fruits arabinogalactan: isolation and structural characterization

Hui Li,Tao Gao,Zhonghao Zhang,Jiangping Lei,Junchao Hu,Zizhong Tang,Shiling Feng,Chunbang Ding,Tao Chen,Yanger Chen,Shu Yuan,Ming Yuan

Polysaccharides were the key ingredients of many herbal medicines, and were responsible for multiple pharmacological activities. In this study, a novel polysaccharide fraction, named SLP-2, was isolated from Stauntonia leucantha fruits, and purified by DEAE-52 and Sephadex G-100 column chromatography . Furthermore, SLP-2 was identified by congo red, methylation , partial acid hydrolysis and NMR . The results indicated that the backbone of SLP-2 was composed of →4)-β-D-Gal p -(1 → 4)-β-D-Gal p -(1→ substituted at C-6 with 1,5-linked arabinan . SLP-2 had good anti-oxidation ability in vitro. Surprisingly, we found that reduction of carboxyl groups and methylation of hydroxyl groups enhanced the ability to scavenge 2,2′-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radicals and inhibit lipid peroxidation , and weakened the activity to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and reduce ferric iron.

Structural elucidation and bioactivities of a novel arabinogalactan from Coreopsis tinctoria

Hang Zhang,Jing Zhong,Qian Zhang,Degang Qing,Chunyan Yan

Coreopsis tinctoria is being widely cultivated in Xinjiang of China, whose consumption is known to prevent diabetes and neurodegenerative diseases. To elucidate the bioactive ingredients responsible for these benefits, the alkaline soluble crude polysaccharide (CTB) was isolated from C. tinctoria . In vitro experiments showed that the inhibition of α -amylase and α -glucosidase by CTB was 13407-fold and 906-fold higher than that by positive control, respectively. Then, a novel arabinogalactan, CTBP-1, was isolated and purified from CTB. Structural analysis showed that CTBP-1 possessed a 1,6-linked β - d -Gal p and 1,5-linked α - l -Ara f backbone with branches substituted at the C-3 position of the 1,6-linked β - d -Gal p , and the side chains included 1,5-linked α - l -Ara f , T-linked β - d -Gal p and T-linked α - l -Ara f . The inhibitory effects of CTBP-1 on α -amylase and α -glucosidase were 2.7 and 17.9 times that of acarbose, respectively. CTBP-1 could avoid indigestion and similar side effects. In addition, CTBP-1 remarkably inhibited the release of nitric oxide (NO), tumor necrosis factor- α (TNF- α ) and interleukin-6 (IL-6) in lipopolysaccharide (LPS)-stimulated BV2 microglial cells. In summary, CTBP-1 is a novel arabinogalactan with great potential as a treatment for type 2 diabetes and Alzheimer’s disease.

Tobacco Arabinogalactan Protein NtEPc Can Promote Banana (Musa AAA) Somatic Embryogenesis

Shu H.,Xu L.,Li Z.,Li J.,Jin Z.,Chang S.

Banana is an important tropical fruit worldwide. Parthenocarpy and female sterility made it impossible to improve banana varieties through common hybridization. Genetic transformation for banana improvement is imperative. But the low rate that banana embryogenic callus was induced made the transformation cannot be performed in many laboratories. Finding ways to promote banana somatic embryogenesis is critical for banana genetic transformation. After tobacco arabinogalactan protein gene NtEPc was transformed into Escherichia coli (DE3), the recombinant protein was purified and filter-sterilized. A series of the sterilized protein was added into tissue culture medium. It was found that the number of banana immature male flowers developing embryogenic calli increased significantly in the presence of NtEPc protein compared with the effect of the control medium. Among the treatments, explants cultured on medium containing 10 mg/l of NtEPc protein had the highest chance to develop embryogenic calli. The percentage of lines that developed embryogenic calli on this medium was about 12.5 %. These demonstrated that NtEPc protein can be used to promote banana embryogenesis. This is the first paper that reported that foreign arabinogalactan protein (AGP) could be used to improve banana somatic embryogenesis.

Interaction between Bacteroidetes species in the fermentation of Lycium barbarum arabinogalactan

Cui Cao,Lilong Wang,Xueqian Zhang,Chunqing Ai,Zhongfu Wang,Linjuan Huang,Shuang Song,Beiwei Zhu

The present study investigated the utilization of an arabinogalactan from Lycium barbarum (LBP-3) by intestinal Bacteroidetes species. The mixed-culture assay showed 58.4 % LBP-3 was utilized, and Bacteroides caccae and Phocaeicola vulgatus utilized more LBP-3 in single-culture compared to others. During in vitro fermentation of LBP-3, P. vulgatus favored arabinose while B. caccae preferred galactose. Moreover, 9 and 25 oligosaccharides were identified by HPLC-MS n in conditioned media (CM) derived from B. caccae and P. vulgatus , respectively. All of 3 tested Parabacteroides species ( P. distasonis , P. goldsteinii , and P. johnsonii ) markedly proliferated in CM of B. caccae and P. vulgatus , and proliferations of B. uniformis , B. finegoldii , B. ovatus and B. thetaiotaomicron also increased significantly in CM of B. caccae. The study suggests that the ability of Bacteroidetes species to degrade LBP-3 and sheds light on cooperative interactions of Bacteroides, Phocaeicola , and Parabacteroides species in the presence of LBP-3.

Larch Arabinogalactan Alleviates Colitis by Modulating Gut Microbiota and Promoting Bacteroides thetaiotaomicron

Shanshan Zhang,Yonggan Sun,Xinyu Bian,Fangwei Liu,Shaoping Nie

Inflammatory bowel disease (IBD) is linked to gut microbiota dysbiosis, and modulating microbiota through diet is a promising IBD management strategy. Larch arabinogalactan (AG) is a natural polysaccharide with potential prebiotic effects; however, its regulatory effects on gut microbiota and the underlying anti-inflammatory mechanisms remain unclear. Here, we demonstrated that AG improves gut homeostasis in dextran sulfate sodium-induced colitis mice, with effects depend on the gut microbiota. Further analysis revealed that AG notably enriched the Bacteroides, particularly Bacteroides thetaiotaomicron. Specifically, B. thetaiotaomicron exhibited anti-inflammatory activity, producing propionate and upregulating colonic G-protein-coupled receptor 41 (GPR41) expression, effects also observed in AG-treated mice. Collectively, our findings reveal the targeted regulatory effects of AG on gut microbiota and demonstrate that the B. thetaiotaomicron-propionate-GPR41 axis contributes to the amelioration of colitis by AG. These results provide new scientific evidence for the application of AG in health foods and offer novel insights for microbiota-targeted nutritional intervention.

Arabinogalactan ameliorates benzo[a]pyrene-induced intestinal epithelial barrier dysfunction via AhR/MAPK signaling pathway

Shaoying Gong,Jiachen Zheng,Junjie Zhang,Jianchun Han

Benzo[ a ]pyrene (B[ a ]P), a kind of pollutant, can disrupt the gut microbiota, but its effects on the function of intestinal epithelial barrier (IEB) is still unclear. Arabinogalactan (AG), a natural polysaccharide, can protect intestinal tract. Thus, the purpose of this study was to evaluate the effect of B[ a ]P on IEB function and the mitigation effect of AG on the IEB dysfunction induced by B[ a ]P using a Caco-2 cell monolayer model. We found B[ a ]P could damage the IEB integrity by inducing cell cytotoxicity, increasing lactate dehydrogenase leakage, decreasing the transepithelial electrical resistance, and increasing fluorescein isothiocyanate-dextran flux. The mechanism of B[ a ]P-induced IEB damage may through induction of oxidative stress, including increasing reactive oxygen species levels, decreasing glutathione levels, reducing the activity of superoxide dismutase, and increasing malonaldehyde levels. Moreover, it can be due to increasing secretion of pro-inflammatory cytokines (interleukin [IL]-1β, IL-6, and tumor necrosis factor [TNF]-α), down-regulated expression of tight junction (TJ) proteins (claudin-1, zonula occludens [ZO]-1, and occludin), and induced activation of aryl hydrocarbon receptor (AhR)/mitogen activated protein kinase (MAPK) signaling pathway. Remarkably, AG ameliorated B[ a ]P-induced IEB dysfunction through inhibited oxidative stress and pro-inflammatory factor secretion. Our study demonstrated B[ a ]P could damage the IEB and AG could alleviate this damage.

Changes in the functional properties of casein conjugates prepared by Maillard reaction with pectin or arabinogalactan

Siqi Yang,Guofang Zhang,Hong Chu,Peng Du,Aili Li,Libo Liu,Chun Li

The aim of this study was to prepare conjugates of casein (CA) with pectin (CP) or arabinogalactan (AG) by the Maillard reaction (wet-heating) and to investigate the effects of CP or AG on the structural and functional properties of casein. The results indicated that the highest grafting degree of CA with CP or AG was observed at 90 °C for 1.5 h or 1 h, respectively. Secondary structure showed that grafting with CP or AG reduced the α-helix level and increased the random coil level of CA. Glycosylation treatment of CA-CP and CA-AG exhibited lower surface hydrophobicity and higher absolute ζ-potential values, further significantly improving the functional properties of CA (e.g., solubility, foaming property, emulsifying property, thermal stability, and antioxidant activity). Accordingly, our results indicated that it is feasible for CP or AG to improve the functional properties of CA by the Maillard reaction.

Impact of Lycium barbarum arabinogalactan on the fecal metabolome in a DSS-induced chronic colitis mouse model

Cui Cao,Linlin Wang,Chunqing Ai,Guiping Gong,Zhongfu Wang,Linjuan Huang,Shuang Song,Beiwei Zhu

Ulcerative colitis (UC) is often accompanied by the dysbiosis of gut microbiota and metabolism. Our previous study indicated that arabinogalactan from Lycium barbarum (LBP-3) could markedly attenuate the symptoms of chronic UC in mice by modulating the structure of gut microbiota. This study explored the impact of LBP-3 on the fecal metabolomic profiling of the same cohort of mice by HPLC-TripleTOF/MS. Untargeted metabolomic analyses indicated that supplementation with LBP-3 markedly reversed 18 of the 48 differential metabolites (mainly belonging to amino acids and organic acids) disturbed by DSS. Targeted metabolomics revealed that the lower levels of tryptophan, lysine, diiodothyronine, kynurenine, and betaine and higher levels of phenylalanine, leucine, glutamine, isoleucine, homoserine, (S)-2-hydroxyglutarate, 2-isopropylmalic acid, ascorbic acid, gluconic acid, and taurine, which were caused by DSS induction, were reversed by LBP-3 treatment. In addition, pathway analysis showed that the pentose phosphate pathway, phenylalanine metabolism, ascorbate and aldarate metabolism, and phenylalanine, tyrosine and tryptophan biosynthesis were strongly affected by LBP-3. More importantly, the above amino acids, organic acids, and metabolic pathways changed by LBP-3 were correlated with the abundance of gut microbiota such as Turicibacter, Lactobacillus, Parasutterella, Odoribacter, Veillonella, Faecalibacterium, and Ruminococcaceae. This study advances our understanding of the interaction between the microbiome and metabolomics in DSS-induced chronic colitis after LBP-3 treatment.

The structure elucidation of novel arabinogalactan LRP1-S2 against pancreatic cancer cells growth in vitro and in vivo

Fei He,Shihai Zhang,Yanan Li,Xia Chen,Zhenyun Du,Chenghao Shao,Kan Ding

The fruit of Lycium ruthenicum Murr is used as traditional medicine and functional food . Previously we reported that one RG-I pectin from this fruit might inhibit pancreatic cancer cells growth. We further hypothesized that there might be other type of polysaccharides in this fruit also have anti-tumor effect. Here, we showed novel structure of a homogeneous polysaccharide named LRP1-S2 from this fruit and its anti-pancreatic cancer effect. Structure analyses suggested that LRP1-S2 was a novel arabinogalactan with the molecular weight ( Mw ) of 17.0 kDa. Bioactivity test showed that LRP1-S2 might attenuate the proliferation of pancreatic cancer cells in vitro and in vivo without significant cytotoxicity to normal pancreatic HPDE6-C7 cells and LO2 liver cells. Mechanism study indicated that it might induce apoptosis of BxPC-3 by inactivating P38 MAPK/NF-κB and GSK-3β/β-Catenin signaling pathways . These results suggested that LRP1-S2 could be a potential anti-tumor leading compound for functional food and new drug development. Chemical compounds arabinogalactan, pectin, galactan , arabinan , RN-1, HH1-1, LRP1-S2, LRP3-S1.

Comparative study of three arabinogalactan-chlorogenic acid complexes: The impact of polysaccharide structure on interaction mechanism and functional performance

Meng Xiao,Xing Jia,Xinru Li,Guochao Wu,Qiaomei Zhu,Huanhuan Liu,Steve W. Cui,Qingbin Guo

Polysaccharide-polyphenol non-covalent complexes have been widely utilized in food and pharmaceutical systems; however, how the intrinsic structural features of polysaccharides govern interaction mechanisms and functional outcomes remains insufficiently resolved. This study investigates how different molecular structures of arabinogalactan—gum arabic (GA), gum ghatti (GG), and peach gum (PG)—modulate the binding behavior, structural organization, and bioaccessibility of chlorogenic acid (CA). Under optimized conditions, the adsorption capacities of the three polysaccharides for CA were, in descending order: PG (82.88 μg/mg) > GG (73.84 μg/mg) > GA (63.96 μg/mg). Multispectral and conformational analyses suggested that hydrophobic interactions may be the primary force driving the GA-CA interaction, whereas electrostatic forces may be the primary force driving the GG-CA interaction. In contrast, PG-CA formation was highly dependent on conditions favoring hydrogen bonding, which also led to a significant conformational transition from a rigid to a more compact structure. Non-covalent interaction significantly enhanced the antioxidant activity of polysaccharide and the storage stability of CA. Furthermore, in vitro gastrointestinal digestion demonstrated that complexes, especially with PG and GG, effectively inhibited the rapid release of CA and improved its bioaccessibility. These results demonstrate that the molecular structural features of arabinogalactans govern their interaction mechanisms with polyphenols, thereby directly controlling the functional performance of the resulting complexes. This work provides a mechanistic framework for the rational design of polysaccharide-based delivery systems for polyphenols in functional food applications.

A structurally distinct arabinose-bridged arabinogalactan from Lycium barbarum as a TLR4-activating adjuvant for nanovaccine-based cancer immunotherapy

Chun-Jie Bao,Meng-Qiu Liu,Ya-Nan Liu,Li-Qiang Zhao,Yu-Xuan Li,Xin-Lan Zhang,Fang Zhang,Sheng Guo,Er-Xin Shang,Jia-Lun Duan,Jin-Ao Duan

Current studies on plant-derived adjuvants focus on heterogeneous or partially characterized polysaccharides, while the adjuvanticity of structurally defined, arabinogalactan (AG)-rich polysaccharide in cancer nanovaccine settings remains unexplored. Here, we identify and characterize NLBPE1, a homogeneous AG-rich polysaccharide isolated from Lycium barbarum , which features an atypical arabinose-bridged structure. Unlike canonical AG-I or AG-II motifs, NLBPE1 contains galactose side chains connected to the rhamnogalacturonan backbone via  → 5)- α -Ara f -(1 → linkages, forming a structurally distinct AG topology. Mechanistic studies demonstrate that NLBPE1 activates dendritic cells through the TLR4–IKK–NF-κB signaling axis, promoting antigen uptake, cross-presentation, and proinflammatory cytokine release. When co-formulated in a PLGA-based nanovaccine with a model antigen, NLBPE1 markedly enhances CD8 + T cell priming, leading to effective tumor control in vivo . Moreover, combination with anti-PD-1 immune checkpoint blockade yields synergistic antitumor responses in a melanoma model. Together, these findings establish NLBPE1 as a structurally and mechanistically distinct polysaccharide adjuvant with translational potential for next-generation cancer nanovaccine strategies.
