Study on colorimetric detection of Hg2+ by gold nanoparticles modified with melamine and arginine

Yingying Kou,Zhuang Wu,Chenling Yan,Jinggang Wang,Fan Liu,Donghai Yuan

Development of a new sensor for the simple detection of Hg 2+ , which improves on the shortcomings of conventional mercury detection. The size of the nanogold(AuNPs) particles was controlled by controlling the amount of sodium citrate and gold salt. AuNPs was also functionalized by arginine and melamine. The functionalized nanogold was sensitive and selective for Hg 2+ . It is also useful in real water samples. The 32 nm nanogold modification had the lowest limit of detection (LOD). The melamine-functionalized 32 nm AuNPs had a LOD of 4.17 μmol/L, which was better than the arginine-functionalized 32 nm AuNPs with a LOD of 5 μmol/L.

Microenvironment-informed carrier-free maltol-arginine nanoparticles harness CAT2-associated arginine transport for spermatogenic disorders

Ming-hui Jin,Ming-han Li,Tiannuo Wu,Xiaojie Yao,Tiantian Ye,Xiaodan Yan,Shujun Wang

Injured testicular microenvironment suggests enhanced arginine transport. • Carrier-free MAS integrate arginine-guided targeting with antioxidant maltol. • MAS show testicular accumulation and CAT2-associated uptake by Leydig cells. • MAS attenuate oxidative stress-pyroptosis and restore spermatogenic function. Spermatogenic disorders are a major cause of male infertility, with oxidative stress-driven Leydig cell dysfunction serving as a key pathological contributor. However, effective antioxidant therapy remains limited by insufficient accumulation in injured testes and by the formulation complexity associated with conventional carrier-based delivery systems. Here, we investigated the pathological microenvironment of di(2-ethylhexyl) phthalate (DEHP)-injured testes and found increased inducible nitric oxide synthase (iNOS) expression together with upregulation of the cationic amino acid transporter 2 (CAT2) in the testicular interstitium, indicating altered arginine metabolism and transport. Based on this pathological feature, we constructed carrier-free maltol-arginine nanoparticles (MAS) through Mannich reaction-mediated covalent conjugation and non-covalent self-assembly. In this system, the arginine moiety functions as a CAT2-recognizable structural cue for lesion-oriented delivery, whereas maltol provides antioxidant activity for redox regulation. MAS preferentially accumulated in injured testes and showed enhanced uptake by Leydig cells in vitro. Mechanistically, MAS promoted Nrf2 nuclear translocation, improved redox homeostasis, and attenuated NF-κB/NLRP3/GSDMD-mediated pyroptotic signaling. In the murine injury model, MAS ameliorated testicular microenvironmental disruption, improved Leydig cell-associated endocrine function, and enhanced sperm quality. Overall, this study presents a functionally engineered, carrier-free biomaterial strategy that redirects pathology-related arginine transport remodeling into a metabolic delivery advantage, providing a viable strategy for targeted intervention in spermatogenic disorders and restoration of testicular homeostasis. Carrier-free maltol-arginine nanoparticles convert pathology-associated arginine transport remodeling into a delivery advantage, enabling targeted redox regulation and attenuation of pyroptosis in DEHP-induced spermatogenic disorders. Exogenous arginine enhances the chilling tolerance in postharvest papaya fruit by regulating arginine and proline metabolism

Qun Huang,Hanliang Song,Yonggui Pan,Zhengke Zhang

Papaya fruit is easily affected by low temperature, causing chilling injury (CI). In this study, the exogenous arginine was applied in the storage of postharvest papaya fruit and it was found that 0.8 mM arginine treatment significantly alleviated CI of papaya fruit. Arginine treatment promoted the accumulation of polyamines (PAs), which might be ascribed to the increased activities of arginase, arginine decarboxylase (ADC), ornithine decarboxylase (ODC), and S-adenosylmethionine decarboxylase (SAMDC). A higher accumulation of proline was found in arginine-treated papaya fruit, which may be due to increased activity of ornithine-δ-aminotransferase (OAT) and Δ 1 -pyrroline-5-carboxylate acid synthase (P5CS) and inhibited activity of proline dehydrogenase (PDH). In addition, arginine treatment increased higher nitric oxide synthase (NOS) activity and promoted the generation of nitric oxide (NO). The results of this study indicate that exogenous arginine may improve the chilling tolerance of postharvest papaya fruit by regulating arginine and proline metabolism. Practical applications In recent years, chilling injury of tropical fruits is a significant problem, including papaya. Many studies on polyamine, proline, nitric oxide, and chilling injury of fruits, but arginine, as a common precursor of polyamine, proline, and nitric oxide, are rarely studied, especially in papaya fruit. Our findings demonstrated that the exogenous arginine can regulate endogenous arginine metabolism, induce polyamine, putrescine, and nitric oxide accumulation, and coordinate to alleviate chilling injury of papaya fruit.

Metabolism of arginine in juvenile largemouth bass (Micropterus salmoides) after oral or intraperitoneal administration of arginine or its substrates

Wang Jiteng,Zhang Jingyi,Li Xinyu,Xu Hanying Y.,Yang Yeshun,Zhang Jiankun,Feng Wenping,Chen Qiang,Dong Fen,Han Tao

The main objective of this experiment was to study the metabolism of arginine in juvenile largemouth bass ( Micropterus salmoides ). A total of 300 healthy fish (average weight of 25 ± 0.5 g) were randomly assigned to ten groups. Experimental fish were orally administered or intraperitoneally injected with 0.9% sodium chloride, arginine, arginine-aspartate, citrulline, and glutamate solutions, respectively. They were euthanized at 10, 30, 60, 120, and 240 min after oral administration or intraperitoneal injection, and various tissue samples were subsequently collected for analysis. The results revealed that serum ornithine and citrulline concentrations of largemouth bass were significantly increased by oral administration of arginine or arginine-aspartate ( P  < 0.05). Intraperitoneal injection of arginine or arginine-aspartate solution significantly elevated the concentrations of ornithine and citrulline in the serum, liver, kidney, and muscles ( P  < 0.05). The concentrations of citrulline, ornithine, and arginine in serum and muscle increased significantly at 4 h after intraperitoneal injection of glutamate ( P  < 0.05). Intraperitoneal injection of citrulline significantly increased the concentrations of ornithine and arginine in the serum and muscles ( P  < 0.05). The research findings demonstrate that both free and small peptide forms of arginine were rapidly degraded to ornithine due to the high arginase activity in various tissues of largemouth bass. Additionally, the pathway of synthesizing citrulline from glutamate and then arginine from citrulline may exist in largemouth bass, but the exact location of this synthesis process may differ from that found in mammals.

Rapid, enantioselective and colorimetric detection of D-arginine

Xianzhe Yu,Binjie Zhang,Cailing Fan,Qianqian Yan,Shenglin Wang,Hui Hu,Qinxi Dong,Gengyu Du,Yanan Gao,Chaoyuan Zeng

D-amino acids are of biological significance yet are not clearly understood due to the lack of powerful analytical tools for their identification. Thus, the specific detection of a single enantiomer of a particular amino acid remains a great challenge due to their structural similarity. Here, we report a strategy to incorporate multiple reaction sites on a chiral 1,1′-bi-2,2′-naphthol-based fluorescent probe. It can respond specifically to D-arginine, while producing no response when in contact with all other amino acids. The probe can report arginine’s concentration, and enantiomeric configuration and colorimetric studies enable its qualitative determination.

Arginine-engineered gold nanoclusters for doxorubicin chemiluminescence sensing

Cai Chuangui,Lin Zhen,Yao Wensong,Lian Shan,Huang Kaiyuan,Deng Haohua,Lin Xinhua,Chen Wei

Arginine surface-engineered 6-aza-2-thiothymine gold nanoclusters (Arg/ATT-AuNCs) via host-guest assembly were synthesized which exhibit exceptional catalytic activity and intense photoluminescence. Arg/ATT-AuNCs enhanced the CL intensity from NaIO 4 -H 2 O 2 system by 1200-fold. The CL mechanistic studies revealed that Arg/ATT-AuNCs not only accelerated decay of H 2 O 2 to form reactive oxygen species, but also acted as efficient energy acceptors of singlet oxygen ( 1 O 2 ) via a CL resonance energy transfer (CRET) pathway. It was observed that doxorubicin (DOX) quenches the CL signal through an inner filter effect that disrupts the CRET process. Based on this, a straightforward, rapid, and highly sensitive biosensor has been developed for the detection of DOX in human serum samples, achieving a low detection limit of 0.029 µM. The biosensor was applied to DOX determination in human serum samples with recoveries ranging from 91.1 to 106.2%. This study not only provides a new method for the detection of DOX, but also broadens the application scope of AuNCs in the field of CL. Graphical

Solubilization of organic compounds by arginine-derived polymers

Xuchen Huang,Yanbin Huang

Poor aqueous solubility of drugs is one of the major challenges in the pharmaceutical science. In this study, a guanidinium-containing polymer based on arginine was designed and synthesized, and was evaluated as a solubility enhancing additive for three model organic compounds (coumarin, pyrene and doxorubicin). At a guanidinium group concentration of 100   mmol/L, the polymer could significantly increase the solubility of pyrene and doxorubicin by 6- and 11-fold respectively, much more effective than arginine (2- and 3-fold, respectively). In contrast, its effect on the solubility of coumarin was less effective than arginine. The solubilizing effect may be explained by the enhanced interaction between the guanidinium group in the polymer and the aromatic compounds.

In Vivo Targeting of Escherichia coli with Vancomycin-Arginine

Lewis F. Neville,Itamar Shalit,Peter A. Warn,Marc H. Scheetz,Jiuzhi Sun,Madeline B. Chosy,Paul A. Wender,Lynette Cegelski,Jacob T. Rendell

The ability of vancomycin-arginine (V-r) to extend the spectrum of activity of glycopeptides to Gram-negative bacteria was investigated. Its MIC towards Escherichia coli, including β-lactamase expressing Ambler classes A, B, and D, was 8 to 16 μg/ml. Addition of 8 times the MIC of V-r to E. coli was acutely bactericidal and associated with a low frequency of resistance (<2.32 × 10−10). In vivo, V-r markedly reduced E. coli burden by >7 log10 CFU/g in a thigh muscle model. These data warrant further development of V-r in combatting E. coli, including resistant forms.

Enrichment of phosphopeptides by arginine-functionalized magnetic chitosan nanoparticles

Junwei Yang,Si Zhou,Haijiao Zheng,Qiong Jia

One of the most crucial and prevalent post-translational modifications is the phosphorylation of proteins. The study and examination of protein phosphorylation hold immense importance in comprehending disease mechanisms and discovering novel biomarkers. However, the inherent low abundance, low ionization efficiency, and coexistence with non phosphopeptides seriously affect the direct analysis of phosphopeptides by mass spectrometry. In order to tackle these problems, it is necessary to carry out selective enrichment of phosphopeptides prior to conducting mass spectrometry analysis. Herein, magnetic chitosan nanoparticles were developed by incorporating arginine, and were then utilized for phosphopeptide enrichment. A tryptic digest of β-casein was chosen as the standard substance. After enrichment, combined with matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF MS), the detection limit of the method was 0.4 fmol. The synthesized magnetic material demonstrated great potential in the detection of phosphopeptides in complex samples, as proven by its successful application in detecting phosphopeptides in skim milk and human saliva samples.

Aspartic acid/arginine enhance the stability of gelatin emulsions

Hui-min Yan,Yu Song,Man-man Yu,Xiu-ning Yao,Wen-wen Zhang,Ying Xu,Xue-qing Li,Yu-qin Xu,Shun Fang,Jun Qi,Guo-yuan Xiong,Chao Li,Jing-min Jia,Yong Hu

The purpose of this study was to use the complexes of gelatin (GA), positively charged arginine (Arg), and negatively charged aspartic acid (Asp) to produce a stable o/w emulsion using a low-energy stirring method. The thermal stability mechanism of this emulsion was investigated by evaluating its particle size, microstructure, differential scanning calorimetry (DSC), rheology, and ζ potential , as well as the secondary structure, surface hydrophobicity, and interaction force of gelatin-amino acid complexes. The results showed that adding Arg and/or Asp significantly decreased the particle size of the emulsion (P < 0.05). Particularly, the emulsion stabilized by gelatin-Arg-Asp showed the smallest particle size of 0.9 μm (D3.2), the highest absolute zeta potential (21.58 mV), the greatest viscosity, and the highest thermal stability. Even after heating at 121 °C for 20 min, this emulsion still had smaller particles than the unheated emulsion with or without Arg or Asp. Asp and Arg enhanced the triple helix content of gelatin through intramolecular hydrogen bonds and protected the surface hydrophobic groups of gelatins through intermolecular hydrophobic interactions of guanidine groups, increasing the depolymerization and ordered structure of gelatin and thus prolonging the temperature corresponding to the endothermic peak from 24.16 °C to 84.35 °C. These findings should contribute to the high thermal stability of the gelatin-Arg-Asp-stabilized emulsion. This study suggested that macromolecular protein-stabilized oil-in-water emulsions with a low oil phase content could be prepared by low-energy emulsification .

Celastrol inhibits Streptococcus suis infection through disrupting arginine metabolism pathway

Hongdou Liu,Shichao Xu,Rui Lei,Xiaoying Yu,Yi Lu,Yandi Pan,Bo Su,Shiqi Lang,Hongkai Ren,Rendong Fang,Jialu Zhang,Lianci Peng

Celastrol exhibits effective anti- S. suis efficacy both in vitro and in vivo. • Celastrol potentiates the anti- S. suis efficacy of antibiotics. • Celastrol potentially targets arginine deiminase (ADI) and disrupts arginine metabolism of S. suis . • Celastrol disrupts cytoplasmic membrane function of S. suis . Objectives : Given the rising prevalence of Streptococcus suis ( S. suis ) infection and the reduced effectiveness of traditional antibiotics, the development of novel alternative therapeutics or synergistic adjuvants to improve antimicrobial efficacy is urgently required. Celastrol displays antibacterial effects, but the specific mechanism by which celastrol exerts its anti- S. suis activity through interference with arginine metabolism remains unexplored. Purpose : This study investigated the antibacterial mechanism of celastrol against S. suis and further demonstrated its potential as an adjuvant to conventional antibiotics. Methods : The in vitro and in vivo anti- S. suis efficacy of celastrol were evaluated through time-killing curves, drug resistance, biofilm formation assays and mice infection model. The synergistic efficacy of celastrol and antibiotics against S. suis was evaluated through checkerboard microdilution assay. Transcriptomic analysis was performed to analyze the potential anti- S. suis pathways of celastrol. Anti- S. suis target of celastrol was elucidated using Δ arcA mutant strain, CETSA and DARTS assays. Celastrol-disrupted membrane integrity and function of S. suis were employed. Results : Celastrol exerted potent anti- S. suis efficacy in vitro and in vivo , and combination with antibiotics exhibited synergistic antibacterial efficacy. We found that celastrol treatment downregulated arginine-metabolism genes including arcA, argF , and argR . Importantly, celastrol physically interacted with recombinant ADI, the key enzyme catalyzing arginine degradation. Meanwhile, celastrol destroyed membrane integrity, induced ROS accumulation, caused proton motive force (PMF) dissipation, impaired ATP production and inhibited efflux pump activity. Conclusion : This study demonstrates that celastrol exerts anti- S. suis efficacy by disrupting arginine metabolism and compromising membrane function.
