Wound healing is a complicated process that can be susceptible toward infection caused by bacteria. The surging drug resistance has become one of the biggest challenges to the treatment of these infections in public health. Herein, a new asymmetrical aza-boron-dipyrromethene (ABDP) probe is prepared and encapsulated with cholesterol-modified polyethylene glycol polymer (Chol-PEG 2000 -NH 2 ) to construct a PTT/PDT dual-modality antibacterial agent (the ABDP NPs) and provide a new material for the treatment of MRSA-based infections. The ABDP NPs show remarkable wide UV–Vis absorption at 300–850 nm and superior bactericidal effects against Staphylococcus aureus ( S. aureus ), Escherichia coli ( E. coli ), and Methicillin-resistant S. aureus (MRSA), which can be controlled by a phototheranostic antibacterial switch (808 nm laser). Histological analysis indicates that wounds treated by the ABDP NPs plus NIR laser irradiation exhibit more complete re-epithelialization, abundant collagen fibers, and higher hair follicle numbers than the vancomycin-treated group. This study provides a promising perspective using aza-BODIPY dye for the treatment of multidrug-resistant bacteria-based infections and clinical applications in the future.

Cancer is one of the major diseases that threaten human being's health and life. Recently, phototherapy has gained great attention due to the advantages of high selectivity and low toxicity. Nonetheless, the development a single photosensitizer capable of simultaneously achieving photothermal and photodynamic effects remains a great challenge. Aza-boron-dipyrromethene (aza-BODIPY) dyes have been widely studied in biomedical area for their significant absorption in the near infrared region and high fluorescence quantum yield . In this contribution, three aza-BODIPY molecules (TAS-BP, TES-BP and TESO-BP) that conjugated with triphenylamine (TPA) or tetraphenylethylene (TPE) were designed and synthesized. The presence of donor-acceptor-donor pairs within compounds enables the intermolecular electron transfer , leading to the augmentation of near-infrared (NIR) absorbance and the generation of non-radiative heat. The nanoparticles were prepared via the assembly of the related organic compounds with amphiphilic polymer (DSPE-PEG 2000 ), which were named as TAS-BP NPs, TES-BP NPs and TESO-BP NPs, respectively. Importantly, three nanoparticles can generate heat and reactive oxygen species (ROS) simultaneously induced by a single laser (808 nm). The photothermal conversion efficiency of TAS-BP NPs, TES-BP NPs and TESO-BP NPs were determined to be 64.5%, 65.7%, and 53.6%, respectively. Furthermore, the cellular experiments demonstrated the good biocompatibility and outstanding photocytotoxicity of three nanoparticles , and the TES-BP NPs exhibited the highest photodynamic/photothermal synergistic effect. Thus, the as-prepared organic nanomaterials were demonstrated as potential candidates for cancer treatment through phototherapy.

Photoimmunotherapy represents an innovative approach to enhancing the efficiency of immunotherapy in cancer treatment. This approach involves the fusion of immunotherapy and phototherapy (encompassing techniques like photodynamic therapy (PDT) and photothermal therapy (PTT)). Boron-dipyrromethene (BODIPY) has the potential to trigger immunotherapy owing to its excellent PD and PT efficiency. However, the improvements in water solubility, bioavailability, PD/PT combined efficiency, and tumor tissue targeting of BODIPY require introduction of suitable carriers for potential practical application. Herein, a disulfide bond-based hollow mesoporous organosilica (HMON) with excellent biocompatibility and GSH-responsive degradation properties was used as a carrier to load a bithiophene Aza-BODIPY dye (B5), constructing a sample chemotherapy reagent-free B5@HMON nanoplatform achieving triple-synergistic photoimmunotherapy. HMON, involving disulfide bond , is utilized to improve water solubility, tumor tissue targeting, and PD efficiency by depleting GSH and enhancing host-guest interaction between B5 and HMO. The study reveals that HMON’s large specific surface area and porous properties significantly enhance the light collection and oxygen adsorption capacity. The HMON’s rich mesoporous structure and internal cavity achieved a loading rate of B5 at 11 %. It was found that the triple-synergistic nanoplatform triggered a stronger anti-tumor immune response, including tumor invasion, cytokine production, calreticulin translocation, and dendritic cell maturation, eliciting specific tumor-specific immunological responses in vivo and in vitro . The BALB/c mouse model with 4T1 tumors was used to assess tumor suppression efficiency in vivo , showing that almost all tumors in the B5@HMON group disappeared after 14 days. Such a simple chemotherapy reagent-free B5@HMON nanoplatform achieved triple-synergistic photoimmunotherapy.

Optical imaging in vivo holds significant implications for disease diagnosis, and nanoprobes with near-infrared (NIR) emission leverage the deep tissue penetration and high spatiotemporal resolution provided by NIR light, demonstrating considerable application potential. This study presents the design and synthesis of three nitrogen-doped boron–dipyrrin (Aza-BODIPY) molecules: Aza–BDP–OCH 3 , Aza–BDP–OH, and Aza–BDP-I. Leveraging the strong electron-accepting properties of the Aza-BODIPY core, we developed a donor–acceptor–donor (D-A-D) structure for Aza–BDP–OCH 3 through modifications with triphenylamine and methoxy groups, resulting in NIR fluorescence. Aza–BDP–OH was obtained via demethylation using boron tribromide, whereas Aza–BDP-I was synthesized by introducing iodine into Aza–BDP–OCH 3 . These three molecules self-assemble with the amphiphilic polymer PMHC 1 8 -mPEG to form nanoparticles (NPs), yielding optical nanoprobes. The resulting NPs exhibit NIR emission, good water solubility, and biocompatibility. At a concentration of 100 μ g ⋅ mL − 1 , these NPs demonstrate low biological toxicity, highlighting their potential for biological applications. Following tail vein injection, Aza–BDP-I NPs accumulate in tumors and effectively illuminate them via the enhanced permeability and retention (EPR) effect. Furthermore, these organic NPs were metabolized by the liver. Therefore, Aza-BODIPY-based NIR fluorescent NPs offer a promising platform for the development of in vivo optical nanoprobes.

As an important noninvasive tumor treatment method, phototherapy has drawn extensive research interest. However, the requirements of separate excitation wavelengths, high degree of electron–hole recombination, and low reactive oxygen species (ROS) production capability are still the major barriers. This work reports the construction of a novel nanoplatform: design and synthesis of an aza-BODIPY (AB) probe-decorated mesoporous black titanium dioxide (TiO2) (MT) nanoparticles (NPs) for enhanced photodynamic therapy and photothermal therapy under single-wavelength near-infrared (NIR) laser irradiation for the first time. AB probe-decorated MT NPs (abbreviated as MTAB) were synthesized through the Al reduction of mesoporous anatase TiO2 NPs and subsequent adsorption of the AB probe. The mesoporous structure of MT ensured AB loading capacity and avoided the complicated modification and synthesis processes. Heterogeneous MTAB, which possessed staggered energy levels, were assessed for their capability for effective separation of photogenerated electrons and holes for the first time. Upon NIR laser light irradiation, MTAB exhibited sufficient ROS generation, resulting in distinct tumor cell killing and tumor tissue elimination. This unique heterogeneous nanoplatform with staggered energy levels provides a new strategy to enhance ROS generation and improve the therapeutic efficacy.

Tumor oxygen spatial heterogeneity is a critical challenge for the photodynamic inhibition of solid tumors. Development of an intelligent nanoagent to initiate optimal therapeutics according to the localized oxygen levels is an effective settlement. Herein, we report an activatable nanoagent (BDP-Oxide nanoparticles (NPs)) to enable the oxygen auto-adaptive photodynamic/photothermal complementary treatment. Upon the nanoagent accumulated in the tumor region, the low extracellular pH could trigger the disassociation of the nanoagent to release the phototheranostic agent, BDP-Oxide, which will subsequently afford the fluorescence imaging-guided photodynamic oxidation after it gets into the outer oxygen-rich tumors. Along with the penetration deepening in the solid tumor, furthermore, BDP-Oxide could be reduced into BDP by the cytochrome P450 (CYP450) enzymes activated in the low oxygen tension regions of inner hypoxic tumors, which will switch on the photothermal and photoacoustic effects. Overall, the BDP-Oxide NPs-enabled photodynamic/photothermal complementary therapy significantly suppressed the solid tumor growth (inhibition rate of 94.8%). This work proposes an intelligent platform to address the oxygen partial pressure for the optimization of cancer phototherapeutics.

Achieving J-aggregation of a molecule is a fascinating way to construct fluorescent imaging and photothermal therapy agents in the second near-infrared window. Modulation of the balance between intermolecular π–π stacking and steric interactions is an elegant method to acquire J-aggregation dyes. Herein, we succeeded in synthesizing an aza-BODIPY dye with J-aggregation characteristics by introducing steric hindrance (TPA) and a π-bridge (thiophene) in the aza-BODIPY skeleton. In aqueous solutions, supramolecular J-aggregates with a regular lamellar stacking structure could quickly be formed by the dye-templated self-assembly and longer absorption (λmax = 939 nm) and emission (λmax = 1039 nm) bands were observed. After co-assembly of the dye and an amphiphilic polypeptide (POEGMA23–PAsp20), the obtained J-aggregation nanoparticles (J-NPs) with good water solubility and smaller size were more suitable for application in living organisms. In addition, the J-NPs exhibited good stability, considerable photothermal conversion capacity (η = 35.6%), and excellent resistance to pH, H2O2, and photobleaching. In vitro and in vivo experiments revealed that the J-NPs show great NIR-II fluorescence imaging capabilities and anti-tumor effects under 915 nm irradiation (1 W cm−2). This is a rare example of using BODIPY dyes to perform NIR-II fluorescence imaging-guided photothermal therapy under NIR-II irradiation.

Photoinduced radical chemistry is pivotal for synthesizing polymeric materials under light irradiation. In this study, carbazole moieties were incorporated at distinct positions within the aza-boron-dipyrromethene (aza-BODIPY) scaffold, yielding three novel push–pull dyes that function as high-performance photoinitiators for the free radical polymerization of acrylates upon exposure to visible-light LED irradiation. When combined with iodonium salts or amines as co-initiators, these aza-BODIPY derivatives form efficient two-component photoinitiating systems that exhibit outstanding initiation performance. Notably, distinct polymerization kinetics were observed for two benchmark acrylate monomers. Through a combination of experimental investigations and theoretical calculations, a comprehensive photochemical mechanism has been elucidated. Furthermore, these newly developed carbazole-substituted aza-BODIPY-based photoinitiating systems enable the fabrication of high-resolution three-dimensional macroscale architectures via 3D printing. This work advances the fundamental understanding and practical application of visible-light-induced free radical polymerization.

Tumor hypoxia limits the efficacy of photodynamic therapy (PDT), necessitating photosensitizers with hypoxia-adaptive therapy and imaging. Here, we present a fluorinated N -oxide aza-BODIPY ( OFBD ) nanoemulsion ( OFBD-NP ) for hypoxia-responsive, multimodal imaging-guided phototherapy. OFBD generates robust singlet oxygen under normoxia for PDT, but is reduced by CYP450 enzymes in hypoxic cells to photothermal-potent FBD , enabling switchable PDT/PTT. Co-assembly with fluorinated oil enhances oxygen delivery, boosts 19 F MRI sensitivity, and promotes J -aggregation, shifting fluorescence into the NIR-II window for deep-tissue imaging. The redox conversion also activates photoacoustic signals, enabling responsive tri-modal imaging (NIR-II FLI/ 19 F MRI/PAI). OFBD-NP shows potent cytotoxicity in vitro under both normoxic and hypoxic conditions via apoptosis. In vivo, it selectively accumulates in tumors, offers high-contrast imaging, and leads to complete tumor regression in subcutaneous A549 models after laser irradiation, without systemic toxicity. This work demonstrates a smart nanoplatform that integrates deep-tissue imaging and hypoxia-triggered therapeutic switching, addressing major limitations of conventional photosensitizers in cancer imaging and phototherapy.

Immune checkpoint blockade (ICB) therapy is a widely favored anti-tumor treatment, but it shows limited response to non-immunogenic “cold” tumors and suffers from drug resistance. Photodynamic therapy (PDT), as a powerful localized treatment approach, can convert a “cold tumor” into a “hot tumor” by inducing immunogenic cell death (ICD) in tumor cells, thereby enhancing tumor immunogenicity and promoting tumor immunotherapy. However, the effectiveness of PDT is largely hindered by the limited penetration depth into tumor tissues. To address these issues, we proposed an all-in-one drug system with NIR-II photo-accelerated PDT effects, efficient immune checkpoint gene silencing, and a facile manufacturing process. The so-called all-in-one drug system comprises a multi-modal designed polymer PPNP and siRNA. PPNP is an amphipathic polymer that includes the near infrared-II (NIR-II) photosensitizer Aza-boron-dipyrromethene (Aza-BODIPY), a glutathione (GSH)-cleavable linker, and a cationic monomer derived from cholesterol. PPNP can self-assemble and efficiently load siRNA. Under laser irradiation, PPNP triggers a potent ICD cascade, causing the on-demand release of siPD-L1, reshaping the tumor's immunosuppressive microenvironment, effectively inhibiting the growth of various tumors, and stimulating the immune memory. This study represents a generalized platform for PDT and gene silencing, designed to modulate immune-related signaling pathways for improved anticancer therapy.
