Determination and Correlation of Solubility of Borneol, Camphor, and Isoborneol in Different Solvents

Jingjing Chen,Jingjing He,Ningxin Li,Huidong Zheng,Suying Zhao

The solubility of borneol, camphor, and isoborneol in four solvents (i.e., acetone, ethanol, p-cymene, and p-xylene) was measured by the static equilibrium method within the temperature range of 296.65–348.85 K under the atmosphere. The fusion points and fusion enthalpies of borneol and isoborneol were determined by differential scanning calorimetry. The Apelblat equation, Wilson model, and NRTL model were employed to correlate the measured solubility data, and the NRTL model fits the measured data better. Mixing properties of each solute–solvent pair are calculated by the NRTL model.

Curdione combined with borneol treats bacterial mixed HPV infection by regulating the crosstalk among immune cells

Jingwei Liu,Tong Shu,Yiheng Mu,Wanlin Zheng,Xiaohuan Lu,Hong Tao

Background: Human papillomavirus (HPV) infection is a worldwide reproductive system disease. Baofukang suppository, a traditional herbal preparation that includes curdione and borneol, has been reported to treat bacterial vaginosis (BV) and HPV infection in China. However, the therapeutic mechanism is still unknown. This study aims to explore the molecular mechanisms of curdione and borneol in treating HPV infection. Methods: We conducted a retrospective cohort analysis of medical records from a single-center study involving 205 HPV patients, focusing on the correlation between HPV clearance and co-infection with other pathogens, confirming the efficacy of Baofukang suppository. Bioinformatics and network pharmacology approaches were employed to identify therapeutic targets of Baofukang suppository for BV/HPV co-infections. qRT-PCR, Western blot, immunofluorescence staining, and flow cytometry were utilized to validate the therapeutic targets of curdione and borneol, along with the associated immune molecular changes. Finally, the molecular mechanisms and therapeutic efficacy of curdione and borneol were confirmed in vivo using an LPS/TC-1 cervical orthotopic injection model. Results: Curdione and borneol selectively inhibit the secretion of interleukin-6 (IL-6) and interleukin-1β (IL-1β) by macrophages. The reduction in IL-6 and IL-1β levels effectively inhibits the expression of CD274 (Programmed death ligand 1, PD-L1) in infected epithelial cells by inhibiting STAT3 phosphorylation, thereby suppressing their immune evasion capabilities. Furthermore, curdione and borneol enhance the expression of tumor necrosis factor α (TNF-α) and caspase 1 (CASP1) in macrophages, as well as the expression of interleukin 12 (IL-12) and interleukin 23 (IL-23) in dendritic cells (DCs). The expression of these inflammatory factors effectively promotes the migration and differentiation of T cells to the site of infection, completing the clearance of infected epithelial cells. Conclusion: The main components of Baofukang suppository, curdione and borneol, inhibit the progression of HPV infection and the occurrence of cervical cancer by modulating the communication between innate and adaptive immunity, promoting the recruitment and recognition of CD8+ T cells to eliminate HPV-infected epithelial cells.

In situ borneol-modified polyester with antibacterial adhesion and long-term fungal-repellent properties

Zixu Xie,Chen Chen,Xinyu Chen,Fanqiang Bu,Guofeng Li,Pengfei Zhang,Xing Wang

Polyester is widely used in biomedical, textile, and food packaging fields. Therefore, enhancing it with antimicrobial properties would be a significant advancement. In this paper, a series of borneol-triazine polyesters (BTPs) with different structures are synthesized through room temperature polycondensation . The structure and composition of BTPs are systematically characterized by 1 H NMR, FTIR and GPC . Antimicrobial results reveal that the ability of BTPs to resist bacterial or fungal adhesion is directly related to the polymer structure . When the polymer chain of BTPs adopts a rigid structure, they exhibit excellent anti-adhesive and inhibitory performances against both Gram-negative bacteria ( Escherichia coli ) and Gram-positive bacteria ( Staphylococcus aureus ). Meanwhile, the as-synthesized BTPs poses a fungal-repelling effect on common fungal strains ( Aspergillus niger ) for up to 30 d. Further studies have shown that a stereochemical structure brought by borneol is key for imparting effective antimicrobial properties to BTPs. In addition, BTPs are non-leaching materials with low cellular cytotoxicity. Taking into consideration, BTP provides a potential strategy for preparing a new class of antimicrobial polyester materials .

Durable anti-superbug adhesion cotton textiles decorated with natural borneol-based finishing agent

Xu Jiangqi,Li Xia,Yu Wenjing,Wang Xing,Ma Jianzhong

There is an urgent need to develop novel antimicrobial cotton textiles (CT) because microbial contamination is one of the greatest threats to public health due to the emerging of infectious diseases and drug-resistant pathogens. In this work, an isocyanate group containing borneol-based antimicrobial finishing agent (NCO-B) was developed which can be covalently grafted to the surface of cotton textiles by a simple one-step method. The obtained NCO-B-finished CT (NCO-B-CT) exhibited broad-spectrum antimicrobial adhesion properties against bacteria ( E. coli ), including superbugs (MRSA), and fungi ( A. niger ). Due to its unique antimicrobial mechanism of borneol stereochemistry, the obtained NCO-B-CT is a non-releasing antimicrobial material which causes no skin sensitization and has no cytotoxicity. The mechanical properties of the CT were also improved after this finishing process. Additionally, it displayed durable washing fastness and antimicrobial adhesion properties by bearing 50 times laundering cycles. Therefore, this material may have great potential for application in multifunctional textiles, flexible skin electronics and medical protection. Graphical abstract

Antibiofilm Activities of Borneol-Citral-Loaded Pickering Emulsions against Pseudomonas aeruginosa and Staphylococcus aureus in Physiologically Relevant Chronic Infection Models

Wen Wang,Xuerui Bao,Mona Bové,Petra Rigole,Xiaofeng Meng,Jianyu Su,Tom Coenye

Phytochemicals are promising antibacterials for the development of novel antibiofilm drugs, but their antibiofilm activity in physiologically relevant model systems is poorly characterized. As the host microenvironment can interfere with the activity of the phytochemicals, mimicking the complex environment found in biofilm associated infections is essential to predict the clinical potential of novel phytochemical-based antimicrobials. In the present study, we examined the antibiofilm activity of borneol, citral, and combinations of both as well as their Pickering emulsions against Staphylococcus aureus and Pseudomonas aeruginosa in an in vivo-like synthetic cystic fibrosis medium (SCFM2) model, an in vitro wound model (consisting of an artificial dermis and blood components at physiological levels), and an in vivo Galleria mellonella model. The Pickering emulsions demonstrated an enhanced biofilm inhibitory activity compared to both citral and the borneol/citral combination, reducing the minimum biofilm inhibitory concentration (MBIC) values up to 2 to 4 times against P. aeruginosa PAO1 and 2 to 8 times against S. aureus P8-AE1 in SCMF2. In addition, citral, the combination borneol/citral, and their Pickering emulsions can completely eliminate the established biofilm of S. aureus P8-AE1. The effectiveness of Pickering emulsions was also demonstrated in the wound model with a reduction of up to 4.8 log units in biofilm formation by S. aureus Mu50. Furthermore, citral and Pickering emulsions exhibited a significant degree of protection against S. aureus infection in the G. mellonella model. The present findings reveal the potential of citral- or borneol/citral-based Pickering emulsions as a type of alternative antibiofilm candidate to control pathogenicity in chronic infection.IMPORTANCE There is clearly an urgent need for novel formulations with antimicrobial and antibiofilm activity, but while there are plenty of studies investigating them using simple in vitro systems, there is a lack of studies in which (combinations of) phytochemicals are evaluated in relevant models that closely resemble the in vivo situation. Here, we examined the antibiofilm activity of borneol, citral, and their combination as well as Pickering emulsions (stabilized by solid particles) of these compounds. Activity was tested against Staphylococcus aureus and Pseudomonas aeruginosa in in vitro models mimicking cystic fibrosis sputum and wounds as well as in an in vivo Galleria mellonella model. The Pickering emulsions showed drastically increased antibiofilm activity compared to that of the compounds as such in both in vitro models and protected G. mellonella larvae from S. aureus-induced killing. Our data show that Pickering emulsions from phytochemicals are potentially useful for treating specific biofilm-related chronic infections.

Enzymatic lipophilization of d-borneol extracted from Cinnamomum camphora chvar. Borneol seed

Zhe Dong,Yu Zhao,Jinliang Chen,Ming Chang,Xiaosan Wang,Qingzhe Jin,Xingguo Wang

Borneol esters have attracted increasing interest due to their potential application in the food industry. In this study, the extraction of d -borneol from Cinnamomum camphora chvar. borneol seed was carried out via steam distillation to gain highly purified d -borneol (99.66%). The lipase-catalyzed lipophilization of extracted d -borneol with oleic acid was performed in a solvent-free system. The influences of lipase type, substrate molar ratio, lipase loading, reaction temperature and reaction time on d -bornyl oleate yield were evaluated. Among four kinds of commercial lipases screened for their esterification efficiency, CRL (lipase from Candida rugose ) displayed the highest catalytic activity in the synthesis of d -bornyl oleate. The yield of more than 95% was obtained under the optimal conditions: substrate molar ratio 1:2 ( d -borneol/oleic acid), lipase loading 8%, reaction temperature 45 °C, reaction time 48 h. The structural identification of d -borneol and product d -bornyl oleate was performed by gas chromatography mass spectrometry (GC-MS) and proton nuclear magnetic resonance spectroscopy ( 1 H NMR). The results showed that the product was confirmed to be d -bornyl oleate and the purity of which was 97.51%. This study will pave the way for d -borneol and its esters application in food industry.

Antifungal Paper Based on a Polyborneolacrylate Coating

Jiangqi Xu,Yujia Bai,Meijiao Wan,Yanhui Liu,Lei Tao,Xing Wang

Paper documents and products are very susceptible to microbial contamination and damage. Fungi are mainly responsible for those biodeterioration processes. Traditional microbicidal strategies constitute a serious health risk even when microbes are dead. Ideal methods should not be toxic to humans and should have no adverse effects on paper, but should own a broad spectrum, good chemical stability and low cost. In this work, we utilize an advanced antimicrobial strategy of surface stereochemistry by applying a coating of a shallow layer of polyborneolacrylate (PBA), resulting in the desired antifungal performance. The PBA-coated paper is challenged with the most common air-borne fungi growing on paper,Aspergillus nigerandPenicilliumsp. Ten percent by weight of the coating concentration or a 19-μm infiltration of PBA is sufficient to keep the paper spotless. The PBA coating also exhibits significant inhibition of spores’ germination. After PBA coating, both physicochemical properties (paper whiteness, pH, mechanical strength) and inking performance display only slight changes, which are acceptable for general utilization. This PBA coating method is nontoxic, rapid and cost-effective, thus demonstrating great potential for applications in paper products. Keywords:antifungal;coating;paper products;polyborneolacrylate;surface stereochemistry

Supramolecular cyclodextrin-based reservoir as nasal delivery vehicle for rivastigmine to brain

Manli Wang,Yuting Lv,Huipeng Xu,Xiangyu Zhao,Guoqing Zhang,Siwen Wang,Caifen Wang,Wenting Wu,Li Wu,Weifeng Zhu,Jiwen Zhang

The purpose of this study involved the synthesis of supramolecular reservoir (i.e. cyclodextrin metal-organic framework, MOF) using cyclodextrins as building blocks, followed by cross-linking to obtain crosslinked CD framework (CDF) using CD-MOF as template and functionalized with borneol (BO) to enhance rivastigmine (RIV) permeation and facilitate brain targeting via intranasal administration. Utilizing BO modified CDF (BO-CDF) with cubic shape as a carrier for the encapsulation of RIV, a nasal RIV delivery system (RIV@BO-CDF) was fabricated. The particle size of RIV@BO-CDF was approximately 250 nm, and the drug loading capacity reached 15 ± 2 %. BO-CDF improved the mucoadhesion and enhanced RIV permeability with the plasma concentration-time curve (AUC), the brain AUC and the peak drug concentration within brain in rats 1.7, 2.3 and 8 times than that of oral RIV solution, respectively. The relative drug targeting efficiency percentage (DTE, 139.4 %) and direct drug transfer percentage (DTP, 28.3 %) of RIV@BO-COF indicated good targeting efficiency and direct nose-to-brain drug delivery. Overall, this study provides a potential application of supramolecular cyclodextrin-based reservoir to enhance the brain targeting and efficacy of the RIV via nasal delivery.

Design, characterization and comparison of transdermal delivery of colchicine via borneol-chemically-modified and borneol-physically-modified ethosome

Yujia Zhang,Nan Zhang,Hui Song,He Li,Jin Wen,Xiaochuan Tan,Wensheng Zheng

Gout is a kind of joint disease characterized by the accumulation of monosodium urate (MSU) crystals in the joint and its surrounding tissue, causing persistent hyperuricemia. Colchicine is the first choice of treatment for acute gout attacks. Due to strong toxicity of colchicines oral tablets, there are high fluctuations of blood drug concentration and serious irritation of gastrointestinal tract, and hence a transdermal preparation can help to slow down the blood drug concentration, reduce the frequency of drug taking, and improve the patients' compliance of the drug. The ethosome is a lipid carrier with high concentration of ethanol and has been proved to promote the penetration of drugs into the skin. Borneol (BO) is an excellent penetration enhancer in Chinese medicine, which can promote the entry of drugs into the skin. This paper prepared the borneol-physically-modified colchicine ethosome (COL-bpES) and used the prepared borneol-dioleoyl phosphoethanloamine (BO-DOPE) to prepare borneol-chemically-modified colchicine ethosome (COL-bcES). Compared to the free colchicine aqueous solution (free COL) and normal colchicine ethosome (COL-ES), the borneol-modified colchicine ethosome (COL-bES) demonstrated better drug penetration effect, while the particle size of the COL-bcES was lower than that of the COL-bpES. Toxicity, in vitro diffusion, pharmacokinetics and pharmacodynamics are superior to those of COL-bpES, providing a better delivery system for the treatment of small molecule inflammatory drugs.

Authentication of Two Different Chemical Types of Cinnamomum Camphora Leaves by Microscopic Technique with GC-MS and GC Analysis

Ying-Jiao Liu,Li-Juan Xie,Lan Xiao,Li-Min Gong,Ta-Si Liu,Nai-Hong Chen

Cinnamomum camphora Chvar. Borneol (CCCB) and Cinnamomum camphora Chvar. Cineol (CCCC) are two different chemical types of C. camphora . The clinical application of these chemical types of CC shows big differences, but sometimes they are confused because of the similarity of morphological features. To identify the two types of CC and to analyze the differences of compositions, their morphological characteristics were investigated by microscopic techniques and the chemical components were determined by GC-MS analysis and GC internal standard method. Distinguishing features of morphological characteristics were the shape, petiol, gland, and smell. Microscopic study of the transverse sections of leaves showed 13 – 15 columns xylem vessel in the leaves of CCCB, 11 – 13 columns xylem vessel, and more obvious and thicker cell walls of fiber cells in the leaves of CCCC. From this research, GC-MS analysis of essential oil of the leaves could provide mainly differential characteristics to distinguish between CCCB and CCCC. The total of 33 compounds were identified in the essential oil of CCCB leaves and the main component was d -borneol (55.11%), while d -borneol in CCCC leaves was not the main component and the amount of d -borneol might be related to the microstructure of gland. L(-)-camphor and d-borneol were quantitatively analyzed by GC with FID detector, using methyl salicylate as the internal standard. Each gram of the total essential oil contains 0.68 g of d -borneol in CCCB, and each gram of the total essential oil contains 0.150 and 0.015 g of L(-)-camphor in CCCB and CCCC, respectively, from Hunan province. Other habitats showed the same trend. This research was aimed to establish a method based on the microscopic, GC-MS, and GC techniques to identify and quantify CCCB and CCCC compositions, for providing scientific data to ensure with accuracy the origin of CCCB and CCCC plant material.

Core–shell alum-borneol fiber for high bioavailability

Lv Yarong,Han Yufen,Yu Zhongxun,Chen Jia,Li Chenxi,Wang Ce,Hu Ping,Liu Yong

Currently, the treatment of burns poses a significant challenge to clinical surgical. The use of nanofibers combined with drugs provides an entirely new option for treating burns. Alum-borneol combination has been shown as a promising alternative in clinical burn treatment. However, the utilization of the alum-borneol combination is not optimistic due to the low solubility of borneol. In this study, alum-borneol incorporated polyvinyl pyrrolidone fibers with a core–shell structure were fabricated through coaxial electrospinning. In vitro Borneol release behavior of fibers with different ratios of alum to borneol was explored. Scanning electron microscopy, transmission electron microscope, Fourier transform infrared spectroscopy, X-ray diffraction, differential scanning calorimeter, in vitro drug release, and in vitro release mechanism were evaluated. The results showed that the fiber membranes maintained an integrated morphology. In vitro dissolution data showed an improved solubility of borneol, which reached more than 82% at 240 min in alum-borneol fibers. It was 4.8 times higher than borneol powder, and the ratio of alum to borneol was 2:1 for the best results. Therefore, alum-borneol incorporated polyvinyl pyrrolidone fibers can significantly improve the dissolution rate of borneol, which opens up a new way for the combined application of the alum and borneol.

Preparation of Alum-borneol-PVP Drug-loaded Fibers by Electrospinning

Huang Libing,Liu Yueqi,Sang Xinyu,Song Jinghui,Hu Ping,Liu Yong

T he alum-borneol nanoemulsion(ABN), which combines the mineral medicine alum and the botanical medicine borneol, has been applied for approximately 40 years in the clinical treatment of burns, scalds, radiation dermatitis and shingles, and has a good curative effect. However, the current formula and dosage form of ABN pose problems of low borneol content and ease of precipitation, which greatly affects the efficacy of the drug. In this study, polyvinylpyrrolidone(PVP) was selected as a carrier mixed with different proportions of alum and borneol to produce alum-borneol-PVP fibers(ABPF) by electrospinning. The results showed that the stable system with good drug dispersion was 2:3(alum:borneol). The dissolution content of borneol from the ABPF was about 80% in 4 h, which was much higher than that of the alum-borneol liquid(ABL) and ABN. The ABPF membrane showed a more significant inhibitory effect on Staphylococcus aureus than the ABL and ABN. The composite fiber markedly increased the drug content of borneol, which was 800 times of that in ABN. The fiber had a higher solubility than the nanoemulsion in vitro , which is of great importance for the development of new forms for the clinical application of alum and borneol.
