Butenolide derivatives from Aspergillus terreus selectively inhibit butyrylcholinesterase

Cui Xiang,Deng Shanshan,Li Guoyin,Zhang Yunxia,Wang Lining,Wu Changjing,Deng Yanru

Two undescribed butenolide derivatives, asperteretal J (1) and K (2), together with 13 known ones (3–15) were isolated from an endophytic fungus Aspergillus terreus SGP-1, the fermentation product of which exhibited selective inhibitory activity toward butyrylcholinesterase. The structures of the new compounds were elucidated based on HRMS and NMR data, and the absolute configurations were determined by specific optical rotation comparison. All compounds were evaluated for cholinesterase inhibitory effects with galantamine as a positive control. Compounds 4–8 selectively inhibited butyrylcholinesterase with IC50 values of 18.4–45.8 µM in a competitive manner, with Ki values of 12.3–38.2 µM. The structure-activity relationship was discussed. Molecular docking and dynamic simulation of the inhibitor-enzyme complex were performed to better understand the interactions.

Environmentally Friendly Antifouling Coatings Based on Biodegradable Polymer and Natural Antifoulant

Chunfeng Ma,Weipeng Zhang,Guangzhao Zhang,Pei-Yuan Qian

Marine biofouling causes huge economic losses and serious problems to maritime industries. Consequently, there is a great demand for environmentally friendly antibiofouling technology. In the present study, we developed novel antifouling coatings by incorporating butenolide derived from marine bacteria into biodegradable poly(ε-caprolactone) based polyurethane. Measurements of mass loss indicated that the polymer was degraded in seawater and that the degradation rate increased in the presence of marine organisms or enzymes. Moreover, measured release rates indicated that butenolide could be released from the biodegradable polymer for at least 3 months and the release rate depended on both the concentration of butenolide and the temperature. Incorporating a naturally occurring resin (rosin) into the biodegradable polymer increased the self-renewal rate and improved the later release rate of butenolide. The field test indicated that the system had excellent antifouling properties.

Biomimetic Butenolide Antifoulants Enable Effective Control of Marine Biofouling on Aquaculture Netting

Jianhua Zhu,Jinye Bi,Huikai Chen,Cailing Zhao,Guoqiang Lu,Jianming Wang,Dandan Li,Jianrong Song,Chunwen Zhang

HighlightsWhat are the main findings?A biomimetic butenolide antifoulant was synthesized via a green route and integrated into silicone coatings. The coatings exhibit strong antibacterial activity against Gram-positive and Gram-negative bacteria, including the marine pathogenic bacteriumVibrio harveyi.Inhibits algal attachment at 8 wt% while preserving planktonic growth, indicating a non-biocidal mechanism.Long-term field tests confirm durable antifouling performance in marine environments. What is the implication of the main finding?Enables sustainable, eco-compatible antifouling coatings for offshore aquaculture applications.Marine biofouling on aquaculture netting compromises mass transfer and structural performance, particularly in high-fouling marine environments. Here, a biomimetic antifoulant based on a butenolide scaffold was synthesized and incorporated into silicone coatings to achieve effective antifouling performance. The antifoulant exhibited bactericidal activity againstEscherichia coli,Vibrio harveyi, andStaphylococcus aureus, while coating characterization showed increased hydrophobicity and reduced water uptake with minimal loss of adhesion. Algal attachment assays revealed significant inhibition ofIsochrysis zhanjiangensis, with an optimal loading of 8 wt%. Notably, planktonic algal growth remained largely unaffected, indicating a surface-mediated mechanism. Mechanistically, the antifoulant is proposed to interfere with quorum sensing-regulated microbial behavior, thereby suppressing initial colonization and biofilm formation. Field immersion tests over six months confirmed sustained antifouling performance with low fouling coverage and stable coating integrity. These findings demonstrate a promising strategy for environmentally compatible biofouling control in offshore aquaculture.

Butenolide Derivatives with α-Glucosidase Inhibitions from the Deep-Sea-Derived Fungus Aspergillus terreus YPGA10

Zhongbin Cheng,Yuanli Li,Wan Liu,Lijun Liu,Jie Liu,Wangjun Yuan,Zhuhua Luo,Wei Xu,Qin Li

Three new butenolide derivatives, namely aspernolides N–P (1–3), together with six known analogues (4–9), were isolated from the ethyl acetate (EtOAc) extract of the deep sea-derived fungusAspergillus terreusYPGA10. The structures of compounds1–3were determined on the basis of comprehensive analyses of the nuclear magnetic resonance (NMR) and mass spectroscopy (MS) data, and the absolute configurations of1and2were determined by comparisons of experimental electronic circular dichroism (ECD) with calculated ECD spectra. Compound1represents the rare example ofAspergillus-derived butenolide derivatives featured by a monosubstituted benzene ring. Compounds6–9exhibited remarkable inhibitory effects againstα-glucosidase with IC50values of 3.87, 1.37, 6.98, and 8.06 μM, respectively, being much more active than the positive control acarbose (190.2 μM).Keywords:Aspergillus terreusYPGA10;deep-sea-derived fungus;butenolide derivatives;α-glucosidase
