An Electrochemical Aptamer-Based (E-AB) Sensor Using Ti3C2Tx(MXene) and cDNA-PAA for Detection of AFB1

Xiaoya Meng,Zhongyu Li,Zili Xia,Dongdong Chen,Zijing Yuan,Yang Han,Heng Zhang,Xia Sun,Jun Seop Lee,Usmanova Surayo Rakhmatzhanovna,Bobokalonov Jamshed Tolehmurodovich,Wenping Zhao

An electrochemical aptasensor based on Ti 3 C 2 T x (MXene) and cDNA-PAA was established for sensitive detection of aflatoxin B1 (AFB1) in peanuts. The high specific surface area of Ti 3 C 2 T x (MXene) contributed to improve electrochemical efficiency and enhancing sensor stability which were characterized by the Randles−Sevcik equation. The self-made polyacrylic acid (PAA) nanogel was added with aptamer complementary chain (cDNA) to form cDNA-PAA nano gel composite, which could realize the synergistic amplification of the difference between the electrochemical signals before and after the addition of AFB1. The sensing effect of cDNA-PAA was validated using DPV. Methylene blue (MB) was modified at the proximal 3′ termini of the aptamer (Apt-MB), meanwhile AuNPs was used to immobilize Apt-MB and improve the efficiency of electrochemical reactions. The competition between AFB1 and cDNA-PAA combined with Apt MB resulted in a significant change in the electrochemical signal current. In addition, the constructed electrochemical aptasensor had a lower detection limit of 1.0 × 10 −3 ng/L for AFB1 under optimal detection conditions, and a detection range of 1 to 1000 ng/L. Moreover, peanuts with different concentrations of AFB1 were used as actual detection samples. Through the constructed sensor detection, the spiked recovery rates within the spiking range of 50 to 500 ng/mL were 97.76% to 101.84% ( n = 3).

Fluorescent biosensor based on aptamer-cDNA hybridization for Cd2+ detection and molecular mechanism investigation

Zhenzhen Xie,Wei Jia,Xingzhen Li,Lei Pan,Shihao Xu,Changlong Jiang

Cadmium ion (Cd 2+ ) pollution poses severe environmental and health risks due to its high toxicity, bioaccumulation potential, and persistence. Although traditional detection methods require bulky instruments and complex procedures, limiting the suitability for rapid on-site analysis. The paper introduces aptamer-based fluorescent probes and double-stranded fluorescent probe based on an aptamer and its complementary DNA (cDNA). The former is composed of aptamers and SYBR Green dyes. The detection range of Cd 2+ is 0–8 μM, and limit of detection (LOD) is 15 nM. The probe consists of a double-stranded structure formed by a 5′-FAM-labeled Cd 2+ aptamer and a 3′-BHQ1-modified cDNA. By combining this probe with a hydrogel matrix, a smartphone-based sensing platform is constructed, which enables highly sensitive and convenient detection of Cd 2+ . It indicated that the detection range of the probe for Cd 2+ is 0–50 μM, with the LOD of 27.6 nM. Molecular dynamics simulations (MD) disclosed the binding mechanism of the aptamer and Cd 2+ : the specific recognition by nucleobases such as G9, C12, and T17 in the Coulomb force-driven double-twisted folding conformation plays a crucial role. Further integrating the probe into the hydrogel matrix constructed the dsDNA@hydrogel platform successfully achieved real-time visual detection of Cd 2+ in environmental samples. It not only offers a highly sensitive and cost-effective new approach for heavy metal detection, but promotes the advancement of portable environmental monitoring technologies through the innovation of the aptamer-hydrogel system.

Characterization of Chitin Synthase A cDNA from Diaphorina citri (Hemiptera: Liviidae) and Its Response to Diflubenzuron

Cong Zhang,Wenfeng Hu,Zhuo Yu,Xian Liu,Jing Wang,Tianrong Xin,Zhiwen Zou,Bin Xia

Simple SummaryThe transcriptional level of chitin synthase A (DcCHSA) was investigated inDiaphorina citriKuwayama at various developmental stages and tissues in this work. The survival and molting ofD. citrinymphs were negatively impacted by high concentrations of DFB, which could also causeDcCHSAto express itself significantly. We also looked into how CHS knockout affected nymph development and eclosion. The findings revealed thatDcCHSAwas critical for the eclosion and molting ofD. citrinymphs. This study provides a solid theoretical foundation forD. citrimanagement in China.AbstractDiaphorina citriKuwayama is the vector of HLB and one of the most common pests in citrus orchards in southern China. One of the most significant genes inD. citri’s growth and development is the chitin synthase gene. In this study, the CHS gene (DcCHSA) ofD. citriwas cloned and analyzed by bioinformatics. According to RT-qPCR findings,DcCHSAwas expressed at many growth processes ofD. citri, with the greatest influence in the fifth-instar nymph. The molting failure rate and mortality ofD. citrirose as DFB concentration increased in this research, as did the expression level ofDcCHSA. Feeding onDcCHSAcaused a large drop in target gene expression, affected nymph molting, caused failure or even death in freshly eclosion adults, increased mortality, and reduced the molting success rate over time. These findings showed thatDcCHSAwas involved in nymph to adult development and may aid in the identification of molecular targets forD. citriregulation. It provided new ideas for further control ofD. citri. Keywords:Diaphorina citri;chitin;diflubenzuron;transcription level;RNA interference

Probe-Type Fiber Optic Biosensor Based on Barium Titanate Glass Microsphere Enhanced SPR Effect for Virus RNA Detection

Xuegang Li,Huiying Sun,Yanan Zhang,Yong Zhao,Xue Zhou

In this paper, a groundbreaking probe-type fiber optic biosensor, harnessing the surface plasmon resonance (SPR) effect enhanced by barium titanate glass microspheres, is unveiled for virus RNA detection. The multi-physics simulation was used to verify the sensitivity improvement achieved by introducing barium titanate microspheres on the surface of the optical fiber microspheres probe. Experiments confirmed this, pushing the sensor's limit to an impressive 6802 nm/RIU. Biomarker capture on the microsphere surface subtly alters the local refractive index, triggering a detectable SPR spectral shift. To showcase its prowess, the sensor tackled the reverse-transcribed complementary DNA (cDNA) of COVID-19 RNA. Chemical modification of the gold film and DNA hybridization technology ensured specific cDNA detection. The limit of detection for this target reached a remarkable 1.58 nmol/L. This sensor not only excels in virus RNA detection but also holds immense potential for diagnosing major diseases by pinpointing the concentrations of key biomarkers.

Dipeptide nanoparticle and aptamer-based hybrid fluorescence platform for enrofloxacin determination

Jin Yuting,Yan Rongfang,Wang Shuo,Wang Xianghong,Zhang Xuemei,Tang Yiwei

A novel fluorescence platform was fabricated for enrofloxacin determination by using cDNA-modified dipeptide fluorescence nanoparticles (FDNP-cDNA) and aptamer-modified magnetic Fe 3 O 4 nanoparticles (Fe 3 O 4 -Apt). The FDNP were prepared via tryptophan-phenylalanine self-assembling. When magnetic Fe 3 O 4 -Apt incubated with standard solution or sample extracts, the target enrofloxacin was selectively captured by the aptamer on the surface of the Fe 3 O 4 nanoparticles. After removing interference by washing with phosphate-buffered saline, the FDNP-cDNA was added, which can bind to the aptamer on the surface of the Fe 3 O 4 nanoparticles not occupied by the analyte. The higher the concentration of the target enrofloxacin in the standard or sample solution is, the less the FDNP-cDNA can be bound with the Fe 3 O 4 nanoparticles, and the more the FDNP-cDNA can be observed in the supernatant. Fluorescence intensity (E x /E m  = 310/380 nm) increased linearly in the enrofloxacin concentration range 0.70 to 10.0 ng/mL with a detection limit of 0.26 ng/mL (S/N = 3). Good recoveries (88.17–99.30%) were obtained in spiked lake water, chicken, and eel samples with relative standard deviation of 2.7–6.2% ( n  = 3). Graphical abstract

Aptamer-cDNA-driven nanozyme strategy: A recyclable dual-signal immunosensor for amplified detection of aflatoxin B1 in edible oil

Xiaolong Li,Wenlin Li,Mengmeng Cai,Haoxin Gong,Xuejiao Wang,Chen Dong,Cuixiang Li,Shangyi Zhao,Mengna Wan,Xueqin Wang

Aflatoxin B 1 (AFB 1 ) poses a serious threat to human health, highlighting theurgent need for sensitive, precise and visual detection methods in daily foods. However, traditional detection methods relying on peroxidase (POD)-mimetic activity are susceptible to external interference, which increases false-positive or false-negative rates and compromise precision and repeatability. To address these challenges, a dual-signal nano-immunosensor was developed using an aptamer-cDNA-assisted colorimetric-fluorescent nanozyme-linked immunosorbent assay for ultrasensitive AFB 1 detection in oils. This competitive enzyme-linked immunosorbent assay (ELISA)-type nanosensor incorporates a Cu 2 O@MnO 2 nanozyme that catalyzes the colorimetric reaction to generate the first signal, while the second (fluorescent) signal is produced via rhodamine B isothiocyanate (RBITC) fluorescence quenching through the inner filter effect. This strategy combines the high specificity of aptamers for AFB 1 with the exceptional oxidase-mimicking activity of cDNA-decorated Cu 2 O@MnO 2 nanozymes. In addition, the integration of aptamer-conjugated Fe 3 O 4 magnetic nanoparticles (MNPs) enables high specificity for AFB 1 , as well as magnetic separation and nanozyme recyclability. Compared with traditional ELISA, the composited nanosensor demonstrated remarkable enzymatic capability, achieving 188-fold and 68-fold changes in colorimetric and fluorescent modes, respectively, with detection limits significantly loweredto 0.016 ng/mL for colorimetry and 0.044 ng/mL for fluorescence. Therefore, this immunosensor provides a highly reliable, accurate, sensitive and visible approach for rapid AFB 1 detection, offering an advanced strategy for future food safety monitoring .

Molecular Cloning and Expression of CYP9A61: A Chlorpyrifos-Ethyl and Lambda-Cyhalothrin-Inducible Cytochrome P450 cDNA from Cydia pomonella

Xueqing Yang,Xianchun Li,Yalin Zhang

Cytochrome P450 monooxygenases (CYPs or P450s) play paramount roles in detoxification of insecticides in a number of insect pests. However, little is known about the roles of P450s and their responses to insecticide exposure in the codling mothCydia pomonella(L.), an economically important fruit pest. Here we report the characterization and expression analysis of the first P450 gene, designated asCYP9A61, from this pest. The full-length cDNA sequence ofCYP9A61is 2071 bp long and its open reading frame (ORF) encodes 538 amino acids. Sequence analysis shows that CYP9A61 shares 51%–60% identity with other known CYP9s and contains the highly conserved substrate recognition site SRS1, SRS4 and SRS5. Quantitative real-time PCR showed thatCYP9A61were 67-fold higher in the fifth instar larvae than in the first instar, and more abundant in the silk gland and fat body than other tissues. Exposure of the 3rd instar larvae to 12.5 mg L−1of chlorpyrifos-ethyl for 60 h and 0.19 mg L−1of lambda-cyhalothrin for 36 h resulted in 2.20-and 3.47-fold induction ofCYP9A61, respectively. Exposure of the 3rd instar larvae to these two insecticides also significantly enhanced the total P450 activity. The results suggested thatCYP9A61is an insecticide-detoxifying P450.Keywords:Cydia pomonella;cytochrome P450;induction;response;insecticides

Highly sensitive aptasensor based on synergetic catalysis activity of MoS2-Au-HE composite using cDNA-Au-GOD for signal amplification

Hai-Yan Song,Tian-Fang Kang,Li-Ping Lu,Shui-Yuan Cheng

Single or few-layer nanosheets of MoS 2 (MoS 2 nanosheets) and a composite composed of MoS 2 nanosheets, Au nanoparticles (AuNPs) and hemin (HE) (denoted as MoS 2 -Au-HE) were prepared. The composites possessed high synergetic catalysis activity towards the electroreduction of hydrogen peroxide. Furthermore, glucose oxidase (GOD) and AuNPs were used as marker of the complementary DNA (cDNA) strand of kanamycin aptamer to prepare a conjugate (reffered as cDNA-Au-GOD) that was designed as the signal probe. Both cDNA-Au-GOD and MoS 2 -Au-HE were applied to fabricate aptasensor for kanamycin. MoS 2 -Au-HE acted as solid platform for kanamycin aptamer and signal transmitters. AuNPs were employed as the supporter of cDNA and GOD which catalyze dissolved oxygen to produce hydrogen peroxide in the presence of glucose. Then cathodic peak current of H 2 O 2 was recorded by differential pulse voltammetry (DPV). The electrochemical reduction of H 2 O 2 was catalyzed by MoS 2 -Au-HE that was modified onto the surface of a glassy carbon electrode (GCE). The cathodic peak current of H 2 O 2 was highly linearly decreased with an increase of kanamycin concentrations from 1.0   ng/L to 1.0×10 5   ng/L, with a detection limit of 0.8   ng/L. This aptasensor can be used to detect kanamycin in milk with high specificity, sensitivity and selectivity .

Sensitive ratiometric electrochemical detection of microRNA-21 based on dual signal amplification of AuNPs@Fc@MgAl-LDH coupled with MB-cDNA@AuNPs

Lan Jiang,Lina Zou,Jing Dong,Ping Zhang,Jian Zhou,Hao Jiang,Huanyu Ren,Huiru Niu,Hao Liao,Xiaojing Zhang,Shanshan An,Fei Ren,Xiuhong Ge,Feiyan Yang,Lang Cheng,Hongzhi Pan,Shengzhong Rong,Hongkun Ma

Non-coding RNAs are critical tumor biomarkers, rendering innovations in their detection technologies highly significant. To overcome the limitations of conventional single-signal electrochemical biosensors (susceptibility to interference, insufficient stability), this study developed a novel ratiometric electrochemical DNA biosensor with dual-signal amplification. Its core novelty is the rational integration of two synergistic nanomaterials: Ferrocene-functionalized MgAl-layered double hydroxide (Fc@MgAl-LDH, as stable immobilization matrix and internal reference signal source) and AuNPs@Fc@MgAl-LDH coupled with MB-cDNA@AuNPs, which construct a cascaded signal amplification pathway. The sensing mechanism involves two-stage amplification: Primary by high-loading-capacity AuNPs@Fc@MgAl-LDH, secondary by MB-cDNA@AuNPs hybridizing with electrode-immobilized microRNA-21 (miRNA-21). The ratiometric signal ( I MB / I Fc ) enables built-in correction, boosting detection accuracy and reproducibility. After systematic optimization, the sensor has a wide linear range (1.0 fM–0.1 nM) and an ultra-low LOD of 0.757 fM, eliminating RNA extraction/enzymatic amplification to simplify operation. Serum validation shows high accuracy (95.3%–101.4% recovery), indicating strong clinical diagnostic application potential.

Sensitive colorimetric detection of ochratoxin A by a dual-functional Au/Fe3O4 nanohybrid-based aptasensor

Rong Huang,Lu Lu Xiong,Hui Hui Chai,Jing Jing Fu,Zhisong Lu,Ling Yu

A novel colorimetric aptasensor based on a Au/Fe3O4 nanohybrid was developed to detect ochratoxin A (OTA). The aptasensor is composed of a free OTA aptamer, a Au/Fe3O4 nanohybrid coated with biotinylated complementary DNA of the OTA aptamer (biotin-cDNA-Au/Fe3O4), and free alkaline-phosphatase-labeled streptavidin (SA-ALP). The Au/Fe3O4 nanohybrid not only immobilizes biotin-cDNA but also magnetically separates SA-ALP from the sample solution. One part of the OTA aptamer sequence hybridizes with biotin-cDNA immobilized on Au/Fe3O4, and the left part of the OTA aptamer sequence covers the biotin and blocks the specific interaction between biotin and SA-ALP. OTA can interrupt the interaction of OTA aptamer binding to biotin-cDNA-Au/Fe3O4 and can inhibit the shielding effect of the OTA aptamer on biotin. The amount of SA-ALP that can be captured by biotin-cDNA-Au/Fe3O4 thus increases with increasing OTA concentration. Through a simple magnetic separation, the collected SA-ALP-linked Au/Fe3O4 can produce a yellow-colored solution in the presence of p-nitrophenyl phosphate (p-NPP). This colorimetric aptasensor can detect OTA as low as 1.15 ng mL−1 with high specificity.

Exploring the performance of multi-channel tetrahedral nucleic acid tweezers platforms for efficient and sensitive biosensing

Jingyang Zhang,Mengmeng Chen,Yuan Peng,Shuang Li,Dianpeng Han,Shuyue Ren,Kang Qin,Sen Li,Tie Han,Yu Wang,Zhixian Gao

The nucleic tweezers-based detection strategies have shown excellent application prospects in molecular detection and diagnosis thanks to the specific target responsiveness and good biocompatibility. However, the limited detection efficiency and unsatisfactory sensitivity make their applications in complex diagnosis difficult, and few reports focused on it. Here, we explored the molecular dynamics simulations and biosensing performance of the Tetrahedral Nucleic Acid Tweezers, including the Antennae like-Tetrahedron Nucleic Acid Tweezer (ATNAT) and the Covered-Tetrahedron Nucleic Acid Tweezer (CTNAT), revealing the molecular dynamics of ATNAT and CTNAT reporters visually and molecularly. After that, we compared the multi-target detection capabilities of DNA tetrahedral tweezers, and combined the CTNAT reporter with better multi-target detection performance with the aptamer and Exponential amplification reaction (EXPAR), developing an efficient and sensitive EXPAR-cDNA-CTNAT strategy. Then we applied the EXPAR-cDNA-CTNAT strategy to detect testosterone, cortisol, and creatine kinase isoenzymes, realizing sensitive and accurate fatigue diagnosis. Compared with the traditional detection strategies, the EXPAR-cDNA-CTNAT strategy showed improved sensitivity and detection efficiency with excellent specificity, and the limits of detection (LODs) for the multi-target detection were as low as 41, 68, and 8 pM, respectively. The EXPAR-cDNA-CTNAT strategy was reliable for multi-target detection, which had great potential in biological science, food safety, and medical diagnosis.

Universal DNA detection realized by peptide based carbon nanotube biosensors

Wenjun Li,Yubo Gao,Jiaona Zhang,Xiaofang Wang,Feng Yin,Zigang Li,Min Zhang

Although DNA recognition has been achieved using numerous biosensors with various sensing probes, the utilization of bio-interaction between DNA and biomolecules has seldom been reported in universal DNA detection. Peptides as natural molecules have the unique ability to bind to universal DNA and excellent selectivity for DNA after being functionalized with specific groups. In this work, we report a peptide based carbon nanotube (CNT) thin-film-transistor (TFT) biosensor, which can achieve sensitive sequence-independent DNA detection. In the presence of DNA, a significant increase of ΔIon could be observed within 5 minutes, which was considered to be due to the electrostatic adsorption between the DNA and peptide of opposite zeta potential. With the gradual increase of the concentration of DNA, the ΔIon signals agree with the Hill–Langmuir model (R2 = 0.98), indicating a negatively cooperative interaction between the peptide and DNA (the Hill coefficient n < 1). Compared with the former reported universal DNA bio-detector and NanoDrop (a spectrometer from Thermo Scientific™), this unique peptide based CNT-DNA sensor demonstrated a broader sensing range from nearly 1.6 × 10−4 to 5 μmol L−1 and a much lower detection limit of approximately 0.88 μg L−1. For the quantification of cDNA from T47D cancer cells, this unique peptide based CNT sensor could achieve efficient cDNA detection. To the best of our knowledge, this is the first report on the utilization of a peptide as a sensing element in the design of CNT based DNA biosensors, which enables highly efficient universal DNA detection.
