Formation and characterization of natural polysaccharide hollow nanocapsules via template layer-by-layer self-assembly

Yuxi Liu,Jing Yang,Ziqi Zhao,Junjie Li,Rui Zhang,Fanglian Yao

With natural polysaccharides carrageenan (Car) and chitosan (Cs) as the polyanion and polycation, respectively, multilayer hollow nanocapsules have been fabricated via sequential layer-by-layer (LbL) electrostatic self-assembly from the sacrificed templates nanospheres (SiO 2 –NH 2 ). The LbL assembly process with the polysaccharides on SiO 2 –NH 2 core was followed by ζ -potential and size analysis. The fabrication of (Car/Cs) x nanocapsules and the removing of the SiO 2 –NH 2 core templates were confirmed by TGA and EDS analysis. The morphology of SiO 2 (Car/Cs) x nanospheres and (Car/Cs) x nanocapsules were observed by TEM analysis. The size analysis of (Car/Cs) x nanocapsules indicated that the cyst wall thickness and cavity volume of the nanocapsules are pH and ionic strength dual responsive. Due to the biocompatibility of the natural polysaccharides carrageenan and chitosan and the responsiveness of nanocapsules to pH and ionic strength, the (Car/Cs) x multilayer nanocapsules are expected to be used as nanoreactors or nanocontainers to control the synthesis, encapsulation, and releasing behaviors of bioactive molecules.

Air permeable alumina-based porous ceramics with high performance prepared by novel gel casting route

Wang Ziyu,Chen Yuanqing,Demir Muslum,Wang Ting,Gu Mengmeng,Wang Cheng

In the present work, porous ceramics with open pores were fabricated using the gel casting method, which utilizes carrageenan as a cross-linking agent. Beyond the foaming agents or templates techniques, this approach employed large-sized Al 2 O 3 particles (~ 45 μm) as the underlying framework material, enabling the direct formation of minute open pores within the green bodies and sintered samples. In order to finely adjust the pore structure and bolster flexural strength, micropowders of α-Al 2 O 3 and Kaolin were incorporated as filling materials, alongside the introduction of rare-earth oxides as sintering additives. The study diligently examined the impact of varying the content of sintering additives as well as the sintering temperatures on the characteristics of the porous ceramics. As a result, remarkable achievements were obtained, culminating in the production of Al 2 O 3 -based porous ceramics measuring 8 mm in thickness, exhibiting flexural strength exceeding 160 MPa, porosity surpassing 30%, and exceptional gas permeability of over 30 L/min/cm 2 under a pressure drop of 100 kPa.

Comparative analysis of carrageenan/ovalbumin and carrageenan oligosaccharide/ovalbumin complexes

Hujun Xie,Qing Cao,Yingying Jiang,Yingying Pan,Sihan Chen,Qingbo Jiao,Gerui Ren,Di Wu,Min Huang

The saccharides ι-carrageenan (ι-CG) and ι-carrageenan oligosaccharides (ι-CO) exhibit different molecular chain lengths and charge intensities, which are pivotal determinants of their interactions with ovalbumin (OVA). This study investigated the interaction mechanisms between OVA and ι-CG as well as between OVA and ι-CO using experimental methods and molecular docking. The results of zeta potential showed that the strongest electrostatic interaction occurred at an OVA/ι-CG ratio of 100:13 (−1.59 ± 0.41 mV) and an OVA/ι-CO ratio of 100:10 (−1.32 ± 0.30 mV). The OVA/ι-CG complexes showed higher turbidity (1.63 ± 0.08 cm −1 ). Furthermore, Fourier transform infrared spectroscopy showed that ι-CG significantly altered the secondary structure of OVA, whereas ι-CO did not affect it. Fluorescence spectroscopy also verified the interactions in the OVA/ι-CG and OVA/ι-CO complexes. Both complexes exhibited shear-thinning behaviour; the largest storage modulus ( G ′) and loss modulus ( G ″) were observed at 100:13 and 100:10 ratios for OVA/ι-CG OVA/ι-CO samples, respectively. ι-CG induced OVA to form more loosely clumped aggregates, whereas OVA and ι-CO formed compact granular aggregates when observed using confocal scanning laser microscopy. Molecular docking results showed that more salt bridges were formed between ι-CG and OVA than between ι-CO and OVA. A schematic model was established to elucidate the effects of ι-CG and ι-CO on OVA. This study provides instructions for designing innovative food systems based on different charge intensities and chain lengths of saccharides.

Mechanical (ball milling) activation to regulate microstructure of κ-carrageenan and improve freeze-thaw stability of κ-carrageenan gels and κ-carrageenan-based emulsion gels

Xinwei Xu,Yating Ding,Huifen Weng,Qiong Xiao,Jun Chen,Yonghui Zhang,Qiuming Yang,Fuquan Chen,Anfeng Xiao

To expand the wide range of applications of κ-carrageenan in the food and biomedical fields, we have physically modified the carrageenan and focused on its freeze-thaw stability. In this study, κ-carrageenan (KC) was modified by mechanically activated to prepare wet ball milling κ-carrageenan (WMKC), and its properties were determined. The research results indicate that mechanical activation helps improve the freeze-thaw stability of KC gels and enables KC to better maintain its gel network structure during the freeze-thaw process. These phenomena are related to the effects of mechanical activation on the particle morphology, molecular structure, molecular interactions, and moisture migration of κ-carrageenan. The particle morphology images demonstrated that the WMKC particles exhibited a more irregular surface, which facilitated greater penetration of water molecules into the interior of the κ-carrageenan particles. Furthermore, the results of molecular structure and molecular interaction analyses showed that mechanical activation resulted in the removal of sulfate groups from the κ-carrageenan molecular chain. This resulted in more free -OH being exposed on the molecular chain, which in turn provided more binding sites for KC molecules with water molecules. Low-field nuclear magnetic resonance (LF-NMR) results also proved that WMKC could better maintain the binding with water molecules during the freeze-thaw process. A better combination of WMKC molecules and water molecules can significantly diminish the irreversible damage to the gel network structure of WMKC during the freeze-thaw process, thereby effectively enhancing the freeze-thaw stability of WMKC. In addition, we further used WMKC as the gel matrix and investigated that the freeze-thaw stability of the WMKC-based emulsion gel was optimal when the mechanical activation duration was 3h, which well meets the demand of κ-carrageenan emulsion gels in the frozen food industry.

Evaluation of the effect of the chitosan/carrageenan ratio on lovastatin release from chitosan/carrageenan based biomaterials

Ha Manh Hung,Vu Quoc Manh,Vu Thi Thu Thao,Dao Thi Phuong Thuy,Pham Tien Dung,Nguyen Thi Bich Viet,Duong Khanh Linh,Nguyen Ngoc Linh,Doan Thi Yen Oanh,Nguyen Thuy Chinh,Thai Hoang,Vu Quoc Trung

The chitosan/carrageenan/lovastatin biomaterials were prepared using a solution technique with 5 % lovastatin and a variable chitosan/carrageenan mass ratio. Fourier transform infrared spectroscopy (FT-IR) and field emission scanning electron microscopy (FESEM) images revealed that the complex's components were well dispersed into each other without changing the chemical structure, forming dipole interactions and hydrogen bonds among them. The abilities to release Lovastatin (Lov) in pH 2 and pH 7.4 solutions, corresponding to the environments in stomach and intestinal fluids, respectively, are both affected by the ratio of polymer components. The best results were obtained with the chitosan/carrageenan/lovastatin (w/w/%w = 10/90/5%) (CsCL195) biomaterial sample in which the particles are evenly dispersed. While the drug release process was poor in the pH 2 solution (gastric juice), a good release observed in the pH 7.4 solution (intestinal fluid) allows facilitating drug absorption through the intestinal mucosa cells into the blood, from which the effects of hypercholesterolemia treatment can be obtained.

Enhancement mechanism of ι-carrageenan on the network structure and gel-related properties of soy protein isolate/λ-carrageenan system

Dongling Qiao,Yuyan Zhang,Farong Sun,Michelle Yoo,Guohua Zhao,Binjia Zhang

Enhancement mechanism of ι-carrageenan on the network structure and gel-related properties of soy protein isolate (SPI)/λ-carrageenan system was investigated. Adding ι-carrageenan decreased SPI particles' size in nano-scale, reduced hydrophobic interactions by suppressing exposure of hydrophobic groups, and increased the disulfide bonds in SPI. With rising ι-carrageenan content (< 0.55 %), the interaction between ι-carrageenan and SPI was enhanced mainly through electrostatic interaction and hydrogen bond. Meanwhile, SPI particles were reduced and their stacking compactness was increased as well. These contributed to the improvement of network structure and thus increased weakly bonded water from 97.7 to 98.5 %, water holding capacity from 55.29 % to 61.70 % and gel hardness from 0.53 to 1.30 N. Higher ι-carrageenan content (> 0.55 %) induced micro-phase separation as shown by CLSM images, and led to reduction of gel hardness. These results favor the rational design and application of SPI-based gel systems with desired practical properties.

A novel composite of collagen-hydroxyapatite/kappa-carrageenan

Wenpo Feng,Shuying Feng,Keyong Tang,Xichan He,Aihua Jing,Gaofeng Liang

This study was to obtain Collagen-Hydroxyapatite/kappa-Carrageenan (COL- HAP/KCAR) composite with similar to those of natural bone. After been extracted from rabbit skins, the collagen was used in combination with Ca(NO 3 ) 2 and (NH 4 ) 2 HPO 4 to obtain COL-HAP composite in situ. kappa-Carrageenan, a kind of plant-polysaccharides, was also employed to improve the properties of the composite. Influences of preparing conditions on the microstructure of the composite and the bonding formation of HAP particles with collagen were characterized by XRD , SEM, and FT-IR. The properties of COL-HAP/KCAR composite, such as mechanical properties , hygroscopicity , degradable properties, and cytotoxicity were studied. XRD and SEM results showed that the inorganic phases of the composite were mainly hydroxyapatite well distributing on the biopolymer . FT-IR indicated that the N H and COO- of collagen may be the nucleation sites in mineralization. kappa-Carrageenan may act as a cross-linking agent, which decreased the hygroscopicity and biodegradability of COL-HAP ( p  < 0.01). The studies on the mechanical properties, hygroscopicity, biodegradability, cytotoxicity and haemolytic phenotype indicated that the material is a potential bone repair material.

Transparent and thermally super-insulating κ-carrageenan aerogels

Yuxun Zhou,Wenxiao Liu,Di Liu,Zhifang Sun

•Zirconium-ion crosslinking effectively reinforced the κ-carrageenan gel network. • •A polarity-gradient solvent-exchange strategy significantly reduced gel shrinkage. • •SAXS revealed the structural evolution of κ-carrageenan fibers during aerogel formation. • •The obtained aerogel combined high transparency with ultralow thermal conductivity. This study presents a facile strategy for fabricating transparent κ-carrageenan aerogels via multivalent metal-ion crosslinking, polarity-gradient solvent exchange, and supercritical carbon dioxide drying. The structural evolution of the κ-carrageenan gel network toward a nanofibrous architecture was further elucidated by SAXS. The resulting aerogels exhibited a unique combination of high optical transparency (up to 79.4%), high specific surface area (442.9–504.9 m 2  g −1 ), ultralow thermal conductivity (14.84–17.22 mW m −1  K −1 ), and low bulk density (54.42–77.33 mg cm −3 ). In addition, the crosslinking-induced structural and chemical changes in κ-carrageenan were systematically investigated by FT-IR, XRD, and XPS, while the thermal stability and compressive mechanical performance of the aerogels were also comprehensively evaluated. Owing to their highly transparent nanostructure and excellent thermal insulation capability, the κ-carrageenan aerogels developed in this work demonstrate a highly competitive property profile among polysaccharide-based aerogels reported to date and hold considerable promise for transparent thermal insulation applications. This study reports a strategy for preparing a transparent nanofibrous κ-carrageenan aerogel through steps including tetravalent metal ion crosslinking, polarity-gradient solvent exchange, and supercritical carbon dioxide drying. The resulting aerogel exhibits high transparency, low haze, ultra-low thermal conductivity, and low density. Its overall performance shows significant competitiveness among currently reported polysaccharide-based aerogels, and it also holds outstanding application potential in the field of transparent thermal insulation. Revealing the role of λ-carrageenan on the enhancement of gel-related properties of acid-induced soy protein isolate/λ-carrageenan system

Dongling Qiao,Yuyan Zhang,Lisong Lin,Kexin Li,Fan Zhu,Genfa Wang,Gaolei Xi,Fatang Jiang,Binjia Zhang,Fengwei Xie

This work explored the pivotal role of λ-carrageenan in fortifying the gel structure, consequently enhancing gel-related attributes within an acid-induced soy protein isolate/λ-carrageenan system (SPI/Car). The model system was prepared by subjecting a thermally treated mixture of these two biopolymers to acidification. Small-angle X-ray scattering (SAXS) and chemical force analysis unveiled the profound influence of hydrogen bonding and electrostatic interactions in the formation of the intricate network architecture of SPI/Car gels. Remarkably, λ-carrageenan interacted with the hydrophilic groups on the surface of agglomerated SPI particles, effectively composing the fundamental framework of SPI/Car gels. Simultaneously, λ-carrageenan emerged as a linchpin through hydrogen bonding, establishing pivotal crosslinking connections. The introduction of λ-carrageenan resulted in a reduction in SPI particle size, the dispersion of previously formed aggregates, and a subtle blurring of the SPI particles. It also significantly heightened the prevalence of hydrogen bonding and electrostatic interactions, ultimately giving rise to a robust yet somewhat coarse gel structure, as evidenced by confocal laser scanning microscopy (CLSM). These profound structural alterations underpinned the enhancement of water-holding capacity (WHC) and gel hardness in SPI/Car gels. These findings hold promise for the rational design and application of SPI-based systems in the production of processed vegetarian and vegan foods.

Carrageenan-based nanoparticles for drug delivery: A systematic review

Elias Guilherme Basilio Silva,Gildacio Pereira Chaves Filho,Ana Paula Ramos

Carrageenans, sulfated polysaccharides isolated from red seaweed, have gained attention as a versatile material for nanoparticle-based drug delivery: they are biocompatible and biodegradable and can form responsive nanostructures. The growing number of studies on carrageenan-based nanocarriers makes a systematic review essential for synthesizing current knowledge, identifying gaps, and guiding future research and clinical translation efforts. This systematic review, conducted under the PRISMA guidelines, evaluates 19 studies that addressed the design, physicochemical properties, and therapeutic applications of carrageenan-based nanoparticles. Most of the studies employed κ-carrageenan, while some used chemically modified forms of carrageenans to enhance drug encapsulation and to control drug release. The carrageenan-based nanocarriers cited herein can entrap drugs, respond to changes in pH, and potentially deliver drugs at target sites, particularly during antimicrobial and anticancer therapies. In general, the carrageenan-based nanoparticles are spherical, with diameters ranging from tens to hundreds of nanometers. Despite promising translational results, carrageenan-based nanoformulations are heterogeneous, which has prevented studies from being directly compared. In addition, most studies have been carried out in vitro. The findings highlight that carrageenans are potential natural nanocarriers and emphasize that further in vivo validation and standardized approaches are needed to advance their clinical translation. An innovative konjac glucomannan/κ-carrageenan mixed tensile gel

Di Wu,Simin Yu,Hongshan Liang,Mohamed Eid,Bin Li,Jing Li,Jin Mao

BACKGROUND Konjac glucomannan (KGM) showed a synergistic interaction with κ-carrageenan (CAR), which led to the formation of a promising compound hydrocolloid gel in the food field (such as jelly). Nevertheless, the mixed gels formed by adding KGM to CAR still displayed defects in gel strength and syneresis, and would hardly meet the quality requirements of some gel foods. However, deacetylated KGM and maltodextrin (MD) have always been used in gel foods and affect their viscosity and rheological properties. RESULTS In our paper, different amounts of MD were first used to alter the textural properties, and the results showed that both tensile strength and elongation exhibited first an increasing and then a decreasing trend with the increasing MD proportion and achieved a maximum at a final maltodextrin proportion of 4 g kg −1 in the KGM/CAR/MD system. Based on the above results, we further explored the effects of deacetylation degree of KGM on the gel properties of mixed gel system. The results revealed that, compared to the native KGM, the partial deacetylated KGM was capable of significantly improving the tensile strength and elongation of KGM/CAR mixed gel. CONCLUSION Our study found that the appropriate addition of MD (0.4%) and DKGM were able to alter the tensile properties of KGM/CAR mixed gel, with potential to meet the needs of consumers and further design innovative tensile gel products in the soft gel industry. © 2021 Society of Chemical Industry

Self-healing carrageenan-driven Polyacrylamide hydrogels for strain sensing

Fan ZiWen,Duan LiJie,Gao GuangHui

Conductive hydrogels have attached considerable attention due to their good stretchability, excellent conductivity when they are applied in soft electronics. However, to fabricate a flexible hydrogel sensor with excellent toughness and good self-healing properties remains a challenge. In this work, we assembled a dual physical-crosslinking (DPC) ionic conductive polyacrylamide/carrageenan double-network (DN) hydrogel. This hydrogel has excellent fracture tensile stress and toughness, and demonstrates rapid self-recovery and self-healing ability due to the unique dual physical-crosslinking structures. Besides, the hydrogel is highly conductive by adding some conductive ions. As a result, the hydrogel-based sensor can stably detect human motions and physiological signals. The work provides novel ideas for the development of flexible sensing devices.
