ChlOR, a GMC family oxidoreductase that evolved independently from the actinomycete, confers resistance to amphenicol antibiotics

Yingying Qian,Minggen Cheng,Lin Lai,Jing Zhou,Gerben J. Zylstra,Xing Huang

Overuse of the amphenicol antibiotics chloramphenicol (CHL) and thiamphenicol (TAP) poses a great threat to ecosystem safety and human health. The strain, Nocardioides sp. LMS-CY, Nocardioides sp. QY071 and Nocardioides sp. L-11A, classified as a gram-positive actinomycete, harbours a complete CHL metabolic pathway. However, the metabolic genes (clusters) involved in the entire pathway in gram-positive actinomycetes are still limited. Here, chlOR LMS , chlOR QY071 and chlOR L-11A completely from the actinomycete Nocardioides spp. were found to act on the C 1 -OH of the CHL/TAP side chain, directly converting CHL/TAP to 4-nitrobenzaldehyde (PNBD)/4-methylsulfonyl benzaldehyde (PMBD) and transforming PNBD/PMBD into 4-nitrobenzyl alcohol (PNBM)/4-methylsulfonyl phenyl methanol (PMBM). Furthermore, oxidoreductases can transform PNBM into 4-nitrobenzoate (PNBA). The oxidoreductases ChlOR LMS , ChlOR QY071 and ChlOR L-11A were all classified as cellobiose dehydrogenases from the glucose methanol choline (GMC) family. Based on the Swiss-Prot database, ChlOR QY071 exhibited a lower identity (27.12%–35.10% similarity) with the reported oxidoreductases. Enzymatic and molecular docking analyses showed that ChlOR QY071 and ChlOR L-11A from the two similar genomes were remarkably more effective in metabolizing CHL than ChlOR LMS . Overall, the detailed resistance mechanism of CHL/TAP by actinomycete strains isolated from soil and livestock manure will provide insights into the occurrence of CHL/TAP resistance genes in the environment, resistance risk and bioremediation of CHL/TAP-contaminated environments.

Degradation kinetics, byproducts formation and estimated toxicity of metronidazole (MNZ) during chlor(am)ination

Shisheng Zhang,Tao Lin,Wei Chen,Hang Xu,Hui Tao

The residues of pharmaceuticals and personal care products (PPCPs) in environmental waters have been widespread concerned. Metronidazole (MNZ), normally employed to treat inflammation and infection, was chosen as one model PPCP. The degradation of MNZ by chlorination could be fitted by pseudo-first-order kinetics as the observed pseudo-first-order rate constants increasing from 0.0302 min −1 to 0.2872 min −1 . However, the kinetics during chloramination of MNZ followed pseudo-second-order reaction, whose estimated half-live was approximately 6–8 times longer than chlorination. The chlor(am)ination of MNZ especially formed chloroform (CF), dicholoacetamide (DCAcAm), tricholoacetamide (TCAcAm) and dichloroacetonitrile (DCAN), and their yields were overall lower under chloramination than chlorination. During chlorination, the yield of CF was increased from 0.35 ± 0.02% to 2.06 ± 0.12% with 1–20 chlorine/MNZ molar ratio, whereas the formations of DCAcAm, TCAcAm and DCAN increased firstly and then decreased. Increasing chloramine dosage promoted the concentrations of scheduled disinfection byproducts (DBPs). CF and TCAcAm kept continuous generation in chlor(am)ination versus reaction time. Compared with the chlorination, the chloramination of MNZ was more dependent on pH value due to the self-degradation of chloramine. Faintly acidic condition favored N-DBPs' formation in MNZ when it was subjected to chlor(am)ination. The chloramination of MNZ produced cytotoxicity and genotoxicity by 10–15 folds lower than chlorination, and DCAN formed during chloramination dominated both DBPs' yields and toxicity contribution. Opposite to chlorination, the integrated toxicity of MNZ during chloramination varied linearly versus N-DBPs' yields.

Detection, identification and control of polar iodinated disinfection byproducts in chlor(am)inated secondary wastewater effluents

Yan Huang,Yangyang Zhang,Qing Zhou,Aimin Li,Peng Shi,Jingfan Qiu,Yang Pan

Disinfection of wastewater effluents could generate numerous toxic disinfection byproducts (DBPs) during wastewater reclamation owing to the complexity of their dissolved organic matter. Recently, iodinated disinfection byproducts (I-DBPs) have attracted increasing attention as a result of their higher toxicity than their brominated and chlorinated analogues. In this study, various new polar I-DBPs in chlor(am)inated wastewater effluents were detected with ultra-performance liquid chromatography/electrospray ionization-triple quadrupole mass spectrometry (UPLC/ESI-tqMS) by setting a precursor ion scan of m/z 126.9. Three new polar I-DBPs were identified as wastewater I-DBPs, including iodoacetic acid, chloroiodoacetic acid and 3,5-diiodo-4-hydroxybenzaldehyde. The remaining new polar I-DBPs were proposed with structures, nine of which were found to be aromatic halogenated DBPs, and six of which were proposed to be in a group because they share closely related m/z values, retention times, and fragmentation pathways. In addition, formation of these new polar I-DBPs with disinfectants and contact times was also studied. Compared with chlorination, chloramination generated more species and higher concentrations of new polar I-DBPs. Furthermore, control of the newly detected polar I-DBPs during chlorination by coagulation, resin adsorption, and combined processes was compared. Resin adsorption showed a higher precursor removal efficiency than coagulation, and thus achieved a better control of these new polar I-DBPs. Coagulation followed by resin adsorption was the most effective compared with single coagulation and single resin adsorption.

Formation and estimated toxicity of trihalomethanes, haloacetonitriles, and haloacetamides from the chlor(am)ination of acetaminophen

Shunke Ding,Wenhai Chu,Tom Bond,Qi Wang,Naiyun Gao,Bin Xu,Erdeng Du

The occurrence of pharmaceuticals and personal care products (PPCPs) in natural waters, which act as drinking water sources, raises concerns. Moreover, those compounds incompletely removed by treatment have the chance to form toxic disinfection byproducts (DBPs) during subsequent disinfection. In this study, acetaminophen (Apap), commonly used to treat pain and fever, was selected as a model PPCP. The formation of carbonaceous and nitrogenous DBPs, namely trihalomethanes, haloacetonitriles, and haloacetamides, during chlor(am)ination of Apap was investigated. Yields of chloroform (CF), dichloroacetonitrile (DCAN), dicholoacetamide (DCAcAm), and tricholoacetamide (TCAcAm), during chlorination were all higher than from chloramination. The yields of CF continuously increased over 48   h during both chlorination and chloramination. During chlorination, as the chlorine/Apap molar ratios increased from 1 to 20, CF yields increased from 0.33   ±   0.02% to 2.52   ±   0.15%, while the yields of DCAN, DCAcAm and TCAcAm all increased then decreased. In contrast, during chloramination, increased chloramine doses enhanced the formation of all DBPs. Acidic conditions favored nitrogenous DBP formation, regardless of chlorination or chloramination, whereas alkaline conditions enhanced CF formation. Two proposed formation mechanisms are presented. The analysed DBPs formed during chlorination were 2 orders of magnitude more genotoxic and cytotoxicity than those from chloramination.

Pervaporation separation of dimethyl carbonate/methanol mixtures with regenerated perfluoro-ion-exchange membranes in chlor-alkali industry

Wan-Zhong Lang,Hai-Yan Niu,Ya-Xin Liu,Chun-Xia Liu,Ya-Jun Guo

The waste perfluoro-ion-exchange membranes (PFIEMs) in chlor-alkali industry were regenerated and used to the separation of dimethyl carbonate (DMC)/methanol (MeOH) mixtures by pervaporation process. The energy-dispersive spectrum (EDS) demonstrates that the impurities on the surfaces of waste PFIEMs can be effectively cleared by the regeneration process. The degree of swelling, sorption, and pervaporation properties of the regenerated PFIEMs with different counter ions were investigated. The results indicate that the counter ions of PFIEMs conspicuously influence the degree of swelling, sorption, and pervaporation properties for DMC/MeOH mixtures. The degree of swelling and solubility selectivity both decreases with the alkali metal counter ions in the sequence: Li + > Na + > K + > Cs + . The degree of swelling increases with MeOH concentration increasing in feed liquid. The pervaporation measurements illustrate that the permeation flux decreases and the separation factor increases with the rising in ion radius of counter ions. The increase of feed concentration (MeOH) and feed temperature is advantageous to improve permeation flux while at the cost of separation factor decreasing. © 2013 Wiley Periodicals, Inc. J. Appl. Sequential recovery of lead and zinc from wastewater coupled with chlor-alkali electrolysis in a membrane reactor

Yu Gao,Yugang Zhou,Shuaicai Wang,Tao Zhang,Ruina Zhang,Yanshuo Li,Wei Fang

The treatment of heavy metal-contaminated wastewater remains a critical challenge due to the coexistence of multiple metal ions and the lack of efficient recovery strategies. Herein, we report an H-type electrochemical membrane reactor that enables the sequential recovery of Pb 2+ and Zn 2+ from multicomponent wastewater while simultaneously driving chlor-alkali electrolysis. The system operates in a two-electrode configuration with a polysulfone-ZrO 2 composite membrane, which prevents cross-contamination between the compartments and stabilizes the local pH environment. In a solution containing 0.01 M Pb 2+ and 0.01 M Zn 2+ , Pb 2+ is preferentially reduced and deposited as fluffy lead with 99.3% recovery efficiency after 10 h of galvanostatic electrolysis at 10 mA cm −2 . When most of Pb 2+ is depleted, the hydrogen evolution reaction becomes dominant, raising the pH and inducing the precipitation of Zn 2+ as Zn(OH) 2 with 99.8% recovery efficiency, which is subsequently converted to ZnO. The membrane improves Pb 2+ /Zn 2+ recovery efficiencies and reduces energy consumption compared with the membrane-free system. The anode produces Cl 2 with Faradaic efficiency >95%, and the cathode generates H 2 (with Faradaic efficiency: from 1.6% at 1 h to 99.2% at 10 h) and NaOH. This integrated strategy offers a sustainable pathway for the stepwise recovery of metal resources from complex wastewater while co-generating valuable chemicals.

A ZrO2-PSU composite membrane reactor for simultaneous Cl2 and H2/NaOH production via chlor-alkali electrolysis

Tao Zhang,Yugang Zhou,Wei Fang

A ZrO 2 -PSU composite membrane with asymmetric structure was developed for chlor-alkali electrolysis. • ZrO 2 addition enhanced hydrophilicity and reduced area resistance but lowered bubble point pressure. • The membrane reactor achieved high Faradaic efficiencies (>96% for HER and > 98% for CER) with considerable NaOH yield. • The zero-gap electrolyzer delivered 3.33 V at 100 mA cm −2 over 20 h with robust structural and chemical stability. The chlor-alkali process is a vital industrial route for the production of chlorine (Cl 2 ), hydrogen (H 2 ), and sodium hydroxide (NaOH). However, conventional ion-exchange membrane-based technologies face challenges related to high cost, limited durability, and susceptibility to fouling and degradation under harsh operating conditions. Here, we report a zirconium dioxide-polysulfone (ZrO 2 -PSU) composite membrane reactor that enables simultaneous and efficient Cl 2 and H 2 /NaOH production via chlor-alkali electrolysis. The composite membrane, fabricated via phase inversion tape casting, integrates ZrO 2 nanoparticles into a porous PSU matrix, achieving a well-defined asymmetric structure with a dense top layer and a finger-like porous bottom layer. The optimized composite membrane ( Z -PSU-70) exhibits lowest area resistance, enhanced hydrophilicity, and effective gas barrier properties. In an H-type electrolysis cell using a nickel mesh cathode and a RuO 2 @carbon paper anode, the reactor delivers stable cell voltages and high Faradaic efficiencies (>96% for H 2 and > 98% for Cl 2 ) with minimal gas crossover. Furthermore, a zero-gap flow cell configuration achieves a low cell voltage of approximately 3.33  V at 100 mA cm −2 , with stable performance over 20 h. Post-electrolysis characterizations confirm the structural and chemical stability of both the membrane and electrodes. This work presents a cost-effective and high-performance membrane reactor design, offering a promising pathway toward more sustainable and efficient chlor-alkali electrolysis.

Comparative analysis of para-nitrophenol degradation and DBPs formation in the UV/chlor(am)ine/Fe(Ⅲ) systems

Qing Wang,Lin Deng,Changbo Zhang,Rajendra Prasad Singh,Gongde Wu

Para-nitrophenol (PNP), a highly toxic compound widely detected in water bodies, was investigated as the target contaminant during the UV/chlor(am)ine/Fe(III) processes to evaluate its degradation kinetics model, and disinfection byproducts (DBPs) formation and pathways. The incorporation of Fe(III) introduced a oxidation-reduction cycle of Fe(II) and Fe(III), which can generate additional hydroxyl radicals, reactive chlorine species, and reactive nitrogen species, thereby accelerating PNP degradation by 21.85 % in the UV/chlorine system and 31.08 % in the UV/chloramine system. PNP degradation followed pseudo-first-order kinetics, with observed rate constants ( k obs ) calculated as 0.0938 and 0.0604 min −1 , in the UV/chlorine/Fe(III) and UV/chloramine/Fe(III) systems, respectively. Radical quenching experiments confirmed the dominant reactive species in each system, supporting the mechanistic interpretation. In the UV/chlorine/Fe(III) system, DBP yields ranked as follows: Halonitromethanes (HNMs) > trihalomethanes (THMs) > haloacetamides (HAcAms) > haloacetonitriles (HANs), while the order was HNMs > THMs > HANs > HAcAms, in the UV/chloramine/Fe(III) system. Factors influencing DBP formation, including Fe(III) dosage, initial pH, and chlor(am)ine dosage, were systematically investigated. The cytotoxicity index (CTI) contributions of each DBP in the UV/chlor(am)ine/Fe(III) systems were ranked as follows: HNMs > HANs > THMs > HAcAms. Based on detected transformation products (TPs) and radical contribution analysis, plausible pathways for PNP transformation and DBPs formation were proposed. These findings provide valuable insights into PNP degradation behavior and the mechanisms of DBPs formation in the UV/chlor(am)ine/Fe(III) systems, supporting their future practical applications.

Designing High-Performance Nanofiltration Membranes for High-Salinity Separation of Sulfate and Chloride in the Chlor-Alkali Process

Qian Wang,Yue Wang,Bo-Zhi Chen,Tian-Dan Lu,Han-Lin Wu,Yi-Qun Fan,Weihong Xing,Shi-Peng Sun

Enormous demand exists for separation of high-salinity solutions across various industries, especially in those high-pollution businesses which are urgently looking for high-throughput and precise-selective nanofiltration (NF) membranes stably operating at elevated concentrations and other harsh environments. This work develops a novel NF membrane (i.e., high-performance thin-film composite (H-TFC)) through straightforward structure design instead of complex post-treatment to achieve high performance during high-salinity separation of sulfate and chloride ions. Through manipulating dope composition and coagulation bath temperature, unique macro- and microstructures of the H-TFC membrane were designed: (1) The substrate possesses the best ratio of the spongy and fingerlike layers, which not only provides low resistance transport channels to improve the membrane permeability (13.0 L m2– h–1 bar–1) but also ensures enough strength and minimum defects to treat high-salinity solutions. (2) The unique nominal pore size (0.364 nm) of the finely tuned selective layer can realize the separation of Cl– and SO42– at relatively high level (separation factor about 9000). The excellent long-term stability and resistance toward chlorine and heat grant the H-TFC membranes great potential for application in high-salinity solution separation of chlor-alkali and other industries.

Impact of pre-oxidation using H2O2 and ultraviolet/H2O2 on disinfection byproducts generated from chlor(am)ination of chloramphenicol

Zhang Yimeng,Chu Wenhai,Xu Ting,Yin Daqiang,Xu Bin,Li Pan,An Na

Hydrogen peroxide with and without activation by ultraviolet irradiation (UV/H 2 O 2 and H 2 O 2 , respectively) has been investigated to degrade emerging contaminants in drinking water. The antibiotic chloramphenicol (CAP) and its analogues (thiamphenicol and florfenicol), referred to collectively as CAPs, can produce a range of disinfection by-products (DBPs) during chlorination or chloramination (chlor(am)ination). The impact of unactivated and activated H 2 O 2 pre-oxidation on the formation of nitrogenous and carbonaceous DBPs from the chlor(am)ination of CAP model precursors was investigated. A theoretical estimated toxicity evaluation was also carried out based on the DBPs that formed. H 2 O 2 pretreatment alone did not affect organic carbon and nitrogen concentrations, but significantly reduced the formation of trichloromethane (TCM) during chlorination and trichloronitromethane (TCNM) during chloramination, and enhanced dichloroacetonitrile (DCAN) yields during chloramination. These significant changes in DBP yields caused by H 2 O 2 pre-oxidation are attributable to the transformation of the three CAPs into more reactive precursor structures. The three CAPs did not form DCAN and TCNM during chlorination, regardless of whether H 2 O 2 pre-oxidation was used. However, UV/H 2 O 2 pretreatment resulted in the unexpected formation of TCNM during chlorination. Furthermore, UV/H 2 O 2 pre-oxidation increased the formation of DCAN and TCNM, whereas it had little effect on TCM formation during chloramination. A preliminary estimated toxicity evaluation demonstrated that UV/H 2 O 2 pre-oxidation at low doses, followed by chlorination, resulted in the lowest estimated toxicity from a range of DBPs. Conversely, the estimated toxicity evaluation showed that unactivated H 2 O 2 (high dose) was preferable to UV activated H 2 O 2 for overall control of these investigated DBPs during chloramination.

Ultra-bubble-repellent sodium perfluorosulfonic acid membrane with a mussel-inspired intermediate layer for high-efficiency chlor-alkali electrolysis

Jianlong Lei,Xundao Liu,Xiaohong Chen,Hanfeng Luo,Wei Feng,Jiangshan Zhang,Feng Liu,Supeng Pei,Yongming Zhang

Compromised performance caused by gas product adhesion remains a major challenge for membrane applications in chlor-alkali electrolysis. Here, an ultra-bubble-repellent sodium perfluorosulfonic acid (PFSA-Na) membrane with hierarchical structures of aggregated silica was successfully synthesized by tethering hydrophilic silica onto a mussel-inspired intermediate layer. The surface chemistry, morphology and loading could be controlled via surface tailoring of the silica by γ-(2,3-epoxypropoxy)propyltrimethoxysilane (KH560). The developed membrane exhibited high hydrophilicity (7.5 ± 1.3°), an ultralow bubble contact area (164.7 ± 2.1°) and negligible bubble adhesion forces (<6 μN) while maintaining efficient NaCl transport and low electric double layer resistance. The superaerophobic-hydrophilic surface enables an excellent cleaning property. When the modified membrane was applied in chlor-alkali electrolysis, the Cl 2 /H 2 gas bubbles were repelled from the membrane surface in time and minimized the negative effects of bubble adhesion, resulting in high-efficiency chlor-alkali electrolysis. Further, the developed membrane exhibited high stability during the continuous 10-h electrolysis process. This result indicates that developing a superaerophobic surface with highly efficient mass transport can lead to high-performance chlor-alkali electrolysis.

Investigation of clofibric acid removal by UV/persulfate and UV/chlorine processes: Kinetics and formation of disinfection byproducts during subsequent chlor(am)ination

Xian Lu,Yisheng Shao,Naiyun Gao,Juxiang Chen,Huiping Deng,Wenhai Chu,Na An,Fangyuan Peng

This study investigated the UV/persulfate (UV/PS) and UV/chlorine processes as alternative method for the removal of clofibric acid (CA). The formation of disinfection byproducts (DBPs) during subsequent chlor(am)ination was also evaluated. The degradation of CA followed the pseudo-first order kinetics. The second-order rate constants of CA with SO4 − , OH and Cl were respectively determined as k SO 4 - ,CA kSO4-,CA   =   (1.73   ±   0.01)   ×   10 9   M −1   s −1 , k OH ,CA kOH,CA   =   (2.72   ±   0.08)   ×   10 9   M −1   s −1 and k Cl ,CA kCl,CA   =   (9.76   ±   0.15)   ×   10 10   M −1   s −1 . The degradation rate constant increased with increasing oxidant dosage in UV/PS and UV/chlorine processes. The degradation rate constant was found to be the highest at pH 9 and decreased dramatically at pH 11 in UV/PS process. For UV/chlorine, the rate constant continuously decreased with increasing pH from 3 to 11. Presence of HCO 3 − and Cl − had different effects (promotion and/or inhibition) on CA degradation in both processes. An inhibition effect was observed in the presence of NOM for the two UV-based processes. The higher CA removal in real water suggested the two processes were suitable for treating water containing CA, and the UV/chlorine was more cost-effective than UV/PS based on the total cost of electrical energy. Compared with the chlor(am)ination of CA, the UV/PS and UV/chlorine pre-oxidation significantly impacted the DBP formation during subsequent chlor(am)ination, which indicated the application of the two UV-based processes needs to be carefully balanced against the downstream effect on DBP formation.
