Baby primrose (Primula forbesii) is a newly cultivated and valuable ornamental plant with great market potential for both indoor and landscape use. As a container plant, baby primrose has long, weak flower stalks that can easily lodge, resulting in poor-quality plants, especially during transportation. To control plant height and subsequently prevent flower peduncle lodging, we investigated the effects of two plant growth regulators (PGRs), chlormequat chloride (CCC) at 0, 250, 500, or 750 ppm and uniconazole (UNI) at 25, 50, or 75 ppm on growth, development, and flowering of two cultivars of baby primrose, Fragrant Luolan and Red Star. Plant growth regulators at the proposed concentrations were applied twice throughout the experiment. Both PGRs significantly suppressed plant height in both cultivars, with a 16% to 27% reduction by CCC and 50% to 59% by UNI compared with untreated plants. Among CCC-treated groups, plants were shortest when CCC was applied at 500 ppm; plant height was suppressed more when treated with UNI. In both cultivars, UNI significantly suppressed the first, second, and third peduncle lengths. Furthermore, CCC affected peduncle length, but to a lesser extent than UNI. Plant growth regulator applications generally had little effect on flower characteristics of baby primrose. Neither PGRs influenced the inflorescence number and flower size; however, PGRs did increase the number of floral whorls and suppressed pedicel length of ‘Red Star’. New leaf growth was suppressed by both PGRs. In addition, peduncle cell length and width were both significantly suppressed by PGR applications. We concluded that two foliar applications of UNI at 25 ppm comprised the most effective method of controlling baby primrose plant height while maintaining desirable flower traits at a relatively low production cost. Results of this study provide guidance for techniques that can be used to effectively control the plant height of potted baby primrose for commercial greenhouse production.

A simple and sensitive method for detection of chlormequat chloride residue in wheat was developed using surface-enhanced Raman spectroscopy (SERS) coupled with chemometric methods on a portable Raman spectrometer. Pretreatment of wheat samples was performed using a two-step extraction procedure. Effective and uniform active substrate (gold nanorods) was prepared and mixed with the sample extraction solution for SERS measurement. The limit of detection for chlormequat chloride in wheat extracting solutions and wheat samples was 0.25 mg/L and 0.25 μg/g, which was far below the maximum residual value in wheat of China. Then, support vector regression (SVR) and kernel principal component analysis (KPCA), multiple linear regression, and partial least squares regression were employed to develop the regression models for quantitative analysis of chlormequat chloride residue with spectra around the characteristic peaks at 666, 713, and 853 cm−1. As for the residue in wheat, the predicted recovery of established optimal model was in the range of 94.7% to 104.6%, and the standard deviation was about 0.007 mg/L to 0.066 mg/L. The results demonstrated that SERS, SVR, and KPCA can provide the accurate and quantitative determination for chlormequat chloride residue in wheat.

Chlormequat chloride (CCC), a widely used plant growth regulator, is a choline analogue that has been shown to have endocrine-disrupting effects. Previous studies have shown that maternal exposure to CCC could induce hyperlipidemia and growth disruption in rat offspring. This study aims to further investigate the effects of peripubertal exposure to CCC on pubertal development and lipid homeostasis, as well as the underlying mechanisms. In vivo , male weanling rats were exposed to CCC (0, 20, 75 and 200 mg/kg bw/day) from post-natal day 21 to 60 via daily oral gavage. The results in rats showed that 75 mg/kg CCC treatment induced hepatic steatosis, predominantly microvesicular steatosis with a small amount of macrovesicular steatosis, in rat livers and 200 mg/kg CCC treatment induced liver damage including inflammatory infiltration, hepatic sinusoidal dilation and necrosis. In vitro , HepG2 cells were treated with CCC (0, 30, 60, 120, 240 and 480 μg/mL) for 24 h. And the results showed that CCC above 120 μg/mL induced an increase in triglyceride and neutral lipid levels of HepG2 cells. Mechanism exploration revealed that CCC treatment promoted the activation of mTOR/SREBP1 signalling pathway and inhibited activation of AMPK in both in vivo rat livers and in vitro HepG2 cells. Treatment with AMPK activator Acadesine (AICAR) could alleviate the lipid accumulation in HepG2 cells induced by CCC. Collectively, the present results indicate that CCC might induce hepatic steatosis by promoting mTOR/SREBP1 mediated lipogenesis via AMPK inhibition.

A sensitive method for detection of chlormequat chloride on cherry tomato peel was developed using surface-enhanced Raman spectroscopy (SERS) coupled with chemometric methods on a portable Raman spectrometer. Firstly, the spectra of ethanol solutions containing 10, 5, 2.5, 1, 0.5 or
0.25 mg/L of chlormequat chloride were collected, and characteristic signals can be detected when concentration was higher than or equal to 0.25 mg/L. Then, quantitative determination was conducted by the regression models developed using partial least-squares regression (PLSR), PLSR with
competitive adaptive reweighted sampling, PLSR with random frog, and random forest with the spectra of 600–1800 cm–1 and characteristic range, and root mean square error of cross-validation of the most remarkable model was 0.0503 mg/L. As for the spiked cherry tomato
samples with 0.5 mg/L to 10 mg/L chlormequat chloride, the predicted recovery rate obtained with the optimal model was in the range of 93.2% to 99.3%, and standard deviation ranged from 0.032 mg/L to 0.878 mg/L.
