A novel aminoclay–curcumin hybrid for enhanced chemotherapy

Suhang Wang,Han Cao,Yiming Zhong,Yuhong Yang,Zhengzhong Shao

Curcumin (Cur) has been demonstrated as an efficacious anti-tumor agent. However, its therapeutic applications are largely limited by its extremely low aqueous solubility, low stability and poor bioavailability. In this work, a simple one-pot synthesis was developed to fabricate a novel aminoclay–curcumin (AC–Cur) hybrid via the in situ loading of Cur over AC. The resultant hybrid was amenable to exfoliation and readily dispersible in aqueous media. Compared with free Cur, the AC–Cur hybrid displayed significantly enhanced solubility of Cur in both acidic and neutral environments, as well as enhanced stability of Cur in both neutral and alkaline environments. More importantly, this hybrid achieved a much better therapeutic effect than free Cur against various tumor cell lines, which was mainly attributed to its significantly improved solubility, stability and cellular uptake. Our results suggest that such an AC–Cur hybrid with high bioavailability can potentially be applied in nanomedicine for the treatment of malignant tumours.

Curcumin/pEGCG-encapsulated nanoparticles enhance spinal cord injury recovery by regulating CD74 to alleviate oxidative stress and inflammation

Chen Tianjun,Wan Li,Xiao Yongchun,Wang Ke,Wu Ping,Li Can,Huang Caiqiang,Liu Xiangge,Xue Wei,Sun Guodong,Ji Xin,Lin Hongsheng,Ji Zhisheng

Spinal cord injury (SCI) often accompanies impairment of motor function, yet there is currently no highly effective treatment method specifically for this condition. Oxidative stress and inflammation are pivotal factors contributing to severe neurological deficits after SCI. In this study, a type of curcumin (Cur) nanoparticle (HA-CurNPs) was developed to address this challenge by alleviating oxidative stress and inflammation. Through non-covalent interactions, curcumin (Cur) and poly (-)-epigallocatechin-3-gallate (pEGCG) are co-encapsulated within hyaluronic acid (HA), resulting in nanoparticles termed HA-CurNPs. These nanoparticles gradually release curcumin and pEGCG at the SCI site. The released pEGCG and curcumin not only scavenge reactive oxygen species (ROS) and prevents apoptosis, thereby improving the neuronal microenvironment, but also regulate CD74 to promote microglial polarization toward an M2 phenotype, and inhibits M1 polarization, thereby suppressing the inflammatory response and fostering neuronal regeneration. Moreover, in vivo experiments on SCI mice demonstrate that HA-CurNPs effectively protect neuronal cells and myelin, reduce glial scar formation, thereby facilitating the repair of damaged spinal cord tissues, restoring electrical signaling at the injury site, and improving motor functions. Overall, this study demonstrates that HA-CurNPs significantly reduce oxidative stress and inflammation following SCI, markedly improving motor function in SCI mice. This provides a promising therapeutic approach for the treatment of SCI.

Electrochemiluminescence platform for curcumin detection based on g-C3N4 of N-rich vacancy and Ti2AlC

Yujia Song,Li Tian,Ruidan Li,Yanjia Guo,Pengfei Han,Guangping Ma,Hanyue Jiang,Wenzhuo Wang,Juan Lu

In this work, g-C 3 N 4 of N-rich vacancy (CNNV) and mesoporous SiO 2 were synthesized. A novel electrochemiluminescence (ECL) sensor for curcumin (Cur) based on CNNV@SiO 2 and Ti 2 AlC was constructed. An initial ECL signal was generated from the CNNV-S 2 O 8 2 − system. SiO 2 significantly adsorbed more S 2 O 8 2 − , thereby enhancing the ECL signal. Ti 2 AlC served as an effective co-reaction promoter, further amplifying the ECL signal of CNNV@SiO 2 -S 2 O 8 2 − system. With the addition of Cur, the ECL signal also decreased, which can be attributed to the resonance energy transfer (RET) mechanism between CNNV (donor) and Cur (acceptor). Under optimal conditions, a strong linear relationship was observed between the change in ECL signal (ΔI) and the logarithm of Cur concentration (lgC Cur ), spanning from 5 × 10 −15 to 5 × 10 −9  mol L −1 with a detection limit of 1.7 × 10 −15  mol L −1 . The constructed ECL sensor was successfully applied to detect Cur in real samples.

Responsive nanoparticles synergize with Curcumin to break the “reactive oxygen Species-Neuroinflammation” vicious cycle, enhancing traumatic brain injury outcomes

Fu Xianhua,Zhang Yongkang,Chen Guojie,Mao Guangyao,Tang Jiajia,Xu Jin,Han Yuhan,Chen Honglin,Ding Lianshu

Traumatic brain injury (TBI) disrupts oxygen homeostasis in the brain, leading to excessive reactive oxygen species (ROS) production and dysregulated antioxidant mechanisms, which fail to clear excess ROS. This ROS overload promotes the expression of pro-inflammatory genes, releasing cytokines and chemokines and creating a vicious “ROS-neuroinflammation” cycle, making it essential to break this cycle for effective TBI treatment. In this study, we developed cysteine-alanine-glutamine-lysine (CAQK) peptide-modified antioxidant nanoparticles (C-PPS/C) for co-delivery of curcumin (Cur) to modulate oxidative and neuroinflammatory disturbances after TBI. In TBI mice, C-PPS/C nanoparticles accumulated in injured brain regions, where poly (propylene sulfide) 120 scavenged ROS, reducing oxidative stress, while Cur release further suppressed ROS and inflammation. C-PPS/C nanoparticles broke the “ROS-neuroinflammation” cycle, protecting the blood-brain barrier (BBB), reducing acute brain edema, and promoting long-term neurological recovery. Further investigation showed that C-PPS/C nanoparticles inhibited the NF-κB pathway, reducing pro-inflammatory gene expression and mitigating inflammation, suggesting a promising approach for TBI treatment.

Natural synergy: Oleanolic acid-curcumin co-assembled nanoparticles combat osteoarthritis

Chen Liu,Wanchun Du,Liang Zhang,Jiacheng Wang

Curcumin (Cur) is a natural polyphenol that is one of the most valuable natural products. However, its use as a functional food is limited by low water solubility, chemical instability and poor bioavailability. In this study, a supramolecular co-assembly strategy was used to construct an oleanolic acid-curcumin (OLA-Cur) co-assembly composite nano-slow-release treatment system. As a co-assembled compound, OLA is a widely present pentacyclic triterpenoid compound with multiple biological activities in the plant kingdom, which is expected to jointly alleviate the damaging effects of papain-induced mouse osteoarthritis model. The OLA-Cur NPs shows the solid core-shell structure, which can effectively improve the water solubility of Cur and OLA, and has good stability and sustained release characteristics. The analysis results show that the two compounds are mainly assembled through hydrogen bonding interactions, hydrophobic interactions, and π - π stacking interactions. The OLA-Cur NPs can inhibit the release of pro-inflammatory cytokines TNF-α, IL-6, and IL-1β induced by LPS in RAW264.7 mouse macrophages, promote the secretion of anti-inflammatory cytokine IL-10, and improve the oxidative stress index of hydrogen peroxide induced human rheumatoid arthritis synovial fibroblasts. In addition, it has a certain improvement effect on cartilage and subchondral bone damage in mouse osteoarthritis models. These findings suggest that constructing co-assembled composite nanoparticles based on pure natural compounds may break through the limitations of a variety of important nutritional ingredients in functional foods.

Study of Zn2+ complexation driven zein-based W/O/W Pickering emulsions for enhancing curcumin bioaccessibility and fluorouracil bioactivity

Long Deng,Junqiu Liao,Weiqi Liu,Shiyuan Liu,Xiaoxiao Liang,Rujin Zhou,Yanbin Jiang

Overcoming gastrointestinal tract barriers to enhance bioavailability is a major challenge for oral delivery. The innovative design of multi-compartment functional Pickering emulsions would provide an ideal option for targeted co-delivery of bioactive components. In this study, pH-responsive water-in-oil-in-water (W/O/W) Pickering double emulsions co-loaded with curcumin and fluorouracil were fabricated by zein/anthocyanin (ACN)/Zn 2+ nanoparticles (ZAZn NPs) via a one-step emulsification process. The unique emulsifier was designed by depositing ACN-Zn 2+ complexes on zein nanoparticles (zein NPs). The binding of ACN to the nanoparticle surface through hydrogen bonding interaction enhanced the hydrophilization and stimuli-response of zein NPs. Furthermore, the co-ordination of Zn 2+ and ACN facilitated the formation of nanoparticles with near-neutral wettability, which served as the dominant driving force for the one-step generation of W/O/W Pickering emulsions. The droplet size, distribution and internal structure of Pickering double emulsions were influenced by nanoparticle concentration and oil fraction. Due to Zn 2+ complexation with curcumin at the oil/water interface, the W/O/W Pickering emulsions obtained exhibited high curcumin encapsulation efficiency (91.2%) and free fatty acid release degree (65.9%), resulting in increased bio-accessibility (71.0%). The simulated gastrointestinal release demonstrated that curcumin and fluorouracil achieved controlled intestinal release, which was driven by a pH-mediated competitive adsorption-desorption mechanism. Moreover, in vitro cell experiments showed that co-encapsulated Pickering double emulsions exhibited 2.5 times greater bio-activity than those containing fluorouracil only. This facile strategy for preparing Pickering double emulsions is promising for the co-delivery of different physico-chemical bio-active compounds, which would improve oral bio-accessibility and bio-activity.

Development of curcumin-loaded methoxy poly(ethylene glycol)-block- poly(caprolactone)-block-poly(1, 4, 8-Trioxa [4.6] spiro-9-undecanone) nanoparticles and studies on their in vitro anti-tumor activities

Yongli Shi,Weiwei Ma,Min Gao,Yaxing Yang

The purpose of this paper was to fabricate a novel methoxy poly(ethylene glycol)-block-poly(caprolactone)-block-poly(1, 4, 8-Trioxa [4.6] spiro-9-undecanone) (mPEG- b -PCL- b -PTOSUO, mPECT) triblock copolymer and study on the in vitro anti-tumor activity of curcumin-loaded mPECT nanoparticles (NPs). The mPEG- b -PCL- b -PTOSUO NPs were fabricated with solvent evaporation. Transmission electron microscope (TEM) and laser particle analyzer were applied to investigate the morphology and size distribution of the obtained mPECT NPs. The cytotoxicity of the copolymer (mPECT) was reflected by cell viability. Curcumin (CUR), as a model drug, was encapsulated into mPECT NPs. The in vitro anti-tumor activity of CUR-loaded mPECT NPs were also studied. 1 H nuclear magnetic resonance ( 1 H NMR), Raman, and Fourier transform infrared spectroscopy (FTIR) spectra confirmed the obtaining of mPECT. TEM photograph showed that most of mPECT NPs were in spherical shapes with a uniform size distribution. High cell viability suggested that the cargo-free mPECT NPs had no obvious cytotoxicity. Fluorescent photographs illustrated that CUR-loaded mPECT NPs could be up-taken by SW1990 cells. The medicated NPs could inhibit the proliferation of SW1990 cells. Therefore, the mPECT NPs could be used as a vehicle to improve the bioavailability and anti-tumor effects of CUR.

Design and evaluation of dual CD44 receptor and folate receptor-targeting double-smart pH-response multifunctional nanocarrier

Chen Daquan,Song Xiaoyan,Wang Kaili,Guo Chunjing,Yu Yueming,Fan Huaying,Zhao Feng

In this article, in order to enhance the bioavailiability and tumor targeting of curcumin (Cur), the oligosaccharides of hyaluronan conjugates, folic acid-oligosaccharides of hyaluronan-acetal-menthone 1,2-glycerol ketal (FA-oHA-Ace-MGK) carried oHA as a ligand to CD44 receptor, double-pH-sensitive Ace-MGK as hydrophobic moieties, and FA as the target of folate receptor. The structure characteristics of this smart response multifunctional dual-targeting nano-sized carrier was measured by fourier-transform infrared (FT-IR) and nuclear magnetic resonance ( 1 H–NMR). Cur, an anticancer drug, was successfully loaded in FA-oHA-Ace-MGK micelles by self-assembly. The measurement results of transmission electron microscopy (TEM) presented that the Cur-loaded micelles were spherical in shape with the average size of 166.3 ± 2.12 nm and zeta potential − 30.07 mV. Much more encapsulated Cur could be released at mildly acidic environments than at pH 7.4, from the Cur-FA-oHA-Ace-MGK micelles. Cytotoxicity assay indicated that non-Cur loaded micelles mostly had no cytotoxicity to MCF-7 cells and A549 cells, and Cur-loaded micelles had significantly lower survival rate than Cur suspension in the same concentration, which proved that the drug-loaded micelles can effectively inhibit tumor cell growth. The targeting of CD44 receptors and folate receptors was proved in vitro cellular uptake assay. These results showed the promising potential of FA-oHA-Ace-MGK as an effective nano-sized carrier for anti-tumor drug delivery.

A stimuli-responsive drug delivery system based on konjac glucomannan, carboxymethyl chitosan and mesoporous polydopamine nanoparticles

Zherui Zhao,Wanting Shi,Yufei Wu,Linxiu Kong,Jun Gao,Yong Kong

A stimuli-responsive drug delivery system is developed for controlled delivery of curcumin (Cur) and chemo-photothermal therapy of breast cancer (BC). Cur is first loaded into mesoporous polydopamine nanoparticles (mPDA NPs) by π-π stacking, and then the Cur loaded mPDA NPs (mPDA NPs@Cur) are encapsulated in the hydrogels prepared through the crosslinking of oxidized konjac glucomannan (oxKGM) and carboxymethyl chitosan (CMCS). Owing to the pH-sensitivity of the hydrogels and the outstanding photothermal conversion capability of mPDA NPs, the release of Cur from the hydrogels can be greatly accelerated in acidic media upon near infrared (NIR) irradiation. Cytotoxicity assay indicates that the hydrogels have significant cytotoxicity against murine breast tumor cell 4 T1 while the drug-free hydrogels (oxKGM/CMCS/mPDA NPs) show good biocompatibility. In addition, the hyperthermia generated upon NIR irradiation can lead to the apoptosis of cancer cells, achieving chemo-photothermal combination therapy of BC. Release kinetics study reveals that the release of Cur from the hydrogels follows zero-order model.

Novel high-strength, recyclable, microbial-resistant, and freeze-thaw dual topological network hydrogel cooling media

Nan Wang,Yi-Miao Zhang,Jun Li,Hua-Feng Mao,Qian Zhou,Hui Yang,Li-Jun Wang,Zhou-Yu Wang,Kun Li,Xiao-Qi Yu

The demand for multifunctional hydrogels, offering high mechanical strength, efficient cooling, and antimicrobial properties, is growing in food preservation. Here, a dual-network (DN) hydrogel PM@Cur , which includes curcumin, is fabricated through chemical crosslinking and hydrogen bonding interactions. The resulting hydrogels can withstand more than five freeze-thaw cycles at −80 °C, and resist brittleness after liquid nitrogen treatment. PM@Cur also exhibits surface hydrophobicity (contact angle >90°) for both water and organic solvents. These properties meet the mechanical, anti-fouling, and recyclable demands for hydrogel coolants. The antimicrobial assays in vitro confirmed that the inclusion of curcumin provided the PM@Cur with photodynamic antimicrobial capacity. Finally, the prepared PM@Cur hydrogel ice cubes have been confirmed to exhibit better anti-melting properties than traditional ice cubes, thus enabling the preservation of strawberries and shrimp. This study presents an innovative solution for producing advanced functional integrated hydrogels, offering a promising and safer option for food coolants.\.

Novel multifunctional triple folic acid, biotin and CD44 targeting pH-sensitive nano-actiniaes for breast cancer combinational therapy

Mengna Liu,Bingjie Wang,Chunjing Guo,Xiaoya Hou,Ziting Cheng,Daquan Chen

In this study, novel multifunctional folic acid, biotin, and CD44 receptors targeted and pH-sensitive “nano-actiniaes” were fabricated with icariin (ICA) and curcumin (Cur) as loaded model drugs for breast cancer therapy. The newly synthesized polymer oligomeric hyaluronic acid-hydrazone bond-folic acid-biotin (Bio-oHA-Hyd-FA) was characterized by 1H NMR spectrogram (proton nuclear magnetic resonance). The obtained drug carrier Bio-oHA-Hyd-FA self-assembled into nanomicelles, named as “nano-actiniaes”, in aqueous media with hydrodynamic diameter of 162.7 ± 5 nm. The size, surface zeta potential, and morphology of the “nano-actiniaes” were observed via TEM. The in vitro release experiment indicated that much more encapsulated icariin (ICA) and curcumin (Cur) were released from the Bio-oHA-Hyd-FA micelles (nano-actiniaes) in the acidic environment. Additionally, the cytotoxicity research demonstrated that the Bio-oHA-Hyd-FA carrier material was completely nontoxic, and the ICA&Cur “nano-actiniaes” had greater cytotoxicity compared with other control groups. In addition, the “nano-actiniaes” were found to significantly inhibit cancer cell invasion by Transwell assay. Moreover, in vivo evaluation of anti-tumor effect illustrated that the ICA and Cur “nano-actiniaes” possessed inhibitory effect on tumors. Consequently, the multi-targeted pH-sensitive “nano-actiniaes” can realize significant tumor targeting and effectively inhibit tumor growth.

Smart nanocomposite hydrogels based on azo crosslinked graphene oxide for oral colon-specific drug delivery

Lin Hou,Yuyang Shi,Guixiang Jiang,Wei Liu,Huili Han,Qianhua Feng,Junxiao Ren,Yujie Yuan,Yongchao Wang,Jinjin Shi,Zhenzhong Zhang

A safe and efficient nanocomposite hydrogel for colon cancer drug delivery was synthesized using pH-sensitive and biocompatible graphene oxide (GO) containing azoaromatic crosslinks as well as poly (vinyl alcohol) (PVA) (GO–N=N–GO/PVA composite hydrogels). Curcumin (CUR), an anti-cancer drug, was encapsulated successfully into the hydrogel through a freezing and thawing process. Fourier transform infrared spectroscopy, scanning electron microscopy and Raman spectroscopy were performed to confirm the formation and morphological properties of the nanocomposite hydrogel. The hydrogels exhibited good swelling properties in a pH-sensitive manner. Drug release studies under conditions mimicking stomach to colon transit have shown that the drug was protected from being released completely into the physiological environment of the stomach and small intestine. In vivo imaging analysis, pharmacokinetics and a distribution of the gastrointestinal tract experiment were systematically studied and evaluated as colon-specific drug delivery systems. All the results demonstrated that GO–N=N–GO/PVA composite hydrogels could protect CUR well while passing through the stomach and small intestine to the proximal colon, and enhance the colon-targeting ability and residence time in the colon site. Therefore, CUR loaded GO–N=N–GO/PVA composite hydrogels might potentially provide a theoretical basis for the treatment of colon cancer with high efficiency and low toxicity.
