Development of MgO-loaded PLA/dECM Antibacterial Nanofibrous Membranes for Enhanced Gingival Regeneration

Shu Sun,Jing Qin,Yifu Zhuang,Pengfei Cai,Xiao Yu,Hongsheng Wang,Xiu-Mei Mo,Jinglei Wu,Mohamed El-Newehy,Meera Moydeen Abdulhameed,Mingyue Fan,Wenhao Qian,Binbin Sun

Clinically, gingival tissue repair is challenging due to the complex oral microbial environment and inflammation. The development of gingival membranes using tissue engineering techniques offers a promising solution to this issue. This study focuses on developing the nanofibrous gingival membrane, combining polylactic acid (PLA), decellularized extracellular matrix (dECM), and magnesium oxide (MgO) nanoparticles. Electrospinning was used to fabricate membranes with varying ratios of PLA, dECM, and MgO, and their mechanical, antibacterial, and cell-proliferation properties were evaluated. NIH-3T3 and rat gingival fibroblasts (RGF) cells were cultured on the membranes to assess biocompatibility. A rat model with gingival defects was used to test in vivo tissue regeneration. It was indicated that the antibacterial nanofibrous membranes with MgO showed enhanced antibacterial effects, reduced inflammation, and promoted gingival tissue repair.

Robust, self-lubricating and injectable dECM-loaded hydrogels based on rapid photo-crosslinking for articular cartilage regeneration

Zhenxi Liu,Dong Zhang,Huili Kang,Xinyi Gu,Fangping Chen,Changsheng Liu

The restoration of articular cartilage poses a significant clinical challenge due to its avascular nature and the limited presence of chondrocytes. Recently, there has been a growing emphasis on developing materials that emulate the properties of cartilage to facilitate high-quality repair. Here, an injectable dECM-loaded hydrogel (HS-dECM) with potent compressive strength and self-lubricating ability was prepared by photo-crosslinking for cartilage regeneration. The HS-dECM hydrogel was formed by integrating cartilage decellularized extracellular matrix (dECM) with hyaluronic acid methacrylate (HAMA) and sulfobetaine methacrylate (SBMA). Once injected into irregular defects, a robust hydrogel was rapidly formed in situ by UV irradiation with negligible heat release. The HS-dECM hydrogel exhibited robust compressive strength (898 ± 86 kPa), excellent fatigue resistance and a low friction coefficient, and had the feasibility of minimally invasive implantation. Besides, HS-dECM hydrogel promoted chondrogenic differentiation of BMSCs in vitro and repaired articular cartilage defects in rats. Therefore, the high-performance injectable HS-dECM hydrogel represents a novel minimally invasive strategy that significantly promote the cartilage injuries repair and regeneration.

The Development of a 3D PET Fibrous Scaffold Modified with an Umbilical Cord dECM for Liver Tissue Engineering

Yang Li,Yang Zhang,Kebo Zhong,Shuguang Liao,Guifeng Zhang

Organ and tissue dysfunction represents a clinically significant condition. By integrating cell biology with materials science, tissue engineering enables the reconstruction and restoration of damaged tissues or organs, offering a noninvasive repair approach. In our study, we replicated the cellular growth environment by utilizing a human umbilical cord-derived decellularized extracellular matrix (dECM) as a modifying agent for the polyethylene terephthalate (PET) polymeric fiber scaffold. This allowed us to create a dECM-coated polyester fiber-based scaffold, PET-dECM, tailored for liver tissue engineering purposes. We effectively produced a decellularized human umbilical cord-derived ECM through a combined decellularization process involving trypsin/EDTA, TritonX-100, and sodium deoxycholate. The application of the dECM coating onto the PET material was accomplished through several steps, such as ester hydrolysis, EDC/NHS-activated crosslinking, and dECM conjugation. The biological performance of the PET-dECM was validated using RG cell culture assays. Notably, the dECM coating significantly improved PET’s hydrophilicity and biocompatibility, thereby aiding cell adhesion, proliferation, and functional differentiation (p< 0.05). It was further found that the hepatocyte function of HepaRG was significantly enhanced on the PET-dECM, which may be attributed to the dECM’s ability to facilitate the restoration of cell polarity. The PET-dECM holds promise as an effective hepatocyte culture carrier and could potentially find application in liver tissue engineering.

Preparation and properties of a 3D printed nHA/PLA bone tissue engineering scaffold loaded with a β-CD–CHX combined dECM hydrogel

Shangbo Li,Zijian Liu,Xiaohan Gao,Lidi Cheng,Zexian Xu,Li Li,Yaru Diao,Liqiang Chen,Yanshan Liu,Jian Sun

Jaw defects, which can result from a multitude of causes, significantly affect the physical well-being and psychological health of patients. The repair of these infected defects presents a formidable challenge in the clinical and research fields, owing to their intricate and diverse nature. This study aims to develop a personalized bone tissue engineering scaffold that synergistically offers antibacterial and osteogenic properties for treating infected maxillary defects. This study engineered a novel temperature-sensitive, sustained-release hydrogel by amalgamating β-cyclodextrin (β-CD) with chlorhexidine (CHX) and a decellularized extracellular matrix (dECM). This hydrogel was further integrated with a polylactic acid (PLA)–nano hydroxyapatite (nHA) scaffold, fabricated through 3D printing, to form a multifaceted composite scaffold (nHA/PLA/dECM/β-CD–CHX). Drug release assays revealed that this composite scaffold ensures prolonged and sustained release. Bacteriological studies confirmed that the β-CD–CHX loaded scaffold exhibits persistent antibacterial efficacy, thus effectively inhibiting bacterial growth. Moreover, the scaffold demonstrated robust mechanical strength. Cellular assays validated its superior biocompatibility, attributed to dECM and nHA components, significantly enhancing the proliferation, adhesion, and osteogenic differentiation of osteogenic precursor cells (MC3T3-E1). Consequently, the nHA/PLA/dECM/β-CD–CHX composite scaffold, synthesized via 3D printing technology, shows promise in inducing bone regeneration, preventing infection, and facilitating the repair of jaw defects, positioning itself as a potential breakthrough in bone tissue engineering.

Constructing Nerve Guidance Conduit using dECM-Doped Conductive Hydrogel to Promote Peripheral Nerve Regeneration

Lizhao Yan,Shuang Liu,Jianwen Wang,Xiaoyue Ding,Yingsong Zhao,Nan Gao,Zishen Xia,Ming Li,Qianqian Wei,Oseweuba Valentine Okoro,Yanfang Sun,Lei Nie,Amin Shavandi,Guohua Jiang,Jianghai Chen,Lihong Fan,Yuxiong Weng

Peripheral nerve injury often leads to the loss of neurological functions due to the slow regeneration rate and inefficient functional reconstruction. Current clinical treatments using nerve guidance conduits (NGCs) still face challenges in providing a biomimetic microenvironment to promote nerve repair. Herein, decellularized extracellular matrix (dECM) is obtained from porcine Achilles tendon and crosslinked with 3-amino-4-methoxybenzoic acid grafted gelatin (PAMB-G) to obtain conductive hydrogels. Then, a novel nerve guidance conduit is developed by assembling poly(vinyl alcohol) (PVA) conduit and conductive ECM@PAMB-G hydrogel. This bioengineered ECM@PAMB-G/PVA conduit demonstrated excellent cytocompatibility, electrical conductivity, mechanical properties, and biodegradability. In vitro experiments confirmed that the ECM@PAMB-G hydrogel significantly promotes the proliferation and migration of PC12 cells and primary Schwann cells, as well as the growth of dorsal root ganglion (DRG) axons. Furthermore, in vivo studies in a rat sciatic nerve model exhibited improvements in axonal regeneration, Schwann cell migration, myelin sheath formation, and functional recovery mediated by the ECM@PAMB-G/PVA conduit. This work demonstrates the synergistic effects of extracellular matrix and electrical cues in enhancing peripheral nerve regeneration. The ECM@PAMB-G/PVA nerve guidance conduit shows potential as an alternative to autografts for supporting peripheral nerve reconstruction.

Lubricating Micro-Interface Assisted General Strategy for Preparing dECM-Microparticle–Based Heterogeneous Granular Inks toward 3D Printing

Kunxi Zhang,Yinan Zhu,Tao Wang,Dawei Niu,Zijie Xiahou,Hongzhong Wang,Peiliang Fu,Haiyan Cui,Jingbo Yin

Decellularized extracellular matrix (dECM) retains the bioactive components to mimic tissue-specific microenvironment. Designing dECM-based inks for 3D printing to prepare scaffolds can enhance the regeneration of corresponding tissues. However, due to the strict requirements for inks, how to efficiently prepare dECM-based inks is a challenge. Inspired by granular lubrication, the present study proposes an effective and general method for preparing granular ink from dECM microparticles (MP) with high content. Through dopamine polymerization, dECM-MPs are coated with polydopamine (PDA). Meanwhile, gelatin methacryloyl (GelMA)-based microspheres (GelMA-MSs) are prepared with available double bonds. dECM-MPs (30–40 wt%) and GelMA-MSs assemble to form a granular ink, exhibiting well-performed extrudability, plasticity, and fidelity. Further inspired by joint lubrication, hyaluronic acid (HA) is introduced into PDA coating and GelMA-MSs to reduce the micro-interface friction, further promoting the printability and broadening the printable window of dECM-MPs content to 30–60 wt%. Osteochondral scaffold and skin wound dressing are fabricated through 3D printing from the corresponding dECM-MPs, enhancing the regeneration of the corresponding tissues in vivo. Especially, the realization of printability of highly rigid dECM-MPs confirms the universality, reliability and efficiency of the present strategy.

dECM restores macrophage immune homeostasis and alleviates iron overload to promote DTPI healing: Dysfunctional iron metabolism in pressure injuries is related to aberrant CD163 and homx-1 signal transduction

Zhang Ju,Shan Hui,Guo Jinglin,Wang Xiaoying,Wang Weiwei

Due to its highly insidious and rapid progression, deep tissue pressure injury (DTPI) is a clinical challenge. Our previous study found that DTPI may be a skeletal muscle injury dominated by macrophage immune dysfunction due to excessive iron accumulation. Decellularized skeletal muscle matrix (dECM) hydrogel promotes skeletal muscle injury repair. However, its role in polarizing macrophages and regulating iron metabolism in DTPI remains unclear. Here, porcine dECM hydrogel was prepared, and its therapeutic function and mechanism in repairing DTPI were investigated. The stimulus of dECM hydrogel towards RAW264.7 cells resulted in a significantly higher percentage of CD206+ macrophages and notably decreased intracellular divalent iron levels. In mice DTPI model, dECM hydrogel treatment promoted M1 to M2 macrophage conversion, improved iron metabolism, and reduced oxidative stress in the early stage of DTPI. In the remodeling phase, the dECM hydrogel remarkably enhanced revascularization and accelerated skeletal muscle repair. Furthermore, the immunomodulation of dECM hydrogels in vivo was mainly involved in the P13k/Akt signaling pathway, as revealed by GO and KEGG pathway analysis, which may ameliorate the iron deposition and promote the healing of DTPI. Our findings indicate that dECM hydrogel is promising in skeletal muscle repair, inflammation resolution, and tissue injury healing by effectively restoring macrophage immune homeostasis and normalizing iron metabolism.

Modulating Cell–Scaffold Interaction via dECM-Decorated Melt Electrowriting PCL Scaffolds

Wenchao Li,Xiang Gao,Peng Zhang

Aligned fibrous scaffolds are essential for directing soft-tissue regeneration, yet synthetic polymers lack native biochemical cues. To bridge this gap, bioactive and anisotropic scaffolds were developed by combining melt electrowriting (MEW) with decellularized extracellular matrix (dECM) decoration to enhance cell–scaffold interactions for soft tissue engineering. Porous polycaprolactone (PCL) scaffolds with aligned microfibers and tunable pore architectures (aspect ratios 1:1, 1:2, and 1:3) were fabricated via MEW and subsequently coated with porcine skeletal muscle dECM using a dip-gelation method. Comprehensive surface characterization confirmed the presence and robust adhesion of the dECM coating on the PCL scaffolds, which concurrently enhanced surface hydrophilicity. Furthermore, mechanical testing demonstrated that the resulting composite scaffold retained the structural integrity required to meet the mechanical demands of tissue regeneration. In vitro studies using L929 fibroblasts demonstrated that dECM decoration significantly improved cell adhesion, proliferation, and alignment along the fiber direction. Notably, scaffolds with 1:1 and 1:2 aspect ratios supported the highest cell density and guided morphological elongation most effectively. These findings highlight the synergistic potential of topographical cues and biochemical signaling in scaffold design for functional tissue regeneration.

Multi-responsive injectable dECM hydrogel for sustained doxycycline delivery in osteoarthritis therapy

Ximing Wang,Ting Zheng,Luyao Cai,Clara Chen,Junjie Xu,Jinzhong Zhao

Osteoarthritis (OA) is the most prevalent degenerative joint disease worldwide. Current therapeutic approaches are limited by rapid drug clearance from the joint cavity, necessitating frequent administrations and failing to provide sustained therapeutic effects. Injectable hydrogels that can persistently remain within the joint space represent a promising strategy for comprehensive OA treatment. Herein, an injectable hydrogel system based on dermal-derived decellularized extracellular matrix (dECM) engineered with N-isopropylacrylamide, tannic acid, and sodium tetraborate was developed to achieve superior lubrication and strong tissue adhesion. The hydrogel exhibits exceptional lubrication properties and robust tissue adhesion, enabling prolonged retention within the joint cavity while providing continuous cartilage protection. Its excellent shear-thinning behavior and rapid self-healing capability facilitate minimally invasive injection and ensure conformal coverage of defect sites. In vitro and in vivo evaluations demonstrated that the hydrogel promotes cell proliferation, modulates anti-inflammatory responses, and significantly preserves cartilage integrity with enhanced proteoglycan and type II collagen (COL-2) expression. The sustained therapeutic efficacy was attributed to the hydrogel’s ability to maintain prolonged residence time while continuously delivering bioactive components and providing mechanical lubrication. These findings demonstrate that this multifunctional dECM-based injectable hydrogel represents a promising therapeutic platform for OA treatment suitable for clinical translation.

Mitochondrial energy reprogramming via dECM/GP@EVs@Gpnmb to prevent post-myocardial infarction heart failure

Na Li,Ying Wang,Ling Yuan,Shuang Peng,Zhansheng Hu,Baoquan Wang,Dan Zhang,Longxiang Su,Huiping Wu

Heart failure following myocardial infarction (MI) remains a major cause of mortality. This study aimed to prevent post-MI heart failure by facilitating macrophage mitochondrial transfer through Engineered Extracellular Vesicles-Dextran-Peptide Hydrogel (dECM/GP@EVs@Gpnmb) to induce reprogramming of myocardial mitochondrial energy metabolism. Key macrophage subsets in MI progression were identified using single-cell RNA sequencing data. A mouse model with Gpnmb knockout in macrophages was developed, showing exacerbated myocardial injury. In vitro experiments demonstrated that Gpnmb+ macrophages could transfer “healthy” mitochondria to cardiomyocytes, regulating mitochondrial energy metabolism. The dECM/GP@EVs@Gpnmb promoted a reparative macrophage phenotype, improving mitochondrial energy metabolism in cardiomyocytes and reducing myocardial damage. In vivo , dECM/GP@EVs@Gpnmb treatment enhanced the accumulation of EVs@Gpnmb in infarcted areas, improving cardiac function in MI mice. Overall, this approach holds promise for preventing and treating post-MI heart failure by mediating macrophage mitochondrial transfer and reprogramming myocardial mitochondrial function.

Bioprinted dECM particles-laden microgels enhance the mesenchymal stromal cell paracrine effect for myocardial infarction treatment

Shiqin Peng,Ying Hao,Bo Li,Jingruo Chen,Hao Zhou,Tailuo Liu,Dingyi Zhang,Lifan Xu,Qiyue Liao,Hanif Ullah,Yuwen Chen,Mao Chen

Despite guideline-directed reperfusion and pharmacological therapies, myocardial infarction (MI) continues to pose substantial clinical challenges, with high incidence and mortality rates. Mesenchymal stromal cell (MSC) transplantation represents a promising therapeutic strategy, yet its efficacy remains limited by poor survival and unstable paracrine function of transplanted cells within the hostile infarct microenvironment. Here, we developed an integrated microcarrier system via digital light processing, which encapsulates MSCs within microgels functionalized with cardiac-derived decellularized extracellular matrix (dECM) particles (termed MSCs@dECMMG). By leveraging the native biochemical composition and structural cues of cardiac dECM, this bioinspired microcarrier was designed not only to enhance MSC viability but also to actively modulate their secretory activity. Conditioned medium from MSCs@dECMMG promoted angiogenic responses in human umbilical vein endothelial cells and attenuated oxidative stress‑induced apoptosis in H9c2 cells. In the rat MI model, local implantation of MSCs@dECMMG markedly improved the left ventricular ejection fraction from 32.03% to 60.77%, reduced the fibrotic area by 17.44%, and increased the left ventricular wall thickness by 2.6-fold. Collectively, this study demonstrates that a dECM‑integrated microgel can serve as a bioactive carrier that directs MSC secretory signaling, offering a novel strategy to advance cell‑based therapy for MI. Statement of significance Low survival and unstable paracrine effects of transplanted MSCs in the hostile infarct microenvironment remain a major clinical challenge. Addressing how to specifically enhance MSC paracrine function is critical for effective cardiac repair. Here, we present a precision-engineered microcarrier system that integrates mechanically ground cardiac dECM particles into DLP-bioprinted microgels to deliver MSCs. Unlike prior studies focusing on proliferation or differentiation behaviors, we further uncovered and harnessed cardiac dECM to potentiate MSC paracrine signaling, markedly improving myocardial repair following MI. This introduces a new dimension to dECM application—from guiding differentiation to paracrine enhancement—while providing a translational-ready platform with scalability and controllability for future cell-based therapies. Tendon dECM Composited with Chitosan with Loading Skin Precursor Stem Cell Exosome for Enhanced Diabetic Wound Healing

Yunguang Chen,Yingying Liang,Yaling Deng,Lei Nie

Diabetic wounds are a common and severe complication of diabetes mellitus, characterized by delayed healing due to persistent inflammation, impaired angiogenesis, and cellular dysfunction. Conventional therapeutic approaches remain limited in efficacy. In recent years, exosomes have attracted considerable attention in wound healing and regenerative medicine because of their crucial role in intercellular communication and tissue repair. However, rapid clearance of exosomes in vivo greatly limits their therapeutic efficacy. To address this critical limitation, we engineered a decellularized extracellular matrix (dECM)-based hydrogel system functionalized with exosomes derived from skin-derived precursor cells (SKPs). This biomimetic scaffold was designed to serve as a local exosome-delivery platform at the wound site, with the aim of improving exosome utilization and augmenting their regenerative effects. Comprehensive in vitro characterization demonstrated that the exosome-loaded composite hydrogels exhibited robust pro-angiogenic activity, as evidenced by enhanced endothelial cell proliferation, migration, and tube formation. Moreover, the hydrogels displayed significant antibacterial effects against wound-relevant pathogens and potent reactive oxygen species (ROS)-scavenging capacity, thereby mitigating oxidative damage. Notably, the composite hydrogels also promoted the phenotypic polarization of macrophages toward the pro-regenerative M2 phenotype. In parallel, in vivo studies using a streptozotocin-induced diabetic rat wound model confirmed that treatment with the composite hydrogels significantly accelerated wound closure rates compared to control groups. Histological and immunohistochemical analyses revealed enhanced angiogenesis, as evidenced by increased CD31-positive microvessel density, as well as improved collagen deposition, re-epithelialization, and an attenuated local inflammatory microenvironment characterized by reduced pro-inflammatory cytokine expression and elevated M2 macrophage infiltration. Collectively, the SKPs exosome-loaded dECM based composite hydrogels developed in this study represent a potential therapeutic strategy for the treatment of diabetic wounds.
