Phlorizin alleviates deltamethrin-induced oxidative stress in brine shrimp Artemia

Ma Dandan,Zhou Qingli,Sui Liying,Guo Qingbin,Liu Huanhuan,Liang Honghe,Li Zhenjing,Sang Zhongna

Deltamethrin (DEL), a commonly used pyrethroid pesticide, results in higher reactive oxygen species (ROS) levels in aquatic animals, which consequently unbalance the redox state. Phlorizin (PHL) is a flavonoid and a natural product promising to prevent or reduce pesticide-induced oxidative stress. Artemia is a micro-crustacean widely used in marine hatcheries and an experimental aquatic organism for environmental toxicology research. This research aimed to evaluate the toxicity of DEL on Artemia and the antioxidative effect of PHL against the toxicity. Results show that 0.08-mg/mL PHL exerted its antioxidative effects on hatching percentage of the cysts in 24-h incubation and on body length and survival rate of Artemia in 12-d culture. After 12-d culture, 12-, 24-, and 36-h DEL exposure showed significant drops in SOD, CAT, and GSH-Px enzyme activities, and significant increases in ROS and malondialdehyde (MDA) levels in Artemia ( P <0.05). On the contrary, 0.08-mg/mL PHL application improved the enzyme activities and decreased the ROS and MDA levels ( P <0.05). Moreover, 0.08-mg/mL PHL significantly increased mRNA expression levels of Cu/Zn SOD, CAT, GST, HO-1, NQO1 , and Nrf2 , and decreased mRNA expression level of Keap1 in the DEL-exposed Artemia ( P <0.05). Therefore, DEL is toxic to Artemia , while PHL alleviates DEL-induced oxidative damage by possibly regulating the Nrf2 signaling pathway. This study provided a theoretical basis for PHL to reduce pesticide-induced toxicity in aquatic animals.

Development of a novel deltamethrin sensor based on molecularly imprinted silica nanospheres embedded CdTe quantum dots

Shenguang Ge,Juanjuan Lu,Lei Ge,Mei Yan,Jinghua Yu

A novel procedure for the determination of deltmethrin (DM) is reported. The water-soluble CdTe quantum dots (QDs) and highly fluorescent silica molecularly imprinted nanospheres embedded CdTe QDs (CdTe–SiO 2 -MIPs) were prepared and characterized by fluorescence spectroscopy, UV–vis spectroscopy, TEM and IR. The fluorescence nanosensor based CdTe–SiO 2 -MIPs is developed. The possible quenching mechanism is discussed by DM. Under optimal conditions, the relative fluorescence intensity of CdTe–SiO 2 -MIPs decreased with increasing DM by a Stern–Volmer type equation in the concentration range of 0.5–35.0   μg   mL −1 , the corresponding detection limit is 0.16   μg   mL −1 . The developed sensor based on CdTe–SiO 2 -MIPs was applied to determine DM in fruit and vegetable samples.

Fermentation and Immobilization of Insect-Derived Deltamethrin-Degrading Strain, Microbacterium sp

Zhengyan Wang,Qiong Luo,Yifan Liu,Tianwei Ye,Yujia Zhang,Wei Xu

In agricultural production, the extensive use of insecticides has led to the persistent presence of residues such as deltamethrin in the environment, posing threats to ecosystems and human health. Utilizing microbes for environmental bioremediation presents a feasible approach. Studies have found that insects with strong insecticide tolerance often harbor gut bacteria capable of degrading insecticides. These gut bacteria are becoming an excellent reservoir of insecticide-degrading bacteria with bioremediation potential. However, in practical applications, the activity of free bacterial cells is easily affected by environmental factors, limiting the stability of their remediation efficacy. This study focused on a Microbacterium sp. strain isolated from insects. By optimizing culture conditions and the sodium alginate immobilization process, the remediation efficiency of the immobilized strain in contaminated soil and water was investigated. The results demonstrated that the immobilized strain significantly outperformed free bacteria in deltamethrin degradation performance in both contaminated soil and water, while also exhibiting good stability and reusability. This research provides an effective method for the remediation of deltamethrin-contaminated environments. Extensive application of deltamethrin on agricultural products results in serious contamination of the environment. Its negative impact on environmental and public health necessitates the development of environmental remediation technologies. Detailed investigations of microbial degradation of deltamethrin may be useful for the development of bioremediation strategies. In this study, the deltamethrin removal capability of a bacterial strain, Microbacterium sp., previously isolated from the gut of Tribolium castaneum (Herbst) (Coleoptera: Tenebrionidae) was first investigated. When 3 mL of the bacterial solution (OD 600 = 1) was inoculated into 97 mL of MS media containing 200 μg/mL deltamethrin, it could remove 45.7% deltamethrin after 64 h of incubation. This strain grew fastest in LB media with an inoculum volume of 3% in pH 7 at 175 rpm, 25 °C. To enhance its environmental tolerance, this strain was immobilized with sodium alginate. Microbacterium sp.-containing calcium alginate microspheres (CAMs) exhibited an enhanced deltamethrin removal capability compared to free bacteria, and CAMs generated by immobilization with 2% sodium alginate and 3% CaCl 2 cross-linking for 4 h possessed the maximum deltamethrin removal capability. The ultrastructure of Microbacterium sp.-containing CAMs prepared under optimal conditions was a three-dimensional mesh structure with pores and dense features, and the bacteria grew well in the immobilized carrier. After being reused five times, the deltamethrin removal rate of immobilized Microbacterium sp. still reached over 50%. When Microbacterium sp. was inoculated into deltamethrin-contaminated water or soil for 48 h, the deltamethrin removal rate of immobilized bacteria was 1.4 times higher than that of free bacteria. These results suggest that Microbacterium sp.-containing CAMs possess an excellent deltamethrin removal capability and good reusability, showing great potential for the remediation of deltamethrin-contaminated environments.

High-Dose Deltamethrin Induces Developmental Toxicity in Caenorhabditis elegans via IRE-1

Chuhong Chen,Ying Deng,Linyan Liu,Zhenyan Zou,Chenzhong Jin,Zhiyin Chen,Shuanghui Wang

Deltamethrin (DM), a Type II pyrethroid, is widely used worldwide in agriculture, household applications, and medicine. Recent studies have shown that DM exerts a variety of toxic effects on organs such as the kidney, heart muscle, and nerves in animals. However, little is known about the effects of high-dose DM on growth and development, and the mechanism of toxicity remains unclear. Using theCaenorhabditis elegansmodel, we found that high-dose DM caused a delay in nematode development. Our results showed that high-dose DM reduced the activation of the endoplasmic reticulum unfolded protein response (UPRER). Further studies revealed that high-dose DM-induced developmental toxicity and reduced capacity for UPRERactivation were associated with the IRE-1/XBP-1 pathway. Our results provide new evidence for the developmental toxicity of DM and new insights into the mechanism of DM toxicity. Keywords:deltamethrin;Caenorhabditis elegans;development;toxicity;endoplasmic reticulum unfolded protein response

miR-29b-triggered epigenetic regulation of cardiotoxicity following exposure to deltamethrin in zebrafish

Jie Wang,Yuanyuan Liu,Yujia Yan,Aoxue Wang,Yuyao Jiang,Zexin Wen,Kun Qiao,Hanqing Li,Tiantian Hu,Yongfang Ma,Shengli Zhou,Wenjun Gui,Shuying Li

Deltamethrin is a classical pyrethroid insecticide that is frequently detected in aquatic environments and organisms. Furthermore, deltamethrin has been detected in samples related to human health and is a potential risk to public health. This study aimed to investigate the mechanism of cardiotoxicity induced by deltamethrin. Zebrafish were exposed to 0.005, 0.05, or 0.5 μg/L deltamethrin for 28 days. The results showed a significant reduction in male reproduction compared to female reproduction. Additionally, the heart rate decreased by 15.75 % in F1 after parental exposure to 0.5 μg/L deltamethrin. To evaluate cardiotoxicity, deltamethrin was administered to the zebrafish embryos. By using miRNA-Seq and bioinformatics analysis, it was discovered that miR-29b functions as a toxic regulator by targeting dnmts . The overexpression of miR-29b and inhibition of dnmts resulted in cardiac abnormalities, such as pericardial edema, bradycardia, and abnormal expression of genes related to the heart. Similar changes in the levels of miR-29b and dnmts were also detected in the gonads of F0 males and F1 embryos, confirming their effects. Overall, the results suggest that deltamethrin may have adverse effects on heart development in early-stage zebrafish and on reproduction in adult zebrafish. Furthermore, epigenetic modifications may threaten the cardiac function of offspring.

Acute and chronic toxicity of deltamethrin, permethrin, and dihaloacetylated heterocyclic pyrethroids in mice

Qiuyan Zhu,Yang Yang,Zhiting Lao,Yingying Zhong,Kun Zhang,Suqing Zhao

BACKGROUND Pyrethroids, a class of insecticides, that act on the nervous system of insects. Frequent consumption of foods with pyrethroid residues increase the risk of developmental and neurological diseases in humans. Assessing the toxicity of novel synthetic pyrethroids to mammals is also critical to the development of agrochemicals. RESULTS Using mice as models, the acute and chronic toxicity of deltamethrin, permethrin, dihaloacetylated pyrethroids to mammals was researched by gavage administration. Acute toxicity assessment displayed that the median lethal dose (LD 50 ) of deltamethrin, permethrin and dihaloacetylated heterocyclic pyrethroids DCA-O, DCA-01, and DCA-11 tested were greater than 500 mg/kg of weight. Furthermore, chronic toxicity assessment demonstrated that deltamethrin, permethrin caused epidermal damage near the genitals, while dihaloacetylated heterocyclic pyrethroids DCA-O, DCA-01, and DCA-11 showed no relevant symptoms. However, both the acute and chronic toxicity assessment suggested that pyrethroids exposure induced mice loss weight. Additionally, the elevated plus maze (EPM) test showed that pyrethroids caused anxiety-like behaviors and no motor defects in Kunming mice. Beside, during the sucrose preference test (SPT), 60-day pyrethroids exposure increased excitatory behaviors in mice. However, the neurochemical studies displayed that pyrethroids exposure increased the total amount of glutamate (Gln), glutamine (Glu) and γ -aminobutyric acid (GABA) in the mice's blood. CONCLUSION Pyrethroids exposure induced weight loss in mice, although the acute oral toxicity of deltamethrin, permethrin and dihaloacetylated heterocyclic pyrethroids DCA-O, DCA-01, and DCA-11 was low. However, regarding chronic toxicity, deltamethrin, permethrin and dihaloacetylated heterocyclic pyrethroids DCA-O, DCA-01, and DCA-11 induced anxiety-like behaviors, excitatory behaviors, Gln-Glu-GABA circulatory dysfunction in blood. Particularly, deltamethrin, though permethrin also had reproductive toxicity. © 2020 Society of Chemical Industry

Photodegradation kinetics, mechanism and aquatic toxicity of deltamethrin, permethrin and dihaloacetylated heterocyclic pyrethroids

Qiuyan Zhu,Yang Yang,Zhiting Lao,Yingying Zhong,Kun Zhang,Suqing Zhao

Photochemical methods attracted much research interests for their high-efficiency and low secondary pollution. Decomposition of synthetic pyrethroids , the fourth major group of insecticides in use worldwide, was also of great significance due to their possible environmental risks. The photodegradation of deltamethrin , permethrin and dihaloacetylated heterocyclic pyrethroids in methanol/acetone = 9/1 (by volume) by a 400 W mercury lamp was examined. The t 1/2 of tested pyrethroids was less than 25 min, except for cis -permethrin with a t 1/2 of up to 50 min. The trans -isomer of permethrin and compound DCA-01 with a smaller t 1/2 might be more susceptible to degradation than their cis -isomer. Besides, the photodegradation of pyrethroids was divided into twelve pathways including isomerization , ester hydrolysis, ester bond cleavage, C O bond cleavage, 3,3-dimethylacrylate formation, double bond break, C1-C3 bond cleavage in cyclopropyl, reductive dehalogenation, decarboxylation , nucleophilic reagents attack on lone pair electrons on oxygen atoms in the phenyl ether, cyano hydrolysis, and halogenated hydrocarbon hydrolysis. The ECOSAR program displayed that pyrethroids and most of their photodegradation products were toxic to fish, daphnid, green algae . Particularly, some photodegradation products were more harmful to aquatic organisms than their parents.

New evidence for neurobehavioral toxicity of deltamethrin at environmentally relevant levels in zebrafish

Lei Lei,Biran Zhu,Kun Qiao,Yuxi Zhou,Xiangping Chen,Jun Men,Lihua Yang,Qidong Wang,Jian Han,Bingsheng Zhou

Deltamethrin, a widely used type II pyrethroid insecticide, was reported with neurotoxicity to aquatic organisms, such as fish. However, the effects and potential mechanisms on the central nervous system remain largely unknown, especially under environmental concentrations. Therefore, we exposed adult female zebrafish to environmentally relevant levels of deltamethrin (30, 100, and 333 ng/L) for 21 days to assess neurobehavioral changes related to the central nervous system and explore the modes of action. Behavioral assays revealed significant increases in the swimming speeds, residence time near other fish and the shoaling cohesion in exposed fish. Transcriptomic results enriched the disrupted neural functions involving the glutamatergic and dopaminergic synapses in the brain. The qRT-PCR confirmed the upregulation of the factors for promoting the glutamate release. The measurement of neurotransmitters showed significantly increased content of the excitatory neurotransmitter glutamate in the brain. Taken together, deltamethrin exposure increased the glutamate level and promoted the release of such an excitatory neurotransmitter between the glutamatergic synapses in the brain, which eventually led to hyperactivity of social behaviors in adult zebrafish.

In vitro study of deltamethrin-induced extracellular traps in hemocytes of Ruditapes philippinarum

Yijing Han,Qianqian Zhang,Lizhu Chen,Jianmin Zhao,Dinglong Yang

Deltamethrin (DLM), a broad-spectrum pesticide, has been proven to have toxic effects on aquatic organisms. Here, we detected the formation of extracellular traps (ETosis) formation in Manila clam ( Ruditapes philippinarum ) hemocytes stimulated by three concentrations of DLM (0.01, 0.1 and 1 μg/mL) in vitro, and explored the underlying mechanisms induced by this pesticide. Extracellular DNA structure observation and quantitative results indicated that DLM exposure could obviously induce hemocytes ETosis, especially under high concentration of DLM induction. Moreover, DLM increased the levels of myeloperoxidase (MPO) and reactive oxygen species (ROS) in a dose-dependent manner, and enhanced the mRNA expression of several ROS-related genes. DPI (NADPH oxidase inhibitor) and ABAH (MPO inhibitor) could substantially inhibit DLM-induced extracellular traps (ETs), suggesting that the induced ETs release was caused by the induction of the ROS burst and MPO production. In addition, three concentrations of DLM-induced ETs were also accompanied by mitochondrial dysfunction, such as increasing the production of mitochondrial ROS, leading to a decrease in mitochondrial membrane potential (MMP) and activation of mitochondrial permeability transition pore (MPTP). Taken together, these results will shed new light on the immunotoxicity of DLM in clams and perhaps lays the foundation for health assessment in bivalves.

Optimised Preparation and Formula of Deltamethrin Nanoemulsion for Enhanced Insecticidal Efficacy and Superior Biosafety

Bingzhang Li,Ning Wei,Yaohai Li,Yang Ci,Quzhen Gesang,Jianguo Feng

Stable nanoemulsions with fine droplets reduce organic solvent use and improve the dispersion of hydrophobic pesticide. However, current studies on deltamethrin nanoemulsion lack systematic formula optimization, performance evaluation and biosafety assessment. This study developed a stable deltamethrin nanoemulsion (Del@Ne) and tested its physicochemical properties, insecticidal activity and non-target safety. In 2025, the effects of surfactant ratio, dosage, preparation temperature and emulsification method on emulsion stability was systematically investigated. The optimal formula contained an active ingredient (2.5% deltamethrin), a surfactant ratio of 8:1 (#601:#500), a 6% surfactant dosage, a 17.25% oil phase (S-100:DMF = 20:3), and deionised water filled to 100%, prepared by adding deionised water to an oil phase containing deltamethrin and surfactants at 40 °C. Del@Ne exhibited small droplet size and good storage stability (TSI ≈ 1), which had better wettability on peach leaves with contact angle falling from 40.4° to 21.6° in 120 s. Del@Ne also gave higher toxicity againstMyzus persicae(LC50= 66.85 mg L−1) than Del@EC (80.69 mg L−1), while showing lower toxicity to zebrafish, earthworms and Harmonia axyridis, as well as better biocompatibility with human L02 hepatocytes. These results provide references for rapid screening of nanoemulsion formulation parameters and also offer insights for the efficient utilization of hydrophobic pesticides.

Two integument-enriched chemosensory proteins contribute to deltamethrin tolerance through cuticular binding in Locusta migratoria

Haihua Wu,Qiaoqiao Xu,Yuemin Ma,Xiaoming Zhao,Jianzhen Zhang,Yichao Zhang

Insect chemosensory proteins (CSPs) are ubiquitously distributed in both chemosensory and non-chemosensory tissues and play multiple physiological roles. Recent studies have demonstrated that CSPs-mediated resistance/tolerance is a novel mechanism of insecticide resistance/tolerance. Two CSP genes ( LmCSP3 and LmCSP4 ) in Locusta migratoria could be induced by deltamethrin. However, it remains unclear whether they function in deltamethrin tolerance. Thus, in this study, we investigated the structural characteristics, spatiotemporal expression patterns, and functional roles of LmCSP3 and LmCSP4 in deltamethrin tolerance. The results showed that the two LmCSPs include the conserved olfactory-specific protein D (OS-D) domain and four invariant cysteine residues arranged in a C1-X8-C2-X18-C3-X2-C4 pattern. Both LmCSPs showed integument-enriched expression patterns. LmCSP3 's peak expression was found in the 5th-instar nymphs while LmCSP4 's expression was observed in the 3rd and 5th instars. RNAi-mediated knockdown of LmCSP3 and LmCSP4 increased locust mortality by 20.86% and 25.96% at a diagnostic deltamethrin concentration of 18 μg/mL, respectively ( P  ≤ 0.05). HPLC-based quantification demonstrated that silencing of LmCSP3 and LmCSP4 significantly increased the cuticular penetration rate of deltamethrin by 20.15% and 22.16%, respectively ( P  ≤ 0.01). In vitro fluorescence competitive binding assays showed that LmCSP3 had moderate binding affinities to deltamethrin while LmCSP4 exhibited weaker interaction to deltamethrin. These results suggested that locust cuticle LmCSP3 and LmCSP4 contributed to the deltamethrin tolerance by binding the insecticides within the cuticular matrix. Our findings elucidate the association between CSPs and deltamethrin tolerance in L. migratoria , providing critical insights for targeted resistance management strategies.

Preparation and adsorption selectivity of deltamethrin molecularly imprinted polymers by two-step seed swelling method

Lei Xie,Liumei Zhou,Lu Li,Xinan Xie,Yan Li

Deltamethrin (DM) is one of the most efficient pyrethroid insecticides, which is widely applied to the control of insect pests in tea. In this work, efficient molecularly imprinted polymers (MIPs) were synthesized using DM as the template and acrylamide as the functional monomer, in conjunction with two-step seed swelling polymerization. Scanning electron microscopy, Fourier transform infrared spectroscopy, and Brunauer–Emmett–Teller surface area analysis confirmed the staggered pore structure and the presence of binding site in DM MIPs. The adsorption properties of the DM MIPs were also investigated based on assessing equilibrium adsorption as well as kinetic modeling and solid-phase extraction. Isothermal equilibrium adsorption experiments show that the adsorption behavior is consistent with the Freundlich and Halsey models, indicating heterogeneous multilayer specific adsorption. Fitting of the kinetic data demonstrates that chemical adsorption could be the rate-limiting step in DM extraction, which is consistent with a pseudo-second-order kinetic model. Using the imprinted polymers as solid-phase extraction filler, the DM recoveries from various teas were greater than 80% and exceeded those of the structural analogs cypermethrin, lambda-cyhalothrin, and bifenthrin. These results confirm that the MIPs exhibit specific adsorption of this analyte. © 2018 Wiley Periodicals, Inc. J. Appl. Polym. Sci. 2019 , 136 , 47415.
