Impact of Frying on Changes in Clam (Ruditapes philippinarum) Lipids and Frying Oils: Compositional Changes and Oxidative Deterioration

Zhong-Yuan Liu,Da-Yong Zhou,Kanyasiri Rakariyatham,Hong-Kai Xie,De-Yang Li,Bei-Wei Zhu,Fereidoon Shahidi

The current study shows the compositional changes and oxidation development of clam ( Ruditapes philippinarum , R. philippinarum ) lipids and frying oils when subjected to different processing conditions. Parameters measured include acid value, peroxide value (POV), thiobarbituric acid-reactive substances (TBARS), total oxidation (TOTOX), lipid classes, fatty acid composition, phosphatidylcholine (PC), and phosphatidylethanolamine (PE) contents together with major glycerophospholipid (GP) molecular species. Deep-fat frying increased triacylglycerol (TAG) content and decreased the contents of PC, PE, and GP molecular species in clam in a time-dependent manner. Meanwhile, minor amounts of free fatty acids, diacylglycerols, monoacylglycerols, and polar lipids were detected in frying oils, indicating lipid migration between the clam and frying oils. The time-dependent increase of POV, TBARS, and TOTOX in fried clams and frying oils with concurrent reduction of docosahexenoic acid and eicosapentaenoic acid indicates extensive oxidative degradation of clam lipids. Moreover, the moisture-rich clam aggravated the deterioration of frying oils. Consequently, deep-fat frying significantly altered the lipid profile and decreased the nutritional value of clams.

Rapid and non-destructive identification of the leather artifacts using FTIR-PCA

Mingrui Zhang,Zonghuan Ba,Jie Liu,Yong Lei,Mǎdǎlina Georgiana Albu Kaya,Keyong Tang

Due to the variations in deterioration and the diversity of tanning processes, the identification of leather artifacts is a challenge. In the present work, a non-destructive and rapid identification method is provided by combining Fourier transform infrared spectroscopy (FTIR) with principal component analysis (PCA). By analyzing a total of 4 types, encompassing with 30 model sets, this method has been proven effective in eliminating interference related to the deterioration and overcoming key obstacles in identifying the leather manufacturing process of the leather artifacts. Two pieces of leather artifacts representing different periods, regions, and animal types were studied using the proposed method. Further high-performance liquid chromatography (HPLC) was performed to verify the identification results and method accuracy. In addition, by evaluating against the models, the alkaline treated sample showed higher reliability compared to the acid treated one. This method effectively addressed the challenges in accurately identifying the leather tanning processes and demonstrated the importance of analyzing and conserving a wider range collagen-based artifacts.

Rapid evaluation of mold deterioration in maize using GC-IMS

Mengyi Liu,Yan Zhao,Yating Xiong,Zixuan Wu,Yucan Qin,Ying Xin,Haoxin Lv

Maize is highly susceptible to mildew during storage due to its rich nutrient content and hygroscopicity, posing significant risks to food safety and quality. This study employed gas chromatography-ion mobility spectrometry (GC-IMS) for the non-destructive detection and early identification of volatile organic compounds (VOCs) in stored maize. Six key biomarkers-propan-1-ol, hexanal, propanal, nonanal, butan-2-one, and isobutanol-were identified as reliable indicators of initial mold development. By integrating VOCs fingerprinting with principal component analysis (PCA) and heatmap clustering, maize samples were categorized into four distinct stages of mold severity. Finally, the GC-IMS classification method was verified by traditional methods, which showed a high degree of consistency with the traditional storage quality parameters, including fatty acid value and total plate count. These results confirm that GC-IMS is an effective tool for the rapid, on-site monitoring of maize storage conditions, enabling timely interventions to reduce postharvest losses and strengthen food safety management.

Fungal deterioration of the bagasse storage from the harvested sugarcane

Peng Na,Yao Ziting,Wang Ziting,Huang Jiangfeng,Khan Muhammad Tahir,Chen Baoshan,Zhang Muqing

Background Sugarcane is an essential crop for sugar and ethanol production. Immediate processing of sugarcane is necessary after harvested because of rapid sucrose losses and deterioration of stalks. This study was conducted to fill the knowledge gap regarding the exploration of fungal communities in harvested deteriorating sugarcane. Experiments were performed on simulating production at 30 °C and 40 °C after 0, 12, and 60 h of sugarcane harvesting and powder-processing. Results Both pH and sucrose content declined significantly within 12 h. Fungal taxa were unraveled using ITS amplicon sequencing. With the increasing temperature, the diversity of the fungal community decreased over time. The fungal community structure significantly changed within 12 h of bagasse storage. Before stored, the dominant genus (species) in bagasse was Wickerhamomyces ( W. anomalus ). Following storage, Kazachstania ( K. humilis ) and Saccharomyces ( S. cerevisiae ) gradually grew, becoming abundant fungi at 30 °C and 40 °C. The bagasse at different temperatures had a similar pattern after storage for the same intervals, indicating that the temperature was the primary cause for the variation of core features. Moreover, most of the top fungal genera were significantly correlated with environmental factors (pH and sucrose of sugarcane, storage time, and temperature). In addition, the impact of dominant fungal species isolated from the deteriorating sugarcane on sucrose content and pH in the stored sugarcane juice was verified. Conclusions The study highlighted the importance of timeliness to refine sugar as soon as possible after harvesting the sugarcane. The lessons learned from this research are vital for sugarcane growers and the sugar industry for minimizing post-harvest losses.

Oral isoniazid causes oxidative stress, oocyte deterioration and infertility in mice

Peipei Qiao,Yingbing Zhang,Ying Yang,Ru Meng,Zhiming Xu,Xianlei Jiang,Yong Zhang,Chengtu Zhang,Jianmin Su

Isoniazid (INH), a synthetic first-line tuberculosis antibiotic, has been widely used in clinical treatment. It has been reported to cause toxic effects at multiple tissue sites and also increases the incidence of adverse pregnancy outcomes; but the mechanism of action of INH on the reproductive system of female mammals remains unclear. Here, we demonstrate that oral INH (40 mg/kg/day every other day for 28 days) severely affects oocyte maturation and fertilization, late blastocyst development and fertility. We found that INH could disrupt standard spindle assembly, chromosome arrangement, and actin filament dynamics, which compromised meiotic progression of mouse oocytes. INH treatment increased the level of reactive oxygen species (ROS) and activated the oxidative stress response pathway, Keap1-Nrf2. It also caused apoptosis of oocytes and mitochondrial dysfunction. Our findings demonstrate that oral INH reduces fertility and damages the mammalian reproductive system by altering cytoskeletal dynamics and Juno expression, inducing oxidative stress and apoptosis, and activating the Keap1-Nrf2 signaling pathway in mouse oocytes.

A nanodrug combining CD47 and sonodynamic therapy efficiently inhibits osteosarcoma deterioration

Ming Gong,Yufeng Huang,Huixiong Feng,Jiaming Lin,Anfei Huang,Jinxin Hu,Qinglian Tang,Xiaojun Zhu,Shisong Han,Jinchang Lu,Jin Wang

Treatments for osteosarcoma (OS) with pulmonary metastases reach a bottleneck with a survival rate of 10–20%. The suppressive tumor associated macrophages(TAMs) and CD47 over-expression greatly lead to the treatment failure. Sonodynamic therapy (SDT) can generate ROS with deep tumor penetration to induce tumor cell apoptosis , which is reported to further induce M1 macrophage polarization. CD47 inhibition combined with SDT to synergistically modulate TAMs may induce superior effects for OS treatment. In this work, for the first time, a biomimetic nanodrug named MPIRx was deveploped by loading IR780 (a sonosensitizer) and RRx-001 (a CD47 inhibitor) in PEG-PCL nanomicelles and then coating with OS cell membranes . After ultrasound activation, the nanodrug significantly inhibited OS proliferation and migration, induced apoptosis and immunogenic cell death in OS cells. Furthermore, MPIRx could guide macrophage migrating towards tumor cells and promote M1-type polarization while increasing the phagocytosis activity of macrophages on OS cells. Ultimately, MPIRx showed good tumor accumulation in vivo and successfully inhibited subcutaneous OS and orthotopic tumor with deterioration of pulmonary metastasis. Overall, by creating a local oxidative microenvironment and modulating the TAMs/CD47 in tumor tissue, the MPIRx nanodrug presents a novel strategy for macrophage-related immunotherapy to successfully eliminate OS and inhibit the intractable pulmonary metastasis.

Detection of Quality Deterioration of Packaged Raw Beef Based on Hyperspectral Technology

Cheng Wu,Yingjie Feng,Jiarui Cui,Zhang Yao,Hailong Xu,Songlei Wang

It is an important measure to ensure food quality and safety that real-time monitoring of the key quality indicators of fresh meat after packaging in the process of storage and transportation. The feasibility of combining hyperspectral imaging (HSI) technology with chemometrics and deep learning to detect the quality deterioration of polyethylene (PE)-packaged raw beef, especially for a key lipid oxidation indicator of malondialdehyde (MDA) content, was explored in this study. The feasibility of filtering to overcome the interference of packaging film on the spectral data was further investigated. Near-infrared HSI (400–1000 nm) was used to collect spectral and spatial data of beef samples during short-term storage. A least squares regression (PLSR) and echo-neural network optimized by vulture optimization algorithms (BES-ESN) models were developed by multivariate data processing methods. The results showed that the performance of models established by PE-packed beef samples was usually inferior to that established by unpacked beef samples. The changes of MDA content in beef were visualized according to the optimal model. In addition, Gaussian filtering was applied to reduce the interference effect caused by the packaging film. The results have demonstrated that HSI combined with Gaussian filter preprocessing and multivariate data processing provided an efficient and reliable tool for detecting the freshness of beef in PE packaging. The best model had a coefficient of determination ( R 2 P ) of 0.8309 and a root mean squared error of prediction (RMSEP) of 0.2180, demonstrating the potential of hyperspectral techniques for real-time monitoring of packaged raw meat quality. The findings can provide some references for the meat industry to develop advanced non-invasive quality assurance systems in the meat industry.

Rapid microchip-based FAIMS determination of trimethylamine, an indicator of pork deterioration

Qi Wang,Yun-Fei Xie,Wei-Jun Zhao,Peng Li,He Qian,Xiao-Zhi Wang

A fast and quantitative method for the detection of biogenic amines based on the microchip-based high-field asymmetric waveform ion mobility spectrometry (FAIMS) technique was established for evaluating the degree of spoilage of pork stored at room temperature. Trimethylamine (TMA) was selected as a good indicator for its volatility and high proton affinity. Analysis of standard TMA showed the same characteristic peak with the pork sample. The limit of detection of the system for TMA dissolved in deionized water, with 100 μL of 8 mol L−1 NaOH, was 3 ng and the measurement was accomplished within 15 s. The relative standard deviation (RSD) was 6.20%. The delivery of the sample gas was based on the dynamic headspace method. The effect of the storage time on the formation of biogenic amines was examined, and not surprisingly, the content of TMA increases over time. The total volatile basic nitrogen (TVB-N) was measured simultaneously based on the classical semi-micro Kjeldahl method. A good correlation coefficient between these two methods was obtained, the correlation coefficient was 0.952. Therefore the microchip-based FAIMS technique provides a quantitative classification of the spoilage or freshness of pork, especially when a rapid detection is needed.

Perfluorodecanoic acid induces meiotic defects and deterioration of mice oocytes in vitro

Shu-Zi Deng,Chang-Long Xu,Zhong-Feng Xu,Li-Ying Zhou,Shu-Juan Xie,Kang-Na Wei,Yuan-Chang Jin,Zhao-Cheng Zeng,Xiang-Jun Yang,Shu-Hua Tan,Hai-Long Wang

Perfluorodecanoic acid (PFDA) is a member of the perfluoroalkyl substances, which are toxic to organic functions. Recently, it has been found in follicular fluid, seriously interfering with reproduction. Follicular fluid provides the oocyte with necessary resources during the process of oocytes maturation. However, the effects of PFDA on the oocyte need investigation. Our study evaluated the impacts of PFDA on the meiosis and development potential of mouse oocytes by exposing oocytes to PFDA in vitro at 350, 400, and 450 μM concentrations. The results showed that exposure to PFDA resulted in the first meiotic prophase arrest by obstructing the function of the maturation-promoting factor. It also induced the dysfunction of the spindle assembly checkpoint, expedited the progression of the first meiotic process, and increased the risk of aneuploidy. The oocytes treated with PFDA had a broken cytoskeleton which also contributed to meiotic maturation failure. Besides, PFDA exposure caused mitochondria defections, increased the reactive oxygen species level in oocytes, and consequently induced oocyte apoptosis. Moreover, PFDA produced epigenetic modifications in oocytes and increased the frequency of mature oocytes with declined development potential. In summary, our data indicated that PFDA disturbs the meiotic process and induces oocyte quality deterioration.

Autophagy-mediated energy charge sustainability alleviated postharvest quality deterioration of strawberry (Fragaria × ananassa)

Yicheng Ren,Quan Ma,Dong Li,Yanpei Chen,Qinyang Cheng,Zisheng Luo

Fluctuations in energy status are critical physiological factors influencing postharvest fruit quality. However, the role of autophagy, a stress-resistant biological process, in regulating postharvest fruit energy status remains unclear. In this study, we treated strawberry fruit with the autophagy inhibitor hydroxychloroquine (HCQ) to investigate the role of autophagy in maintaining energy status. The results demonstrated that HCQ treatment inhibited autophagosome formation, reducing relative autophagic activity by 73 % on day 6. HCQ treatment also reduced the energy charge and the NADH/NAD + ratio by 5 % and 61 %, respectively, on day 2. Corresponding to the decline in energy status, HCQ treatment led to a reduction in cytochrome C oxidase (CCO) and succinate dehydrogenase (SDH) activities by 47.48 % and 56.34 %, respectively, on day 3. Additionally, fluctuations in energy charge led to abnormalities in ripening and decreased stress resistance in autophagy-inhibited fruit. In conclusion, this study provides evidence that autophagy-mediated energy charge sustainability mitigates postharvest quality deterioration in fruit.

Effect of Ferrous Ion on Heat-Induced Aroma Deterioration of Green Tea Infusion

Ying Gao,Jie-Qiong Wang,Jian-Xin Chen,Fang Wang,Gen-Sheng Chen,Jun-Feng Yin,Yong-Quan Xu

Aroma deterioration is one of the biggest problems in processing tea beverages. The aroma of tea infusion deteriorates fast during heat sterilization and the presence of ferrous ion (Fe2+) aggravates it. The underlying mechanism remains unveiled. In this study, Fe2+was verified to deteriorate the aroma quality of green tea infusion with heat treatment. Catechins were necessary for Fe2+-mediated aroma deterioration. By enhancing the degradation of catechins, Fe2+dramatically increased the production of hydrogen peroxide (H2O2). Fe2+and H2O2together exacerbated the aroma of green tea infusion with heat treatment. GC-MS analysis revealed that the presence of Fe2+enhanced the loss of green/grassy volatiles and promoted the formation of new volatiles with diversified aroma characteristics, resulting in a dull scent of green tea infusion. Our results revealed how Fe2+induced aroma deterioration of green tea infusion with heat treatment and could help guide tea producers in attenuating the aroma deterioration of tea infusion during processing. Keywords:green tea infusion;aroma deterioration;ferrous ion;catechins;hydrogen peroxide

Preparation of aldehyde-recognition nanofiber label for monitoring oxidative deterioration in processed meat products

Mengjie Li,Chong Sun,Shenjun Chen,Huiwen Liu,Juan Xu,Yongzhi Zhu,Daoying Wang

In this study, an oxidation label for the quantitative identification of aldehydes was developed using polyvinyl alcohol (PVA) as the carrier polymer, blueberry anthocyanin (BA) as the chromogenic agent, and hydroxylamine sulfate (HAS) as the coupling agent. Electrospinning technology was employed in this process. Based on aldehyde-specific recognition, the PVA-BA-HAS oxidation label displayed noticeable changes from white to rose pink corresponding to varying degrees of oxidation in processed meat products, with a detection time of less than 1 min. The label showed exceptional morphology and performance, including hydrophobicity, thermal stability, and biocompatibility, etc. Moreover, the assessment of oxidation levels obtained from the PVA-BA-HAS labels were in good agreement with the data measured through POV analysis, confirming practicability of the oxidation label. These results indicated that the prepared oxidation label could be utilized for real-time monitoring of oxidation levels with high accuracy and efficiency, expanding new possibilities for meat quality grading.
