Comparative toxicity and bioaccumulation of fenvalerate and esfenvalerate to earthworm Eisenia fetida

Xiaoqing Ye,Kang Xiong,Jing Liu

More attention is being paid to the enantioselective toxicity of chiral pesticides. However, limited investigations have been done to assess the ecological risks of chiral pesticides to soil community. Fenvalerate (FV), an extensively used synthetic pyrethroid, is a typical chiral pesticide. The most insecticidally active enantiomer of FV, esfenvalerate (ESFV), also has been marketed and widely used. In this study, the toxicological sensitivity and bioaccumulation of FV and ESFV in earthworms were assessed. The results showed that FV was less toxic than ESFV, but more accumulated in earthworms. ESFV was at least 4 times more toxic to earthworms than FV according to the filter paper contact toxicity test and the artificial soil test. Enantiospecific induction in oxidative stress was observed in earthworms exposed to FV and ESFV. The bioaccumulation of FV and ESFV in earthworm tissues was also enantioselective, preferentially accumulating FV. The uptake of ESFV by earthworms was lower than that of FV, so that the biota to soil accumulation factor (BSAF) value of ESFV was lower than that of FV. Our findings suggest that the enantioselective toxicity and bioaccumulation of chiral pesticides should be considered for evaluating ecological risks of these compounds to non-target organisms.

Two single mutations in carboxylesterase 001C improve fenvalerate hydrolase activity in Helicoverpa armigera

Jing-jing Xu,Yong-mei Chang,Mei Lu,Yuan Tie,Yan-ling Dong,Guang-you Chen,Zhi-qing Ma,Xi-li Liu,Yong-qiang Li

Carboxylesterases (CarEs) usually play critical roles in the detoxification of toxic chemicals and therefore may be involved in insecticide resistance in agricultural pests. Previous work has shown that CarE 001C from Helicoverpa armigera was able to metabolize the isomers of cypermethrin and fenvalerate . In this study, seven mutants of CarE 001C with single amino acid substitution were produced and expressed in the Escherichia coli. Enzyme kinetic analysis indicated that all seven mutations dramatically reduced enzymatic activities toward the generic substrate α -naphthyl acetate, but in vitro metabolism assay showed that two of the mutations, H423I and R322L, significantly improved hydrolase activities toward fenvalerate, with their recorded specific activities being 3.5 and 5.1 nM·s −1 ·mg −1 proteins, respectively. Further, thermostability assay showed that the stability of one mutant enzyme was enhanced. This study will help us better understand the potential of CarEs in insecticide detoxification and resistance in H. armigera .

Ultrasensitive aptasensor based on SERS-coupled immunochromatography for rapid detection of fenvalerate in food samples

Wangyu Nan,Qingdan Ye,Jie huang,Pei Liang,Qianmin Dong,Kun Pang,Zihong Ye,Kai Sun,Yufeng Wang

In this study, we developed an aptamer-mediated surface-enhanced Raman scattering (SERS)-coupled immunochromatographic assay (ICA) for the quantitative detection of pyrethroids. For this approach, core-shell gold nanoparticles (Au@Au NPs) were used as SERS probes by conjugating with aptamers, and fluorescent Raman reporter molecules (MPBN) were incorporated to yield stable, reproducible spectral signals. Leveraging the aptamers' specific recognition of the target analyte, Raman-labeled Au@Au NPs could be effectively captured by the test line of the immunochromatographic strip, thereby enabling the quantitative analysis of fenvalerate-with a limit of detection (LOD) as low as 0.4 ppb. Comparative experiments revealed that the detection sensitivity of this SERS-ICA method was 476 times higher than that of conventional lateral flow assays (LFA), showcasing a remarkable sensitivity advantage. To validate its practicality, spiked recovery experiments were conducted on three real samples, namely welsh onions, cowpeas, and kidney beans. The results showed that the spiked recoveries ranged from 78.3% to 112.5%, with relative standard deviations (RSD) between 1.7% and 20.1%. This fully confirms that the method possesses favorable specificity and accuracy, making it suitable for real-sample detection. This detection platform offers a new technical route for the rapid on-site monitoring of trace contaminants in food safety and environmental surveillance, holding broad application prospects.

Identification of cis-acting elements in response to fenvalerate in the CYP6B7 promoter of Helicoverpa armigera

Yun Huang,Peizhuo Wu,Junyue Zheng,Lihong Qiu

Cytochrome P450-mediated detoxification plays an important role in the development of insecticide resistance . Previous studies have shown that cytochrome P450 CYP6B7 was induced by fenvalerate and involved in fenvalerate detoxification in Helicoverpa armigera . However, the transcriptional regulation of CYP6B7 induced by fenvalerate remains unclear. Here, a series of progressive 5′ deletions of CYP6B7 promoter reporter genes were constructed, and the relative luciferase activities were detected. The results revealed that the relative luciferase activity of plasmid p (−655/−1) was significantly induced by fenvalerate. Further deletion of the region between −655 and −486 bp showed that the highest luciferase activity induced by fenvalerate was observed in plasmid p (−528/−1), while p (−485/−1) had the lowest fenvalerate-induced luciferase activity. Moreover, internal deletion and mutation in the region between −508 and −486 bp resulted in a significant reduction in fenvalerate-induced CYP6B7 promoter activity, suggesting that the cis -acting element responsible for fenvalerate in the CYP6B7 promoter was located between −508 and −486 bp. These results promote an understanding of the expression regulation mechanism of P450 genes that conferring resistance to insecticides .

Atmospheric Cold Plasma Degradation of Fenvalerate Residues on Shiitake Mushrooms: Mechanisms, Toxicity Evolution, and Quality Effects

Hu Shi,Ziwen Cheng,Shiwei Dong,Yang Jiao,Hongru Liu

Fenvalerate residues on edible mushrooms pose significant risks to food safety and aquatic ecosystems. This study investigated the efficiency, degradation mechanisms, toxicity evolution, and quality effects of atmospheric cold plasma (ACP) for removing fenvalerate from shiitake mushrooms. Fenvalerate degradation increased with ACP treatment voltage and exposure time, reaching a maximum efficiency of 82.5% at 80 kV for 15 min. Quantum chemical calculations based on Fukui functions and frontier molecular orbitals identified phenoxy and chlorophenyl moieties as primary reactive sites. High-performance liquid chromatography–tandem mass spectrometry revealed degradation pathways dominated by hydroxylation, ester bond cleavage, and oxidative transformations. Toxicity assessment using ECOSAR predictions and yeast bioassays demonstrated substantial reductions in acute and chronic toxicity by ACP treatment, although some intermediates retained residual toxicity. In addition, ACP preserved mushroom quality during refrigerated storage. Overall, ACP represents a promising non-thermal strategy for pesticide detoxification while preserving edible mushroom quality.

Fenvalerate toxicity in Haliotis discus hannai: Gill damage and organ responses under food and water exposure

Xiaohan Liu,Guiqing Wu,Shaomeng Hua,Huawei Qin,Xuan Li,Zhenman Xie,Xiangquan Liu,Lizhu Chen,Dinglong Yang,Jianmin Zhao

Fenvalerate (FEN), a widely used synthetic pyrethroid pesticide that poses substantial risks to aquatic ecosystems. This study systematically compared the toxic effects of FEN exposure through food and water pathways in the abalone Haliotis discus hannai , with specific focus on the gills. Our results demonstrated detectable FEN accumulation in the gills under both exposure routes, and water exposure led to significantly higher accumulation, particularly at higher concentrations. Proteomic analysis revealed that both exposure routes induced substantial molecular alterations, with food exposure primarily disrupting Metabolic processes, while water exposure caused more extensive disruptions, including Genetic Information Processing pathways. Both exposure routes induced oxidative stress in the gills, evidenced by significantly elevated superoxide dismutase (SOD) and glutathione S-transferase (GST) activities and increased malondialdehyde (MDA) levels, though acetylcholinesterase (AChE) activity remained unchanged across all groups. Molecular analysis showed that both food and water exposure upregulated genes related to antioxidant defense (Nuclear factor erythroid 2-related factor 2 , Nrf2 ), apoptosis (BCL2-Associated X Protein, BAX ; Cysteine-Aspartic Acid Protease 3, Caspase-3 ), immune inflammation (Toll-Like Receptor 2, TLR2 ; Toll-Like Receptor 4, TLR4 ; Tumor Necrosis Factor Alpha, TNF-α ), and cellular signaling (Phosphatidylinositol 3-Kinase, PI3K ), while downregulating the anti-apoptotic gene (B-cell Lymphoma 2, Bcl-2 ), with these effects being most pronounced under water exposure. Integrated biomarker response (IBR) and principal component analysis (PCA) consistently demonstrated that both exposure routes disrupted gill physiological homeostasis, with water exposure causing the most severe toxicological effects. These findings establish that while food exposure induces significant toxicity in abalone gills, water exposure represents the predominant risk factor through interconnected oxidative, immune, and apoptotic pathways.

Synergistic Enhancement Effect of Ag/rGO as SERS Platform for Capture and Trace Detection of Fenvalerate Molecules

Minghui Yu,Chongyang Qin,Zhi Yu,Biao Sun,Dejiang Ni,De Zhang,Pei Liang

Surface-enhanced Raman scattering (SERS) provides an alternative rapid detection method for pesticide residues in food, but fenvalerate possesses poor affinity to the novel metal substrate, thus restricting its analysis. To break this bottleneck, a SERS-active platform with an Ag/rGO composite structure was engineered using a facile method for fenvalerate detection. Ag nanoparticles with a 60 nm diameter can grow evenly on the top and bottom of rGO layers under intense ultrasonic oscillation, and rGO in hybrid material acts as an ideal hotspot holder between the gaps of Ag nanoparticles, not only allowing the interaction area to be enhanced both electromagnetically and chemically but also enabling the capture and enrichment of fenvalerate pesticide molecules into the “hotspot” area to improve detection sensitivity. Ag/rGO composite substrate possesses superior SERS performance with an ultralow detectable concentration of 4-aminothiophenol (10−10M) and good reproducibility, endowing the material with a better enhancement effect than pure Ag nanoparticles. When used as the SERS substrate for fenvalerate detection, Ag/rGO composite material showed excellent performance in both experiments and theoretical calculation, with the limit of detection (LOD) of fenvalerate being as low as 1.69 × 10−5mg/kg and a detection model with an R2of 99.2%, demonstrating its exciting potential as a SERS substrate for pesticides detection.

Identification and characterization of both cis- and trans-regulators mediating fenvalerate-induced expression of CYP6B7 in Helicoverpa armigera

Yun Huang,Li Xu,Junyue Zheng,Peizhuo Wu,Yu Zhang,Lihong Qiu

As we known, inducibility is an important feature of P450 genes. Previous studies indicated that CYP6B7 could be induced and involved in fenvalerate detoxification in Helicoverpa armigera . However, the regulatory mechanism of CYP6B7 induced by fenvalerate is still unclear. In this study, CYP6B7 promoter of H. armigera was cloned and the cis -acting element of fenvalerate was identified to be located between −84 and − 55 bp of CYP6B7 promoter. Subsequently, 33 candidate transcription factors ( CYP6B7 -fenvalerate association proteins, CAPs) that may bind to the cis -acting element were screened and verified by yeast one-hybrid. Among them, the expression levels of several CAPs were significantly induced by fenvalerate. Knockdown of juvenile hormone-binding protein-like (JHBP), RNA polymerase II-associated protein 3 (RPAP3), fatty acid synthase-like (FAS) and peptidoglycan recognition protein LB-like (PGRP) resulted in significant expression inhibition of CYP6B7 , and increased sensitivity of H. armigera to fenvalerate. Co-transfection of reporter gene p (−84/−55) with pFast-CAP showed that JHBP, RPAP3 and PGRP could significantly increase the activity of CYP6B7 promoter. These results suggested that trans -acting factors JHBP, RPAP3 and PGRP may bind with cis -acting elements to regulate the expression of CYP6B7 induced by fenvalerate, and play an important role in the detoxification of H. armigera to fenvalerate.

Identification of insect cuticular protein genes LCP17 and SgAbd5 from Helicoverpa armigera and evaluation their roles in fenvalerate resistance

Junyue Zheng,Peizhuo Wu,Yun Huang,Yu Zhang,Lihong Qiu

Insect cuticular protein (ICP) plays an important role in insect growth and development . However, research on the role of ICP in insecticide resistance is very limited. In this study, insect cuticular protein genes LCP17 and SgAbd5 were cloned and characterized in Helicoverpa armigera based on previous transcriptome data. The functions of LCP17 and SgAbd5 genes in fenvalerate resistance were assessed by RNA interference (RNAi), and their response to fenvalerate was further detected. The results showed that LCP17 and SgAbd5 were overexpressed in the fenvalerate-resistant strain comparing with a susceptible strain. The open reading frames of LCP17 and SgAbd5 genes were 423 bp and 369 bp, encoding 141 and 123 amino acids , respectively. LCP17 and SgAbd5 genes were highly expressed in the larval stage, but less expressed in the adult and pupal stages. The expression level of LCP17 and SgAbd5 genes increased significantly after fenvalerate treatment at 24 h. When the cotton bollworms larvae were exposed to fenvalerate at LD 50 level, RNAi-mediated silencing of LCP17 and SgAbd5 genes increased the mortality from 50.68% to 68.67% and 63.89%, respectively; the mortality increased to even higher level, which was 73.61%, when these two genes were co-silenced. Moreover, silencing of these two genes caused the cuticle lamellar structure to become loose, which led to increased penetration of fenvalerate into the larvae. The results suggested that LCP17 and SgAbd5 may be involved in the resistance of cotton bollworm to fenvalerate, and LCP17 and SgAbd5 could serve as potential targets for H. armigera control.

Functional study on candidate regulators mediating the expression of CYP6B7 induced by fenvalerate in a susceptible strain of Helicoverpa armigera

Yun Huang,Junyue Zheng,Peizhuo Wu,Yu Zhang,Lihong Qiu

Transcription factors play an important role in regulating the expression of detoxification genes (e.g. P450s ) that confer insecticide resistance. Our previous study identified a series of candidate transcription factors ( CYP6B7 -fenvalerate association proteins, CAPs) that may be related to fenvalerate-induced expression of CYP6B7 in a field HDTJ strain of H. armigera . Whether these CAPs can mediate the transcript of CYP6B7 induced by fenvalerate in a susceptible HDS strain of H. armigera remains unknown. Further study showed that the expression levels of multiple CAPs were significantly induced by fenvalerate in HDS strain. Knockdown of CAP19 [fatty acid synthase-like (FAS)], CAP22 [polysaccharide biosynthesis domain-containing protein 1 (PBDC1)], CAP24 [5-formyltetrahydrofolate cycloligase (5-FCL)], CAP30 [peptidoglycan recognition protein LB-like (PGRP)] and CAP33 [NADH dehydrogenase [ubiquinone] 1 alpha subcomplex subunit 11 (NDUFA11)] resulted in significant inhibition of CYP6B7 and some other P450 genes expression; meanwhile, the sensitivity of HDS strain larvae to fenvalerate was significantly increased. In addition, PBDC1, PGRP and NDUFA11, either alone or in combination, could significantly enhance the activity of CYP6B7 promoter in HDS strain, as well as the expression level of CYP6B7 gene in Sf9 cells line. These results suggested that PBDC1, PGRP and NDUFA11 may be involved in the transcript regulation of key detoxifying genes in response to fenvalerate in HDS strain of H. armigera .

A fluorescent molecularly imprinted polymer sensor synthesized by atom transfer radical precipitation polymerization for determination of ultra trace fenvalerate in the environment

Wenjuan Han,Lin Gao,Xiuying Li,Liang Wang,Yongsheng Yan,Guangbo Che,Bo Hu,Xue Lin,Minshan Song

In this study, novel fluorescence molecularly imprinted polymers (FMIPs) were prepared via atom transfer radical precipitation polymerization (ATRPP) for the optical detection of trace fenvalerate (FE). The FMIPs were characterized by FT-IR, SEM, LCSM, which demonstrated that the composite material exhibited a regular morphology and excellent monodispersity. The FMIPs showed high fluorescence intensity and highly selective recognition. Utilizing fluorescence quenching as a detecting tool, FMIPs were successfully used as a sensor to selectively and sensitively detect FE with a good linear relationship and a correlation coefficient of 0.9946 by the Stern–Volmer equation was obtained. Moreover, the experimental result also indicated good reproducibility via regeneration experiments and FMIPs as a type of fluorescent recognition sensor for FE was satisfactorily applied for environmental protection. Overall, this study demonstrated a good potential of FMIPs for the detection of FE in the environment.

A novel microfluidic origami photoelectrochemical sensor based on CdTe quantum dots modified molecularly imprinted polymer and its highly selective detection of S-fenvalerate

Yanhu Wang,Dejin Zang,Shenguang Ge,Lei Ge,Jinghua Yu,Mei Yan

Driven by the urgent demand of high-selectively point-of-care testing device for pesticide, molecular imprinting-photoelectrochemistry (MI-PEC) was introduced into microfluidic paper-based analytical strategy to design a novel paper-based photoelectrochemical (paper-based PEC) protocol. The MI-PEC strategy was constructed based on CdTe quantum dots dotted molecular imprinted polymers (CdTe QDs@MIPs), and triggered by a common ultraviolet lamp (∼365   nm, 50$). The paper-based PEC sensor was fabricated by immobilizing CdTe QDs@MIPs on paper-based screen-printed working electrodes (WEs) via gold nanoparticles (Au NPs), which was electrodeposited on the surface of WE to improve the electron transfer efficiency for high sensitivity. Using S-fenvalerate as model analyte, the produced photocurrent from the proposed paper-based MI-PEC sensor upon ultraviolet radiation decreased with the increasing concentrations of S-fenvalerate solution, and the quenched paper-based MI-PEC showed a low detection limit of 3.2   ×   10 −9   mol   L −1 . This study has made a successful attempt in the development of highly selective and sensitive photoelectrochemical sensor for S-fenvalerate monitoring.
