Zn2+ enhanced the anti-breast cancer activity of fucoidan based nanoparticles by activating the cGAS-STING pathway

Pengyu Xue,Yinghan Wang,Xuefei Wang,Shilin Zhou,Xiaochen Wu,Lin Long,Chuanlong Guo

The incidence rate of breast cancer remains high, and lung metastasis often leads to treatment failure. Immunotherapy based on the cyclic guanosine monophosphate synthesis (cGAS) − interface gene stimulatory factor (STING) pathway is expected to bring benefits to the treatment of breast cancer. This study provided a delivery system based on fucoidan (Fu) and polyvinylpyrrolidone (PVP) for the potential molecule resveratrol (Res). The study confirmed that Fu and PVP were assembled into nanoparticles through hydrogen bonding and effectively loaded Res (Fu/PVP/Res). Interestingly, the adsorption of Zn 2+ (Fu/PVP/Res Zn 2+ ) was expected to provide impetus for the treatment of breast cancer. The results in vitro confirmed that Fu/PVP/Res Zn 2+ significantly inhibited the proliferation of breast cancer 4T1 cells, induced the accumulation of mitochondrial reactive oxygen species (ROS) and the damage of mitochondrial membrane potential (MMP), and further induced the cytoplasmic release of mtDNA, which was the direct cause of the activation of cGAS-STING pathway. In vivo studies confirmed that Fu/PVP/Res Zn 2+ treatment inhibited the growth of 4T1 transplanted tumors. Excitingly, Fu/PVP/Res Zn 2+ treatment also prevented lung metastasis of in situ tumors. The in vivo mechanism confirmed that Fu/PVP/Res Zn 2+ treatment activated the cGAS-STING pathway and induced abnormal expression of tumor apoptosis factors such as Bax/Bcl-2. In conclusion, our research provided potential for the treatment of breast cancer.

Application of Fucoidan in Caco-2 Model Establishment

Qiong Yang,Maochen Xing,Ke Wang,Qiang Wei,Jiarui Zhao,Yuan Wang,Kai Ji,Shuliang Song

The Caco-2 model is a common cell model for material intestinal absorption in vitro, which usually takes 21 days to establish. Although some studies have shown that adding puromycin (PM) can shorten the model establishment period to 7 days, this still requires a long modeling time. Therefore, exploring a shorter modeling method can reduce the experimental costs and promote the development and application of the model. Fucoidan is an acidic polysaccharide with various biological activities. Our study showed that the transepithelial electrical resistance (TEER) value could reach 600 Ω·cm2on the fourth day after the addition of fucoidan and puromycin, which met the applicable standards of the model (>500 Ω). Moreover, the alkaline phosphatase (AKP) activity, fluorescein sodium transmittance, and cell morphology of this model all met the requirements of model establishment. Fucoidan did not affect the absorption of macromolecular proteins and drugs. The results indicate that fucoidan can be applied to establish the Caco-2 model and can shorten the model establishment period to 5 days.Keywords:fucoidan;Caco-2 model;absorption of macromolecules

Fucoidan restores age-associated intestinal homeostasis in aged mice

Xie Shufu,McClements David Julian,Hong Qing,Liu Zhenmin,Zhang Zipei,Qiu Chao,Zhao Jianwei,Jin Zhengyu,Chen Long

With global aging, intestinal homeostasis disruption contributes to inflammation, oxidative stress, and metabolic disorders. This study assessed fucoidan at doses of 100, 250 and 500 mg/kg in aged mice. The high dose (500 mg/kg) improved food intake, body weight and intestinal transit, and elevated α-amylase activity. It also relieved oxidative stress, strengthened intestinal tight junctions and restored mucosal morphology. Multi-omics analyses revealed that fucoidan reshaped the gut microbiome—enriching beneficial taxa (e.g., Akkermansia ) while suppressing opportunistic pathogens—and increased short-chain fatty acid production. Untargeted metabolomics indicated enrichment of antioxidant and barrier-protective metabolites and clearance of cytotoxic compounds, with key impacts on amino acid, lipid, and energy metabolism. Overall, fucoidan systematically restores aging intestinal homeostasis via a “microbiota–metabolite–host” axis, supporting its potential as an anti-aging functional food.

Emulsification properties of ovalbumin-fucoidan (OVA-FUC) binary complexes

Xiefei Li,Yingmei Wu,Wenshan Duan,Lei Chen,Lujie Cheng,Junmei Liu,Yan Zhou,Chao Ai,Xin Li,Qun Huang

The poor thermal stability and emulsifying properties of ovalbumin (OVA) limit its functional performance, but these limitations may be overcome by forming binary complexes. We prepared binary complexes of OVA and fucoidan (FUC) through electrostatic self-assembly and investigated the emulsifying properties of the complex by measuring the particle size, interfacial membrane thickness, zeta potential, and stability of the emulsion prepared with camellia oil and the complex. The OVA-FUC emulsions have a thicker interfacial membrane, lower mobility, higher viscosity, and better stability compared with the OVA emulsions. The emulsion prepared with 1.5 % OVA-FUC remained stable and homogeneous during storage. They tended to become unstable with freeze–thaw, but the oil encapsulated did not leak after coalescence occurred. With the addition of Ca 2+ , the OVA-FUC emulsion will be converted into a gel state. These findings indicate that OVA-FUC binary complexes can be used to prepare high-performance emulsions with great potential for development.

Study on Absorption Mechanism and Tissue Distribution of Fucoidan

Xu Bai,E Zhang,Bo Hu,Hao Liang,Shuliang Song,Aiguo Ji

Fucoidan exhibits several pharmacological activities and is characterized by high safety and the absence of toxic side effects. However, the absorption of fucoidan is not well-characterized. In the present study, fucoidan were labeled with fluorescein isothiocyanate (FITC) and their ability to traverse a monolayer of Caco-2 cells was examined. The apparent permeability coefficients (Papp × 10−6) of FITC-labeled fucoidan (FITC-fucoidan) were 26.23, 20.15, 17.93, 16.11 cm/sec, respectively, at the concentration of 10 μg/mL at 0.5, 1, 1.5 and 2 h. The absorption of FITC-fucoidan was suppressed by inhibitors of clathrin-mediated endocytosis, chlorpromazine, NH4Cl, and Dynasore; the inhibition rates were 84.24%, 74.61%, and 63.94%, respectively. This finding suggested that clathrin-mediated endocytosis was involved in fucoidan transport. Finally, tissue distribution of FITC-fucoidan was studied in vivo after injection of 50 mg/kg body weight into the tail vein of mice. The results showed that FITC-fucoidan targeted kidney and liver, reaching concentrations of 1092.31 and 284.27 μg/g respectively after 0.5 h. In summary, the present work identified the mechanism of absorption of fucoidan and documented its tissue distribution, providing a theoretical basis for the future development of fucoidan applications. Keywords:fucoidan;fluorescent labeling;clathrin;absorption;tissue distribution.

Fucoidan ameliorates diabetic skeletal muscle atrophy through PI3K/Akt pathway

Caixia Li,Yaping Liu,Mingzhi Yang,Haoyue Huang,Lulu Tang,Yufan Miao,Wenjie Li,Xing Li

It has been reported that fucoidan plays a beneficial role in type 2 diabetic mellitus (T2DM). However, it remains obscure if fucoidan has the protective effects on diabetes induced skeletal muscle atrophy. Here, we investigated the effect and mechanism of fucoidan on diabetic muscle atrophy. The results revealed that fucoidan ameliorated the general symptoms of diabetes, alleviated insulin resistance, as well as improved glucose metabolism disorders in vivo and vitro . Moreover, fucoidan also improved the atrophy of skeletal muscle by resisting the reduction of muscle fiber diameter and balance the protein anabolism and catabolism. It is worth noting that these beneficial effects of fucoidan on skeletal muscle atrophy and insulin resistance were abolished by inhibiting PI3K/Akt pathway. Taken together, fucoidan ameliorates diabetic skeletal muscle atrophy at least partially by the PI3K/Akt pathway.

Hypoxia responsive fucoidan-based micelles for oxidative stress-augmented chemotherapy

Li Li,Bingjie Wang,Qi Zhang,Panpan Song,Tianze Jiang,Xia Zhao

Enhancing tumor reactive oxygen species (ROS) is a promising strategy for improved cancer chemotherapy. However, the high level of glutathione (GSH) in hypoxic tumor cells can neutralize ROS, restricting the application of ROS boosted therapy. Herein, hypoxia-responsive polymer micelles ( [email protected] ) were constructed to amplify intra-tumor oxidative stress to enhance the chemotherapeutic efficacy of doxorubicin (DOX). The amphiphilic polymer consists of hydrophilic fucoidan (Fuc) linked by azobenzene with hydrophobic linolenic acid (LA), and can self-assemble into micelles to encapsulate DOX. Under hypoxia, [email protected] decreased intratumoral GSH by consuming nicotinamide adenine dinucleotide phosphate (NADPH), and caused accumulation of lethal ROS and lipid peroxides, resulting in mitochondrial damage and enhanced anti-tumor performance of DOX. [email protected] (IC 50 1.685 μg/mL) improved the effect of DOX (IC 50 7.847 μg/mL) on hypoxic tumor cells by nearly 4.6-fold. The current work provides a novel strategy for enhancing the antitumor efficacy via oxidative stress augmented chemotherapy.

Fucoidan-modified PLGA nanoparticles for integrated diagnosis and treatment of thrombosis

Wentao Ma,Shiwen Gong,Xianbiao He,Die Hu,Bo Cheng,Jing Yang,Binbin Li,Xinyu Wang

Thrombosis is a critical marker of cardiovascular diseases such as stroke and myocardial infarction. Timely diagnosis and dissolution of thrombi can significantly reduce patient mortality risk. Theranostic nanoparticles with targeting capabilities offer a promising solution to this issue. We developed fucoidan-modified PLGA nanoparticles loaded with indocyanine green (ICG) and lumbrokinase (LK) (FCP-ICG@LK NPs) to achieve integrated diagnosis and treatment of thrombotic diseases while minimizing hemorrhagic risks. Targeting experiments demonstrated the affinity of fucoidan for P-selectin. In a mouse model of carotid thrombosis, FCP-ICG@LK NPs demonstrated excellent diagnostic and monitoring capabilities while effectively dissolving thrombi under near-infrared light. Compared with free LK, FCP-ICG@LK NPs showed better thrombolysis effects at the same dose. Furthermore, bleeding risk assessment experiments indicated that FCP-ICG@LK NPs do not induce hemorrhagic risks. FCP-ICG@LK NPs showed no significant cytotoxicity or hemolysis. These results indicate that fucoidan-modified PLGA drug-loaded nanoparticles are a promising thrombus-targeted theranostic agent.

Design optimization of Fucoidan-coating Cationic Liposomes for enhance Gemcitabine delivery

Silli Epiphane K.,Zheng Zhenjiang,Zhou Xintao,Li Mengfei,Tang Jiali,Guo Ruizhe,Tan Chunlu,Wang Ying

Obstacles facing chemotherapeutic drugs for cancers led scientists to load Gemcitabine (GEM) into nanocarriers like liposomes, known for their nontoxicity profile and targeting capacity. The liposomal nanostructures containing GEM were coated with Fucoidan (FU) due to its anti-tumor properties by targeting cancer cells. Thus four different cationic liposomes formulations were prepared by thin-film hydration method in optimal conditions: DOTAP (formulation A); DPPC/DOTAP (4:1 molar ratio, formulation B), DPPC/DMPC/DOTAP (4:1:1 molar ratio, formulation C) and DPPC/DMPC/DOTAP/DSPE-mPEG2000 (4:1:1:0.1 molar ratio, formulation D). They were studied to identify lipid-compositions offering effective GEM-entrapment and successful coating of FU on the liposome surface. Additional qualitative characteristics, such as particle size, polydispersity index, zeta potential, stability and in vitro drug release were then evaluated. Formulation C gave the best GEM-entrapment efficiency (EE) but formed aggregates when coated with FU, giving non-homogenous large size particles then not suitable for effective delivery. It was the same situation with formulation A and B. Only the formulation D showed a good GEM-EE (> 80%) and affinity by successful coating FU from three different algae species. The PEGylated formulation D coated of FU, with regard to storage stability and drug release studies, revealed to be a promising approach on design of optimal drug delivery system.

Structural characterization and effect on leukopenia of fucoidan from Durvillaea antarctica

Yingjie Yang,Ting Hu,Jianjie Li,Meng Xin,Xia Zhao

Fucoidans from brown seaweed shows various bioactive properties and promising prospects in biomedical field. Here, a novel fucoidan (F-4) was extracted and purified from Durvillaea antarctica . The structure of F-4 was characterized by HPLC, HPGPC, GC–MS, together with IR and NMR spectral analysis. F-4 is a sulfated polysaccharide mainly composed of fucose (Fuc), galactose (Gal), and glucose (Glc) in a molar ratio of 26.4: 7.1: 1.0. The backbone of F-4 is composed of (1→3) and (1→4)-linked- α -L-Fuc p residues, which sulfated at C-4 or C-2 positions and branched with α-L-Fuc, β-D-Gal, and β-D-Glc residues. Furthermore, F-4 can effectively promote the growth of leukocyte in a mouse model induced by cyclophosphamide, possibly by activating hematopoietic progenitor cells and regulating the hematopoietic microenvironment of bone marrow. Our data provide useful information for further investigation of fucoidan in the treatment of chemotherapy-induced leukopenia.

Fucoidan hydrogels induced by κ-carrageenan: Rheological, thermal and structural characterization

Nan Wang,Jie Tian,Linlin Wang,Shuang Song,Chunqing Ai,Srinivas Janaswamy,Chengrong Wen

Fucoidan (FUC) is a non-gelling polysaccharide but could interact with κ-carrageenan (KC) to form a stable gel blend. However, their interaction mechanism is unclear. Herein, FUC and KC blended gels are prepared by mixing FUC (10 and 20 mg/mL) and KC (6, 7 and 8 mg/mL) solutions, and characterized through LF-NMR, rheology , DSC , Cryo-SEM, and FTIR. The FTIR analysis confirms the formation of hydrogen bonds between FUC and KC chains. The KC addition to FUC significantly improves the water retention and frost resistance. The viscoelastic measurements reveal higher gelling nature of the FUC-KC binary mixtures, and the DSC results confirm the higher thermal stability . The Cryo-SEM images clearly reveal the gel network structure. The outcome of this study deemed to further the FUC use in food and non-food applications.

Probiotic-loaded calcium alginate/fucoidan hydrogels for promoting oral ulcer healing

Xue Dou,Guotai Li,Shuang Wang,Dan Shao,Danyang Wang,Xuyang Deng,Yanli Zhu,Pengyu Gao,Jia Liu,Na Deng,Changqing Yuan,Qihui Zhou

Probiotics are beneficial bacteria located in the oral cavity which exhibit antimicrobial properties and contribute to the regulation of immune function and the modulation of tissue repair. Fucoidan (FD), a marine prebiotic, may further enhance the ability of probiotics to promote ulcer healing. However, neither FD nor probiotics are attached to the oral cavity and neither are well-suited for oral ulcer healing owing to the wet and highly dynamic environment. In this study, probiotic-loaded calcium alginate/fucoidan composite hydrogels were developed for use as bioactive oral ulcer patches. The well-shaped hydrogels exhibited remarkable wet-tissue adhesion, suitable swelling and mechanical properties, sustained probiotic release, and excellent storage durability. Moreover, in vitro biological assays demonstrated that the composite hydrogel exhibited excellent cyto/hemocompatibility and antimicrobial effects. Importantly, compared to commercial oral ulcer patches, bioactive hydrogels show superior therapeutic capability for promoting ulcer healing in vivo by enhancing cell migration, inducing epithelial formation and orderly collagen fiber deposition, as well as facilitating neovascularization. These results demonstrate that this novel composite hydrogel patch demonstrates great potential for the treatment of oral ulcerations.
