A ratiometric fluorescent probe based on Lu-Eu-DPA nanoparticles for sensitive and visual detection of gentamicin sulfate, neomycin, and erythromycin in food

Jie Mingsha,Zhu Amei,Zhu Bing,Guo Ruipeng,Lan Shengkai,He Jiaxin,Lang Xingang,Bai Yanhong

The overuse of antibiotics leads to significant health risks and environmental challenges. Therefore, selective and sensitive detection of antibiotic residues is a critical issue. Here, a double-emission ratiometric fluorescence probe (Lu-Eu-DPA) was constructed for the selective detection of gentamicin sulfate (GEN), neomycin (NEO), and erythromycin (EM). The Lu-Eu-DPA coordination polymer nanoparticles composed of luminol, the coordinated nuclear metal Eu 3+ , and 2,6-pyridinedicarboxylic acid (DPA). The coordination reaction between luminol molecules and Eu 3+ ions produce the aggregation-induced emission (AIE) effect, while DPA acts as a ligand for the antenna, absorbing energy from the excitation light and then transferring the energy to Eu 3+ . Based on the AIE effect and antenna effect (AE), the fluorescence intensity of Lu-Eu-DPA at 426 nm and 617 nm was correlated with the concentration of GEN, NEO, and EM, and the detection limits of GEN, NEO, and EM were 20, 25, and 48 nmol/L, respectively. The detection capability of the proposed method in pork samples was also studied. In addition, semi-quantitative determination of GEN, NEO, and EM was conveniently and efficiently performed using a smartphone-based visualization method. These results suggested that this probe with good selectivity and specificity was a promising method for swiftly detecting multiple antibiotic residues.

DETERMINATION OF AMINOGLYCOSIDE ANTIBIOTICS BY A COLORIMETRIC METHOD BASED ON THE AGGREGATION OF GOLD NANOPARTICLES

RUIYONG WANG,SHUMIN FAN,RUIQIANG WANG,RUI WANG,HUANJING DOU,LVJING WANG

A sensitive and selective colorimetric biosensor for determination of gentamicin, amikacin and tobramycin was proposed with the unmodified gold nanoparticles (GNPs) as the sensing element. Gentamicin, amikacin and tobramycin can rapidly induce the aggregation of gold nanoparticles and is accompanied by a color change from red to blue. The concentration of gentamicin, amikacin and tobramycin can be determined by using UV-Vis spectrometer. The experimental parameters were optimized with regard to pH, incubation time and the concentration of the GNPs. Under optimal experimental conditions, the linear range of the colorimetric sensor for gentamicin/amikacin/tobramycin were 2.67–33.93 ng mL -1 , 13.33–66.67 ng mL -1 and 20–180 ng mL -1 , respectively. The corresponding limit of detection (3σ) was 0.354 ng mL -1 , 0.999 ng mL -1 and 0.579 ng mL -1 , respectively. This assay was simple and used to detect aminoglycoside antibiotics in milk and medicine products.

Lysozyme Associated Liposomal Gentamicin Inhibits Bacterial Biofilm

Yilin Hou,Zhaojie Wang,Peng Zhang,Hu Bai,Yuelin Sun,Jinyou Duan,Haibo Mu

Bacteria on living or inert surfaces usually form biofilms which make them highly resistant to antibiotics and immune clearance. Herein, we develop a simple approach to overcome the above conundrum through lysozyme-associated liposomal gentamicin (LLG). The association of lysozyme to the surface of liposomes can effectively reduce the fusion of liposomes and undesirable payload release in regular storage or physiological environments. The LLG was more effective at damaging established biofilms and inhibiting biofilm formation of pathogens including Gram-positive and Gram-negative bacteria than gentamicin alone. This strategy may provide a novel approach to treat infections due to bacterial biofilm. Keywords:liposome;lysozyme;gentamicin;biofilm

Chitosan-Gentamicin Conjugate Hydrogel Promoting Skin Scald Repair

Tingting Yan,Songzhi Kong,Qianqian Ouyang,Chengpeng Li,Tingting Hou,Yu Chen,Sidong Li

Our earlier research indicated that chitosan-gentamicin conjugate (CS-GT) possesses superior antimicrobial activity and good water solubility. To develop CS-GT-based scald dressings, the antibacterial properties of CS-GT were further studied, and the biosafety of CS-GT and the healing mechanism of CS-GT hydrogel was systematically explored in this article. It was found that cell viability shows a declined inclination with the prolonged culture time and the increased concentration of CS-GT. After three day’s culture, the cell viability could still remain at 79.72% when CS-GT concentration was as high as 1000 μg/mL. On the other hand, the hemolysis rate of CS-GT was lower than 5% when its concentration is 800 μg/mL. Therefore CS-GT has good cytocompatibility and hemocompatibility. A wound-healing experiment has shown that the skin healing rate of CS-GT hydrogel was the highest at 99.61%, followed by the positive control (wet burn ointment) 94.98%, GT hydrogel 87.50%, and matrix 77.39%. The blank control group, however, possessed the lowest healing rate of 75.45%. Further analysis indicated that CS-GT hydrogel could promote the synthesis of total protein (TP) in skin granulation tissue, resulting in the enhanced hydroxyproline (HYP) content, which facilitated collagen fibrogenesis, reduced cytokine expression in an inflammatory response, and, ultimately, accelerated wound healing. To sum up, CS-GT hydrogel is a promising scald dressing.Keywords:chitosan-gentamicin conjugate;antimicrobial;anti-inflammatory;scald repair

Overcoming gentamicin resistance in MRSA by a polyphenol-assembled nanoplatform

Shuailin Liu,Mengna Wang,Yujie Shen,Yinghui Song,Baojun Yang,Jifeng Liu,Xia Gao

OG/ICG, an intelligent nano-prodrug, is engineered by conjugating OPC with GEN, and ICG loading. • OG/ICG remains inert systemically but switches charge and dissociates in acidic infections. • The efficacy of GEN against MRSA was restored through the synergistic action of OPC and photothermia. • Combined chemical, physical, and targeting actions enable full eradication of GEN-resistant MRSA. The waning efficacy of aminoglycosides, like gentamicin (GEN), against methicillin-resistant Staphylococcus aureus (MRSA) necessitates strategies that transcend the protracted pipeline of de novo antibiotic discovery. Herein, we present a “resensitization” strategy enabled by a carrier-free, supramolecular nano-prodrug (OG/ICG NPs), engineered via the oxidative co-assembly of oligomeric proanthocyanidin (OPC) and GEN, followed by indocyanine green (ICG) loading. Distinct from conventional inert delivery systems, this bioactive platform remains stable physiologically but undergoes charge reversal and rapid dissociation within the acidic infection microenvironment. Mechanistically, the released OPC functions as a potent resistance-breaking adjuvant by disrupting cell wall integrity and inhibiting efflux pumps, thereby facilitating massive intracellular GEN accumulation. Under near-infrared (NIR) irradiation, localized hyperthermia synergizes with this chemical attack to compromise membrane permeability and trigger pronounced metabolic collapse, characterized by disrupted fatty acid degradation and ATP depletion, as revealed by both transcriptomic and phenotypic profiling. Furthermore, the nanoplatform promotes regulating macrophage polarization from M1 to M2 phenotype, accelerating abscess resolution and tissue regeneration. This work establishes a robust, dual-responsive paradigm that not only eradicates resistant pathogens through metabolic and structural disruption but also orchestrates an immune-favorable microenvironment for wound healing.

Gentamicin decorated phosphatidylcholine-chitosan nanoparticles against biofilms and intracellular bacteria

Yuanhao Qiu,Dan Xu,Guoqing Sui,Dongdong Wang,Ming Wu,Lipeng Han,Haibo Mu,Jinyou Duan

Biofilms and intracellular bacteria often cause a series of overwhelming public health issues due to their strong drug resistance. Hence, chitosan nanoparticles (CS NPs), phosphatidylcholine and gentamicin were used to synthesize a novel nanodrug delivery system (GPC NPs). Dynamic light scattering (DLS) demonstrated that the surface zeta-potential of GPC NPs was −19.5 mV. The morphology of GPC NPs was observed by scanning electron microscopy (SEM). The gentamicin adsorption and release behaviors of GPC NPs were also investigated. The GPC NPs could effectively damage and remove the biofilm formed by pathogens through permeation of the antibiotic into the biofilm. In addition, the nanoparticles were readily engulfed by macrophages which facilitated the killing of intracellular bacteria and had neglectable cytotoxicity. Our study indicated that GPC NPs could be used as a promising nanoantibacterial agent against biofilms and intracellular bacteria.

A gentamicin-thioctic acid multifunctional hydrogel for accelerating infected wound healing

Yamei Gao,Xiang Zhan,Shaohu Huo,Ling Fu,Zhen Tang,Keke Qi,Chunchun Lv,Chengyuan Liu,Yulin Zhu,Shenggang Ding,Yongmei Lv

Bacterial infections remain a major concern during wound healing and tissue bonding. The excessive proliferation of bacteria will seriously hinder the repair of the wound and even lead to death. Generally, surgical sutures might cause damage to the surrounding tissues and inevitable infection due to the unfixed shape of the wound. Thus, it is urgent to develop novel antibacterial skin dressing with self-healing and strong adhesion properties. Herein, we prepared an antibacterial and self-healable hydrogel with strong adhesion activity through natural small molecules, including thioctic acid TA and gentamicin (GM). The rapid ring-opening-polymerization of the TA (PTA) forms the backbone of macromolecules, and the functional hydrogel was constructed with the crosslinking of GM, termed as G-PTA, which offers hydrogen bonding interactions between the amino and hydroxyl groups of GM and carboxylic group side chains of poly(TA). The synthesized hydrogel exhibited rapid self-healing ability and strong tissue adhesion due to the internal dynamic disulfide bonds and multiple hydrogen bonds. Importantly, the introduction of GM enabled the G-PTA hydrogel to sustainably release antibiotics and exhibit a durative antibacterial effect with the degradation of PTA, which further shorten the therapeutic time and enhance tissue regeneration in a wound infection model. The in vitro and in vivo experiments demonstrate that the G-PTA hydrogel has potential as a surgical antibacterial biological adhesive, especially for bacterial wound infections.

Fluorescent molecularly imprinted sensor for rapid detection of gentamicin in Milk

Guo Hui,Jing Ding,Jiaxin Li,Shenglan Zhao,Yiting Deng,Peilong Ma,Longteng Wang

Gentamicin, a widely used veterinary antibiotic in animal husbandry, poses significant food safety risks due to its persistence in livestock products, soil, and water systems. Chronic exposure to gentamicin residues is associated with severe health effects, including ototoxicity, nephrotoxicity, and neuromuscular toxicity. To address this challenge, we developed a fluorescent sensing strategy by synthesizing a core–shell nanocomposite, Fe₃O₄@SiO₂@N-GQDs@MIPs (termed the MIPs probe). This probe was constructed by integrating nitrogen-doped graphene quantum dots (N-GQDs) with magnetic Fe₃O₄ nanoparticles, and the overall detection platform is referred to as the MIP sensor. The sensor operates via static quenching, enabling specific and sensitive detection of gentamicin with a linear range of 3–900 nM and a detection limit of 0.274 nM. The sensor was successfully applied to detect gentamicin in milk samples, demonstrating excellent recovery rates (94.0–103.3%) and precision (RSD < 2.2%). This work presents a robust, efficient, and field-deployable platform for monitoring antibiotic residues in food products, offering significant improvements over conventional chromatographic and immunological methods.

Porous magnesium loaded with gentamicin sulphate and in vitro release behavior

Qiuyan Li,Guofeng Jiang,Dong Wang,Huang Wang,Liang Ding,Guo He

Our aim was to develop a biocompatible bone repair material that has the advantage of preventing postoperative infections. Finally, the porous magnesium (p-Mg) loaded with gentamicin sulphate (GS-loaded Mg-G) was fabricated. The GS release behavior of the GS-loaded Mg-G in phosphate buffer saline (PBS) was investigated. The effective release time of GS reached to 14   days. In addition, the effects of porosity and pore diameter of p-Mg on the GS release behavior of the GS-loaded Mg-G were studied. In the initial burst release stage, the GS release rate of the GS-loaded Mg-G increased with the increasing porosity or the increasing pore diameter of p-Mg. The GS-loaded Mg-G with larger original pore diameter has higher burst release of GS. Moreover, the in vitro antibacterial test of the GS-loaded Mg-G indicated that this biomaterial has obvious antibacterial effect. This study can provide information for p-Mg loaded with drug(s) as functional bone repair materials with drug-delivery capabilities.

Carvacrol potentiates gentamicin against Staphylococcus aureus via disrupting bacterial respiratory bioenergetics

Sijie Wu,Xiaomin Ren,Ronger Wu,Jia Cheng,Xiaoyong Huang

The emergence of methicillin-resistant Staphylococcus aureus (MRSA) calls for novel antibiotic adjuvants. Carvacrol (Car), a natural monoterpene phenol derived from the essential oils of aromatic plants, exhibits membrane-active properties and has potential as an antibacterial adjuvant. Here, we found that Car markedly enhanced the bactericidal activities of Gentamicin (Gen) against MRSA. Mechanistically, Car dissipated membrane potential, inhibited respiratory chain dehydrogenase activity, and depleted intracellular ATP. Moreover, Car promoted membrane lipid peroxidation, ultimately leading to structural and functional membrane damage and bacterial death. Notably, pharmacological perturbation of respiratory and metabolic pathways further supported that this synergistic effect depends on bacterial respiratory bioenergetics. In addition, the combination showed favorable in vivo antibacterial efficacy in both Galleria mellonella and murine infection models, without apparent toxicity under the tested conditions. Collectively, these findings indicate that Car may serve as a natural antibacterial adjuvant for combination therapy against drug-resistant infections.

Systematic engineering of Micromonospora echinospora cell factory for gentamicin C1a overproduction

Feng Xu,Hao Gao,Rong Ben,Kaihao Hu,Yuan Wang,Ali Mohsin,Yuanxin Guo,Xu Li,Haifeng Hang,Ju Chu,Xiwei Tian

The clinical importance of gentamicin C1a as a broad-spectrum aminoglycoside antibiotic underscores the need for efficient biomanufacturing strategies. In this study, we developed a systematic engineering framework to enhance gentamicin C1a production. First, a genome-scale metabolic model (iFX1172) was reconstructed to pinpoint critical bottlenecks in both regulatory and biosynthetic pathways. Guided by model predictions and experimental validation, we identified gen C, met K, and Bld D as synergistic targets. Coordinated overexpression of these genes increased gentamicin C1a titers to 198.1 mg/L, representing a 34.3% improvement over the parental strain, and also enhanced the titers of other aminoglycoside antibiotics by up to 1.6-fold, demonstrating the universality of the strategy. Metabolic flux analysis and targeted metabolomics revealed that redox homeostasis and ATP availability are pivotal for biosynthesis. Finally, process optimization in a fed-batch bioreactor using a Bayesian framework, coupled with in situ resin adsorption, yielded 964.1 mg/L gentamicin C1a with a yield of 24.1 mg/g glucose and a productivity of 6.7 mg/L/h.
