A ratiometric fluorescent probe based on Lu-Eu-DPA nanoparticles for sensitive and visual detection of gentamicin sulfate, neomycin, and erythromycin in food

Jie Mingsha,Zhu Amei,Zhu Bing,Guo Ruipeng,Lan Shengkai,He Jiaxin,Lang Xingang,Bai Yanhong

The overuse of antibiotics leads to significant health risks and environmental challenges. Therefore, selective and sensitive detection of antibiotic residues is a critical issue. Here, a double-emission ratiometric fluorescence probe (Lu-Eu-DPA) was constructed for the selective detection of gentamicin sulfate (GEN), neomycin (NEO), and erythromycin (EM). The Lu-Eu-DPA coordination polymer nanoparticles composed of luminol, the coordinated nuclear metal Eu 3+ , and 2,6-pyridinedicarboxylic acid (DPA). The coordination reaction between luminol molecules and Eu 3+ ions produce the aggregation-induced emission (AIE) effect, while DPA acts as a ligand for the antenna, absorbing energy from the excitation light and then transferring the energy to Eu 3+ . Based on the AIE effect and antenna effect (AE), the fluorescence intensity of Lu-Eu-DPA at 426 nm and 617 nm was correlated with the concentration of GEN, NEO, and EM, and the detection limits of GEN, NEO, and EM were 20, 25, and 48 nmol/L, respectively. The detection capability of the proposed method in pork samples was also studied. In addition, semi-quantitative determination of GEN, NEO, and EM was conveniently and efficiently performed using a smartphone-based visualization method. These results suggested that this probe with good selectivity and specificity was a promising method for swiftly detecting multiple antibiotic residues.

Gentamicin Sulfate Grafted Magnetic GO Nanohybrids with Excellent Antibacterial Properties and Recyclability

Xing Wu,Jingya Zhou,Zeng Liu,Wei Shao

In this study, magnetic graphene oxide (MGO) nanohybrids were first prepared by loading Fe3O4NPs onto graphene oxide (GO). Then, GS-MGO nanohybrids were prepared by grafting gentamicin sulfate (GS) onto MGO directly using a simple amidation reaction. The prepared GS-MGO had the same magnetism as MGO. They exhibited excellent antibacterial ability against Gram-negative bacteria and Gram-positive bacteria. The GS-MGO had excellent antibacterial performance againstEscherichia coli(E. coli),Staphylococcus aureus(S. aureus), andListeria monocytogenes(L. monocytogenes). When the addition concentration of GS-MGO was 1.25 mg/mL, the calculated bacteriostatic ratios againstE. coliandS. aureusachieved 89.8% and 100%, respectively. ForL. monocytogenes, only 0.05 mg/mL of GS-MGO had an antibacterial ratio as high as 99%. In addition, the prepared GS-MGO nanohybrids also exhibited excellent non-leaching activity with good recycling antibacterial ability. After eight times antibacterial tests, GS-MGO nanohybrids still exhibited an excellent inhibition effect onE. coli,S. aureus, andL. monocytogenes. Therefore, as a non-leaching antibacterial agent, the fabricated GS-MGO nanohybrid had dramatic antibacterial properties and also showed great recycling ability. Thus, it displayed great potential in the design of novel recycling antibacterial agents with non-leaching activity. Keywords:magnetic;graphene oxide;gentamicin sulfate;antibacterial;recyclable activity

Dynamic Release of Antibiotic Drug Gentamicin Sulfate from Novel Zirconium Phosphate Nano-Platelets

Ai Jing,Chen Yan,Jing Hui,Gao Xuechuan,Liu Jiayin,Ma Kexin,Suo Jian,Yu Shiyong,Song Shuyan

The intercalation of gentamicin sulfate (GS) into novel zirconium phosphate (ZrP) nano-platelets has been achieved using a hydrated ZrP phase known as the -ZrP (Zr(HPO4)2·6H2O) which possesses six water molecules per formula unit. This paper revealed the successful intercalation of the GS into -ZrP at different molar ratios of GS: -ZrP (1:10,1:5,1:2), which were demonstrated by the emergence of a new phase in the X-ray powder diffraction (XRD) pattern with an interlayer distance of 13.1 Å. The morphologies and sizes of the samples were characterized by scanning electron microscopy (SEM) and transmission electron microscopy (TEM) which showed the characteristic platelet-like hexagonal shape of the ZrP nanoparticles. Compared to spherical shape particles the hexagonal shape may show better margination, adhesion, and binding properties. The average particle size of GS: -ZrP nano-platelets was 120 nm in diameter and the average thickness of the particles was 10 nm. The drug release in vitro on the obtained GS: -ZrP nano-platelets was carried out in pH 7.4 buffer solution at 37 °C. The cytotoxicity cell viability experiments in vitro for the GS and GS: -ZrP nanoparticles showed that GS:ZrP had low cytotoxicity. Overall, our study demonstrated that the GS: -ZrP nanoparticles possessed a moderate GS loading efficiency, low cytotoxicity and favorable biocompatibility.

Coaxial electrospray of gentamicin sulfate-loaded micro/nano particles for sustained antibacterial PMMA bone cement

Xiaoyan Li,Xinyu Zhang,Jialong Yi,Chenhu Mei,Jixiang Zhu,Ming Li,Longqi Zhao,Deng-guang Yu

Polymethyl methacrylate (PMMA) bone cement is widely used in orthopedic surgery, and enhancing its antibacterial properties is crucial for preventing perioperative infections. In this study, unique “berry-shaped” micro/nano composite particles were prepared via coaxial electrospraying for the development of antibacterial PMMA bone cement. These particles consist of PMMA microspheres uniformly embedded with nanosilica (SiO 2 ) drug carriers coated with quaternary ammonium chitosan (HACC) and loaded with gentamicin sulfate (GS). Using a polylactic acid (PLA) nanofiber mat as the collection substrate, uniform assembly of the micro/nano composite particles and their strong anchoring to the matrix were achieved by tailoring the core-sheath solution properties and leveraging capillary-driven interactions. This hierarchical design not only improved interfacial adhesion to the PMMA bone cement matrix and improved flexural toughness, but also reduced the curing temperature. The corresponding antibacterial PMMA composite bone cement released GS sustainably for over five days, demonstrating potent antibacterial activity against both Escherichia coli ( E. coli ) and methicillin-resistant Staphylococcus aureus ( MRSA ), as well as good fibroblast biocompatibility. This study proposed an integrated “material-structure-assembly” strategy for the one-step fabrication of functional micro/nano composite particles, providing a new approach for the development of advanced biomedical materials.

Hierarchical antibiotic delivery system based on calcium phosphate cement/montmorillonite-gentamicin sulfate with drug release pathways

Lei Chen,Xiuying Lin,Min Wei,Bo Zhang,Yani Sun,Xi Chen,Shitong Zhang,Hao Zhang,Jieyu Zhang,Xiaojiao Yu,Binghua Yao,Kang Zhao,Yufei Tang,Quanchang Tan,Zixiang Wu

Antibiotic-loaded calcium phosphate cement (CPC) has emerged as a promising biomaterial for drug delivery in orthopedics. However, there are problems such as the burst release of antibiotics, low cumulative release ratio, inappropriate release cycle, inferior mechanical strength, and poor anti-collapse properties. In this research, montmorillonite-gentamicin (MMT-GS) was fabricated by solution intercalation method and served as the drug release pathways in CPC to avoid burst release of GS, achieving promoted cumulative release ratios and a release cycle matched the time of inflammatory response. The results indicated that the highest cumulative release ratio and release concentration of GS in CPC/MMT-GS was 94.1 ± 2.8 % and 1183.05 μg/mL, and the release cycle was up to 504 h. In addition, the hierarchical GS delivery system was divided into three stages, and the kinetics followed the Korsmeyer-Peppas model, the zero-order model, and the diffusion-dissolution model, respectively. Meanwhile, the compressive strength of CPC/MMT-GS was up to 51.33 ± 3.62 MPa. Antibacterial results demonstrated that CPC/MMT-GS exhibited excellent in vitro long-lasting antibacterial properties to E. coli and S. aureus . Furthermore, CPC/MMT-GS promoted osteoblast proliferation and exhibited excellent in vivo histocompatibility. Therefore, CPC/MMT-GS has favorable application prospects in the treatment of bone defects with bacterial infections and inflammatory reactions.

Novel CdS/MOF Cathodic Photoelectrochemical (PEC) Platform for the Detection of Doxorubicin Hydrochloride and Gentamicin Sulfate

Wenxia Dong,Zhongping Li,Wen Wen,Bin Liu,Guangming Wen

Nanomaterial selection is critical for photoelectrochemical (PEC) sensing. In this report, a novel cathodic photoelectrochemical (PEC) strategy was proposed for the detection of doxorubicin hydrochloride (Dox) and gentamicin sulfate (CN). The photocathode was synthesized by noncovalently coupling cadmium sulfide (CdS) to the porphyrin-derived metal–organic framework (CdS@PCN-224). This type of assembly created a pleasant interface for the combination of doxorubicin hydrochloride and gentamicin sulfate, resulting in a good CdS@PCN-224 donor–acceptor system. When compared to a single optoelectronic material, its photocurrent is enhanced by unprecedented nine times. This research could pave the way for the realization of PCN-224’s enormous potential in PEC sensing.

Antibacterial and anticancer activities of asymmetric lollipop-like mesoporous silica nanoparticles loaded with curcumin and gentamicin sulfate

Yajun Cheng,Yudi Zhang,Weijun Deng,Jing Hu

Asymmetric mesoporous silica nanoparticles with anisotropic geometry and dual-compartments are highly desired for loading and release of dual-drugs in separated storage spaces. In this study, an asymmetric lollipop-like mesoporous silica nanoparticle Fe 3 O 4 @SiO 2 &EPMO (EPMO = ethane bridged periodic mesoporous organosilica) was successfully developed via an anisotropic epitaxial growth strategy. The asymmetric nanoparticles show a uniform lollipop shape with a head of spherical Fe 3 O 4 @SiO 2 core-shell that is 200 nm in diameter and a tail of EPMO nanorods with a length of ∼90 nm, and a specific surface area of ∼650.3 m 2 g −1 . Most importantly, the asymmetric nanoparticles possess the unique dual independent (hydrophilic/hydrophobic) spaces with good loading capacities and are significantly more efficient for cancer cell killing than pure drug based on in vitro studies. Additionally, the dual-drug-loaded nanoparticles exhibited excellent antibacterial activity.

Green and Sustainable Self-Assembly Nanocomposite from Gentamicin Sulfate/Lignosulfonate with Efficient Antibacterial and Wound-Healing Activity

Jiafeng Zou,Dandan Bao,Rui Ma,Zhihong Zhu,Xiaojie Chen,Jingjing Zhu,Xudong Fan,Ke Zhang,Hongyue Zheng,Fanzhu Li,Ji-Gang Piao

In recent years, developments of drug delivery systems of antibiotics have been needed for improving antibacterial performance due to massive usage of antibiotics. With the global growing concept of being green, lignosulfonate (LS) has been deemed as an ecofriendly stabilizer and carrier that holds promise in preparing nanocomposites for antibacterial treatment. At the same time, LS and gentamicin sulfate (GS) have been proven to form a nanocomposite (LS-GS) with highly uniform size and roundness in solution. It also possesses a good loading capacity of GS. Density functional theory (DFT) verified that the interaction forces in LS-GS, a nanocomposite with self-assembly characteristics, were mainly combined via hydrogen bonds, van der Waals forces, and repulsion. The obtained LS-GS has the biocompatible, biodegradable, and low-cost merits of LS. It also exhibited excellent in vitro antibacterial activities against Escherichia coli and Staphylococcus aureus due to the antibacterial properties of GS. Moreover, an in-depth antibacterial study revealed the highly efficient antibacterial and wound-healing applications of LS-GS in vivo. Collectively, LS, a carrier material with low cost, which strikingly increases the antibacterial and wound-healing ability of GS in vitro and in vivo, will contribute to the production of large-scale antibacterial nanocomposites.

Preparation and performance of porous hydroxyapatite/poly(lactic-co-glycolic acid) drug-loaded microsphere scaffolds for gentamicin sulfate delivery

Cao Xing,Dai Lingjun,Sun Shichang,Ma Rui,Liu Xiangli

The prevention/treatment of osteomyelitis infection by combining the local antibiotic delivery system with the bone regeneration scaffold can effectively overcome the drawbacks of systemic antibiotic administration and realize the full cycle control of drug release. Herein, by introducing sodium carboxymethyl cellulose (SCMC) cross-linking agent to improve the binding force of poly(lactic-co-glycolic acid) (PLGA) microspheres and porous hydroxyapatite (HAp) bone scaffold. The porous HAp/PLGA drug-loaded microsphere bone scaffold for gentamicin sulfate (GS) delivery was successfully prepared. The optimal preparation parameters, drug release characteristics, SCMC enhancement mechanism, antibacterial properties and bone cell activity of porous HAp/PLGA drug-loaded microsphere bone scaffolds were comprehensively investigated. The results showed that the 0.1% SCMC-modified porous HAp/PLGA drug-loaded microsphere bone scaffold has a cumulative drug release of 45.0 ± 0.90% on the first day, which is about 20% lower than that of pure PLGA drug-loaded microspheres. Moreover, its drug release can be sustained and stably released for more than 17 d, which is attributed to the enhancement of the binding force between the microspheres and HAp by SCMC (combination for more than 3 weeks). Meanwhile, the diameter of the antibacterial ring expanded from the initial 10 ± 0.50 to 28 ± 1.2 mm after 14 d, which also indicated the sustained and stable release of GS. Alamar Blue analysis results showed that 0.1% SCMC-modified composite bone scaffold is beneficial to the proliferation activity of bone cells, and its 14 day activity increased by 20%. The above results indicate that the SCMC-modified composite bone scaffold has the potential to treat/prevent osteomyelitis.

Protein-reduced gold nanoparticles mixed with gentamicin sulfate and loaded into konjac/gelatin sponge heal wounds and kill drug-resistant bacteria

Yini Zou,Ruiqi Xie,Enling Hu,Ping Qian,Bitao Lu,Guangqian Lan,Fei Lu

Timely antibacterial treatment of wounds reduces the probability of wound infection and promotes wound healing. However, the materials used to treat wounds often fail to provide both sterilization (especially for super bacteria) and moisture, and some may even cause secondary injury to the wound. In this study, gold nanoparticles (Au NPs) of average grain diameter of 3 ± 1 nm were prepared using egg white as the reductant. These particles showed no aggregation and pink fluorescence. Au NPs were mixed with gentamicin sulfate (GS) and loaded into a mixture of konjac glucomannan (KGM) and gelatin as wound dressing (KGM/ [email protected] NPs/GS). Antibacterial experiments showed that the Au NPs amplified the antibacterial activity of GS; Au NPs/GS efficiently eliminated bacteria, especially super bacteria. Cytotoxicity tests indicated that KGM/ [email protected] NPs/GS showed basically no cytotoxicity to L929 cells. In addition, KGM/ [email protected] NPs/GS possesses good water absorption, water retention, and enhanced mechanical properties, which can provide a moist environment for wounds and promote healing. In conclusion, our study showed that the antibacterial activity of KGM/ [email protected] NPs/GS is better than that of only GS and that it efficiently eliminated super bacteria. Therefore, KGM/ [email protected] NPs/GS can be used for killing superbugs, inhibiting bacterial growth, and promoting wound healing.
