Human NAD(P)H: quinone oxidoreductase (hNQO1) is an important biomarker for human malignant tumors. Detection of NQO1 accurately is of great significance to improve the early diagnosis of cancer and prognosis of cancer patients. In this study, based on the covalent assembly strategy, hNQO1-activated fluorescent probes 1 and 2 are constructed by introducing coumarin precursor 2-cyano-3-(4-(diethylamino)-2-hydroxyphenyl) acrylic acid and self-immolative linkers. Under reaction with hNQO1 and NADH, turn-on fluorescence appears due to in-situ formation of the organic fluorescent compound 7-diethylamino-3-cyanocoumarin, and fluorescent intensity changes significantly. Probe 1 and 2 for detection of hNQO1 are not interfered by other substances and have low toxicity in cells. In addition to quantitative detection of hNQO1 in vitro, they have also been successfully applied to fluorescent imaging in living cells.

Human NAD(P)H:quinone oxidoreductase (hNQO1) can be used as a biomarker for the early diagnosis of cancer. In this article, a novel fluorescent probe 1 for detecting hNQO1 was designed and synthesized by using 2-dicyanomethylene-3-cyano-4,5,5-trimethyl-2,5-dihydrofuran (TCF) derivative (TCF-OH) as a fluorophore and quinone propionic acid (QPA) as a recognition group. The probe, which has high selectivity for hNQO1 and a Stokes shift of about 117 nm, shows a linear relationship with hNQO1 concentrations in the range of 0.25–3 μg ml−1 with the color changes from yellow to red and was successfully applied to intracellular hNQO1 imaging.

Identifying cancer at the cellular level during an early stage offers the hope of greatly improved outcomes for cancer patients. As potential cancer biomarkers, nitroreductase (NTR) and human quinine oxidoreductase 1 (hNQO1) are overexpressed in many type of cancer cells. Simultaneous detection of these two biomarkers would benefit diagnostic precision in related cancers without yielding false positive results. Herein, based on a dye generated in situ strategy, a dual-enzyme-responsive probe, CNN, was rationally designed and synthesized by installing p-nitrobenzene and trimethyl-locked quinone propionic acid groups, which are specific for NTR and hNQO1, respectively, into a single fluorophore. This probe is only activated in the presence of both NTR and hNQO1 and produces a large fluorescence response, enabling the detection of both endogenous NTR and hNQO1 activity in living cells. The imaging results indicate that the CNN probe differentiates cancer cells (HeLa, MDA-MB-231 and HepG2 cells) from normal liver HL-7702 cells owing to the existence of relatively high endogenous levels of both biomarkers in these cancer cells.

Human NAD(P)H:quinone oxidoreductase 1 (hNQO1), a proteinase that engages in detoxification of quinones and capable of activating anti-tumor drugs, has drawn increasing attention as tumor biomarker and drug target. Despite significant advances, the detection of hNQO1 up to now primarily relies on stimulus-responsive probes involving metabolization of a complicated synthetic quinone-functionalized probe, which, however, remains challenging to improve the sensing sensitivity, and are lack of sufficient stability. Herein, we report a highly sensitive electrochemiluminescent detection of hNQO1 and associated drug screening by straightforward sensing of the metabolin, enabled by a futile redox cycle reaction. Thanks to the intrinsic circulatory amplification and the luminol-modified nickel foam electrode, the electrochemiluminescent (ECL) sensor exhibited a record-level detection limit (10 ng/mL) and SNR (∼460) for hNQO1. The same format was also successfully applied to rapidly ranking hNQO1-directed anti-tumor drugs down to several-ten minutes. This new strategy would open a new era for hNQO1 detection with uncompromised high performance and also stimulate ECL as a promising tool that combines both diagnostic and drug screening functions for the popularization of proteinases in cancer management in a future application.
