Low Concentrations of Antibiotics Alter Microbial Communities and Induce High Abundances of Antibiotic-Resistant Genes in Ornamental Water

Lingyun Tian,Hao Fang,Qianbo Mao,Yi Bai,Zirui Ye,Dingjun Hu,Xiaoheng Wang,Yiyu Hou,Nan Ye,Shuai Zhang,Yan Ma

Antibiotic resistance poses a significant threat to the public health domain. A favorable platform for generating and disseminating antibiotic-resistant genes (ARGs) and antibiotic-resistant bacteria (ARB) is provided by landscaped fish ponds created by urbanization. This research delved into the effects exerted by different concentrations of specific antibiotics, namely tetracycline and ciprofloxacin, on the microbial community composition present in water samples obtained from a landscape pond. Additionally, we analyzed the abundance of ARGs and the class 1 integron-integrase gene (intI1), and identified potential hosts of ARGs. The results indicated that the consistent administration of antibiotics significantly influenced the microbial community structure, resulting in variations within both bacterial communities and functionalities. Furthermore, the absolute quantities of ARGs, includingtetA,tetC,qnrA, andqnrS, as well as the integrase geneintI1, exhibited augmentation in response to varying types and concentrations of antibiotics. Notably, the regular input of low concentrations of antibiotics produced higher levels of abundance of ARGs than the regular input of higher concentrations of antibiotics. The use of different types of antibiotics led to diverse host bacteria structures.

Inactivation of sulfonamide antibiotic resistant bacteria and control of intracellular antibiotic resistance transmission risk by sulfide-modified nanoscale zero-valent iron

Yuwei Wang,Jingfeng Gao,Wanjun Duan,Wenzhi Zhang,Yifan Zhao,Jie Liu

The inactivation of a gram-negative sulfonamide antibiotic resistant bacteria (ARB) HLS.6 and removal of intracellular antibiotic resistance gene (ARG, sul1 ) and class I integrase gene ( intI1 ) by nanoscale zero-valent iron (nZVI) and sulfide-modified nZVI (S-nZVI) with different S/Fe molar ratios were investigated in this study. The S-nZVI with high sulfur content (S/Fe = 0.05, 0.1, 0.2) was superior to nZVI and the treatment effect was best when S/Fe was 0.1. The ARB (2 × 10 7 CFU/mL) could be completely inactivated by 1.12 g/L of S-nZVI (S/Fe = 0.1) within 15 min, and the removal rates of intracellular sul1 and intI1 reached up to 4.39 log and 4.67 log at 60 min, respectively. Quenching experiments and flow cytometry proved that reactive oxygen species and adsorption were involved in the ARB inactivation and target genes removal. Bacterial death and live staining experiments and transmission electron microscopy showed that the ARB cell structure and intracellular DNA were severely damaged after S-nZVI treatment. This study provided a potential alternative method for controlling the antibiotic resistance in aquatic environment.

Elucidating the effect of microbial inoculum and ferric chloride as additives on the removal of antibiotic resistance genes from chicken manure during aerobic composting

Honghong Guo,Jie Gu,Xiaojuan Wang,Mubasher Nasir,Jing Yu,Liusheng Lei,Qianzhi Wang

This experiment investigated the effect of adding a microbial inoculum (M) and ferric chloride (F) on the fate of antibiotic resistance genes (ARGs) during chicken manure composting. Adding M and F improved the microbial activity in the compost and facilitated the removal of ARGs, whereas the combined treatment achieved the best results, especially in reducing the enrichment of sul resistance genes. Tn916/1545 and intI1 were important genetic elements that affected the transfer of ARGs, and Tn916/1545 was closely related to the transfer of tetM , tetW , and ermQ in Firmicutes. Kyoto Encyclopedia of Genes and Genomes functional predictions indicated that M and F could reduce the abundance of membrane transport and signal transduction molecules in the compost products. Thus, these findings suggest that the combined application of M and F is a promising strategy that could potentially inhibit the transfer of ARGs during composting.

Antibiotic-resistant genes and antibiotic-resistant bacteria in the effluent of urban residential areas, hospitals, and a municipal wastewater treatment plant system

Li Jianan,Cheng Weixiao,Xu Like,Strong P. J.,Chen Hong

In this study, we determined the abundance of 8 antibiotics (3 tetracyclines, 4 sulfonamides, and 1 trimethoprim), 12 antibiotic-resistant genes (10 tet , 2 sul ), 4 antibiotic-resistant bacteria (tetracycline, sulfamethoxazole, and combined resistance), and class 1 integron integrase gene ( intI 1) in the effluent of residential areas, hospitals, and municipal wastewater treatment plant (WWTP) systems. The concentrations of total/individual targets (antibiotics, genes, and bacteria) varied remarkably among different samples, but the hospital samples generally had a lower abundance than the residential area samples. The WWTP demonstrated removal efficiencies of 50.8 % tetracyclines, 66.8 % sulfonamides, 0.5 logs to 2.5 logs tet genes, and less than 1 log of sul and intI 1 genes, as well as 0.5 log to 1 log removal for target bacteria. Except for the total tetracycline concentration and the proportion of tetracycline-resistant bacteria ( R 2  = 0.330, P  < 0.05), there was no significant correlation between antibiotics and the corresponding resistant bacteria ( P  > 0.05). In contrast, various relationships were identified between antibiotics and antibiotic resistance genes ( P  < 0.05). Tet (A) and tet (B) displayed noticeable relationships with both tetracycline and combined antibiotic-resistant bacteria ( P  < 0.01).

Occurrence and removal of antibiotics and the corresponding resistance genes in wastewater treatment plants: effluents’ influence to downstream water environment

Li Jianan,Cheng Weixiao,Xu Like,Jiao Yanan,Baig Shams Ali,Chen Hong

In this study, the occurrence of 8 antibiotics [3 tetracyclines (TCs), 4 sulfonamides, and 1 trimethoprim (TMP)], 12 antibiotic resistance genes (ARGs) (10 tet , 2 sul ), 4 types of bacteria [no antibiotics, anti-TC, anti-sulfamethoxazole (SMX), and anti-double], and intI 1 in two wastewater treatment plants (WWTPs) were assessed and their influences in downstream lake were investigated. Both WWTPs’ effluent demonstrated some similarities, but the abundance and removal rate varied significantly. Results revealed that biological treatment mainly removed antibiotics and ARGs, whereas physical techniques were found to eliminate antibiotic resistance bacteria (ARBs) abundance (about 1 log for each one). UV disinfection did not significantly enhance the removal efficiency, and the release of the abundantly available target contaminants from the excess sludge may pose threats to human and the environment. Different antibiotics showed diverse influences on the downstream lake, and the concentrations of sulfamethazine (SM2) and SMX were observed to increase enormously. The total ARG abundance ascended about 0.1 log and some ARGs (e.g., tet C, intI 1, tet A) increased due to the high input of the effluent. In addition, the abundance of ARB variation in the lake also changed, but the abundance of four types of bacteria remained stable in the downstream sampling sites.

Action of oxytetracycline (OTC) degrading bacterium and its application in Moving Bed Biofilm Reactor (MBBR) for aquaculture wastewater pre-treatment

Sicheng Shao,Yongyou Hu,Jianhua Cheng,Yuancai Chen

In this study, the characteristics of biodegradation of oxytetracycline (OTC) by strain Ochrobactrum sp. KSS10 were studied under various environmental conditions, including initial OTC concentrations, variable temperature, initial pH, and diverse carbon sources. The capability of this bacterial strain for performing simultaneous OTC degradation and nitrate reduction was also explored under aerobic conditions . An OTC degradation ratio of 63.33% and a nitrate removal ratio of 98.64% were obtained within 96 h. In addition, removal of OTC and ammonia from synthetic aquaculture wastewater by a Moving Bed Biofilm Reactor (MBBR) and changes in the resistant genes of microbial communities were also investigated. The results demonstrated that the strain KSS10 was the dominant contributor in OTC and ammonia removal in the MBBR chamber. It removed almost all ammonia and approximately 76.42% of OTC. The abundances of genes tet L, tet X and intI 1 were reduced by the MBBR, but the abundance of tet G and tet M were increased due to horizontal and vertical gene transfers. Such a result can potentially be used by the strain KSS10 for removing antibiotics and nitrogen from aquaculture wastewater during pre-treatment.

The removal of antibiotic, antibiotic resistant bacteria and genes in persulfate oxidation system via activating by antibiotic fermentation dregs derived biochar

Yafei Wang,Chenhao Wang,Shen Yan,Yingchun Li,Chen Cai,Huiling Liu,Peng Ren,Mengmeng Wang,Shaoping Kuang

To reduce the adverse effects of antibiotics and antibiotic resistance genes (ARGs) in the environment, nitrogen-doped biochar (NLBH), derived from lincomycin fermentation dregs (LFD), was employed as an activator in a persulfate (PDS) oxidation system. The system was evaluated for its ability to degrade tetracycline (TC), inactivate antibiotic resistant bacteria (ARB) and remove ARGs. The degradation efficiency of TC in the NLBH/PDS system (70.1%) was higher than that in the pristine biochar/PDS system (10.3%). It was confirmed that the defects and edge pyridinic nitrogen generated in the nitrogen doping process were the reactive sites for PDS activation. According to electron paramagnetic resonance (EPR) and radical quenching experiments, the major mechanism for PDS activation was a non-radical pathway dominated by singlet oxygen ( 1 O 2 ). Radical pathways involving sulfate (SO 4 · - ) and hydroxyl radicals (·OH) were also at play in the NLBH/PDS system, but their role was minor. TC was principally degraded by hydroxylation , demethylation, and decarboxylation , and within 90 min, the NLBH/PDS system effectively inactivated 71.5% of ARB ( Pseudomonas sp. HLS-6). Intracellular ARGs (iARGs; sul 1, sul 2) and intI 1 had log reduction efficiencies of 2.73–4.04 and 2.70, respectively, whereas, extracellular ARGs ( sul 1 and sul 2) and intI 1 accumulated noticeably by 1.52–4.18-and 4.92-log, respectively. This work highlights a promising alternative technique for the removal of antibiotics, ARB and iARGs in future advanced wastewater treatment systems.

Removal of antibiotic resistance genes and pathogenicity in effluent from municipal wastewater treatment plant by plasma oxidation

Rongwei Zhong,Hu Li,Yangyang Wang,Ying Zhang,Jian Zhou,Tiecheng Wang

Municipal wastewater treatment plants (MWTP) are the main gathering places for antibiotic resistance genes (ARGs) and pathogenic bacteria. In this study, plasma oxidation was applied to remove the ARGs and inhibit pathogenicity in the effluent. The experimental results demonstrated that approximately 1.32 log, 1.34 log, 2.39 log, and 2.80 log of aac (3)-II , bla TEM-1 , Tet W, and Tet C were removed within 25 min of the plasma treatment, respectively. OH, 1 O 2 , O 3 , H 2 O 2 , NO 2 − , and NO 3 − all contributed to ARG removal. Coexisting dissolved organic matter was also oxidized, which was positively related to ARG removal. Plasma oxidation led to a decrease in the diversity and richness of microbial communities; Parcubacteria and Proteobacteria were the main potential hosts of ARGs and mobile genetic elements. Integron gene IntI 1 decreased by 2.46 log after plasma treatment, which was positively related to ARG removal. Gene expression related to ATP binding, DNA repair, gene transfer, and the cell membrane was reduced after plasma treatment, resulting in effective ARG removal. Furthermore, pathogenic genes related to lumpy skin disease, Niemann-Pick disease, polycystic kidney and hepatic disease were all down-regulated by plasma treatment. Overall, the plasma treatment inhibited the transmission risks of ARGs and pathogenicity in the effluent of MWTP.
