A Novel Method to Extract Juglone from Juglans mandshurica Waste Branches Using a Water-in-Oil Microemulsion

Li Chunying,Meng Xianming,Tian Mengfei,Li Shen,Tian Yao,Wang Tingting,Zhao Chunjian

Pruning of Juglans mandshurica produces many branches, which are potentially a rich source of juglone although they are usually discarded as waste. Herein, we describe a water-in-oil microemulsion-based microwave-assisted extraction (MBMAE) method to isolate the juglone found in waste branches from J. mandshurica . In our experiment, J. mandshurica waste branch powder was added to the MBMAE system. Using the optimized microemulsion system comprising Tween 80: n -propanol: n -hexane:water at 27:13.5:4.5:55 (w/w/w/w), and a pH of 5.6, at an operating temperature of 40 °C, operating time of 63 s, and operating power of 400 W, the juglone yield was 4.58 mg/g from a ratio of microemulsion to branch power of 20:1 (mL/g). The juglone extraction yield using the MBMAE method was 1.86- and 6.65-fold better than that obtained from microwave-assisted extraction by ethanol (Ethanol-MAE) and heat reflux extraction by ethanol (Ethanol-HRE), respectively. The MBMAE method may be useful as an alternative to traditional extraction methods to isolate juglone. Graphic Abstract

Juglone Suppresses Inflammation and Oxidative Stress in Colitis Mice

Chen Shuai,Wu Xin,Yu Zengli

Juglone (JUG), a natural product found in walnut trees and other plants, shows potent antioxidant, antimicrobial, and immunoregulatory activities. However, it remains unknown whether JUG can alleviate ulcerative colitis. This study aims to explore the effect of JUG on dextran sulfate sodium (DSS)-induced colitis in mice. The mice were randomly assigned into three groups: the vehicle group, the DSS group, and the JUG group. The experiments lasted for 17 days; during the experiment, all mice received dimethyl sulfoxide (DMSO, 0.03% v/v)-containing water, while the mice in the JUG group received DMSO-containing water supplemented with JUG (0.04 w/v). Colitis was induced by administering DSS (3% w/v) orally for 10 consecutive days. The results showed that the JUG treatment significantly ameliorated body weight loss and disease activity index and improved the survival probability, colon length, and tissue damage. JUG reversed the DSS-induced up-regulation of proinflammatory cytokines, including interleukin (IL)-6, 12, 21, and 23, and tumor necrosis factor-alpha, and anti-inflammatory cytokines, such as IL-10 and transforming growth factor-beta, in the serum of the colitis mice. Additionally, the activation of mitochondrial uncoupling protein 2 and phospho-Nuclear Factor-kappa B p65 and the inhibition of the kelch-like ECH-associated protein 1 and NF-E2-related factor 2 induced by DSS were also reversed under JUG administration. Although the JUG group possessed a similar microbial community structure as the DSS group, JUG enriched potential beneficial microbes such as Lachnospiraceae_NK4A136_group but not pathogens such as Escherichia Shigella, which was dominative in DSS group, at the genus level. In conclusion, our results demonstrated that JUG could be a promising agent for UC prevention to regulate inflammatory cytokines and oxidative stress.

Surface modification of FeⅢ-juglone coating on nanofiltration membranes for efficient biofouling mitigation

Jialin Song,Daliang Xu,Yonghui Han,Xuewu Zhu,Zihan Liu,Guibai Li,Heng Liang

Nanofiltration membranes have increasingly played a vital role in the purification of surface water and the recycling of wastewater. However, the problem of membrane biofouling, which leads to shortened service life and increased energy consumption, has hindered the widespread application of nanofiltration membranes. In this study, we developed functionalized nanofiltration membranes with anti-adhesive and anti-biofouling properties by coordinating Fe III and juglone onto commercial nanofiltration membranes in a facile and viable manner. Due to the hydrophilic nature of the Fe Ⅲ -juglone coating as well as its ultra-thin thickness and minimal impact on the membrane pores, the permeance of the optimally modified membrane even increased slightly (14 %). The outstanding anti-adhesive property of the Fe Ⅲ -juglone coating was demonstrated by a significant reduction in the adsorption of proteins and bacteria. Furthermore, the modified membranes exhibited lower flux decline amplitude and reduced biofilm deposition during dynamic fouling experiment, further supporting the outstanding anti-biofouling performance of the nanofiltration membrane after the modification with Fe Ⅲ -juglone coating. This study presents a novel and feasible approach for simultaneously improving the water permeance, anti-adhesive property and anti-biofouling property of commercial nanofiltration membranes.

Eco-friendly antifouling membranes via green coordination chemistry: Bioinspired CuII-Juglone@PDA modification membranes

Xinyi Sun,Mei Chen,Minliang Zhang,Yanli Li,Xingran Zhang,Ruobin Dai,Xin Wang

An ecofriendly modification mitigates membrane flux decline and extends operation lifespan. • Cu 2+ –juglone coating imparts efficient antibacterial activity to commercial membrane. • PDA layer enhances membrane hydrophilicity and prolongs antibacterial activity. • The optimal modified membrane exhibits significantly enhanced antifouling performance. Fouling remains a critical challenge limiting the widespread application of membrane technologies in water treatment. This study introduces a synergistic interface engineering strategy for polyvinylidene fluoride (PVDF) membranes by integrating Cu II –juglone coordination chemistry with polydopamine (PDA) coating. Conducted under ambient conditions (25 °C, pH 7.4), this green assembly process constructs a robust Cu II -Juglone network that facilitates dual-mode antibacterial activity while minimizing copper dependency. Subsequent PDA deposition further enhances surface hydrophilicity and ensures sustained antibacterial durability, concurrently mitigating organic foulant adhesion. Comprehensive characterization revealed that the optimized PVDF/Cu II -Juglone@PDA membrane exhibited an ∼8° decrease in water contact angle and a marked increase in surface electronegativity compared with PVDF membrane. These modifications collectively improved resistance to organic foulant adhesion, as evidenced by a 63.4% lower total organic carbon (TOC) accumulation than the pristine membrane. The optimized membrane achieved 95% antibacterial efficiency against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ), together with a 39.1% reduction in flux decline under dynamic filtration. This performance arose from synergistic antifouling effects: the PDA layer enhanced hydrophilicity and electrostatic repulsion via a hydration barrier, while the Cu II -juglone layer provided sustained antibacterial activity through metabolic disruption induced by Cu II /juglone release. These combined effects highlight an effective strategy for the eco-friendly fabrication of multifunctional antifouling membranes for sustainable water purification. Surface Deposition of Juglone/FeIII on Microporous Membranes for Oil/Water Separation and Dye Adsorption

Hui Yu,Qi-Zhi Zhong,Tian-Geng Liu,Wen-Ze Qiu,Bai-Heng Wu,Zhi-Kang Xu,Ling-Shu Wan

Deposition of dopamine and tannic acid has received great attention in the fields of surface and interface science and technology. The deposition behaviors of various metal–phenolic systems have been investigated, and it is generally accepted that at least one catechol group is essential to the formation of the coatings. Herein, we report a novel and effective surface-coating system based on the coordination complexes of FeIII ions with a natural product juglone that contains only one phenolic hydroxyl. We investigated the deposition behaviors of this novel system on various substrates. Microporous polypropylene membrane modified with juglone/FeIII coatings is superhydrophilic and underwater superoleophobic, showing high separation efficiency and good reusability for various oil/water emulsions. In addition, the modified membrane can adsorb anionic dyes and selectively remove them from dye mixtures with high efficiency. We further demonstrated that the coating is a result of the synergetic effect of juglone/FeIII coordination and FeIII hydrolysis. This work not only provides new insights into surface deposition systems but also expands the polyphenol family for surface coatings of multifunctional materials.

Synthesis, Biological Evaluation and Structure–Activity Relationship of Juglone Derived Naphthoquinones as Potential Antipsoriatic Agents

Tong Bu,Zile Gong,Yudong Ma,Lixia Dai,Yuchao Ma,Xiaoyan Yu,Xiaorong Yang,Xiaolou Miao,Xiaofei Shang

Psoriasis is a chronic, immune-mediated inflammatory skin disease for which the development of structurally novel and accessible small-molecule candidates remains of considerable interest. In this study, a series of juglone-derived naphthoquinone analogs was synthesized to explore the influence of substitution pattern on anti-inflammatory activity and cytotoxicity. Their biological profiles were first evaluated in LPS-stimulated HaCaT cells by combining cytotoxicity assessment with nitric oxide (NO) screening. Most derivatives showed reduced cytotoxicity compared with juglone, and preliminary structure–activity relationship analysis indicated that retention of a free hydroxyl group at the C-2 position was generally favorable for both reduction in NO release and cellular safety, whereas C-3 alkyl substitution tended to weaken activity and increase cytotoxicity. Among the tested compounds, compound11showed the most favorable balance between reduction in NO release and low cytotoxicity. Further evaluation showed that compound11reduced the protein levels of several inflammatory mediators in the culture supernatants of LPS-stimulated HaCaT cells, including TNF-α, IL-6, IL-1β, IL-17A, and IL-23, under the tested conditions. In an imiquimod-induced psoriasis-like mouse model, topical administration of compound11partially alleviated IMQ-induced psoriasis-like skin lesions, improved histopathological changes to some extent, and reduced selected inflammatory cytokine levels in serum and skin tissues under the tested conditions. Exploratory target prediction, molecular docking, and in silico ADMET analyses provided supportive computational insight into the biological profile of compound11. Overall, these findings suggest that juglone-derived naphthoquinones may serve as useful natural-product-inspired scaffolds for further anti-inflammatory optimization, and compound11warrants further investigation in psoriasis-related experimental models.

Yue Wu-heng,Qin Lan-qun,Cai Juan,Mei Rui,Qian Han-qing,Zou Zheng-yun

Objective To verrify the anti-tumor efficacy and toxicity between juglone (Jug) and Jug-loaded PLGA nanoparticles (Jug-PLGA-NPs). Methods Jug-PLGA-NPs were prepared by ultrasonic emulsification. The anti-tumor activity of Jug (2, 3, 4 µg/mL) and Jug-PLGA-NPs (Jug: 2, 3, 4 µg/mL) in vitro was measured by MTT assay and cell apoptosis analysis. The distribution, anti-tumor effect and biological safety in vivo was evaluated on A375 nude mice. Results With the advantage of good penetration and targeting properties, Jug-PLGA-NPs significantly inhibited proliferation and migration of melanoma cells both in vitro and in vivo ( P <0.05 or P <0.01) with acceptable biocompatibility. Conclusions Jug can inhibit the growth of melanoma but is highly toxic. With the advantage of sustained release, tumor targeting, anti-tumor activity and acceptable biological safety, Jug-PLGA-NPs provide a new pharmaceutical form for future application of Jug.

A synergistic study on the synthesis of juglone via photooxidation in a UV–Vis LED based photomicroreactor

Mohsin Pasha,Saier Liu,Minjing Shang,Min Qiu,Yuanhai Su

Photochemical transformations are mobilized by traceless photons of narrow or broad visible light wavelength and have been proven as a green and sustainable synthesis route for organic compounds. However, most of such investigations were restricted to only parametric studies, lacking in examining a synergistic involvement of actinometric, kinetics and mass transfer mechanisms on product yield. To address this vital issue, we executed the synthesis of natural product juglone via the photooxidation of 1,5-dihydroxynapthalene (DHN) mediated through singlet oxygen, which was produced by triplet–triplet energy transfer from three porphyrin-based photosensitizers to triplet oxygen upon irradiation from UV–Vis LED. Compared to the other two photosensitizers, m-TcPP ( meso -tetracarboxyphenylporphyrine) showed a higher molar extinction coefficient and remarkable energy transfer, resulting in higher DHN conversion and juglone yield. Full DHN conversion and 82% yield were obtained at the m-TcPP loading of 2.0 mol% and the residence time of 22.1 min. Actinometric investigation showed that the photon flux was significantly improved by rising m-TcPP and DHN concentrations. The maximum radiant power absorbed by the DHN photooxidation could reach 17.3% of the total input power of UV–Vis LED strip. The detailed reaction mechanism of the DHN photooxygenation was discussed through the Jablonski diagram and rate constants of intermediate steps were determined. A positive relationship between the photon flux and the apparent rate constant was found by rising m-TcPP concentration from 0.01 to 0.05 mM, while the rise in DHN concentration from 5.0 to 10.0 mM resulted in a negative correlation between these parameters. Consequently, a synergistic improvement of photon flux and apparent rate constant was necessitated to attain complete DHN conversion and high juglone yield.

A Sensitive and Selective UHPLC–MS/MS Method Based on Mercaptopropionic Acid Derivatization for Quantification of Juglone in Biological Matrices

Xinyue Zheng,Yue Deng,Si Cheng,Di Lu,Yibo Wang,Zichang Tian,Hongyu Xue,Lei Yin,Meiyun Shi

In this study, a highly sensitive and selective ultra-high performance liquid chromatography–tandem mass spectrometry (UHPLC–MS/MS) method was developed for the quantification of juglone, a poorly ionizable naphthoquinone, in biological matrices. To enhance ionization efficiency, pre-column derivatization was employed using sulfhydryl nucleophiles (β-mercaptoethanol and 3-mercaptopropionic acid) via Michael addition. Derivatization with mercaptopropionic acid significantly improved mass spectrometric response (50-fold increase) and minimized matrix effects compared to mercaptoethanol. The method, validated per FDA guidelines, exhibited excellent linearity ( r ≥ 0.995) from 3 to 150 ng/mL, with accuracy within −10.6% to 4.3% and precision between 1.29% and 5.34%. The lower limit of quantification was 3 ng/mL. Extraction recovery and matrix effects ranged from 99.84% to 102.06% and 92.75% to 98.98%, respectively, with derivatives demonstrating good stability. Cellular uptake studies in MCF-7 cells revealed time-dependent but limited intracellular accumulation of juglone, with the majority retained in the culture medium. This robust derivatization-based UHPLC–MS/MS approach provides a reliable tool for investigating juglone's pharmacokinetics and toxicological mechanisms.

A novel sono-activatable nanoreactor induces precision intratumoral juglone generation and caspase 3/GSDME-mediated pyroptosis for treatment of bladder cancer

Zhang Yuanfeng,Yang Enguang,Zhang Xinyu,Shi Yibo,Fan Guangrui,Wang Yuhan,Tian Junqiang,Wang Hanzhang,Dong Zhilong,Wang Yingru,Cheng Liang,Zhao Zhiwen,Zhang Yonghai,Wang Baodui,Wang Zhiping

Background Juglone, a natural naphthoquinone compound, exhibits potent anticancer activity but faces clinical limitations due to poor solubility, low bioavailability, and systemic toxicity. While tumor microenvironment-responsive drug activation strategies offer a promising solution, achieving precise spatial control over the conversion of non-toxic precursors into cytotoxic agents remains a significant challenge. Methods We developed DHN@Pt/PtMBCPsNSs, a platinum (Pt)-methylene blue (MB)- based nanoscale sonosensitizer, to enable ultrasound-triggered on-demand juglone synthesis within tumors. The system was evaluated in bladder cancer models including cell lines, patient-derived organoids (PDOs) and patient-derived tumor xenograft (PDX) models, assessing tumor uptake, intracellular distribution, hypoxia modulation (“H 2 O 2 -O 2 -¹O 2 ” cascade), oxidative stress markers (SOD, GSH, MDA, GSSG), and cell death pathways (apoptosis/pyroptosis). Transcriptome sequencing was performed to elucidate molecular mechanisms. Results DHN@Pt/PtMBCPsNSs were internalized by UMUC-3 and T24 cells and primarily accumulated within the mitochondrial compartments. The platinum-based components catalytically exhausted GSH via redox cycling. DHN@Pt/PtMBCPsNSs triggered the production of O 2 and ¹O 2 after local US irradiation, alleviating hypoxia and selectively converting dihydroxynaphthalene (DHN) to juglone in tumors, which reducing the systemic toxicity of juglone. The nanosystem further disrupted cellular redox balance by triggering depletion of antioxidant defenses (SOD and GSH) and reducing the ratio of GSH/GSSG. In vitro cell experiments and PDOs experiments demonstrated that DHN@Pt/PtMBCPsNSs combined with US irradiation can significantly inhibit bladder cancer cell proliferation, induce apoptosis, and regulate pyroptosis via the ROS-mediated caspase 3/GSDME signaling pathway. In the PDX model, DHN@Pt/PtMBCPsNSs accumulated at the tumor site and significantly inhibited tumor growth under US irradiation. Conclusions As a novel sonosensitizer and pyroptosis inducer, DHN@Pt/PtMBCPsNSs achieve a precise synergistic effect of chemotherapy and sonodynamic therapy within cells, while minimizing adverse effects on normal cells and overcoming the limitations associated with natural chemotherapeutic agents. This approach may advance clinical translation of natural anticancer agents. Graphical

Back extraction combined with magnetic solid phase extraction based on bubble column for separation and purification of juglone from Juglans mandshurica Maxim. coproducts

Mengfei Tian,Xianming Meng,Naveed Ahmad,Chunying Li,Zhanyu Yuan,Zidan Luo,Yu Zhang,Chunyu Liang,Chunjian Zhao

Juglone was first preliminary purified by back extraction (BE) which also completed the sustainability of microemulsion as a solvent for extraction of juglone in J. mandshurica Maxim. branches. Then, magnetic solid phase extraction (MSPE) based on bubble column (BC) was proposed to further purify the juglone, GO/ND/Fe 3 O 4 as adsorbent, which broke through the limitation that MSPE is only suitable for trace enrichment. Thus, a BE combined with MSPE based on BC (BE-MSPE/BC) technology to separate and purify juglone from J. mandshurica Maxim. was developed. Under the optimal conditions (optimal adsorption conditions: 0.5 h of adsorption time, 30 mg/mL of solid-liquid ratio, 5.5 of pH, 60 L/min of air flow; optimal desorption conditions: 0.4 h of desorption time, methanol of desorption agent, 100% of desorbent volume fraction, 8 of pH, 40 mg/mL of solid-liquid ratio, 80 L/min of air flow rate), the recovery yield and purity of juglone obtained by BE-MSPE/BC were 80.57% and 72.38%, respectively. BE-MSPE/BC, as a green separation and purification technology that can easily achieve large-scale and sustainable production, is expected to be an alternative method to separate and purify active components in biomass materials and products.
