Kasugamycin (KSM), an aminoglycoside antibiotic, has been widely used as a natural fungicide to control plant diseases, particularly for managing rice blast. However, its uptake mechanism and transport in rice remain to be explored. In this article, rice seedlings were treated by the foliar spraying method, and the content of KSM in rice leaves, stems and roots under different treatments was detected by high-performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS), respectively. Results showed that leaf-applied KSM could be transported to the phloem and migrate to roots and stems after uptake by leaves. Concentration, temperature and pH had significant effects on the uptake of KSM. Compared with the control, the competitive inhibitors d -glucose and phlorizin both inhibited the uptake of KSM, demonstrating that sugar transporter proteins were involved in the uptake process. The energy inhibitors dinitrophenol (DNP) and carbonyl cyanide chlorophenylhydrazone (CCCP) also significantly inhibited the uptake of KSM, indicating that the uptake of KSM required energy consumption. Thus, the uptake of KSM by rice was an active process involving sugar transporter proteins, and it could migrate downward through the phloem. This study contributes to the promotion of the scientific application of antibiotics and the biological control of crop diseases. It will also provide a theoretical basis for the development of root-targeted pesticides and transport pesticides with phloem mobility.

The discovery of novel chiral selectors always fascinates us. This work describes the chiral separation performances of a new chiral selector (kasugamycin, KAS) in capillary electrophoresis (CE) for six pairs of stereoisomers, including ephedrine and pseudoephedrine, quinine and quinidine, cinchonine and cinchonidine, and amlodipine, promethazine and ofloxacin enantiomers. Kasugamycin, an aminoglycoside antibiotic in agriculture, shows significant biological activity against rice blast with low toxicity. As it turns out, this new chiral selector possesses good CE compatibility and stereoselectivity towards model analytes. In this work, we systematically investigated several separation parameters including kasugamycin concentration, buffer pH, separation voltage and the composition of the buffer solution. A detailed discussion about the chiral recognition mechanism was made based on Statistical Product and Service Solution (SPSS) analysis, NMR experiments (1D and 2D) and molecular modeling. This is the first time that kasugamycin is utilized as a chiral selector in CE, and the development of new chiral selectors from agricultural or veterinary antibiotics deserves more attention.

Kasugamycin (KSM), an aminoglycoside antibiotic, has been widely used for the management of plant diseases, especially for the control of rice blast in Asia. However, its uptake mechanism and transport in plants are still obscure. The castor bean (Ricinus communis L.) seeding, a model plant for phloem transport, was used to study the mechanism of uptake and transport of KSM. Results showed that cotyledon-applied KSM could transport into the phloem and distributed in root and shoot of plant. The temperature, concentration, and pH had significant effects on the uptake of KSM, indicating that the uptake of KSM was mediated by an active carrier system. Compared with the control, competitive inhibitors of sugar transporters D-glucose, D-chiro-inositol, and phloridzin inhibited 71.03%, 67.95%, and 61.73% uptake of KSM, respectively. Energy inhibitor dinitrophenol (DNP) and carbonyl cyanide chlorophenylhydrazone (CCCP) also affected the uptake of KSM, and the inhibition rates were 34.23% and 48.06%. All the results showed that the uptake of KSM was mediated by a sugar transporter, and it could transport from shoot to root in plants via the phloem. The study preliminary elucidated the plant–microbe interactions in the context of the transport of microbial secondary metabolites in plants. It has certain significance for scientific application of antibiotics and biological control of plant diseases and provides theoretical basis for the development of bidirectional transport pesticides.

Controlled release formulations can reduce the losses of pesticides and prolong their effective duration. In this work, a novel pH responsive pesticide delivery system based on kasugamycin-conjugated ZnO quantum dots (KAS-ZnO QDs) was established via a cleavable benzoic–imine covalent bond. The as-prepared KAS-ZnO QDs could protect KAS against photodegradation effectively due to the UV shielding effect of KAS-ZnO QDs. The KAS and Zn 2+ released from KAS-ZnO QDs in the acid condition could achieve a synergistic antimicrobial activity. The greenhouse experiment confirmed that application of KAS-ZnO QDs had a longer duration and a better antimicrobial activity against bacterial fruit blotch compared to KAS and NH 2 -ZnO QDs treatments, and showed no any toxicity to plants. Thus, pH-responsive KAS-ZnO QDs could achieve a synergistic antimicrobial effect and offer a new approach to manage plant diseases.

Membrane biofouling remains a major obstacle to the long-term efficiency of nanofiltration (NF) processes in water treatment. In this study, a novel dual-mechanism antibacterial NF membrane was fabricated by integrating an antibiotic monomer and silver nanoparticles (Ag NPs). Kasugamycin, an environmentally compatible aminoglycoside antibiotic, was employed as a functional aqueous monomer to replace traditional amines. The hydrophilic polyester-amide separation layer was constructed via interfacial polymerization, simultaneously incorporating the antibacterial groups. Ag NPs were subsequently immobilized in situ onto the membrane surface through a polydopamine interlayer, constituting the dominant antibacterial component. The resulting Kas-TMC-PDA-Ag membrane exhibited a high Na 2 SO 4 rejection of 98.81% and a competitive permeance of approximately 15 L m −2 h −1 bar −1 . Owing to the dominant bactericidal action of Ag NPs via silver ion release and reactive oxygen species (ROS) generation, complemented by the auxiliary role of kasugamycin in disrupting bacterial protein synthesis, the membrane achieved 100% antibacterial efficiency against both E. coli and S. aureus , while effectively suppressing biofilm formation. Furthermore, it displayed superior biofouling resistance compared to a commercial NF270 membrane, as evidenced by a higher flux recovery ratio (FRR) after bacterial fouling, and maintained excellent dynamic antifouling performance during the filtration of complex lake water with a FRR of 89.1%. With low silver leaching, exceptional operational and chemical stability, this study provides a high-efficiency approach for fabricating biofouling-resistant NF membranes for advanced water treatment.

Kasugamycin (KSM) is easily affected by photolysis, acid–base destruction, and oxidative decomposition in the natural environment, leading to its poor durability and low effective utilization rate, which affects its control effect on plant bacterial diseases. Nanomaterials modified with environment-responsive agents enable the control of the release of pesticides through intelligently responding to external stimuli, thereby improving efficacy and reducing environmental impact. In this study, a pH-responsive controlled release system was constructed using zeolitic imidazolate frameworks (ZIF-93) for the sustained and targeted delivery of KSM. The synthesized KSM@ZIF-93 exhibited a diameter of 63.93 ± 11.19 nm with a drug loading capacity of 20.0%. Under acidic conditions mimicking bacterial infection sites, the Schiff base bonds and coordination bonds in ZIF-93 dissociated, triggering the simultaneous release of KSM and Zn2+, achieving a synergistic antibacterial effect. Light stability experiments revealed a 34.81% reduction in UV-induced degradation of KSM when encapsulated in ZIF-93. In vitro antimicrobial assays demonstrated that KSM@ZIF-93 completely inhibitedErwinia amylovoraat 200 mg/L and had better antibacterial activity and persistence than KSM and ZIF-93. The field experiment and safety evaluation showed that the control effect of KSM@ZIF-93 on pear fire blight at the concentration of 200 mg/L was (75.19 ± 3.63)% and had no toxic effect on pollen germination. This pH-responsive system not only enhances the stability and bioavailability of KSM but also provides a targeted and environmentally compatible strategy for managing bacterial infections during the flowering period of pear trees.

A Cu–N-centered DT–Cu nanozyme enables dual-channel KSM readout. • Chromatography-free sensing enables on-site screening and FL confirmation. • For the first time, B-CDs-referenced readouts cross-validate KSM in food matrices. • The rapid method achieves a low LOD and 90.0–103.6% recoveries in food samples. Rapid screening and quantitative confirmation of kasugamycin (KSM) residues in foods remain challenging without chromatographic instruments. Herein, a low-cost colorimetric/dual-excitation ratiometric fluorescent platform was developed using 3,5-diamino-1,2,4-triazole-coordinated copper nanozymes (DT–Cu). Characterization verified Cu–N-associated active sites and homogeneous elemental distribution. DT–Cu catalyzed H 2 O 2 -mediated oxidation of Ampliflu Red to resorufin, whereas KSM inhibited its peroxidase-like activity through coordination/hydrogen-bonding-associated interactions, causing color fading and red fluorescence attenuation. Blue carbon dots were introduced as an internal reference to construct an I 435 /I 584 readout, improving resistance to matrix and instrumental fluctuations. Under optimized conditions, KSM was detected over 0.001–50 μg mL −1 with a detection limit of 2.31 pg mL −1 within 40 min. The probe showed good 7-day storage stability. Recoveries of 90.0%–103.6% with RSDs of 0.11%–5.89% were obtained in real samples, demonstrating its potential for rapid on-site screening. The construction of an environmentally responsive bactericidal platform enables intelligent and synergistic regulation of bacterial diseases. A pH-responsive pesticide nano delivery system (ZnO@PDA-KAS) was prepared by self-polymerizing a polydopamine (PDA) coating on the surface of ZnO nanospheres synthesized via a hydrothermal method, followed by conjugation with kasugamycin (KAS) in this work. The results showed that ZnO@PDA-KAS exhibited excellent cumulative release under acidic conditions, with a pesticide loading content of 9.4%. The presence of PDA significantly enhanced the adhesion and retention of nanoparticles on rice leaves, whereas fluorescein isothiocyanate-labeled ZnO@PDA exhibited good translocation capacity in rice plants. Moreover, the PDA coating effectively improved the photostability of KAS. In vivo and in vitro antibacterial tests demonstrated that ZnO@PDA-KAS displayed superior antibacterial activity against Xanthomonas oryzae pv. Oryzicola ( Xoc ) due to the synergistic antibacterial effects of Zn 2+ and KAS. This enhanced activity was attributed to the nanoparticles' ability to severely damage bacterial microstructures, enhance rice enzyme activity, and more effectively inhibition of bacterial biofilm formation and induced more severe DNA damage in Xoc cells. Finally, the ZnO@PDA-KAS nanoparticles showed excellent biocompatibility with rice plants and L02 cells. Therefore, the environmentally responsive bactericide ZnO@PDA-KAS provides effective agricultural guidance for efficient and precise release for the management of bacterial plant diseases.
