Aquatic photolysis of strobilurin fungicide kresoxim-methyl: kinetics, pathways, and effects of adjuvants

Xuewei Zhang,Jing Ye,Zhigang Ni,Xuerui Yang,Yuefei Ji,Jean-Marc Chovelon,Guangli Xiu,Lei Zhou

Kresoxim-methyl (KM), an extensively used strobilurin fungicide, displays broad-spectrum fungicidal activity. Despite its prevalent application in agriculture and frequent detection in the environment, specific studies on the photolytic fate of KM in natural surface waters are quite limited. To address this, the present study delves into the photochemical transformation of KM under light irradiation using a high-pressure mercury lamp. Photodegradation of KM followed pseudo-first-order kinetics, with a degradation rate constant of 0.067 ± 0.005 h−1, corresponding to a half-life of approximately 10.3 ± 0.7 h, and the quantum yield was estimated to be 6.95 × 10−3. Transient species identification experiments demonstrated the involvement of singlet oxygen (1O2) and singlet (1KM*) and triplet (3KM*) states. Four different photodegradation pathways, i.e., photoisomerization, hydrolysis, hydroxylation, and ether cleavage, were proposed. Toxicity assessments demonstrated that photodegradation effectively reduced the ecotoxicity of KM. Two typical pesticide adjuvants, namely, Tween 20 and sodium dodecylbenzenesulfonate (SDBS), were selected to evaluate their impacts on KM photolysis. While Tween 20 exhibited a remarkable increase in the photodegradation of KM, SDBS showed a concentration dependent behavior, with an inhibitory effect at low concentrations (<200 μM) and promotional effects at higher concentration levels. The present study elucidated the photolytic KM degradation mechanism, as well as its possible inhibition and enhancement, which is of significant importance for evaluating the environmental fate and ecotoxicological risks of KM in sunlit surface waters.

Residue levels and dietary risk evaluation of prothioconazole-desthio and kresoxim-methyl in cucumbers after field application in twelve regions in China

Chao Dong,Jiye Hu

A commercial formulation called prothioconazole·kresoxim-methyl 50% water dispersible granule (WG), used to control the powdery mildew, is under registration for use on cucumbers. Therefore it is urgent to validate the reliability of the recommended good agricultural practices (GAP) conditions (187.5 g a.i. ha−1, three sprays with a 7-d interval, and a pre-harvest interval of 3 d) by conducting field trials in 12 regions in China, as required by national regulations, with a subsequent risk assessment. The residues of prothioconazole-desthio and kresoxim-methyl in field samples were determined using QuEChERS coupled with high-performance liquid chromatography – tandem mass spectrometry (HPLC-MS/MS). On the suggested pre-harvest interval (PHI, 3 d), the residual concentrations of prothioconazole-desthio (no maximum residue limit set in China) and kresoxim-methyl (maximum residue limit = 0.5 mg/kg) in cucumbers were 0.01–0.020 and 0.01–0.050 mg/kg, respectively. The acute risk quotients of prothioconazole-desthio in cucumbers were no higher than 0.079% for Chinese consumers. The chronic dietary risk quotient of kresoxim-methyl and of prothioconazole-desthio for different groups of consumers in China ranged from 2.3% to 5.3% and from 1.6% to 4.6%, respectively. Thus, prothioconazole·kresoxim-methyl 50% WG sprayed on cucumbers under the recommended GAP information can pose a negligible risk for Chinese consumers.

Histology and metabonomics reveal the toxic effects of kresoxim-methyl on adult zebrafish

Nan Fang,Changpeng Zhang,Haoze Hu,Yanjie Li,Xiangyun Wang,Xueping Zhao,Jinhua Jiang

Studies have shown that kresoxim-methyl (KM) and other strobilurin fungicides have toxic effects on aquatic organisms. However, the potential deleterious effects of kresoxim-methyl (KM) on adult zebrafish regarding the ecological risk of environmental concentration remain unclear. Here, the histology and untargeted metabonomics was used to investigate the adverse effect on female zebrafish after exposure to KM at environmental concentration, aquatic life benchmark and one-half LC 50 of adult zebrafish. Results demonstrated KM affected zebrafish liver, ovary and intestine development, blurred the boundary between hepatocytes or caused hepatic vacuoles, increased the percentage of perinucleolar oocyte and cortical alveolus oocyte, decreased intestinal goblet cells and disturbed villus and wall integrity after 21 d exposure. Metabonomics showed different concentrations of KM simultaneously influenced the metabolites annotated to vitamin digestion and absorption, serotonergic synapse, retinol metabolism, ovarian steroidogenesis and arachidonic acid (AA) metabolism in zebrafish liver. Results showed the decreased triglyceride and cholesterol levels, as well as the metabolic alterations in amino acid, lipid, vitamin and retinol metabolism caused by KM, might disturb the energy supply for normal liver development and oocyte maturation. In addition, KM altered the transcription of Tdo2a , Tdo2b , Ido1 , Cxcl8b , Cyp7a , Cyp11a , Cyp11b , Cyp17a , Cyp19a , Hsd3β , Hsd17β , Pla2 , Ptgs2a and Ptgs2b , the level of TG, TC, MDA, IFN, IL6 and Ca 2+ , and the activity of CAT , SOD Ca 2+ -ATPase in zebrafish liver. Moreover, cytoscape analysis suggested the disturbed AA metabolism caused by KM, might interconnect multiple metabolic pathways to share implicated function in the regulation of oocyte maturation and immune response. Current study brought us closer to an incremental understanding of the toxic mechanism of KM on adult zebrafish, indicated there was crosstalk among different regulatory pathways to regulate the metabolic disorders and biologically hazardous effects induced by KM.

The Disappearance Behavior, Residue Distribution, and Risk Assessment of Kresoxim-Methyl in Banana (Musa nana Lour.) Based on a Modified QuEChERS Procedure Using HPLC-MS/MS

Wang Siwei,Sun Haibin,Liu Yanping,Wang Xiaonan,Chang Hong

A modified-QuEChERS method to determine kresoxim-methyl in banana and soil was developed and validated via high-performance liquid chromatography–tandem mass spectrometry (HPLC-MS/MS). The dissipation behavior, residue distribution, and risk assessment of kresoxim-methyl in banana were further investigated based on this method. The dissipation behavior of kresoxim-methyl in banana and soil was described by using first-order kinetics and its half-life, 4.8–5.7 days and 5.5–6.5 days, respectively. The concentrations of kresoxim-methyl were 0.03–0.08 mg kg−1, 0.06–0.17 mg kg−1, and <0.01 mg kg−1 for a whole banana, peel, and pulp, respectively, on the basis of spraying three times the recommended dosage and pre-harvest interval (PHI, 42 days). The results showed that the risk quotient of kresoxim-methyl in banana to people exhibited an acceptable low dietary risk. This current study could help in guiding the scientific and proper usage of this formulation.

The Dissipation Kinetics, Residue Level and Dietary Risk of Kresoxim-Methyl in Rosa roxburghii and Soil Based on the QuEChERS Method Coupled with LC–MS/MS

Zheng Handinghong,Wu Qiong,Wu Xiaomao

This study aimed to investigate the dissipation, residues and dietary assessment of kresoxim-methyl in the application of Rosa Roxburghii and soil field using liquid chromatography-tandem mass spectrometry (LC–MS/MS). The results show that kresoxim-methyl in R. roxburghii samples was extracted by acetonitrile and purified by ethyl enediamine-N-propylsilane (PSA), while kresoxim-methyl in soil samples was extracted by acetonitrile and purified by octadecylsilyl solid phase dispersant (C 18 ). 0.1% formic acid (v/v)-water–methanol solution was used as the mobile phase, LC–MS/MS exhibited a good linearity in the range of 0.001–10 mg L −1 . The recoveries of R. roxburghii and soil matrix were 82.48%–102.55%, and the relative standard deviation ( RSD) were 1.13%–4.21%. The limit of detection ( LOD ) and quantification ( LOQ ) of kresoxim-methyl in R. roxburghii and soil samples was 0.50 and 0.60 µg kg −1 , respectively. The dissipation dynamics of kresoxim-methyl in R. roxburghii and soil followed the first-order kinetics, with the half-life of 4.28 and 4.41 days, respectively. The terminal residual amount of kresoxim-methyl in R. roxburghii and soil samples was 0.003–1.764 and 0.007–2.091 mg kg −1 , respectively. The dietary intake risk assessment indicates that a risk quotient ( RQ ) for kresoxim-methyl based on the national estimated daily intake ( NEDI ) of 0.1995 mg was 0.79%, suggesting that the use of kresoxim-methyl on R. roxburghii at recommended dosage was safe to consumers. This study provides the theoretical basis for guiding the rational use of kresoxim-methyl in the production of R. roxburghii .
