A heat shock protein protects against oxidative stress induced by lambda-cyhalothrin in the green peach aphid Myzus persicae

Bao Dong,Xi-Ya Liu,Bin Li,Mao-Ye Li,Shi-Guang Li,Su Liu

Lambda-cyhalothrin (LCT) is a pyrethroid insecticide widely used to control insect pests . Insect exposure to LCT may cause abnormal accumulation of reactive oxygen species (ROS) and result in oxidative damage. Heat shock proteins (HSPs) may help protect against oxidative stress . However, little is known about the role of HSPs in response to LCT in the green peach aphid , Myzus persicae . This insect is an important agricultural pest causing severe yield losses in crops. In this study, we characterized a cDNA sequence ( MpHsp70 ) encoding a member of the HSP70 family in M. persicae . MpHsp70 encoded a 623 amino acid protein putatively localized in the cytosol. The highest expression level of MpHsp70 occurred in fourth-instar nymphs. Treatment of M. persicae with LCT resulted in oxidative stress and significantly increased H 2 O 2 and malondialdehyde levels. This led to an elevated transcription level of MpHsp70 . Injection of H 2 O 2 into M. persicae also upregulated the MpHsp70 expression level, suggesting that MpHsp70 is responsive to ROS, particularly H 2 O 2 , induced by LCT. Recombinant MpHSP70 protein was expressed in Escherichia coli . E. coli cells overexpressing MpHSP70 exhibited significant tolerance to H 2 O 2 and the ROS generators, cumene hydroperoxide and paraquat . This indicated that MpHSP70 protects against oxidative stress. Furthermore, knockdown of MpHsp70 by RNA interference resulted in increased susceptibility in apterous adults of M. persicae to LCT. These findings indicate that MpHsp70 plays an important role in defense against LCT-induced oxidative stress and insecticide susceptibility in M. persicae .

Identification of G Protein-Coupled Receptors (GPCRs) Associated with Lambda-Cyhalothrin Detoxification in Cydia pomonella

Yu-Xi Liu,Chao Hu,Yu-Ting Li,Ping Gao,Xue-Qing Yang

While previous studies have reported G protein-coupled receptor (GPCR)-mediated insecticide resistance in various arthropods, the understanding of GPCR-associated resistance mechanisms in Cydia pomonella remains limited. In this study, a total of 95 CpGPCR genes categorized into four families were identified in C. pomonella. Results revealed high expression levels of the majority of the CpGPCRs during the first larval stage and in the head of C. pomonella. Exposure to lambda-cyhalothrin significantly increased the expression of 15 CpGPCRs, including CpGPCR70, which is highly expressed in all larval stages and shows the highest expression in the midgut. RNA interference (RNAi) demonstrated that downregulation of CpGPCR70 leads to reduced expression of key resistance-related genes and a decreased tolerance of larvae to lambda-cyhalothrin. These findings indicate that CpGPCR70 plays a crucial role in regulating the expression of detoxifying genes involved in lambda-cyhalothrin resistance, offering valuable insights for the development of more effective pest control strategies.

Temperature-Dependent Nanogel for Pesticide Smart Delivery with Improved Foliar Dispersion and Bioactivity for Efficient Control of Multiple Pests

Yue Shen,Changcheng An,Jiajun Jiang,Bingna Huang,Ningjun Li,Changjiao Sun,Chong Wang,Shenshan Zhan,Xingye Li,Fei Gao,Xiang Zhao,Haixin Cui,Ravi Gooneratne,Yan Wang

The use of nanomaterials and nanotechnology to construct a smart pesticide delivery system with target-oriented and controlled-release functions is important to increase the effective utilization rate and minimize environmental residue pollution. A temperature-dependent delivery system can modulate the release of pesticide with temperature to improve the efficacy and precision targeting. A series of poly(N-isopropylacrylamide) (PNIPAM)-based nanogels with high deformability and tunable structure were successfully constructed for smart pesticide delivery and effective pest control. A lambda-cyhalothrin (LC)-loaded Pickering emulsion (LC@TNPE) with a stable gel-like network structure was further formed by the temperature-dependent nanogel to encapsule the pesticide. The foliar wettability, photostability, and controlled-release property of LC@TNPE were effectively enhanced compared to the commercial formulation because of the encapsulation and stabilization of nanogel. The release rate of LC positively correlated with temperature changes and thereby adapted to the trend of pest population increase at higher temperature. The LC@TNPE displayed improved control efficacy on multiple target pests including Plutella xylostella, Aphis gossypii, and Pieris rapae compared with the commercial suspension concentrate and microcapsule suspension, and it showed marked efficacy to control Pieris rapae for an extended duration even at a 40% reduced dosage. Furthermore, the safety was evaluated systematically on cells in vitro and with a nontarget organism. Studies confirmed that the system was relatively safe for HepG2 cells and aquatic organism zebrafish. This research provides an insight into creating an efficient and environmentally friendly pesticide nanoformulation for sustainable agriculture production.

Fabrication and evaluation of molecularly imprinted multi-hollow microspheres adsorbents with tunable inner pore structures derived from templating Pickering double emulsions

Jianming Pan,Yijie Yin,Mengyin Gan,Minjia Meng,Xiaohui Dai,Runrun Wu,Weidong Shi,Yongsheng Yan

In this work, lignin was simply tailored by chemical modification with oleic acid, and then surfactant-free water-in-oil-in-water (W 1 /O/W 2 ) Pickering double emulsions were formed by stabilizing two types of lignins (original and modified) at both interfaces. Based on as-prepared W 1 /O/W 2 emulsions, three kinds of molecularly imprinted multi-hollow microspheres (denoted as MIMMs1, MIMMs2 and MIMMs3) were fabricated and evaluated as adsorbents to selective removal of lambda-cyhalothrin (LC). MIMMs exhibited tunable pore structures (size and number of inner pores) by adjusting the volume ratio of internal water phase to the medium oil phase (W 1 O), and MIMMs2 showed the larger adsorption capacity than those of MIMMs2 and MIMMs3, resulting from the common action mechanism of more and larger inner pores. Static adsorption experiments showed that equilibrium time was about 3.0   h, and the equilibrium adsorption amount was 24.79   mg   g −1 for MIMMs2. The selective recognition experiments exhibited the obviously selective adsorption effect of the MIMMs2 for target LC, and MIMMs2 could be effectively regenerated and recycled at less seven times without significant loss of adsorption capacity. Overall, batch mode data suggested that MIMMs2 may be useful for the selective removal of LC from aqueous media.

The effect of lambda-cyhalothrin nanocapsules on the gut microbial communities and immune response of the bee elucidates the potential environmental impact of emerging nanopesticides

Dezheng Guo,Zhongyu Li,Yiwen Zhang,Wei Zhang,Chen Wang,Da-xia Zhang,Feng Liu,Zheng Gao,Baohua Xu,Ningxin Wang

Emerging nanopesticides are gradually gaining widespread application in agriculture due to their excellent properties, but their potential risks to pollinating insects are not fully understood. In this study, lambda-cyhalothrin nanocapsules (LC-NCs) were constructed by electrostatic self-assembly method with iron mineralization optimization, and their effects on bee gut microbial communities and host immune-related factors were investigated. Microbiome sequencing revealed that LC-NCs increase the diversity of gut microbial communities and reduce the complexity of network features, disrupting the overall structure of the microbial communities. In addition, LC-NCs also had systemic effects on the immune response of bees, including increased activity of SOD and CAT enzymes and expression of their genes, as well as downregulation of Defensin1 . Furthermore, we noticed that the immune system of the host was activated simultaneously with a rise in the abundance of beneficial bacteria in the gut. Our research emphasizes the importance of both the host and gut microbiota of holobiont in revealing the potential risks of LC-NCs to environmental indicators of honey bees, and provides references for exploring the interactions between host-microbiota systems under exogenous stress. At the same time, we hope that more research can focus on the potential impacts of nanopesticides on the ecological environment.
