Linalool and 1,8-Cineole as Constitutive Disease-Resistant Factors of Norway Spruce Against Necrotrophic Pathogen Heterobasidion Parviporum

Kai Wang,Wenzi Ren,Liang Hong,Qingao Wang,Rajendra Ghimire,Matti Haapanen,Minna Kivimäenpää,Pengfei Wu,Xiangqing Ma,Fred O. Asiegbu

Norway spruce is an important coniferous species in boreal forests. Root and stem rot diseases caused by the necrotrophic pathogen Heterobasidion parviporum threaten the wood production of Norway spruce which necessitates the search for durable control and management strategies. Breeding for resistant traits is considered a viable long-term strategy. However, identification of potential resistant traits and markers remains a major challenge. In this study, short-term disease resistance screening was conducted using 218 Norway spruce clones from 17 families. Disease resistance was evaluated based on the size of necrosis lesion length following infection with the pathogen. A subset of needles/branches from clones with small (partial resistant) or large (susceptible) lesions were used for terpene analysis and transcriptomic profiling. The results revealed that the content of monoterpene linalool and 1,8-cineole and their respective encoded genes were significantly more abundant and highly expressed in the partial resistant group. Furthermore, linalool and 1,8-cineole were demonstrated to have inhibitory effect on the growth of the pathogen H. parviporum , with morphological distortion of the hyphae. RNAseq analysis revealed that transcript of pathogen genes involved in the regulation of carbohydrate metabolism and stress responses were significantly decreased in presence of the terpenes. The results suggest the relevance of monoterpenes together with jasmonic acid precursor and some genes involved in phenylpropanoid biosynthesis, as constitutive tolerance factors for Norway spruce tolerance against necrotrophic pathogen. The high level of necrosis related cell death gene expression might be factors critical for host susceptibility and disease development.

Oxidized amylose with high carboxyl content: A promising solubilizer and carrier of linalool for antimicrobial activity

Ying Zhou,Youxin Ye,Wenwen Zhang,Songling Li,Jing Chen,Shiting Wang,Defu Li,Changdao Mu

The oxidized amyloses with different carboxyl content were prepared to include linalool for antimicrobial activity in aqueous environment. The results show that linalool can be effectively reserved from volatilization through encapsulation into amylose and oxidized amyloses. The inclusion ability of oxidized amyloses towards linalool is decreasing with the increase of oxidation level due to the depolymerization of amylose. However, the solubilization effect of oxidized amyloses to linalool is enhanced efficiently owning to the high water solubility of oxidized amyloses. It is interesting that the inclusion complexes have good antimicrobial activity in aqueous environment. Linalool solubilized by oxidized amyloses presents better antimicrobial performance than that solubilized by amylose, mainly resulting from that amylose-linalool inclusion complex would aggregate and retrograde fast in aqueous solution, which is disadvantageous for the release of linalool. The study suggests that oxidized amylose is a promising solubilizer and carrier of linalool for antimicrobial activity in aqueous environment.

Evaluation of the phytotoxic effect of the essential oil from Artemisia absinthium

Chunyu Jiang,Shixing Zhou,Lin Liu,Zokir Toshmatov,Ling Huang,Kai Shi,Chi Zhang,Hua Shao

The chemical profile and the phytotoxicity of Artemisia absinthium essential oil (EO) were investigated to evaluate its potential value as a biopesticide for food safety purposes. A total of 54 compounds were identified in A. absinthium EO, with the most abundant constituents being eucalyptol (25.59%), linalool (11.99%), and β-myrcene (10.05%). The EO, linalool, and a mixture of three major components exhibited potent suppressive activity against four receiver species; however, eucalyptol and β-myrcene showed a much weaker effect. Bioassay-guided fractionation led to the isolation of linalool as the major active compound responsible for the EO’s phytotoxicity. Subsequent scanning electron microscopy (SEM) analysis revealed that linalool significantly inhibited root-hair formation and metaxylem development. This is the first report on the determination of linalool as the major active phytotoxic compound in A. absinthium EO, as well as the elucidation of its mechanism of phytotoxicity from the perspective of root structure changes in the receiver species. Our results suggest that both the EO and its major constituents have potential value as environmentally friendly herbicides.

Enzyme Catalytic Efficiencies and Relative Gene Expression Levels of (R)-Linalool Synthase and (S)-Linalool Synthase Determine the Proportion of Linalool Enantiomers in Camellia sinensis var. sinensis

Ying Zhou,Rufang Deng,Xinlan Xu,Ziyin Yang

Linalool is abundant in tea leaves and contributes greatly to tea aroma. The two isomers of linalool, (R)-linalool and (S)-linalool, exist in tea leaves. Our study found that (R)-linalool was the minor isomer in nine of Camellia sinensis var. sinensis cultivars. The (R)-linalool synthase of tea plant CsRLIS was identified subsequently. It is a chloroplast-located protein and specifically catalyzes the formation of (R)-linalool in vitro and in vivo. CsRLIS was observed to be a stress-responsive gene and caused the accumulation of internal (R)-linalool during oolong tea manufacture, mechanical wounding, and insect attack. Further study demonstrated that the catalytic efficiency of CsRLIS was much lower than that of (S)-linalool synthase CsSLIS, which might explain the lower (R)-linalool proportion in C. sinensis var. sinensis cultivars. The relative expression levels of CsRLIS and CsSLIS may also affect the (R)-linalool proportions among C. sinensis var. sinensis cultivars. This information will help us understand differential distributions of chiral aroma compounds in tea.

Improved linalool production in Saccharomyces cerevisiae by combining directed evolution of linalool synthase and overexpression of the complete mevalonate pathway

Pingping Zhou,Yi Du,Nannan Xu,Chunlei Yue,Lidan Ye

With special fragrance and various biological properties, linalool has wide applications in food, pharmaceuticals and cosmetics industries. Microbial biosynthesis has become a promising approach to linalool production, however its efficiency is limited by the limited precursor supply and poor efficiency of linalool synthase. With the goal of enhancing heterologous linalool production in Saccharomyces cerevisiae , protein engineering and metabolic engineering were simultaneously employed to solve these bottlenecks. Linalool-producing yeast was first constructed by introduction of t67OMcLIS selected out from a collection of linalool synthases. The efficiency of linalool biosynthesis was then improved by strengthening the supply of geranyl pyrophosphate (GPP) as precursor via overexpression of the complete mevalonate (MVA) pathway and a GPP synthase variant. To accelerate the conversion of the accumulated GPP to linalool, t67OMcLIS was engineered by means of directed evolution after development of a competition-based color-indicated high-throughput screening method. Expression of the t67OMcLIS variant in the engineered S. cerevisiae with enhanced precursor supply yielded 53.14 mg/L of linalool in biphasic shake-flask culture. This study delivered an efficient linalool-producing yeast cell factory and meanwhile provided efficient strategies for biosynthesis of other valuable monoterpenes.

Linalool Induces Resistance Against Tobacco Mosaic Virus in Tobacco Plants

Yue Jiang,Xiaoyu Pan,Yiming Li,Yizhou Yang,Yina Jia,Bin Lei,Juntao Feng,Zhiqing Ma,Xili Liu,He Yan

The essential oil of Cinnamomum camphora is the most widely consumed and used spice in the world today. It has therapeutic effects in medicine and has been shown to have good antibacterial and bacteriostatic effects in agriculture. This study found that C. camphora oil significantly induced plant disease resistance activity. Linalool, its main active component, significantly induced plant disease resistance activity (67.49% at a concentration of 800 μg/ml) over the same concentration of the chitosan oligosaccharide positive control but had no direct effect on tobacco mosaic virus (TMV). In this study of its antiviral mechanism, linalool induced hypersensitive reaction (HR); the overexpression of related defense enzymes SOD, CAT, POD, and PAL; and the accumulation of H 2 O 2 and SA content in N. glutinosa . Besides, linalool induced crops resistance against Colletotrichum lagenarium , Botrytis cinerea , Sclerotinia sclerotiorum , and Phytophthora capsica . Taken together, the anti-TMV mechanism of linalool involved the induction of plant disease resistance through activation of a plant immune response mediated by salicylic acid. Linalool-induced plant disease resistance activity has a long duration, broad spectrum, and rich resources; linalool thus has the potential to be developed as a new plant-derived antiviral agent and plant immune activator.

Prediction of Linalool Content in Osmanthus fragrans Using E-Nose Technology

Chenchen Zhou,Junjun Fan,Ruinan Tan,Qin Peng,Junhuo Cai,Wangxiang Zhang

Linalool is a well-known volatile compound. It is usually extracted from plants or synthesized by chemical methods. However, there is still no rapid screening method for plants with high linalool content. Therefore, we aimed to establish a method determining linalool content in Osmanthus fragrans by electronic nose (E-nose). The volatile gases of three cultivars of O. fragrans were characterized by gas chromatography-mass spectrometry (GC-MS) and E-nose. Different concentrations of linalool were measured by E-nose. Then, the E-nose data were subjected to principal component analysis (PCA) and linear discriminant analysis (LDA) to establish a discriminant model for determining linalool concentrations. Prediction models based on principal component regression (PCR) and multiple linear regression (MLR) were developed from all sensors and feature sensors to determine linalool concentrations in the three cultivars of O. fragrans flowers. Finally, the linalool concentrations determined by gas chromatography were used to verify the accuracy of the prediction model. The results show that linalool had the highest abundance among the volatile compounds of O. fragrans, and the MLR prediction model had an of 0.992 in calibration sets and an of 0.895 in prediction sets using 10 sensors. This study describes a rapid and accurate method for the detection and quantification of linalool concentrations.

Unveiling the mechanisms for plant-derived linalool control of anthracnose in sorghum

Wangdan Xiong,Xinfeng Jia,Tianyu Wang,Guo Wei,Yanjun Guo

BACKGROUND Sorghum anthracnose, caused by Colletotrichum sublineola , poses a major threat to sorghum production, causing substantial damage globally. Linalool, a terpenoid synthesized by plants, plays a crucial role in defense against pathogens. This study explored the dual defense roles of the plant-synthesized terpenoid linalool through both host resistance potentiation and direct antifungal action. RESULTS Following fungal infection, linalool levels increased significantly in sorghum leaves, with resistant genotypes exhibiting higher linalool accumulation than susceptible ones. A strong positive correlation between linalool content and disease resistance suggested its importance in fungal defense. Transcriptomic profiling identified Sobic.004G019400 as the key terpene synthase (TPS) gene responsive to fungal infection. This was validated functionally through heterologous expression and gas chromatography–mass spectrometry (GC–MS) analysis. Physiological assessments demonstrated that linalool application reduced lesion areas and alleviated declines in photosynthetic activity and transpiration in infected plants. The study also indicated that linalool inhibited C. sublineola spore germination and hyphal growth. Mechanistic investigations demonstrated that linalool effectively inhibited hyphal growth by inducing malondialdehyde overaccumulation, which caused significant damage to the fungal cell membrane and structure. This damage was evidenced by increased electrical conductivity and nucleic acid leakage. Transcriptomic analyses further revealed that linalool disrupted pathways in C. sublineola , including oxidative phosphorylation and the tricarboxylic acid (TCA) cycle. CONCLUSION These findings not only identify Sobic.004G019400 as a molecular target for breeding anthracnose-resistant sorghum varieties, but also establish linalool as a functional antimicrobial agent with potential applications for integrated pest management strategies. © 2025 Society of Chemical Industry.

Chitosan oligosaccharide-g-linalool polymer as inhibitor of hyaluronidase and collagenase activity

Shuifang Mao,Xiaoli Liu,Wenshui Xia

In this study, chitosan oligosaccharide (COS) was modified by grafting Linalool (Lin) on its backbone to improve its anti-inflammatory activity. By changing the molar ratios of COS to Lin, three different degrees of substitution COS-g-Lin1-3 were prepared. The degrees of substitution of derivatives were 0.65, 0.80 and 1.14 respectively. The structure of COS-g-Lin1-3 were characterized by UV–vis, FT-IR, 1 H NMR and elemental analysis in order to show the COS-g-Lin1-3 successfully synthesized. Besides, the thermal stability , solubility, pH stability as well as crystallinity were also investigated. The results revealed that the derivatives exhibited higher thermal stability and more remarkable anti-inflammatory property against hyaluronidase and collagenase than that of COS. The good biocompatibility made this novel material a promising and effective compound for anti-inflammatory applications.

Alleviating Effects of Linalool Fumigation on Botrytis cinerea Infections in Postharvest Tomato Fruits

Qing Shen,Haosen Li,Qifang Wang,Jianquan Wang,Jiarui Ge,Xiaoyu Yang,Xiaoyun Wang,Xiuming Li,Yan Zhang,Ruimin Zhang,Qinghua Shi

Gray mold is a disease that often occurs in postharvest tomato fruits, leading to a significant decline in the fruits’ quality. In production, pesticides are mostly used to control gray mold, which negatively affect both the environment and food safety. The purpose of this article is to study the effects of linalool fumigation on controlling gray mold in tomato fruits and to further investigate the mechanism of linalool function, so as to provide technical support and a theoretical basis for the application of linalool in the green control of tomato gray mold. The results of the in vitro experiments showed that linalool fumigation had a strong inhibitory effect on the mycelial growth ofBotrytis cinereaand that the fumigation of linalool inhibited the expansion of pathogens on tomato fruits. The disease index of tomato fruits in the linalool treatment was always lower than that of the control within 72 h after inoculation withBotrytis cinerea; at the end of experiment, the disease index of the control reached 100.0, which was only 8.0 with the 30 μL/L linalool treatment. Linalool fumigation increased the antioxidant capacity of the tomato fruits underBotrytis cinereainfection through regulating activities of SOD, POD and CAT as well as the ascorbic acid (AsA) content, which could be responsible for the lower malondialdehyde (MDA) accumulation. Linalool fumigation increased the activities of polyphenol oxidase (PPO), and phenylalanine ammonia-lyase (PAL) in tomato fruits, indicating that secondary metabolism was involved in the function of linalool in the regulation of tomato fruit resistance to pathogens. As the main enzymes related to cell structure, polygalacturonase (PG), cellulase (CL) andβ-galactosidase (β-GAL) were inhibited by linalool, which could protect cell wall structures from damage, and strengthened the mechanical barrier against pathogen access to fruit flesh. Keywords:tomato fruit;Botrytis cinerea;linalool;anti-fungal activity;postharvest

Antibacterial activity and metabolomic analysis of linalool against bovine mastitis pathogen Streptococcus agalactiae

Tingyu Liang,Guiguo Huo,Lele Chen,Ling Ding,Jianping Wu,Ji Zhang,Rongmin Wang

Streptococcus agalactiae is among the major causative pathogens of bovine mastitis, as well as crucial pathogen leading to human morbidity and mortality. Being a promising natural antibacterial agent, linalool has been broadly applied in medicine and food processing. However, its antibacterial effect against S. agalactiae has barely been elucidated. This study is the first to investigate the antibacterial activity and action mechanism of linalool against S. agalactiae causing bovine mastitis. Linalool exhibited significant antibacterial activity against S. agalactiae , with an inhibition zone diameter of 23 mm and a minimum inhibitory concentration of 1.875 μL/mL. In addition, linalool damaged cell structural integrity of S. agalactiae , leading to the leakage of intracellular components (alkaline phosphatase, nucleic acids and protein). Linalool also exhibited a scavenging effect on biofilm. Moreover, untargeted metabolomics analysis revealed that linalool stress substantially disrupted intracellular metabolism of S. agalactiae . Linalool caused energy metabolism disorder, and obstructed nucleic acid synthesis in S. agalactiae . Furthermore, downregulation of amino acids (e.g., proline, alanine) and upregulation of saturated fatty acids provide strong evidence for linalool induced cell wall and membrane damage. Overall, linalool exhibited strong antibacterial activity against S. agalactiae by destroying the cell structure and disrupting intracellular metabolism. This study provides a new insight and theoretical foundation for linalool application in preventing S. agalactiae infection.
