Background Chemiluminescence (CL) bioassay is one of the most advanced and used detection method in clinical diagnosis and biomedical research because of the advantages of low background, easy operation, and wide-field imaging without a light source or microscope. The luminol/hydrogen peroxide/horseradish peroxidase (luminol/H 2 O 2 /HRP) system is the most popular CL system, but its application in high-throughput imaging detection is challenged due to its low luminescence efficiency and flash-type emission which is difficult in ensuring the reproducibility and consistency of detection results. Results We reported a glow-type CL system of luminol@CD/H 2 O 2 /HRP by using a supramolecular enhancer of cyclodextrin (CD). This luminol@CD/H 2 O 2 /HRP system exhibited a luminescence lifetime of 41 min for sensitive and accurate imaging analysis. The long-lasting CL emission was attributed to the formation of a 1:1 host–guest complex between luminol and CD, which could stabilize the emitter and effectively reduce nonradiative relaxation. The formation of luminol@CD complex was determined through NMR experiments and theoretical analysis. Under optimum conditions, the luminol@CD/H 2 O 2 /HRP system showed higher sensitivity and much better precision than classical luminol/H 2 O 2 /HRP system for imaging detection of HRP. Especially, this glow-type luminol@CD/H 2 O 2 /HRP system realized CL imaging of microwell arrays on microfluidic chips. In addition, the luminol@CD/H 2 O 2 /HRP system was successfully applied for point-of-care detection of 17 β- estradiol based on a competitive mechanism of host–guest recognition. Significance An efficient CL system is crucial for obtaining reproducible and consistent results for accurate detection. Our luminol@CD/H 2 O 2 /HRP system emitted strong and persistent luminescence, resulting in reliability and efficiency at both CL macroscopic and microscopic imaging detection. We expected the luminol@CD/H 2 O 2 /HRP CL system to be applied in various detection fields.

This paper reports for the first time the chemiluminescence (CL) of luminol in the absence of added oxidant with carbon dots (CDs) as the catalyst. By means of CL spectra, UV–vis spectra, electron spin resonance spectra, Fourier transform infrared and X-ray photoelectron spectra, the possible CL reaction mechanism is proposed. The CL luminophor was still 3-aminophthalate, the CL enhancement of luminol system was supposed to originate from the intrinsic catalytic activity of CDs, which can efficiently catalyze the decomposition of dissolved oxygen to generate superoxide radical anion in luminol solution. Then, the resulting superoxide radical anion reacted with luminol to cause CL emission. Furthermore, the effects of size and surface states on the catalytic activity of CDs were investigated. The results suggested that the catalytic activity of CDs are mainly affected by the surface states, and the size of CDs display little influence on the catalytic activity of CDs. Therefore, the catalytic activity of CDs can be tuned by modulating functional groups on the surface of CDs, which sheds new light on the characteristic of CDs.

ZnSe quantum dots (QDs) with a homogeneous size distribution and good stability were synthesized by the reaction of zinc acetate and NaHSe in the presence of glutathione (GSH). The obtained ZnSe QDs were characterized using transmission electron microscopy (TEM) and X-ray powder diffraction (XRD). Simultaneously, UV-visible spectroscopy (UV-vis) was performed and the fluorescence properties of ZnSe QDs were investigated. The effect of ZnSe QDs on the luminol–potassium periodate (KIO4) chemiluminescence (CL) system, a popular model CL system, was researched. It was found that as-prepared ZnSe QDs could strongly enhance the chemiluminescence (CL) of the luminol–KIO4 system. To discuss the mechanism of the CL enhancement, we studied the UV-vis and CL spectra of the luminol–KIO4–ZnSe QDs system. The CL enhancement should be attributed to the oxidation of luminol to its excited state by KIO4, under the catalysis of ZnSe QDs, which caused strong light emission. It was found that rutin, p-t-butylprocatechol, catechol, paracetamol, resorcinol and tryptophan could markedly inhibit the CL of the luminol–KIO4–ZnSe QDs system among 17 kinds of amino acids and 23 other kinds of organics. The above phenomena presented good linear relations. Thus, effective methods to detect them were developed.

A method is presented for electrochemiluminescent (ECL) detection of the food additive curcumin via an energy transfer strategy and by using luminol-doped silica nanoparticles (luminol-NPs). The ECL emission of the luminol-NPs (peaking at 425 nm) is reduced in the presence of curcumin due to spectral overlap. The assay can be performed within 1 min, response is linear in the 0.1 to 100 µM curcumin concentration range, and the limit of detection is 32 nM. The method is selective over many ions, adenosine triphosphate, ascorbic acid, cysteine and folic acid. It was successfully applied to the determination of curcumin in spiked human serum and urine. The average recoveries range from 99.0 to 102.6%.
Electrochemiluminescence (ECL) “turn-off” detection of curcumin at levels as low as 32 nM via energy transfer using luminol-doped silica nanoparticles. No hydrogen peroxide (H 2 O 2 ) is used in ECL detection which makes the luminol-NPs ECL system more stable than the conventional luminol-H 2 O 2 ECL system.

An ultrasensitive sandwich electrochemiluminescence (ECL) biosensor was designed for determination of concanavalin A (ConA) through the specific carbohydrate-ConA interactions. Three-dimensional porous metal-organic framework (Zn-MOF) was synthesized, which loaded a large amount of luminescent reagents as luminol by encapsulating into its pores to form Zn-MOF@luminol complex. Interestingly, Zn-MOF also acted as the coreactant accelerator in the luminol-H 2 O 2 ECL system. This Zn-MOF@luminol complex was used as the signal probe to achieve a super strong and stable ECL signal. In addition, three-dimensional hierarchical molybdenum disulfide nanoflower and multiwalled carbon nanotubes complex (MoS 2 NF@MWCNTs) with peroxidase-mimicking enzyme property were used as a substrate to modify the glassy carbon electrode to further enhance the ECL signal of luminol by promoting decomposition of H 2 O 2 into reactive oxygen species (ROSs). In addition to the horseradish peroxidase (HRP) catalysis effect on the luminol ECL signal, a triple amplified ConA sandwich ECL sensor with high sensitivity sensor was constructed. The linear range for ConA detection was from 0.5 pg/mL to 100 ng/mL with a detection limit of 0.3 pg/mL ( S / N = 3). The recovery test for ConA in human serum samples was performed with satisfactory results.

A fluorescent dual-emitting platform for fluorescent “turn-on” ratiometric detection of ascorbic acid in beverages was developed utilizing luminol-embedded iron-based metal–organic frameworks (luminol@Fe-DOBDC MOFs). Luminol@Fe-DOBDC MOFs with fluorescent emissions at 430 nm and 540 nm under excitation wavelength of 365 nm were applied to detect ascorbic acid on the basis of ascorbic acid triggering the reduction of Fe 3+ into Fe 2+ . In the presence of ascorbic acid, fluorescent intensity at 540 nm was increased significantly while fluorescent intensity at 430 nm was changed slightly. Two emission peaks separated by 110 nm can eliminate environmental interferences by built-in self-calibration of ratiometric signal, enhancing the sensitivity and accuracy. The increasing ratiometric fluorescent intensity ( I 540 nm / I 430 nm ) has linear relationship with the concentration of ascorbic acid from 0.2 to 30 μM with limit of detection of 70 nM. It is an efficient, sensitive and accurate platform to detect ascorbic acid in commercial beverages using transition-metal-based MOFs.

Bimetallic Pt/Ag nanoparticles (Pt/Ag NPs) were synthesized using 3-hydroxyphenyl-boronic acid as the reducing and stabilizing agent by a simple, one-step hydrothermal method. The bimetallic Pt/Ag NPs were demonstrated to possess intrinsic peroxidase mimetic catalytic activity for the luminol/H2O2 reaction to generate chemiluminescence (CL). The catalytic properties of the bimetallic Pt/Ag NPs were thoroughly investigated. The results showed that the combination of Pt and Ag greatly enhanced the chemiluminescence which is attributed to their synergy. Moreover, the prepared bimetallic Pt/Ag NPs exhibited superior peroxidase mimetic activity to horseradish peroxidase and were stable under a wide range of pH values and temperatures. Using the bimetallic Pt/Ag NPs as the catalyst, a novel chemiluminescence sensor was developed for the determination of glucose with a linear range from 1 to 500 µM and a detection limit of 0.35 µM. The developed method was successfully employed to determine glucose in human serum with satisfactory results. Bimetallic Pt/Ag NPs with peroxidase mimetic catalytic activity have promising applications for determination of glucose in clinical samples.

A signal-on solid-state electrochemiluminescence (ECL) sensor based on luminol@carbon nanotubes (luminol@CNTs) and CdTe-ZnS@hydroxyapatite (CdTe-ZnS@HAP) was constructed for the detection of paclitaxel (PTX). In this sensing platform, luminol was used as the main luminophore and CdTe-ZnS was used as a co-reaction accelerator. CNTs and HAP were acted as carriers to adsorb more luminophores and co-reaction promoters. In addition, due to the enhanced effect of PTX on the ECL intensity of the luminol-O 2 system, PTX was introduced as the final detection target. Under the optimal experimental conditions, the ECL intensity change values (ΔI) of the sensor showed a good linear relationship with the logarithm of PTX concentration in the range of 3 × 10 -12 − 3 × 10 -7 molL -1 , and the limit of detection was 1 × 10 -12 mol L -1 (S/N = 3). This method was successfully applied to the determination of trace PTX concentration in serum.

A Fe-porphyrin covalent organic framework (Fe-PorCOF) was prepared through a postmodification strategy and characterized using different techniques. Fe-PorCOF exhibits an inherent peroxidase/oxidase mimetic catalytic activity and sharply accelerates chemiluminescence (CL) reactions between luminol and hydrogen peroxide (H 2 O 2 ) or dissolved oxygen (O 2 ) under alkaline conditions. The catalytic role was attributed to a significant increase in production of reactive oxygen species. Using the imminent peroxidase mimetic catalytic activity of Fe-PorCOF, a new CL method was developed for determination of H 2 O 2 over a linear range from 0.01 to 10.0 μmol·L −1 and with a limit of detection of 5.3 nmol·L −1 . The combination of the peroxidase mimetic catalytic activity of Fe-PorCOF with the catalytic activity of glucose oxidase on glucose oxidation presents a sensitive CL method for glucose assay. The linear range and the detection limit for glucose were 0.05–8.0 μmol·L −1 and 4.0 nmol·L −1 , respectively. The practicability of this method was assessed by determination of glucose in human sera. As a peroxidase/oxidase mimetic, Fe-PorCOF is easy to prepare and exhibits good catalytic efficiency in the luminol reaction. We believe that this strategy will promote the development of a CL field with functional COFs as a catalyst.

Ratiometric electrochemiluminescence (ECL) assays have attracted widespread attentions in biosensing owing to their precise measurements by eliminating the environmental interferences. However, they mostly needed two eligible luminophors, increasing the complexity of the systems and limiting their practical applications. Herein, using luminol as single luminophor, a dual-potential ratiometric ECL strategy was proposed to detect carcinoembryonic antigen (CEA). The luminol exhibited cathodic and anodic emissions on graphene-ionic liquid-platinum (GR-IL-Pt) composites and Ti 3 C 2 MXenes-Au NPs hybrids, respectively. Then, a sandwich ECL sensor was fabricated using GR-IL-Pt composites as matrix to immobilize the primary antibodies of CEA and Ti 3 C 2 MXenes-Au NPs hybrids as platform to load the secondary antibodies. With the presence of CEA, the ratio of anodic ECL to cathodic ECL (ECL anodic /ECL cathodic ) increased obviously, realizing sensitive ratiometric detection of CEA. In addition, the ECL anodic /ECL cathodic was independent with the concentrations of H 2 O 2 , greatly improving the test reliability. The developed ECL sensor exhibited a sensitive detection toward CEA, performing a wide linearity in the range of 0.1 pg mL −1 - 10 ng mL −1 with a low detection limit of 34.58 fg mL -1 (S/N = 3). Furthermore, this strategy exhibited a good practicality to detect CEA in human serums, providing a promising strategy in ECL bioanalysis.

A metal (Co)–Organic Framework (Co-MOF) was first found to catalyze the chemiluminescence (CL) of luminol. On the basis of X-ray photoelectron spectroscopy, powder X-ray diffraction, CL spectral, UV-visible absorption spectral, and electron spin resonance (ESR) spectral studies, as well as the research of the influence of various free radical scavengers, a possible CL mechanism was proposed. The enhanced CL might be attributed to the formation of a peroxide analogous complex between the oxygen-related radicals and the active metal site of the Co-MOF material. The established Co-MOF–luminol CL system was successfully applied to determine L-cysteine (CySH), based on the selective and sensitive enhancing effect of CySH on this CL system. Under the optimized conditions, CySH was selectively detected in the range 0.1–10 μM with a detection limit of 18 nM. This novel CL system obviously gives impetus to the new research field of metal–organic frameworks (MOFs) in chemiluminescence.

An efficient method has been developed for visualizing latent fingerprints by spatially selective electropolymerization of luminol on the ITO electrode. The fingerprint deposited on the electrode surface functions as a mask so that polyluminol is formed on the bare electrode surface. Therefore, electrochemiluminescence is only generated between the ridges of fingerprint, eventually yielding a negative ECL image. This method is easy to operate and safe to examiners. In addition, the second- and third-level details of fingerprints can be well resolved, indicating the potential application for personal identification and forensic investigation.
