As a crucial mechanobiological regulator, the tension of the cell membrane plays a vital role in governing cellular adhesion, proliferation, and differentiation processes. Additionally, it displayed a dynamic response to mechanical microenvironmental changes. This research systematically examines the mechanoresponsive behaviors of mouse bone marrow mesenchymal stem cells (mBMSCs) that are cultured on poly(dimethylsiloxane) (PDMS) substrates which are functionalized with methyl (-CH 3 ), amino (-NH 2 ), and carboxyl (-COOH) groups. Under both static and stretching conditions, it is found that compared with the -CH 3 surface, static culture on the -NH 2 and -COOH functionalized surfaces significantly promotes the proliferation of mBMSCs and upregulates the expression of extracellular matrix adhesion-related genes, especially focal adhesion kinase (FAK) and integrin β1. Morphometric analysis reveals that there are concomitant increases in the cell spreading area and the number of pseudopods on these modified surfaces. Mechanical stretching stimulation not only amplifies these cellular responses but also leads to more uniform FAK distribution. The assessment by atomic force microscopy (AFM) shows that both chemical functionalization (-NH 2 /-COOH) and stretch stimulation reduce the deformability of the cell membrane, and the -NH 2 modification exhibits a greater membrane-stiffening effect than -COOH.

Both silicon (Si) and zinc (Zn) ions are essential elements to bone health and their mechanisms for promoting osteogenesis have aroused the extensive attention of researchers. Thereinto, the mechanism by which dual ions promote osteogenic differentiation remains to be elucidated. Herein, the effects of Si and Zn ions on the cytological behaviors of mBMSCs were firstly studied. Then, the molecular mechanism of Si-Zn dual ions regulating the osteogenic differentiation of mBMSCs was investigated via transcriptome sequencing technology. In the single-ion system, Si ion at the concentration of 1.5 mM (Si-1.5) had better comprehensive effects of cell proliferation, ALP activity and osteogenesis-related gene expression levels (ALP, Runx2, OCN, Col-I and BSP); Zn ion at the concentration of 50 μM (Zn-50) demonstrated better combining effects of cell proliferation, ALP activity and same osteogenic genes expression levels. In the dual-ion system, the Si (1.5 mM)-Zn (50 μM) group (Si1.5-Zn50) synthetically enhanced ALP activity and osteogenesis genes compared with single-ion groups. Analysis of the transcriptome sequencing results showed that Si ion had a certain effect on promoting the osteogenic differentiation of mBMSCs; Zn ion had a stronger effect of contributing to a better osteogenic differentiation of mBMSCs than that of Si ion; the Si-Zn dual ions had a synergistic enhancement on conducting to the osteogenic differentiation of mBMSCs compared to single ion (Si or Zn). This study offers a blueprint for exploring the regulation mechanism of osteogenic differentiation by dual ions. Graphical Abstract

Cellular behavior is a result of homologous responses to biomaterials, especially material surfaces. An effective way to elicit a desired biological response is to employ micro/nanostructured constructions for biomaterial surface design; however, the brittleness of inorganic materials usually hinders structure regulation. In this study, a micro/nanostructured surface was fabricated using a hydrothermal treatment that did not require crystal regulation reagents. The precursor, β-tricalcium phosphate, was mixed with CaSiO3. The resulting micro/nanostructured surface had a hybrid structure containing nanowires, microrods, and rod clusters. In vitro cellular proliferation, cytotoxicity and osteodifferentiation experiments indicated that the micro/nanostructured surfaces alleviated material cytotoxicity, improved cell proliferation, and effectively promoted osteogenic differentiation of mouse bone mesenchymal stem cells. These results suggest that this simplified surface treatment is an efficient way to improve cell biological response to inorganic biomaterials.

Modifying substrates through mineralization is a popular way to improve the osteogenic performance. Screening of the best mineralization characteristics on specific substrates for stem cells is meaningful but not fully studied. In this paper, poly(lactic-co-glycolic acid)/hydroxyapatite (PLGA/HA, PH) microsphere scaffolds with superficial pores were fabricated by a low-temperature fusion method. After the mineralization in the 5× stimulated body fluid (SBF) for 0, 7, 12 and 24 h, four mineralized scaffolds (MPH-0, MPH-7, MPH-12 and MPH-24) with different apatite topographies were obtained. It was found that the surface of MPH-7 was evenly decorated with abundant micro-pores, MPH-12 with dense and plain apatite layer, and MPH-24 with small spherical bumps. The responses of mouse bone mesenchymal stem cells (mBMSCs) to the four scaffolds were further studied. The results showed that MPH-7 and MPH-24 had more obvious effects on mBMSCs attachment, proliferation and differentiation than MPH-0 and MPH-12. This work indicated that to obtain the maximum improvement, the mineralization characteristics had to be carefully chosen. This was noteworthy in the chemical modification of surfaces to form the functionalized scaffolds for bone repair.
