Asiaticoside (AS) and madecassoside (MS) are two key triterpenoid saponin compounds found in Centella asiatica . Despite their structural similarities, they exhibit distinct biological activities. AS is known for its anti-inflammatory properties and its effectiveness in treating rheumatoid arthritis, while MS is recognized for its ability to promote wound healing and significantly improve burn scars. Additionally, AS is widely utilized in the cosmetics industry due to its antioxidant and moisturizing properties. Currently, separating and purifying these two compounds simultaneously using existing methods is challenging. This study focuses on synthesizing molecularly imprinted polymers (MIPs) through precipitation polymerization with AS as the template. By optimizing various preparation parameters, such as the molar ratios of functional monomers, porogens, and cross-linking agents, high selectivity was achieved. The maximum adsorption capacity reached 19.16 mg/g, with an imprinting factor of 2.86. The MIPs were characterized using techniques like scanning electron microscopy (SEM), TA, Brunauer–Emmett–Teller (BET) adsorption, and Fourier-transform infrared (FT-IR). Their effectiveness was assessed through static and dynamic adsorption, desorption, and reusability tests. When applied in molecularly imprinted solid-phase extraction (MISPE) of crude extracts from C. asiatica , the recovery of AS was 39.90%, and its purity increased by 3.01 times, whereas the purity of MS improved by 6.51 times. These findings indicate that the preparation of MIPs is cost-effective, highly selective, reproducible, and user-friendly, making it a promising method for enriching and isolating AS from complex mixtures.

Madecassoside (MA) exhibits excellent therapeutic effects in wound healing and scar management. However, its high hydrophilic nature and low permeability through skin tissue limits its topical application. Liposomes are widely used to deliver drugs due to their high structural similarity and biocompatibility with cell membranes. However, normal liposome formulations are too fluid to maintain sufficient adhesion to the wound surface. In this study, in order to make an MA formulation conducive to topical administration, poly(ethylene glycol)-poly( ε -caprolactone)-poly(ethylene glycol) (PEG-PCL-PEG, PECE), a biodegradable and temperature-responsive copolymer material, was synthesized and applied to improve the adhesion properties of MA liposomes. The mean particle size of the PECE-modified MA liposomes was 213.43±4.68 nm, and the zeta potential was −23.80±15.37 mV under the optimal conditions of EPC (egg yolk lecithin) to PECE at a mass ratio of 1:1. Additionally, PECE-modified MA liposomes maintained a hydrogel state for better adhesion until the temperature reached 43°C. Furthermore, the PECE-modified MA liposomes showed superior wound contraction effects relative to the MA liposomes in second-degree burn experiments using a rat model. These results indicated that PECE-modified MA liposomes have better surface adhesion performance and healing effects than unmodified MA liposomes.

Previously, we reported that oral administration of madecassoside, the main active triterpene in Centella asiatica L., exerted anti-arthritis effect by inducing the generation of regulatory T (Treg) cells in small intestine. This study investigates the action site and mechanism of madecassoside to induce Treg cells. In collagen-induced arthritis (CIA) of rats, oral administration of madecassoside significantly alleviated arthritis symptoms, but its main metabolite madecassic acid did not, suggesting that madecassoside functions in the parent form. Madecassoside was shown to increase the number of Treg cells and promote the expression of Foxp3 and IL-10 in rat ileum rather than duodenum and jejunum, as detected using the immunohistochemistry assay and quantitative PCR assay, respectively. Unexpectedly, madecassoside was absent of significant effect on in vitro Treg cell differentiation and the expression of Foxp3 and IL-10. A combined use of broad-spectrum antibiotics resulted in significant reduction of the anti-arthritis effect of madecassoside in CIA rats. The 16S rRNA gene sequence showed that madecassoside could reverse the changes of gut microbiota under arthritis condition, and enrich several bacteria such as Lachnospiraceae, Butyricicoccus, Faecalibacterium, Butyricicoccus pullicaecorum and so on. GC-MS assay showed that madecassoside elevated the levels of acetic acid and butyric acid, but not other short chain fatty acids (SCFAs) in the cecum contents of CIA rats. Butyric acid rather than acetic acid could induce the in vitro differentiation of Treg cells and the expression of Foxp3 and IL-10. Accordingly, when madecassoside was co-administered with heptanoyl CoA, the competitive inhibitor of butyrate synthase, its effect on butyric acid production, Treg cell proportion and arthritis nearly disappeared. These findings indicate that oral madecassoside induces the generation of Treg cells and therefore displays anti-arthritis effect in the parent form but not metabolites, and the ileum is the main action site. The mechanism of madecassoside can be summarized as: expansion of the richness of butyrate-producing bacteria-up-regulation of intestinal butyrate level-induction of Treg cell differentiation and IL-10 expression.
