Enzyme-free “on-off-on” photoelectrochemical biosensor based on cascaded quadratic amplification strategy for miRNA 141 detection

Fangjing Mo,Jingling Wu,Min Chen,Hui Meng,Qian Han,Yingzi Fu

MicroRNAs (miRNAs) assay is of great significance for early diagnosis of diseases, so an enzyme-free “on-off-on” PEC biosensor has been developed for sensitive miRNA 141 determination. Manganese-doped cadmium sulfide coupled with zinc sulfide quantum dots (Mn: [email protected] QDs) and manganese porphyrin (MnPP) have been used as photoelectric material and photosensitizer, respectively. And a high photocurrent of approximately 70.0 μA has been obtained. Cascaded quadratic amplification strategy has been applied in the system. Mn: [email protected] QDs was characterized by transmission electron microscopy (TEM), scanning electron microscope (SEM), X-ray photoelectron spectrum (XPS) and energy dispersive X-ray spectroscopy (EDX). Photoelectrochemical and electrochemical technologies were used to monitor the fabrication process of the biosensor. The sensing platform exhibits recommendable stability and good selectivity, miRNA 141 can be accurately quantified with a linear range of 1.00 × 10 −14 to 1.00 × 10 -8 mol·L -1 and the detection limit of 3.30 fmol·L -1 . This method provides promising potential to explore sensitive detection models for various biological molecules.

Ultrasensitive Faraday cage-type electrochemiluminescence assay for femtomolar miRNA-141 via graphene oxide and hybridization chain reaction-assisted cascade amplification

Jing Lu,Lin Wu,Yufang Hu,Sui Wang,Zhiyong Guo

In this study, a novel electrochemiluminescence (ECL) biosensor for sensitive detection of femtomolar miRNA-141 was constructed on the basis of Faraday cage-type strategy via graphene oxide (GO) and hybridization chain reaction (HCR)-assisted cascade amplification. A capture probe (CP) was immobilized on Fe 3 O 4 @SiO 2 @Au nanoparticles as capture unit, which could catch the miRNA-141, and the immobilization of the signal unit (Ru(phen) 3 2+ -HCR/GO) was allowed via nucleic acid hybridization. The prepared biosensor exhibited two advantages for signal amplification : firstly, GO could lap on the electrode surface directly, extending Outer Helmholtz Plane (OHP) of the sensor due to the large surface area and good electronic transport property; secondly, HCR-assisted cascade amplification was designed by anchoring all HCR products on the GO surface, then embedding Ru(phen) 3 2+ as a signal readout pathway. All these signal molecules could take part in electrochemical reactions , thus further enhancing the ECL signal drastically. Therefore, the proposed sensor constructed by integrating HCR with Faraday cage-type strategy displayed an ultrasensitive detection platform for the miRNA-141 with a low detection limit of 0.03 fM. In addition, this proposed biosensor provides a universal platform for analysis of other microRNAs.

Sensitive Detection of miRNA-141 Using an Endogenous Dual-Switch Activated Bipedal DNAzyme Walker Biosensor

Beibei Zhang,Xinyi Wang,Min Pan,Xinhao Li,Hong Wang,Yingwei Zhang

In this study, a novel dual-switch fluorescent biosensor based on a bipedal DNAzyme walker was developed for the highly sensitive detection of the tumour biomarker miRNA-141. By exploiting the typically elevated expression levels of miRNA-141 and apurinic/apyrimidinic endonuclease 1 (APE1) in tumour cells, an endogenous dual-gating system was constructed. The presence of both targets was required to initiate an enzyme-free strand displacement reaction, enabling target recycling and the subsequent release of bipedal DNAzyme walker components (D1/D2), which were efficiently isolated using streptavidin-modified magnetic beads. In the presence of Mg2+, the activated DNAzyme walker specifically cleaved fluorophore-labelled substrate strands on gold nanoparticles (AuNPs). Through repeated“walking–cleavage”cycles, fluorescence signals were progressively restored and amplified. By integrating AuNPs as carriers and magnetic separation technology, a cascade dual-amplification system based on non-enzymatic strand displacement and bipedal DNAzyme walking was established, markedly enhancing the detection sensitivity. The biosensor exhibited dual-switch-specific activation, effectively suppressing interference from non-target RNAs and other nucleases, and maintained a low background signal. It demonstrated excellent discrimination between tumour cells and normal cells, and showed good anti-interference performance in spiked serum samples. This study provides a new strategy for the development of high-precision tumour biomarker detection, offering potential for trace analysis in complex biological samples.

Ultrasensitive photoelectrochemical detection of cancer-related miRNA-141 by carrier recombination inhibition in hierarchical Ti3C2@ReS2

Lei Liu,Yue Yao,Kejian Ma,Changjian Shangguan,Songlong Jiao,Songyang Zhu,Xiaoxuan Xu

Elucidation of the photoelectrochemical (PEC) detection mechanism and exploring highly efficient photoactive materials hold both a challenge and an opportunity to construct high-performance biosensors. In this work, we present a strategy to enhance the charge separation efficiency by fabricating hierarchical Ti 3 C 2 @ReS 2 via the vertical anchoring flaky ReS 2 on the Ti 3 C 2 backbone. According to the experimental results, the Ti 3 C 2 @ReS 2 sample containing 45 wt % of ReS 2 exhibits a 2.48-time promotion in the photocurrent as compared to ReS 2 owing to the synergistic effects of its photoactive and conductive counterparts, indicating its significant advantages in separating photo-induced carriers and suppressing carrier recombination. In essence, the Ti 3 C 2 @ReS 2 Schottky junction formed at the interface can capture the photogenerated electrons generated by ReS 2 , thereby suppressing carrier recombination. As a result, an ultrasensitive PEC detection platform based on the optimized Ti 3 C 2 @ReS 2 can offer a log-linear detection window from 0.1 fM to 1 nM with an estimated detection limit of 2.4 aM (S/N = 3) for cancer-related miRNA-141 without any other amplification.

SERS and electrochemical dual-mode detection of miRNA-141 by using single Au@Ag nanowire as a new platform

Luo Xianzhun,Dai Qingshan,Qiu Xia,Wang Dongmei,Li Yongxin

As biomarkers of cancer, the accurate and sensitive detection of microRNAs is of great significance. Therefore, we proposed a surface-enhanced Raman scattering (SERS)/electrochemical (EC) dual-mode nanosensor for sensitively detecting miRNA-141. The nanosensor uses Au@Ag nanowires as a novel SERS/EC sensing platform, which has the advantages of good biocompatibility, fast response, and high sensitivity. The dual-mode nanosensor can not only effectively overcome the problem of insufficient reliability of single signal, but also realize the amplification and stable output of the detection signal, to ensure the reliability and repeatability of miRNA detection. With this sensing strategy, the target miRNA-141 can be detected over a wide linear range (100 fM to 50 nM) (LOD of 18.4 fM for SERS and 16.0 fM for electrochemical methods). In addition, the process shows good selectivity and can distinguish miRNA-141 from other interfering miRNAs. The actual analysis of human serum samples also proves that our strategy has good reliability, repeatability, and has broad application prospects in the field of analysis and detection. Isothermal and enzyme-free MicroRNA assay based on catalytic hairpin assembly and rare earth element labeled probes

Jiawei Shen,Tingting Li,Min Wang,Bo Yao

MicroRNAs (miRNAs) are a category of short non-coding RNA molecules which play essential roles in regulating gene expression. Correspondingly, the aberrant expression of miRNAs is commonly related to the development of various diseases including cancers and cardiovascular diseases. Hence, miRNA is considered as an ideal biomarker for disease diagnosis and its quantification is highly demanded. Herein, an isothermal and enzyme-free miRNA assay based on catalytic hairpin assembly (CHA) amplification coupled with inductively coupled plasma mass spectrometry (ICP-MS) detection is proposed. Two hairpin probes are employed in CHA reaction with rare earth element tag on H1 for ICP-MS detection and biotin on H2 for immobilization on magnetic beads. In the presence of target miRNA, the two independent probes open up their stem-loop structure and hybridize. The target miRNA will be released after the hybridization to trigger another CHA reaction. Ultimately, one target miRNA molecule promotes a number of CHA cycles and produces several copies of hybridized complex leading to the enhanced ICP-MS signal. Based on this approach, miRNA-141 was successfully analyzed with high sensitivity and specificity. The limit of detection (LOD) was 88 fmol with a linear range 0.1–2 pmol. Compared with fluorescence mediated CHA method and ICP-MS mediated basic sandwich method, the current strategy displayed the lowest LOD. The developed method was also applied to analyze miRNA-141 and miRNA-21 simultaneously to demonstrate its multiple detecting capability and practical applicability. Furthermore, this miRNA assay has the potential to become a high-throughput method by employing more distinct rare earth elements.
