Growing evidence suggested that long non-coding RNA (lncRNA) played a crucial role in the progression of ulcerative colitis (UC). Therefore, the purpose of this study is to understand how the lncRNA CBR3-AS1, which has been found to be up-regulated in UC, contributes to the bio-progression of the disease. To determine the concentration and relationship of the lncRNA CBR3-AS1, miRNA-145-5p, and FN1 in the LPS-induced Caco-2 model cells, qRT-PCR was employed in this study. Starbase was used to predict the target sites of the lncRNA CBR3-AS1 and the miRNA-145-5p, and Targetscan was used to predict the probable linking points of the FN1 and the miRNA-145-5p, which was confirmed by a twofold luciferase reporter test. The vitality of Caco-2 cells was determined using the CCK-8 and FCM tests. Using the ELISA kit, TNF, IFN, IL-6, and IL-17 were identified. The results of the experiment show that in Caco-2 cells treated with 10 ng/mL LPS, LncRNA CBR3-AS1 was up-regulated. Additionally, Caco-2 cells' LPS-induced apoptosis and inflammatory response were inhibited by lncRNA CBR3-AS1 inhibition. Dual-luciferase reporter experiments demonstrated that miRNA-145-5p and lncRNA CBR3-AS1 might connect. Moreover, miRNA-145-5p, which was shown to be poorly expressed in UC, was found to suppress inflammatory and apoptotic responses in Caco-2 cells activated by LPS. It's significant that FN1 was confirmed to be miRNA-145-5p's downstream target. Sh-CBR3-AS1's inhibitory effects were reversed by miRNA-145-5p knockdown, and the effects of the miRNA-145-5p inhibitor were reversed by sh-FN1. In conclusion, LncRNA CBR3-AS1 may offer a unique method for treating UC by suppressing the function of miRNA-145-5p, which is implicated in the development of UC.

A novel confinement-induced ECL enhancement (CIECL) sensing system has been constructed based on copper clusters (Cu NCs) and nano-vesicles in this work. The CIECL strategy resolved the shortcoming of poor stability and low luminous intensity of Cu NCs. Firstly, nano-vesicles were prepared through liquid-liquid self-assembly with amyl gemini quaternary ammonium salt and anionic surfactant. Subsequently, Cu NCs were confined in the nano-vesicles by electrostatic interaction. Due to the restriction on the intramolecular vibration and rotation of the ligands of Cu NCs, the Cu NC/nano-vesicle-based CIECL system showed the significant luminescence enhancement, good stability and high biocompatibility. Additionally, the Au nanoparticles-array with significant surface plasmon resonance effect has been prepared to further amplify the ECL intensity. Finally, the CIECL-based biosensor has been successfully used to detect miRNA-145 in clinic samples in the linear range of 100 fM to 100 nM, which displayed the potential value in the vascular calcification analysis.

Recently, miRNAs have become a promising biomarker for disease diagnostics. miRNA-145 is closely related to strokes. The accuracy determination of miRNA-145 (miR-145) in stroke patients still remains challenging due to its heterogeneity and low abundance, as well as the complexity of the blood matrix. In this work, we developed a novel electrochemical miRNA-145 biosensor via subtly coupling the cascade strand displacement reaction (CSDR), exonuclease III (Exo III), and magnetic nanoparticles (MNPs). The developed electrochemical biosensor can quantitatively detect miRNA-145 ranging from 1 × 102to 1 × 106aM with a detection limit as low down as 100 aM. This biosensor also exhibits excellent specificity to distinguish similar miRNA sequences even with single-base differences. It has been successfully applied to distinguish healthy people from stroke patients. The results of this biosensor are consistent with the results of the reverse transcription quantitative polymerase chain reaction (RT-qPCR). The proposed electrochemical biosensor has great potential applications for biomedical research on and clinical diagnosis of strokes.Keywords:electrochemistry;miRNA-145;stroke;CSDR;Exo III;MNPs

The self-powered sensing platform uses redox reactions to convert chemical nanogenerators into electrical energy, powering the system for target detection. In this work, a multi-dimensional hollow C@SnO 2 nanocomposite is designed as the electrode substrate and combined with the catalytic hairpin assembly (CHA) amplification strategy. This platform enables highly sensitive detection of the breast cancer marker miRNA-145. C@SnO 2 provides a large specific surface area and excellent electron transport properties, offering abundant enzyme active sites and improving electron transport efficiency. In the presence of miRNA-145, the CHA reaction cycle is triggered, anchoring CHA products to the electrode surface. These signal molecules then participate in the electrochemical reaction, amplifying the signal. Under optimized conditions, the platform demonstrates a linear response range from 1 fM to 10 nM, with a detection limit as low as 0.78 fM. The self-powered sensor enabling sensitive detection of miRNA-145 at low concentrations which is crucial for the early diagnosis of breast cancer. Additionally, this work integrates the self-powered sensing platform with commercial chips, ensuring stable performance and minimal signal fluctuations during long-term continuous monitoring, enabling portable and real-time target monitoring. This design expands the application range of self-powered biosensors and offers new approaches for field detection of other targets.

Mesoporous silica nanoparticles (MSN) can load and deliver potentially synergistic anticancer agents such as small molecule cytotoxics (like doxorubicin, DOX) and nucleic acids (like microRNA, miRNA). However, these cargos have different underlying chemical properties so overcoming respective intracellular delivery barriers is a key consideration. Strategies to deliver DOX from MSN frequently employ pH-driven mechanisms that are restricted to the acidic environment of lysosomes. Conversely, strategies to deliver miRNA make use of approaches that deliberately compromise lysosomal membrane integrity to enable cytosolic delivery of the payload. To reconcile these two needs (lysosomal delivery of DOX and intracellular delivery of miRNA), a new methodology by “weaving” polyethylenimine on the MSN surface through disulfide bonds to achieve superior delivery of chemotherapy (DOX) and miRNA therapy (using miRNA-145) is developed. Furthermore, an active targeting strategy based on a peptide ligand with affinity to glucose-regulated protein 78 (GRP78), a cell surface protein overexpressed in colorectal carcinoma, is developed. The active targeting approach results in enhanced synergistic antitumor effect both in vitro and in vivo in an orthotopic murine model of colorectal cancer. Taken together, this work demonstrates the capability and advantages of “smart” MSN delivery systems to deliver anticancer cargo appropriately to targeted cancer cells.
