Shao Su,Wenfang Cao,Wei Liu,Zaiwei Lu,Dan Zhu,Jie Chao,Lixing Weng,Lihua Wang,Chunhai Fan,Lianhui Wang

The detection of microRNA plays an important role in early cancer diagnosis. Herein, a dual-mode electronic biosensor was developed for microRNA-21 (miRNA-21) detection based on gold nanoparticle-decorated MoS 2 nanosheet (AuNPs@MoS 2 ). A classical DNA “sandwich” structure was employed to construct MoS 2 -based electrochemical sensor , including capture DNA, target miRNA-21 and DNA-modified nanoprobe. [Fe(CN) 6 ] 3-/4- and [Ru(NH 3 ) 6 ] 3+ were selected as electrochemical indicators to monitor the preparation process and evaluate the performance of MoS 2 -based electrochemical biosensor by electrochemical impedance spectroscopy (EIS) and differential pulse voltammetry (DPV), respectively. Such MoS 2 -based biosensor exhibited excellent performance for miRNA-21 detection in the range from 10 fM to 1   nM with detection limit of 0.78   fM and 0.45   fM for DPV and EIS technique, respectively. Furthermore, the proposed MoS 2 -based biosensor displayed high selectivity and stability, which could be used to determine miRNA-21 in human serum samples with satisfactory results. All data suggested that such MoS 2 -based nanocomposite may be a potential candidate for biosensing ranging from nucleic acid to protein detection.

Synthesis of Palladium Nanowires on Flagella Template for Electrochemical Biosensor Detection of microRNA-21

Kuo Yang,Jueyu Wang,Ying Zhang,Daizong Cui,Min Zhao

Simple SummaryThis study proposed a new method for extractingE. coliflagella, which served as a template for synthesizing palladium (Pd) nanowires. By improving the flagella extraction process, we successfully produced Pd nanowires with better structure and function, which could have important applications in technology and medicine. Traditional methods often led to impurities that interfered with nanomaterial synthesis, but our method provided a cleaner and more efficient way of producing high-quality nanowires. UsingE. coliflagella as a template offered environmental benefits, as it eliminated the need for toxic chemicals and reduced energy consumption. This eco-friendly approach was also safer and more reliable for practical applications. Our research also showed how the functional groups on the flagella surface helped form strong bonds with metals, improving the nanowire formation process. This study highlighted the potential of using biological templates for nanomaterial synthesis, contributing to the development of greener and more efficient technologies. In recent years, the use of bacterial flagella as biomimetic templates has gained increasing attention in nanomaterial synthesis due to their unique structural and functional properties. In this study, we optimized the flagella extraction method and achieved a high concentration of flagella solution. Flagella were isolated fromEscherichia coli. Surface characterization revealed that the flagella had abundant functional groups, such as amino and carboxyl groups, which can serve as nucleation sites for the controlled nucleation and growth of metal nanomaterials. Using bacterial flagella as a template, we synthesized one-dimensional palladium nanowires (Fla-Pd NWs). The results of morphological and phase analyses showed that the synthesized palladium nanoparticles were uniformly and densely distributed on the surface of the flagella. Moreover, the Fla-Pd nanowires exhibited superior electrocatalytic activity, which was applied to develop an electrochemical biosensor. This biosensor was used to detect the early breast cancer biomarker microRNA-21 and exhibited a linear range of 0.66–1.98 µmol/L and a detection limit of 0.78 µmol/L. The method demonstrated high selectivity and reusability, making it a promising strategy for early cancer diagnosis.

Ultrasensitive microRNA-21 detection based on DNA hybridization chain reaction and SYBR Green dye

Zhi Li,Bingchen Li,Yunlei Zhou,Huanshun Yin,Jun Wang,Shiyun Ai

It is extremely important for quantifying trace microRNAs in the biomedical applications. In this study, an ultrasensitive, rapid and efficient label-free fluorescence method was proposed and applied for detecting microRNA-21 in serum of gastric cancer patients based on DNA hybridization chain reaction (HCR). DNA H1 and DNA H2 were designed and used as hairpin probes, the HCR was proceeded in the presence of target microRNAs. Amounts of SYBR Green І dyes were used as signal molecules to intercalate long DNA concatemers from HCR, which guaranteed the model of label-free fluorescence and strong fluorescence density. The detection method showed a wide linear region from 1 fM to 10 5  fM, and the limit of detection was 0.2554 fM (at S/N = 3) for microRNAs. The results showed that this method had an excellent specificity and reproducibility. Furthermore, the label-free fluorescence strategy exhibited a sensitive response to microRNA-21 in real serum samples of gastric cancer patients and the results obtained were in accordance with reference method (R 2  = 0.994). Overall, the proposed strategy could be satisfactory for rapid, ultrasensitive and efficient detection of microRNA-21, and held great potentials in clinic diagnosis of gastric cancer.

Construction of an ultrasensitive electrochemical sensing platform for microRNA-21 based on interface impedance spectroscopy

Tianjiao Meng,Dan Zhao,Huimin Ye,Yue Feng,Huan Wang,Yufan Zhang

A hybridization chain reaction (HCR) amplification-based electrochemical impedimetric biosensor is fabricated for the quick, sensitive, and specific detection of miRNA-21 (miR-21) via monitoring of electrode interfacial property changes in real-time. Two sequences of H1 and H2 are adopted to trigger HCR amplification. A large amount of linear DNA concatemer are formed which could change the interfacial properties of the electrode. Interfacial charge transfer resistance difference ( R ct ) is probed via electrochemical impedance spectroscopy (EIS) and Randles equivalent circuit. After amplifying via HCR, oligonucleotides with negatively charged repelling [Fe(CN) 6 ] 3−/4− ions can form a spatial blockage. HCR amplification strategy markedly enhanced the electrochemical signal with a limit of detection (LOD) down to 4.63 fM (S/N = 3). This strategy exhibited excellent selectivity for three different miRNAs: miR-199a, miR-141, and miR-155. Moreover, results show that the proposed method can be applied to miR-21 detection in the total RNA extracted from five cells. This work presents an enzyme-free and label-free EIS nucleic acid sensor for sensitively and selectively detecting miR-21, offering a promising approach in early diseases diagnosis.

Circular RNA circ_0068,888 protects against lipopolysaccharide-induced HK-2 cell injury via sponging microRNA-21–5p

Wei Wei,Yuanyuan Yao,Hongyuan Bi,Wenxin Xu,Yan Gao

Our previous findings revealed that hsa_circ_0068,888 was markedly down-regulated in the plasma of patients with sepsis-associated acute kidney injury (AKI). However, its molecular mechanism in AKI remains unclear. Herein, we explored the role of hsa_circ_0068,888 in AKI. Human renal proximal tubular cell line HK-2 was stimulated with lipopolysaccharide (LPS) to mimic AKI in vitro . Decreased hsa_circ_0068,888 expression was observed in AKI cell model. The overexpression of hsa_circ_0068,888 significantly increased the viability of LPS-stimulated HK-2 cells, whereas hsa_circ_0068,888 downregulation showed the opposite effect. Furthermore, LPS triggered inflammatory response and oxidative stress , which was inhibited by hsa_circ_0068,888 overexpression and enhanced by hsa_circ_0068,888 down-regulation. Hsa_circ_0068,888 overexpression suppressed the activation of nuclear factor-κB (NF-κB) pathway triggered by LPS as evidenced by decreased p-p65 protein level and nuclear translocation of p65 in hsa_circ_0068,888 overexpressed cells. Additionally, we proved that hsa_circ_0068,888 targeted microRNA-21–5p (miR-21–5p). The expression of miR-21–5p was markedly increased and was negatively regulated by hsa_circ_0068,888 in LPS-stimulated HK-2 cells. Furthermore, we demonstrated that miR-21–5p overexpression reversed the effects on cell viability , inflammatory response, oxidative stress, and NF-κB pathway induced by hsa_circ_0068,888 overexpression in LPS-stimulated HK-2 cells. Overall, these results implied that hsa_circ_0068,888 shows a protective effect on AKI by sponging miR-21–5p. Hence, up-regulation of hsa_circ_0068,888 might be a potential strategy in treatment for AKI.

Pd nanoparticles-DNA layered nanoreticulation biosensor based on target-catalytic hairpin assembly for ultrasensitive and selective biosensing of microRNA-21

Tianjiao Meng,Huixian Jia,Siying An,Huan Wang,Xinjian Yang,Yufan Zhang

Herein, a Pd nanoparticles-DNA layered nanoreticulation (Pd NPs-DNA LNR) sensor was assembled to a graphene oxide (GO)-modified electrode by catalytic hairpin assembly (CHA) and a self-assembly process. In this process, GO was modified on the electrode surface to provide -COOH sites for DNA bonding. Moreover, with the CHA circular amplification strategy, many single DNA1 were exposed to be able to be bound with Pd-DNA2 for the formation of the Pd NPs-DNA LNR, which enabled the ultrasensitive detection of microRNA-21 (miR-21) with a low limit of detection 63.1 aM. This strategy combines the amplification technology of CHA and Pd NPs-DNA LNR with Pd NPs. Such a layered nanoreticulation material can support abundant Pd NPs to avoid aggregation and obtain enhanced electrochemical signals. Significantly, this method can be successfully applied in tumor cell lysates and provides a neoteric channel for early disease diagnosis.

Ultra-sensitive detection of microRNA-21 based on duplex-specific nuclease-assisted target recycling and horseradish peroxidase cascading signal amplification

Weipan Peng,Qian Zhao,Jiafang Piao,Miao Zhao,Yiwen Huang,Bo Zhang,Weichen Gao,Dianming Zhou,Guiming Shu,Xiaoqun Gong,Jin Chang

The detection and analysis of microRNA (miRNA) can provide critical information for biomedical research and clinical diagnosis. In this paper, we constructed a simple method to achieve ultra-sensitive and rapid detection of miRNA based on the double-specific nuclease (DSN) assisting target recovery and horseradish peroxidase (HRP) cascading signal amplification. In the existence of target microRNA-21 (miR-21), the DSN enzyme specifically cut the single-stranded DNA (ssDNA) hybridized with the target miRNA but still kept the target miRNA intact, so there were more free HRP released from the polystyrene microparticles-ssDNA-HRP conjugates (PS-ssDNA-HRP) because of the target recycle and signal amplification. The free HRP in the supernatant were collected to react with the substrate 3,3',5,5'-tetramethylbenzidine (TMB). The color and absorbance changes of HRP-TMB system further resulted in qualitative screening by naked eyes and quantitative detection by the microplate reader with the detection limit as low as 3.1 fM. This assay protocol not only showed high sensitivity and selectivity, but also well distinguished the difference between the miRNA family members, as well as the simple operation, fast detection, naked eye qualitative and microplate reader quantification. It has great application value in the biomedical research and clinical diagnosis.

Ag Nanocubes Coupled with Heating-Enhanced DSN-Assisted Cycling Amplification for Surface-Enhanced Raman Spectroscopy Detection of MicroRNA-21

Xing-Yuan Du,Shao-Hua Wu,Xiao-Bin Huang,Jian-Jun Sun

Surface-enhanced Raman scattering (SERS)-based biosensing has strong potential owing to its high sensitivity and good selectivity. In this study, a SERS biosensing platform for microRNA-21 detection was proposed by combining heating-enhanced duplex-specific nuclease (DSN)-assisted target recycling amplification and silver nanocube (AgNC)-based SERS enhancement. The activity of DSN was greatly enhanced by increasing the surface temperature of a heated Au electrode (HAuE) during the DSN-assisted target recycling process, resulting in dramatic miRNA-21 amplification detection. 4-Mercaptobenzoic acid (4-MBA) acting as a Raman reporter was immobilized onto the surface of AgNC to form a AgNC/4-MBA SERS tag with AgNC providing significantly larger electric field enhancement and thus excellent SERS applications compared with conventional silver or gold nanospheres. This enhancement was attributed to the well-known “lighting rod” effect derived from the sharp geometrical feature of the edges and corners of AgNCs. These factors lead to the high sensitivity of the proposed SERS biosensor for the miRNA-21 measurement. A limit of detection of 2.88 fM (S/N = 3) was achieved at an electrode temperature of 55 °C, which was decreased by more than three magnitudes compared with that at 25 °C.

Enzyme-free colorimetric detection of MicroRNA-21 using metal chelator as label for signal generation and amplification

Jiafang Piao,Qian Zhao,Dianming Zhou,Weipan Peng,Weichen Gao,Minghui Chen,Guiming Shu,Xiaoqun Gong,Jin Chang

MicroRNAs (miRNAs) were reported to be potential tumor markers for early diagnosis of cancer. Due to its short sequence, low expression level and high susceptibility to degradation, the stable and sensitive detection method of miRNAs is arduous to establish. In this work, we designed a metal chelator (ethylenediamine tetraacetic acid disodium salt, EDTA•2Na) labeled oligonucleotides as the plasmonic signal supraregulator probe to control the generation of gold nanoparticles (AuNPs). Based on another complementary oligonucleotides of target miRNA labeling SiO 2 microparticles (SiO 2 MPs) as the detecting platform, EDTA•2Na labeled oligonucleotide probes were immobilized on the SiO 2 platform through the sandwich structure in the presence of target miRNA. The sandwich chelating device could further chelate Au 3+ to regulate the generation of AuNPs, resulting in colorimetric signal to qualitatively and quantitatively detect the concentration of microRNA-21 (miR-21). The results indicate that the proposed metal chelator -labeled signal amplification method has outstanding sensitivity (LOD = 8.9 fM) and excellent stability, which will be benefit for the early accurate diagnosis of miRNAs.

A label-free electrochemical platform based on a thionine functionalized magnetic Fe–N–C electrocatalyst for the detection of microRNA-21

Xiangyu Ma,Kun Qian,Onome Ejeromedoghene,Martha Kandawa-Schulz,Wei Song,Yihong Wang

Taking a composite of a nanomaterial and a signal molecule as a substrate material can provide a label-free electrochemical platform. Besides, the nanomaterial with a high catalytic activity towards the signal molecule can improve the sensitivity of the platform. Herein, a thionine functionalized Fe–N–C nanocomposite was employed as the substrate. Firstly, the electrocatalytic activity of Fe–N–C towards the electroreduction of thionine was explored. Then, an immobilization-free and label-free electrochemical platform for the determination of microRNA-21 based on Fe–N–C–thionine/Fe3O4@AuNPs was constructed. A magnetic glassy carbon electrode (MGCE) was used to keep the magnetic Fe–N–C–thionine/Fe3O4@AuNPs modified onto the surface of the MGCE. Fe–N–C and Fe3O4 nanoparticles can co-catalyze the electroreduction of thionine and a strong electrochemical reduction signal of thionine could be realized in the differential pulse voltammetry (DPV) test. Also, a catalytic hairpin assembly (CHA) reaction was utilized to enhance the sensitivity of the developed electrochemical biosensor. Besides, the developed biosensor shows excellent specificity and reproducibility in the test of human serum samples, indicating its wide application prospects in the early-stage diagnosis of tumors.

An innovative “unlocked mechanism” by a double key avenue for one-pot detection of microRNA-21 and microRNA-141

Weipan Peng,Qian Zhao,Minghui Chen,Jiafang Piao,Weichen Gao,Xiaoqun Gong,Jin Chang

The accurate and quantitative detection of microRNAs (miRNAs) as next-generation, reliable biomarkers will provide vital information for cancer research and treatment. However, their unique, intrinsic features pose quite a challenge for miRNA profiling, especially for multiplexed detection. Thus, there is a strong and an ever-growing need to develop an accurate, simple, sensitive and specific miRNA sensing method. Methods: In this study, a simple and novel sensor is presented that uses a flow cytometry (FCM) method based on the double key “unlocked mechanism” and a fluorescence enrichment signal amplification strategy. The “unlocked mechanism” was cleverly designed via using hairpin DNA probes (HDs) labeled by fluorescent particles (FS) as the lock to block part of them, which can specifically hybridize with the probe on polystyrene microparticles (PS). The target miRNA and duplex-specific nuclease (DSN) forming the double key can specifically open the HDs and cleave a single-stranded DNA (ssDNA) into DNA/RNA dimers circularly in order to unlock the special part of the HDs to be specially enriched further on the PS. Results: The designed sensor with a hairpin structure and DSN special performance was found to have a high specificity. The circularly unlocking fluorescent probes and fluorescent signal enrichment can be beneficial for achieving a high sensitivity with a detection limit of 3.39 fM for miRNA-21. Meanwhile, the performance of multiplexing was estimated by simultaneous detection of miR-21 and miR-141, and the method also allowed for miR-21 detection in breast cancer blood samples. Conclusion: The designed sensor based on an “unlocked mechanism” and a signal enrichment strategy resulted in a one-pot, highly specific and sensitive detection of multiplex miRNAs. The whole detection without the need for a complex purification process is based on a FCM and is expected to have a great value in cancer diagnosis and biomedical research.

An enzyme-free electrochemical biosensor based on target-catalytic hairpin assembly and Pd@UiO-66 for the ultrasensitive detection of microRNA-21

Tianjiao Meng,Ningzhao Shang,Anaclet Nsabimana,Huimin Ye,Huan Wang,Chun Wang,Yufan Zhang

MicroRNA-21 (miR-21) has been widely investigated as important biomarkers for cancer diagnosis and treatment. Herein, a highly sensitive nonenzymatic electrochemical biosensor based on [email protected] frameworks ( [email protected] ) and target-catalytic hairpin assembly (CHA) with target recycling approach has been proposed for the detection of miR-21. The proposed biosensor integrates the efficient CHA strategy and excellent electrocatalytic performance of [email protected] nanocomposites. The concentration of miRNA-21 is related to the amount of the adsorbed electrocatalyst , leading to the different electrochemical signals for readout towards paracetamol (AP). This biosensor shows a low limit of detection of 0.713 fM with the dynamic range of 20 fM −600 pM under the optimal experimental conditions, providing a powerful platform for detecting miR-21. Furthermore, the designed biochemical self-assembly strategy of this electrochemical biosensor is promising candidate for potential applications in the analysis of other important genetic biomarkers for early diagnosis of cancers.
