BACKGROUND Sclerotinia stem rot caused by Sclerotinia sclerotiorum seriously endangers oilseed rape production worldwide, and the occurrence of fungicide-resistant mutants of S. sclerotiorum leads to control decline. Thus, it is critical to explore new green substitutes with different action mechanisms and high antifungal activity. Herein, the activity and the action mechanism of natamycin against S. sclerotiorum were evaluated. RESULTS Natamycin showed potent inhibition on the mycelial growth of S. sclerotiorum , and half-maximal effective concentration (EC 50 ) values against 103 S. sclerotiorum strains ranged from 0.53 to 4.04 μg/mL (mean 1.44 μg/mL). Natamycin also exhibited high efficacy against both carbendazim- and dimethachlone-resistant strains of S. sclerotiorum on detached oilseed rape leaves. No cross-resistance was detected between natamycin and carbendazim. Natamycin markedly disrupted hyphal form, sclerotia formation, integrity of the cell membrane, and reduced the content of oxalic acid and ergosterol, whereas it increased the reactive oxygen species (ROS) and malondialdehyde content. Interestingly, exogenous addition of ergosterol could reduce the inhibition of natamycin against S. sclerotiorum . Importantly, natamycin significantly inhibited expression of the Cyp51 gene, which is contrary to results for the triazole fungicide flusilazole, indicating a different action mechanism from triazole fungicides. CONCLUSION Natamycin is a promising effective candidate for the resistance management of S. sclerotiorum . © 2023 Society of Chemical Industry.

Summary Antibiotic-producing microorganisms have evolved several self-resistance mechanisms to prevent auto-toxicity. Overexpression of specific transporters to improve the efflux of toxic antibiotics has been found one of the most important and intrinsic resistance strategies used by many Streptomyces strains. In this work, two ATP-binding cassette (ABC) transporter-encoding genes located in the natamycin biosynthetic gene cluster, scnA and scnB , were identified as the primary exporter genes for natamycin efflux in Streptomyces chattanoogensis L10. Two other transporters located outside the cluster, a major facilitator superfamily transporter Mfs1 and an ABC transporter NepI/II were found to play a complementary role in natamycin efflux. ScnA/ScnB and Mfs1 also participate in exporting the immediate precursor of natamycin, 4,5-de-epoxynatamycin, which is more toxic to S. chattanoogensis L10 than natamycin. As the major complementary exporter for natamycin efflux, Mfs1 is up-regulated in response to intracellular accumulation of natamycin and 4,5-de-epoxynatamycin, suggesting a key role in the stress response for self-resistance. This article discusses a novel antibiotic-related efflux and response system in Streptomyces , as well as a self-resistance mechanism in antibiotic-producing strains.

In this study, natamycin nanoparticles were prepared by anti-solvent method, and natamycin nanoparticles/chitosan coating was prepared and investigated for the preservation of sweet cherry. The results of particle size showed that the prepared natamycin nanoparticles were nano-sized, with a particle size of 209 ± 10 nm. The results of scanning electron microscope (SEM) showed that the apparent morphology was a uniform spherical structure. Fourier transform infrared spectroscopy (FTIR) proved that the prepared natamycin after the combination of Polyvinylpyrrolidone (PVP) and natamycin still had its own functional groups without new chemical bond. Compared with the control samples the solubility of natamycin increased to 1 mg/mL, leading to the enhanced antifungal activity of natamycin nanoparticles/chitosan. Natamycin nanoparticles/chitosan effectively maintained the color of sweet cherry fruits and delayed the decline of firmness, total soluble solids (TSS) and titratable acidity (TA) during storage. In particular, it effectively delayed the decay of sweet cherries, with the lowest decay rate of 10.1 %, which was 66.4 % lower than that of the control group, and there wasn't decay of sweet cherry in 35 d storage. Therefore, natamycin nanoparticles/chitosan was effectively to delay cherry fruit senescence and improved the quality of sweet cherry during the storage period.

A new detection method of natamycin via HPLC–MS/MS was developed that uses an Agilent 6460 HPLC–MS/MS instrument and an Agilent RP18 (2.1 mm × 50 mm, 1.8 μm); the LOD was 1 μg/L, the LOQ was 3.34 μg/L, the recovery was 70–94%, the RSD was approximately 2–4%, and the total run time was 5 min. In the new detection method of natamycin via HPLC–MS/MS, processing of the sample before detection is necessary, which can be performed using solid phase extraction. Surprisingly, natamycin was not detected in any the sampled retail commercial wines from the Chinese market. Natamycin is very unstable in wine because many process steps, including the malo-lactic fermentation, clarification and storage, could result in degradation of natamycin. Regarding the clarifying agents, bentonite exhibited the strongest effect on natamycin. During the storage period, natamycin is very sensitive to light.

The preparation of active packaging loaded with antimicrobial, antioxidant, and other functional agents has become a hot topic for food preservation in recent years. In this field, active fiber films based on spinning methods have attracted the interest of researchers owing to their high specific surface area, high porosity, high loading capacity, and good controlled release capacity. In the present work, neatly arranged ethyl cellulose (EC)/polyvinyl-pyrrolidone (PVP) fibrous films loaded with natamycin as an antimicrobial agent were prepared by microfluidic spinning. The encapsulation efficiency of natamycin was more than 90% in each group and the loading increased with increasing natamycin content. According to the characterization results of the natamycin-loaded EC/PVP fibrous films, hydrogen bonding was formed between natamycin and EC and PVP in the fibrous films. Meanwhile, the water contact angle of the fibrous films was increased, suggesting the improved hydrophobicity of the films. In the in vitro bacterial inhibition experiments, the active fiber films loaded with natamycin showed good antimicrobial activity, which could significantly inhibit the growth of gray mold. In conclusion, N-EC/PVP fibrous films with antimicrobial activity prepared by microfluidic spinning showed good potential in the field of active packaging.

In this study, natamycin-loaded zein/casein composite nanoparticles (Nata-Z/C NPs) were fabricated and incorporated into gelatin film to improve the film's physicochemical and antifungal properties. The mean particle diameter (MPD) and zeta potential (ZP) values of the Nata-Z/C NPs were characterized by dynamic light scattering (DLS) with 68.5 ± 1.2 nm and −33.1 ± 0.8 mV, respectively. The Nata-Z/C NPs were well dispersed in the gelatin film and improved its mechanical properties, including the tensile strength (from 30.5 ± 3.2 MPa to 51.4 ± 3.0 MPa) and percentage elongation at break (from 0.225 ± 0.087 to 0.685 ± 0.103). The NPs as hydrophobic filler also improved the water barrier properties to 0.335 × 10 −9 g ˑ m/m 2 ˑ Pa ˑ s and water contact angle to 122.55 ± 1.54° of the gelatin film. The possible interactions between the Nata-Z/C NPs and gelatin were studied by differential scanning calorimetry (DSC), Fourier-transform infrared spectroscopy (FTIR), and scanning electron microscopy (SEM) techniques. The sustained release of natamycin from the film was achieved by its encapsulation, and the release kinetics were assessed with the Avrami model. Furthermore, the nanocomposite films exhibited strong antifungal activities against Aspergillus niger , Botrytis cinerea , and Penicillium citrinum . This study revealed that the Nata-Z/C NPs could be used as both nanofillers and functional additives to improve the physicochemical and bacteriostatic properties of gelatin films.

The separation of chiral amino acids (AAs) and their derivatives has always been a research difficulty in the field of biochemistry due to the high similarity of enantiomeric structures. In this work, a simple and quick method using natamycin (Nat) as chiral selector has been developed to simultaneously separate chiral AAs and their derivatives of carbobenzoxy/benzyl-AAs (Cbz/Bzl-AAs) by trapped ion mobility spectrometry-mass spectrometry (TIMS-MS). Specifically, 12 groups of the Cbz-AAs and Bzl-AAs can get baseline mobility separation by simple mixing with Nat to form binary diastereomeric complex ions [Nat+(Cbz-D/L-AA)+H] + and [Nat+(Bzl-D/L-AA)+H] + . While for the remained 5 groups of Bzl-D/L-AAs and 16 groups of D/L-AAs with unsatisfying separation, by further adding P-toluenesulfonic acid (PTS), the formed ternary complexes can allow their baseline chiral separation . Specifically, Bzl-D-AAs and Bzl-L-AAs get much improved separation effect by the formed diastereomeric complexes of [Nat+(Bzl-AA) 2 +PTS 2 +H] + , which the R p-p was improved from 0 to 2.40; while the D/L-AAs can get baseline separation by the formed diastereomeric complexes of [Nat + AA + PTS + H] + , [Nat + AA+(PTS) 2 +H] + , and [Nat+(AA) 2 +(PTS) 2 +H] + , with the R p-p ranged from 0.44 to 3.53. Definitely, PTS is the first time reported as the ligand to improve the separation effect for the enantiomers, and with the higher assembly of chiral analyte, Nat, and PTS might enable better chiral separation for the chiral amino acid and their derivatives. Moreover, method validation of relative quantification and accuracy for the D/L-AA and their derivatives were measured in a mixture, yielding R 2 greater than 0.99 and RSD% ≤ 2.68%. Overall, Nat and PTS as chiral selector and ligand can be widely used for chiral AAs and their derivatives mobility separation, and potentially for the separation of other AA-related chiral molecules.

HPCS-stabilized lignin nanoparticles enhanced natamycin solubility and stability. • Gelatin/ZnO films incorporated with LNPs showed superior mechanical and barrier properties. • Films exhibited synergistic photothermal and ROS-mediated antibacterial activity. • Lignin shielded natamycin from degradation, balancing stability and antibacterial efficacy. • Coating extended sweet potato shelf life and showed favorable biodegradability. To address the poor photostability and low aqueous solubility of natamycin (N), a hydroxypropyl chitosan (HPCS)-stabilized lignin nanoparticle (LNP) delivery system (LNP-N/HPCS) was developed in this study. The results showed that the nanocarriers exhibited a high encapsulation efficiency (89.17 ± 2.8%) and a moderate loading efficiency (11.63 ± 0.1%), while significantly increasing the solubility of natamycin to 13.5 times that of its free form at room temperature. Structural analysis indicated that LNP-N/HPCS formed stable core–shell-like nanostructures through hydrogen bonding, van der Waals forces, and π–π interactions, enabling efficient loading and stable dispersion. Furthermore, the delivery system was incorporated into gelatin/ZnO composite films (Gel/ZnO/LNPs) to construct multifunctional active packaging materials. The resulting composite film exhibited good antioxidant activity, mechanical properties, and gas barrier properties. After 10 min of xenon lamp irradiation, the composite film rapidly heated to 54.6 °C. After 150 min of irradiation, 67.55% of the initial natamycin content remained in the Gel/ZnO/LNPs5 composite film, whereas the retention rate of free natamycin was only 24.72%, indicating that the LNP delivery system improved the photostability of natamycin. In addition, the composite system exhibited good antibacterial activity in both film-forming solutions and solid films. In particular, Gel/ZnO/LNPs5 exhibited nearly complete growth inhibition against Escherichia coli , Staphylococcus aureus , and Dickeya dadantii E36 after 10 min of light irradiation, and achieved an inhibition rate of approximately 85% against Fusarium solani-melongenae 24–3. After 150 min of continuous irradiation, the Gel/ZnO/LNPs5 film still exhibited an inhibition rate of 55.67% against Fusarium solani-melongenae 24–3. Practical application results showed that the composite film could delay quality deterioration of sweet potatoes during storage to some extent and gradually disintegrated within approximately 6 days in soil. In addition, no significant inhibitory effect was observed in the seed germination test. Overall, this study provides an effective strategy for developing multifunctional food packaging materials with photothermal responsiveness, light-enhanced antimicrobial activity, and postharvest preservation functions. Blueberries and grapes are highly susceptible to fungal infections during pre-harvest, transport, and postharvest stages, leading to substantial economic losses. Conventional control methods often involve high equipment costs and contribute to the emergence of drug-resistant pathogens. Recently, the antifungal potential of volatile organic compounds (VOCs) has gained increasing attention as a sustainable alternative. Bacillus amyloliquefaciens HJ-B8 was successfully screened for its dual functionality of direct contact inhibition and volatile-mediated inhibition against Penicillium expansum . The VOCs produced by HJ-B8 were analyzed revealing a complex profile with several compounds that may contribute to its antifungal effect. The interaction between natamycin (Nata) and the HJ-B8 strain was also explored, showing a synergistic effect on fungal inhibition. A novel antifungal coating system was developed by combining HJ-B8 and Nata-loaded zein nanoparticles (HJ-B8@Nata-Zein NPs@Amy-Fe²⁺) to enhance the preservation of blueberries and grapes. The antifungal activities of this coating were evaluated in vitro and in vivo . The coating was further characterized by evaluating particle size, zeta potential, encapsulation efficiency, and light stability and assessed the antifungal efficacy. The in vivo tests demonstrated that the coating effectively reduced the decay rate, weight loss, soluble solids content and anthocyanin levels in treated blueberries and grapes. Furthermore, scanning electron microscopy (SEM) revealed improved preservation of fruit surface and flesh integrity. This opens up new possibilities for post-harvest preservation of berries using a synergistic antifungal coating system and provides a theoretical basis for related applications.

The β-cyclodextrin-acrylamide (CDM-AM) copolymer was prepared from acrylamide and β-CD maleate (CDM) using K 2 S 2 O 8 as initiator. The effects of the CDM-AM copolymer on the solubility and fungicidal activity of natamycin (NM) and carbendazim (MBC) were investigated. The stability constant of NM·CDM-AM and MBC·CDM-AM complexes at 303 K were of 10,725.45 M −1 and 3000.89 M −1 , respectively. The complexes were characterized using phase solubility diagrams, NMR spectra and FT-IR spectra. The analysis of the biological activities of these two complexes indicated that they possessed enhancing fungicidal activities compared to NM and MBC alone.

Brown rot caused by Monilinia fructicola ( M. fructicola ) severely affects Shengzhou nane ( Prunus salicina var. taoxingli ). Natamycin (NAT), a natural antimicrobial preservative, has demonstrated good efficacy in mitigating post-harvest decay and maintaining fruit quality. This study investigated antifungal efficacy and underlying mechanism of NAT against M. fructicola in vitro and in vivo settings. In vitro assay demonstrated that NAT exhibited potent antifungal activity against mycelial growth and spore germination of M. fructicola , with efficacy increasing in correlation with higher concentrations of NAT. Specifically, 20.0 mg L −1 NAT induced notable mycelial and spore deformation, characterized by twisting and breakage, while 25.0 mg L −1 NAT completely inhibited mycelial growth and spore germination. NAT exposure suppressed antioxidative enzyme activity, for example, 20 mg L −1 NAT decreased SOD activity by 61.49% of the control at 24 h, and downregulated related gene expression, leading to intracellular accumulation of ROS and oxidative stress. Furthermore, NAT disrupted the integrity of cellular membrane, resulting in leakage of intracellular contents. For instance, DNA leakage increased by 3.2-fold after 24 h. NAT exposure damaged mitochondrial structural and impaired its function, as indicated by the decreased adenosine triphosphate content, by 58.54% of the control, lower energy charge levels, and the reduced activities of malate dehydrogenase, succinate dehydrogenase, Ca 2+ /Mg 2+ -ATPase, and Na + /K + -ATPase by 79.67%, 77.57%, 81.34%, and 89.79% of the control, respectively, following a 24 h exposure. In vivo experiments indicated that NAT treatment resulted in 93.30% reduction in disease incidence at concentration of 200 mg L −1 , as well as a decrease in lesion diameter in Shenzhou nane fruit. Additionally, NAT enhanced the activities of antioxidative enzymes, including superoxide dismutase, catalase, and peroxidase a significant reduction in malondiadehyde levels. NAT also promoted the activity of phenylalanine ammonia-lyase, chitinase, β-1, 3-glucanase, and polyphenol oxidase. The alleviation of oxidative stress and enhancement of defense enzyme activity together increased fruit resistance to M. fructicola . Consequently, higher fruit quality was achieved during storage, thus, 200 mg L −1 NAT increased total soluble solids by 2.42 folds of the control. In conclusion, our findings demonstrated that NAT exhibited a dual-action effect against antibacterial M. fructicola , acting both as a direct antifungal agent and an inducer of host fruit's innate defense mechanism. These findings highlight the potential of NAT treatment as an effective strategy for controlling brown rot caused by M. fructicola in Shengzhou nane fruit

In this study, solution blow spinning (SBS) was used to rapidly develop gelatin/zein/polyurethane (PU) complex nanofibers for application in food antimicrobial packaging. The mean diameters of the gelatin/zein side were between 569.1 nm and 681.7 nm. The thermal stability of the nanofibers was improved caused by the formed hydrogen bonds between natamycin and the proteins. The water contact angle was increased with natamycin mixed in, suggesting that the hydrophobicity of the fibers improved. The addition of PU improved the elongation at break of the nanofibers from 13.58 % to 531.38 %. Moreover, the nanofibers showed a continuous release of natamycin for 240 h, and possessed good antimicrobial activity against Botrytis cinerea and Alternaria alternate . In addition, the use of SBS technology to fabricate nanofibers obtained a rapid feed rate of 3 mL/h, indicating that SBS is a promising technology to rapidly develop nanofibers for food packaging.
