Developing a functional media for precise identification of trace shellfish toxin would underpin the effective assessment of marine pollution. Herein, a novel monolithic column with a dual-mode strategy integrating antifouling and aptamer bionic affinity recognition was proposed for online specific identification of the marine toxin okadaic acid (OA). The zwitterionic monomer 2-(methacryloyloxy)ethylphosphorylcholine (MPC) and aptamers were synergistically employed to enable efficient reduction of matrix interferences and selective capture of target OA. Preparation optimization, characterization, and fouling-resistant mechanism of the dual-mode bionic monolith were evaluated. The zwitterion phosphorylcholine MPC introduced into the monolith significantly improved the fouling resistance to biomass substrates, meanwhile the aptamers were able to provide a high specific recognition capacity. Coupled with LC-MS, the as-prepared monolith provided an effective approach for highly selective and sensitive identification of OA. Good recovery yields of over 90% in shellfish tissue extracts and human serum were achieved with a sensitive limit of detection (LOD) as low as 0.1 ng/mL, as well as excellent specificity and low interference from proteins, fatty acids and analogues. Applied to popular shellfishes (such as clams, mussels, and oysters) and serum samples, trace OA toxin was accurately distinguished and quantified with satisfactory recoveries as 93.8 ± 2.2% – 99.8 ± 2.0% (n=3). Compared to the traditional HLB cartridge and other materials in the LC-MS method, the resulting anti-fouling aptamer monolith provided a more advanced online analysis mode with higher sensitivity and better resolution of OA in biological samples. It might provide an attractive access to an online bionic recognition platform with LC-MS for efficient, anti-interference and sensitive specific detection of trace marine toxin OA in biological samples.

Shellfish toxins are common marine toxins that have attracted worldwide attention due to their serious threats to food safety and human health. In this work, DNA hydrogel based on rolling circle amplification (RCA) and click reaction was developed for surface enhanced Raman scattering (SERS) detection of okadaic acid (OA) combined with target-responsive DNAzyme signal amplification. The RCA strands containing azido-dCTP were combined with cross-linked strands with ethynyl groups via click reaction to form a DNA hydrogel framework. The click reaction strengthened the DNA hydrogel network, making it more rigid and easier to manipulate during the detection process. The aptamer and DNAzyme were introduced into the DNA hydrogel as functionalized DNA components. Upon the specific recognition of OA by the aptamer, the DNAzyme released via the displacement reaction would continuously cleave the enzyme cutting site within the substrate strand to liberate the SERS signal tag. The functionalized DNA hydrogel concurrently achieved the functions of target recognition, signal conversion and signal amplification. Under optimal conditions, this strategy could detect OA with limits of detection as low as 0.2 nmol/L within 30 min. The SERS signal intensity showed good linear relationship with OA concentration in the range of 1.0–300.0 nmol/L. The practical application was validated for detecting trace amounts of OA in aquatic product samples. The stable DNA hydrogel combined with target-responsive DNAzyme signal amplification offers a universal strategy for rapid detection of trace toxins in complex matrixes.

Abstract Okadaic acid (OA) is a marine toxin which inhibits protein phosphatases. Its toxicities in several tissues have been well established, but the influence of exposure to low OA concentrations in male reproductive system is poorly understood. Here we aimed to explore the potential reproductive toxicity of OA and the mechanism behind it. The GC-1 cells were exposed to OA, and the changes of proliferation rates were detected. TUNEL assay was also conducted to determine the GC-1 cell apoptosis. Effects of OA on cytoskeleton and cyclin expression were investigated by fluorescein conjugated phalloidin. Potential molecular regulation on proliferation was analyzed based on transcriptomic analysis. OA inhibited the proliferation of GC-1 cells apparently with loss of cell-cell junctions and focal aggregated around the cells. The cyclin D/E mRNA and protein levels significantly decreased and the phosphorylation of ERK1/2 was blocked concordantly in OA treated cells. PPI network analysis pointed out ERK1 as a key node of responsive gene to the injury. In conclusion, OA shows apparent cytotoxicity on spermatogonial cells of male reproductive system and the cytotoxicity present was revealed by affecting the activation of ERK1/2 MAPK pathway which hinders the cyclin expression and actin stress fiber formation further.

Okadaic acid (OA) is one of the most common marine biological toxins, which can cause human poisoning or even death. It is desirable and essential to develop novel approaches for OA detection in a convenient, practical and low-cost manner. In this study, a novel piezoelectric aptasensor was designed for the detection of OA. The aptamers specifically binding with OA were used as the sensing elements. Quartz crystal microbalance (QCM) device was used as the transducer for responsive signal detection. Gold nanoparticles (AuNPs) were used to amplify the detection signals. The results showed that the linear detection range of OA was 0.5 ∼ 200 nM with a detection limit of 0.32 nM. This biosensor provides a novel approach for inexpensive, ultrasensitive, well-specific and label-free detection of OA as well as other marine biological toxins with minor modifications.

In this work, we present a novel dual-emissive fluoroimmunoassay for synchronous monitoring of okadaic acid (OA) and saxitoxin (STX) using multicolor fluorescent labels composed of sulfur, phosphorous co-doped graphene quantum dots (S, P-GQDs), and ovalbumin (OVA)-coated gold nanoparticles (OVA-AuNPs). The novel OVA-AuNPs were prepared by the reduction of chloroauric acid under alkaline conditions using OVA as a reducing agent. Both S, P-GQDs and OVA-AuNPs exhibit bright fluorescence, more importantly, a large emission wavelength difference (Δλ = 156 nm) under an excitation of 400 nm and relatively independent fluorescence behavior, which are essential to realizing the dual-signal marks in a directly mixing system. Using a competitive fluorescence-linked immunosorbent assay (cFLISA) format, the dual-emissive cFLISA was successfully utilized to measure OA and STX contents in Alectryonella plicatula (commonly named as fingerprint oyster) and the detection results were in good agreement with the commercial enzyme-linked immunosorbent assay (ELISA) kits.

Membrane-based micro-solid phase extraction (M-μSPE) has garnered great attention in sample pretreatment, suffering an inherent contradiction between permeability and adsorption capacity. In this study, a pure microporous organic network (TEB-DIB-MON) fiber membrane was prepared by combining electrostatic spinning technology, Sonogashira-Hagihara reaction and template sacrifice method. The prepared TEB-DIB-MON membrane exhibited a large specific surface area with a hollow and porous structure, thereby providing excellent solvent permeability and high adsorption capacity for okadaic acid (OA, an algal toxin). Under the optimized conditions, a sensitive analytical method was established by coupling M-μSPE with high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). The established method has a low detection limit (0.5 pg mL −1 ), a wide linear range (1.5–1000 pg mL −1 , R ≥ 0.9991), and good reproducibility (RSD ≤ 9.4 %, n = 6), which was then successfully applied for OA detection in marine organisms. Trace amounts of OA (59.3–89.0 pg mL −1) was detected in the oyster and prawn samples. This work demonstrated that the excellent application potential of MON membranes in sample pretreatment, while also presents a novel synthesis strategy for MONs membranes.

This work describes an immunosensor for the determination of okadaic acid (OA) using a Love-wave sensor with the immunogold staining method. In this assay, OA-BSA was captured by the anti-BSA antibody (BSA-MAb) bond with staphylococcal protein A (SPA) on the sensor surface via Fc regions. Cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) were employed to monitor the fabrication steps of immunosensor using the redox probe of K 3 Fe(CN) 6 . An indirect competitive immunoassay format was employed to detect OA after the immunosensor was prepared and gold nanoparticles (AuNPs) were introduced to amplify the phase signal. The results demonstrated that developed immunosensor was able to specifically respond to OA ranging from 10 to 150 ng/mL, providing limit of detection of 5.5 ng/mL. When applied to determine OA concentration in real sample, the results agreed well with those obtained by the commercial ELISA assay. The developed immunosensor is therefore suitable and has great application potential in detection of OA in shellfish.

Okadaic acid (OA) biotoxin acts a well-established inhibitor of protein phosphatase even a tumor promoter of human being, arouse great attention in safety monitoring. However, the powerful and convenient nanosensing technologies for addressing the demands such as rapidity, high sensitivity, and stability in the in-field test of OA shellfish toxin is still scarce. Herein, a high-performance magnetic biometric nanosensor (MBNS) integrating oriented aptamers and ultrasensitive laser-induced fluorescence (LIF) was firstly proposed for the in-field detection of trace OA in seafoods. High-density aptamers hybridized with FAM-labeled cDNA were tethered to the surface of AuNPs on magnetic MIL-101@Fe 3 O 4 , and then finely regulated by mercaptohexyl alcohol (MCH) to be orderly assembled, as was successfully utilized to engineer an active biological nanosensor for highly specific recognition of OA. Aptamers anchored on magnetic Fe 3 O 4 @MOF@AuNPs activate a biometric microreactor of OA, in which the superior LIF properties, conformation regulation of aptamer, and the specific recognition using aptamer genes were adopted. The magnetic nanosensor with an excellent specificity and super sensitivity for OA analysis was achieved within 20 min. Moreover, the content of captured OA could facilely be recorded by measuring the fluorescence intensity , and the limit of detection (LOD) and limit of quantitation of OA (LOQ) reached 0.015 and 0.050 ng/mL respectively, which was far better than most aptamer-based biometric sensing methods. The feasibility for accurate test of trace OA toxin in the fortified shellfish samples was validated with the recovery yields of 88.2–107.5% and RSD of 0.5–7.6%, respectively. The result demonstrated that the oriented-aptamer encoded MNS had significant practical values in rapid and ultrasensitive detection of OA biotoxin and the related safety applications.

Algal toxins can cause neurovirulence, hepatotoxicity, and cytotoxicity in humans through the consumption of contaminated water and food. In this work, we presented a novel aptasensor for the simultaneous detection of two algal toxins, microcysin-LR (MC-LR) and okadaic acid (OA). This system employed green and red upconversion nanoparticle (UCNP) luminescence as the donors and two quenchers (BHQ 1 and BHQ 3 ) as the corresponding acceptors. The two donor–acceptor couples were fabricated by hybridizing the aptamers with their corresponding complementary DNA. The results indicated that the green and red upconversion luminescence could be quenched by the quencher probes because of their highly overlapping spectrum. In the presence of MC-LR and OA, the aptamers preferred to bind to their corresponding analytes and de-hybridize with the complementary DNA. This effect became sufficiently large to prevent green and red luminescence quenching. Under the optimized experimental conditions, the relative luminescence intensity increased as the algal toxin concentrations increased, allowing for the quantification of MC-LR and OA. The relationships between the luminescence intensity and plotting logarithms of algal toxin concentrations were linear in the range from 0.1 to 50 ng mL −1 for MC-LR and OA. As a practical application, this type of dual fluorescence resonance energy transfer (FRET) aptasensor was used to monitor the MC-LR and OA levels in naturally contaminated food samples such as fish and shrimps.

Background Shellfish toxin residues (e.g., okadaic acid, OA) in seafood are a global concern, and the development of precise, sensitive portable test kits is therefore necessary. Magnetic nanoparticle-aptasensors (MNAS) are attractive for specific recognition and sensitive assay, yet direct epitaxial aptamer immobilization on Fe 3 O 4 causes thin, scattered coatings, leading to limited probe capacity and non-specific adsorption. Fabricating high-efficiency MNAS with dense, ordered aptamer arrays for precise detection thus remains a challenging yet promising goal. Results We propose a high-performance magnetic nanoparticle-aptasensor (MNAS) mediated by UiO-66-NH 2 MOFs prepared with a secondary growth strategy (denoted as UiO/UiO-MOFs), in which AuNPs were uniformly anchored on MOF and facilitated the orientation-ordered nucleic acid aptamers enabling specific test of trace OA. The UiO/UiO-MOFs served as a scaffold with ordered amino group arrays, facilitating the construction of a homogeneous AuNPs interface and thus the tethering of high-density aptamers (up to 12.5 μmol/kg). This sensing platform synergistically integrated the advantages of ordered UiO/UiO-MOF self-assembly, robust AuNPs bridging, and the spherically ordered configuration of dense aptamers, thereby achieving efficient target OA recognition with minimal non-specific cross-reactivity. Coupled with laser-induced fluorescence (LIF) detection, the sensor exhibited excellent specificity, ultra-high sensitivity (limit of detection, LOD = 30 pg/mL), and outstanding stability. Notably, the sensor was successfully applied for the accurate on-site detection of OA residues in real mussel and clam samples, with satisfactory recovery rates ranging from 83.0±3.3% to 107.7±1.3%. Significance This study posed an affordable yet powerful strategy capable of simultaneously realizing efficient UiO/UiO-MOFs surface mediation surface mediation and high capacity of spherical orientation-ordered aptamers. It expands the toolkit for epitaxial aptamer immobilization on MNAS, facilitates the ordered anchoring of aptamers, and thus enables the construction of advanced MNAS tailored for shellfish toxin detection.

Okadaic acid (OA), a potent diarrhetic shellfish toxin produced by marine dinoflagellates, accumulates in shellfish and poses a significant threat to human health by causing severe gastrointestinal illness. Consequently, the highly sensitive detection of OA is of crucial importance. In this study, an electrochemiluminescence (ECL) biosensor for OA detection was developed based on a Tb(Ⅲ)-Cbatpy-MOGs complex and a nucleic acid amplification strategy. The sensor integrates a cyclic DNA walker with the enzymatic assistance of Exonuclease III for accurate target quantification. The ECL intensity showed a strong linear relationship with the logarithm of OA concentration over a wide range, achieving a detection limit as low as 0.16 pg/mL, attesting to its high sensitivity. Furthermore, the biosensor exhibited excellent selectivity, reproducibility and stability, confirming its reliability for practical applications. This work provides a robust and highly sensitive method for OA detection and holds great promise for the practical monitoring of marine toxins.

Okadaic acid (OA) is a lipophilic marine biotoxin. In this study, OA was coupled with the carrier proteins keyhole limpet hemocyanin and bovine serum albumin as immunity and detection antigens by an active ester method. The polyclonal antibody against OA was prepared successfully, an indirect competitive ELISA (ciELISA) developed for the detection of OA in shellfish, and the reactive conditions of ciELISA optimized. The LOD (15% inhibition concentration) for the microwell plates was 1.28±0.38 ng/mL, corresponding to 12.8±3.8 ng/g. Two extraction methods were used to remove shellfish matrix interference with high recovery of spiked samples, and the methanol extraction of shellfish mussel was analyzed after dilution in phosphate-buffered saline. For validation of the optimized ciELISA, spiked and natural samples were analyzed by ciELISA, and HPLC with fluorescence detection. The correlation of linear regression equation was y=1.0064x–10.234, and the correlation coefficient was 0.9347. From the results of the comparative study, the established ciELISA assay using polyclonal antibody against OA could be used in preliminary screening of suspicious shellfish samples.
