Screening drugs and compounds to fight against Cronobacter sakazakii (C. sakazakii), one of the most common pathogens that can cause fatal necrotizing enterocolitis, septicema and meningitis, is still needed. We found that p-coumaric acid (pCA) has an inhibitory effect on C. sakazakii in vitro and in vivo. Proteomic changes of C. sakazakii BAA-894 exposed to pCA were studied to reveal the antibacterial mechanisms involved. A total of 1,553 proteins were identified in C. sakazakii BAA-894 by label-free proteomics analysis. Fuzzy cluster analysis showed that 33 were up-regulated, and 110 were down-regulated with pCA treatment. Gene Ontology (GO) analysis concluded that pCA caused the change of metabolic state of bacteria and generally in the state of metabolic inhibition. KEGG Enrichment Analysis (KEGG) analysis showed that pCA inhibited energy metabolism and distorted the balance of amino acid metabolism. Posttranslational modification analysis showed that pCA affected the deamidation of three proteins, including Elongation factor Tu, one of the vital proteins in bacteria. Molecular docking suggested the hydrogen bond between the pCA carboxyl group and Elongation factor Tu Asn-64 might contribute to deamidation. Overall, we found that pCA interfered with cellular energy and amino acid metabolism and promoted elongation factor Tu deamidation, suggesting that pCA can be an effective natural substitute to control C. sakazakii.

Background: P-coumaric acid (p-CA) is a kind of phenylpropionic acid derived from aromatic amino acids, which is widely distributed in many plants and human diets. It has strong pharmacological and inhibitory effects on a variety of tumors. However, the role of p-CA in osteosarcoma, a tumor with a poor prognosis, is still unknown. Therefore, we aimed to evaluate the effect of p-CA on osteosarcoma and explore its potential mechanism. Objective: This study aimed to investigate whether p-CA has an inhibitory effect on the growth of osteosarcoma cells and explore its potential mechanism. Methods: MTT assay and clonogenic assay were used to detect the effect of p-CA on the proliferation of osteosarcoma cells. The effect of p-CA on apoptosis of osteosarcoma cells was detected by the Hoechst staining and flow cytometry. The effects of p-CA on the migration and invasion of osteosarcoma cells were detected by scratch healing assay and Transwell invasion assay. Western blot and PI3K/Akt pathway activator 740Y-P were used to detect the anti-tumor mechanism of p-CA on osteosarcoma cells. The effect of p-CA on osteosarcoma cells in vivo was verified by an orthotopic osteosarcoma tumor animal model in nude mice. Results: MTT assay and clonogenic assay showed that p-CA inhibited the proliferation of osteosarcoma cells. Hoechst stain and flow cytometry showed that p-CA could induce apoptosis of osteosarcoma cells and lead to G2 phase arrest of osteosarcoma cells. Transwell assay and scratch healing assay found that p-CA could inhibit the migration and invasion of osteosarcoma cells. Western blot showed that p-CA could inhibit the activity of the PI3K/Akt signaling pathway in osteosarcoma cells, and 740Y-P could reverse its inhibitory effect. In vivo mouse models, p-CA has an antitumor effect on osteosarcoma cells in vivo, and at the same time, it has less toxic side effects on mice. Conclusion: This study demonstrated that p-CA could effectively inhibit the proliferation, migration and invasion of osteosarcoma cells and promote apoptosis. p-CA may play an anti-osteosarcoma role by inhibiting PI3K/Akt signaling pathway.

Developing novel bilayer food packing film having the ability to prevent bacterial infections and capable of inhibiting oxidation is utmost important, since bacterial infections and oxidation can cause food spoilage. Ag-Metal−organic framework loaded p-coumaric acid modified chitosan (P-CS/Ag@MOF) or chitosan nanoparticles (P-CSNPs/Ag@MOF) and polyvinyl alcohol/starch (PVA/ST) were used as the upper film and lower layer film to successfully prepare a bilayer composite film. The microscopic morphology, water resistance, oil resistance, oxidation resistance, optical properties, cytotoxicity and antibacterial properties of the composite films were compared. The results showed that the surface of P-CS/Ag@MOF bilayer was relatively smooth and its tensile strength (TS) was higher (27.67 MPa). Among them, P-CS/Ag@MOF bilayer films had better oil resistance and oxidation resistance activity. In addition, the P-CS/Ag@MOF bilayer film had good UV-blocking properties and transparency. P-CSNPs/Ag@MOF bilayer film had higher antibacterial activity and cytotoxicity.

Trans-p-hydroxycinnamic acid and its esters in the leaves ofLigustrum robustummight be a new resource to prevent diabetes and its complications. In the present study, a new HPLC-UV method using hydrolyzation with sodium hydroxide for quantitation oftrans-p-hydroxycinnamic acid and totaltrans-p-hydroxycinnamic acid esters in the leaves ofL. robustumwas developed, since it was difficult and troublesome to analyze no less than 34trans-p-hydroxycinnamic acid esters by usual HPLC. The extract ofL. robustumwas hydrolyzed with sodium hydroxide at 80 °C for 2 h, and then, hydrochloride was added. HPLC analysis was performed in reverse phase mode using a C-18 column, eluting with methanol-0.1% acetic acid aqueous solution (40:60,v/v) in isocratic mode at a flow rate of 1.0 mL·min−1and detecting at 310 nm. The linear range fortrans-p-hydroxycinnamic acid was 11.0–352.0 μg·mL−1(r2= 1.000). The limit of detection and limit of quantification were 2.00 and 6.07 μg·mL−1, respectively. The relative standard deviations of intra-day and inter-day variabilities fortrans-p-hydroxycinnamic acid were less than 2%. The percentage recovery oftrans-p-hydroxycinnamic acid was 103.3% ± 1.1%. It is the first HPLC method using hydrolyzation for quantification of many carboxylic esters. Finally, the method was used successfully to determinetrans-p-hydroxycinnamic acid and totaltrans-p-hydroxycinnamic acid esters in various extracts of the leaves ofL. robustum. The 60–70% ethanol extracts ofL. robustumshowed the highest contents of freetrans-p-hydroxycinnamic acid (3.96–3.99 mg·g−1), and the 50–80% ethanol extracts ofL. robustumdisplayed the highest contents of totaltrans-p-hydroxycinnamic acid esters (202.6–210.6 mg·g−1). The method can be applied also to the quality control of the products ofL. robustum.Keywords:trans-p-hydroxycinnamic acid;esters;Ligustrum robustum;HPLC-UV;hydrolyzation;sodium hydroxide;quantification

Commercial epoxy resins are significantly limited in their applications in structural materials, packaging materials, and other fields due to inherent drawbacks such as the non-renewability of synthetic raw materials, high brittleness, and flammability. During the modification process, challenges related to the compatibility of additives and the “see-saw” issue between mechanical performance and flame retardancy persistently arise. This study presents the first synthesis of sustainable flame-retardant epoxy resins (p-CAIE) using biomass-derived p-coumaric acid (p-CA) and isophorone diisocyanate (IPDI) as raw materials. p-CAIE is utilized as a modifier for commercial epoxy resins, resulting in thermosetting materials (p-CAIE-E51/DDM) with excellent thermal stability, superior mechanical properties, and enhanced flame retardancy. The compatibility between the components is significantly improved, and the introduction of extensive hydrogen bonding within the crosslinked network leads to remarkable enhancements in performance. Specifically, the tensile strength and elongation at break of the 15 % p-CAIE-E51/DDM cured resin system increased by 119 % and 360 %, respectively, while the impact strength improved by 151 % compared to pure E51/DDM. The limiting oxygen index (LOI) reached 29.5 %, and the UL-94 test achieved a V-0 rating. Additionally, the peak heat release rate (PHRR), total heat release (THR), total smoke production (TSP), and carbon monoxide production (COP) were reduced by 54.2 %, 55.5 %, 53.6 %, and 40.5 %, respectively, effectively suppressing thermal and smoke release. The material also exhibited high transparency within the visible light range (400 nm-760 nm). Thus, this research provides an environmentally friendly strategy for the multifunctional manufacturing of bio-based epoxy resins, offering a promising solution for simultaneously enhancing the flame retardancy, toughness, and transparency of thermosetting epoxy materials.

Skeletal muscle atrophy is a prevalent complication in chronic kidney disease (CKD), and its pathogenesis is closely related to inflammation and oxidative stress. P-Coumaric acid (PCA) is a phenolic acid with anti-inflammatory and antioxidant pharmacological actions. This research aims to investigate the effect of PCA on CKD-induced muscle atrophy and its underlying mechanism. In our study, in vivo and in vitro models were established by using 5/6 nephrectomized rats and LPS-induced C2C12 myoblasts. The experimental results showed that PCA ameliorated kidney injury in CKD rats and increased skeletal muscle weight and the cross-sectional area of muscle fibres. In both CKD rats and LPS-induced C2C12 myoblasts, PCA also exhibited anti-inflammatory and antioxidant effects, reduced the levels of pro-inflammatory cytokines and enhanced the activity of antioxidant enzymes. Network pharmacology studies have identified 165 common targets between PCA and skeletal muscle atrophy. Furthermore, the experimental results also demonstrated that PCA decreased the expression of TLR4, MyD88, NF-κB p65, MurF1 and MAFbx at both the protein and mRNA levels. Additionally, in vitro experiments showed that the use of TLR4 agonists could reverse the muscle-protective effect of PCA. In summary, this study illustrated that PCA ameliorated skeletal muscle atrophy in CKD rats by inhibiting the TLR4/MyD88/NF-κB pathway.

The fabrication of multi-component film with colloidal particles could be inconvenient. A novel “swell-permeate” (SP) strategy was proposed to form homogeneous multi-component films. The SP strategy allows colloidal particles to fit into the polymer network by stretching the polymer chains assisted by water. We demonstrated the strategy by creating films with polysaccharide substrates as β-cyclodextrin grafted chitosan (CS) with nanocellulose. The addition of nanocellulose significantly increased the mechanical properties and the barrier performance of the films. The size of nanocellulose particles in affecting mechanical properties was investigated by applying different length of cellulose nanocrystal (CNC), the longer of which, due to denser physical entanglements, showed a better increase to the film in the elastic modulus and tensile strength to 4.54-fold and 5.71-fold, respectively. The films were also loaded with ethyl- p -coumarate (EpCA) and had an enhanced performance in anti-microbial for Altenaria alternata , Salmonella typhi , and Escherichia coli . The anti-oxidative property was increased as well, and both effects were valid both in vitro and in ready-to-eat apples. The strategy provides a practical and convenient method for fabricating colloidal particle containing films, and the novel idea of “swell-permeate” is potentially regarded as a new solution to the challenge of ready-to-eat food quality maintenance.

Novel p -coumaric acid (pCA)-loaded nanoliposomes were prepared by the thin-film hydration method, assisted with ultrasonic treatment, and optimized by the response surface methodology. The characterization showed that the fabricated pCA-loaded liposomes were nanosized spherical vesicles (83.55 ± 0.34 nm), exhibiting favorable dispersibility and encapsulation efficiency (55.70 ± 0.10 %). Fourier transform infrared spectroscopy analysis indicated that pCA was encapsulated into phospholipid bilayer through hydrophobic interaction and hydrogen bonding. Tests of temperature stability and centrifugal stability suggested that pCA-loaded nanoliposomes were less sensitive to aggregation and fusion during storage. Incubation experiments revealed that pCA-loaded nanoliposomes had a good inhibitory effect on the expansion of disease area on fresh pod pepper fruit caused by Botrytis cinerea . Additionally, pCA-loaded nanoliposomes effectively extended shelf life of fresh pod peppers by reducing weight loss and naturally-infected decays. The findings presented a viable strategy for designing liposomal encapsulation technology to efficiently enhance antimicrobial activity of pCA in food preservation.
