Redox/pH/NIR-responsive degradable silica nanospheres with fluorescence for drug release and photothermal therapy

Xinli Li,Mengru Chen,Yanbao Zhao,Lei Sun

In this work, S S bond-doped porous silica nanoparticles with CuS nanocores (CuS/OS) were prepared as drug carriers for chemical-photothermal synergistic therapy and redox/pH/light triple-response drug release. The synthesized CuS/OS carriers have dendritic pore structure with particle size of about 100 nm. Tiopronin (MPG) was used as model drug to evaluate the release performance of the carrier. Chitosan (CS) and green fluorescent curcumin (Cur) were used as plugs to seal the loaded drug, which endows the CuS/OS carrier with pH-responsive release and monitoring properties. The obtained MPG-loaded CuS/OS-CS-Cur has pH/GSH/NIR triple responsive release and redox-responsive degradation. The introduction of CuS nanocores gives the CuS/OS system a good photothermal therapy function. The CuS/OS-CS-Cur and its degradation products have good biocompatibility.

In-situ self-assembly of sandwich-like Ti3C2 MXene/gold nanorods nanosheets for synergistically enhanced near-infrared responsive drug delivery

Beibei Zhu,Jun Shi,Chongchong Liu,Jingguo Li,Shaokui Cao

As a novel two-dimensional material, Ti 3 C 2 MXenes has attracted lots of attention in biomedical filed for its large surface area and excellent near-infrared (NIR) responsiveness. In this paper, an in-situ growth and self-assembly approach was employed to combine gold nanorods (GNRs) with Ti 3 C 2 nanosheets to prepare intelligent sandwich-like Ti 3 C 2 @GNRs/PDA/Ti 3 C 2 nanohybrids . Compared with Ti 3 C 2 nanosheets, in-situ growth Ti 3 C 2 @GNRs possessed excellent photothermal conversion efficiency (45.89%), caused by the distinguished photothermal synergy between GNRs and Ti 3 C 2 nanosheets. Moreover, the high specific surface area of Ti 3 C 2 MXene and outstanding adhesion performance of PDA endowed Ti 3 C 2 @GNRs/PDA/Ti 3 C 2 nanohybrids with superior drug loading ability for doxorubicin hydrochloride (DOX) (95.88%). Besides, Ti 3 C 2 @GNRs/PDA/Ti 3 C 2 nanohybrids displayed distinct pH/NIR responsive drug release properties upon NIR irradiation owing to the strong π-π stacking interaction between Ti 3 C 2 @GNRs/PDA/Ti 3 C 2 and DOX, along with the excellent NIR-responsiveness of Ti 3 C 2 @GNRs/PDA/Ti 3 C 2 . This paper offers a practicable method to prepare Ti 3 C 2 MXene-based nanoplatform with synergistically enhanced NIR drug release behavior, brilliant biocompatibility and high drug loading efficiency, which is expected to be applied in remote cancer therapy.

Gold nanorods/graphene oxide nanosheets immobilized by polydopamine for efficient remotely triggered drug delivery

Qi Zeer,Shi Jun,Zhu Beibei,Li Jingguo,Cao Shaokui

Abstract In this paper, a novel one-pot strategy was utilized to immobilize gold nanorods (GNRs) onto the surface of PEGylated graphene oxide (GO-PEG) via polydopamine (PDA) to fabricate GNRs/GO@PDA hybrid nanosheets. The GNRs/GO@PDA nanosheets possessed excellent photostability and photothermal conversion efficiency (36.10%), due to the robust interfacial immobilization of PDA between GNRs and GO-PEG. Besides, GNRs/GO@PDA also could be utilized to be an efficient drug nano-vehicle that possessed an excellent loading ability for anticancer drug doxorubicin hydrochloride (DOX) (86.16%). Furthermore, the drug release of hybrid nanosheets could be triggered by pH value and near-infrared (NIR) light irradiation, which could be mainly ascribed to the strong π – π stacking interaction between GNRs/GO@PDA and DOX, as well as the brilliant NIR-responsive property of GNRs/GO@PDA. It was found that GNRs/GO@PDA possessed extremely low cytotoxicity to MCF-7 cells even at 250 μg mL −1 , while GNRs/GO@PDA(DOX) showed obvious cytotoxicity at 100 μg mL −1 . Thus, this paper presents a feasible strategy to fabricate GNRs/GO hybrid nanosheets with high drug loading capacity, excellent photothermal conversion property and pH/NIR-stimuli-responsive drug delivery performance, which possesses brilliant potentials to be an efficient platform for remotely triggered drug delivery. A multifunctional nanoplatform based on MoS2-nanosheets for targeted drug delivery and chemo-photothermal therapy

Yanbo Yang,Jianrong Wu,David H. Bremner,Shiwei Niu,Yu Li,Xuejing Zhang,Xiaotian Xie,Li-Min Zhu

Synergistic tumor treatment has recently attracted more and more attention due to its remarkable therapeutic effect. Herein, a multifunctional drug delivery system based on hyaluronic acid (HA) targeted dual stimulation responsive MoS 2 nanosheets (HA-PEI-LA-MoS 2 -PEG, HPMP) for active interaction with CD44 receptor positive MCF-7 cells is reported. Melanin (Mel), a new type of photothermal agent and doxorubicin (DOX) are both loaded onto the HPMP nanocomposite and can be released by mild acid or hyperthermia. The prepared HPMP nanocomposite has a uniform hydrodynamic diameter (104 nm), a high drug loading (944.3 mg.g −1 HPMP), a remarkable photothermal effect (photothermal conversion efficiency: 55.3%) and excellent biocompatibility. The DOX release from HPMP@(DOX/Mel) can be precisely controlled by the dual stimuli of utilizing the acidic environment in the tumor cells and external laser irradiation. Meanwhile, loading of Mel onto the surface can enhance the photothermal effect of the MoS 2 nanosheets. In vitro experiments showed that the HPMP@(DOX/Mel) nanoplatform could efficiently deliver DOX into MCF-7 cells and demonstrated enhanced cytotoxicity compared to that of the non-targeted nanoplatform. In vivo experiments in a breast cancer model of nude mice further confirmed that the HPMP@(DOX/Mel) significantly inhibited tumor growth under near infrared (NIR) laser irradiation, which is superior to any single therapy. In summary, this flexible nanoplatform, based on multi-faceted loaded MoS 2 nanosheets, exhibits considerable potential for efficient pH/NIR-responsive targeted drug delivery and chemo-photothermal synergistic tumor therapy.

A novel theranostic nanoplatform for imaging-guided chemo-photothermal therapy in oral squamous cell carcinoma

Mengqin Gu,Li Jiang,Liying Hao,Junzhuo Lu,Zhenqi Liu,Zixue Lei,Yijun Li,Chengge Hua,Wei Li,Xiyu Li

Oral squamous cell carcinoma (OSCC) is highly malignant and invasive, and current treatments are limited due to serious side effects and unsatisfactory outcomes. Here, we reported the terbium ion-doped hydroxyapatite (HATb) nanoparticle as a luminescent probe to encapsulate both the near-infrared (NIR) photothermal agent polydopamine (PDA) and anticancer doxorubicin (DOX) for imaging-guided chemo-photothermal therapy. The morphology, crystal structure, fluorescence, and composition of HATb–PDA–DOX were characterized. HATb–PDA showed a high DOX loading capacity. A theranostic nanoplatform showed pH/NIR responsive release properties and better antitumor outcomes in OSCC cells than monomodal chemotherapy or photothermal therapy, while keeping side effects at a minimum. Also, the luminescence signal was confirmed to be tracked and the increase of the red/green (R/G) ratio caused by the DOX release could be used to monitor the DOX release content. Furthermore, HATb–PDA–DOX plus NIR treatment synergistically promoted in vitro cell death through the overproduction of reactive oxygen species (ROS), cell cycle arrest, and increased cell apoptosis. Overall, this work presents an innovative strategy in designing a multifunctional nano-system for imaging-guided cancer treatment.

Multimodal theranostics augmented by transmembrane polymer-sealed nano-enzymatic porous MoS2 nanoflowers

Huiling Jiang,Yilin Du,Leilei Chen,Min Qian,Yafeng Yang,Taotao Huo,Xueying Yan,Tao Ye,Bing Han,Yi Wang,Rongqin Huang

Developing an all-in-one multimodal theranostic platform that can synergistically integrate sensitive photoacoustic (PA) imaging, enhanced photothermal therapy (PTT) and photodynamic therapy (PDT) as well as the nano-enzyme activated chemodynamic therapy (CDT) presents a great challenge for the current nanomedicine design. Herein, a simple hydrothermal method was used to prepare porous molybdenum disulfide (MoS 2 ) nanoflowers. These nanoflowers were assembled by three dimensional (3D)-stacked MoS 2 nanosheets with plentiful pores and large surfaces, which thus exhibited enhanced photothermal conversion via light trapping and peroxidase (POD)-like activity via active defects exposure. Consequently, this 3D-MoS 2 nanostructure could be well-sealed by polyethylene glycol-polyethylenimine polymer modified with nucleolar translocation signal sequence of the LIM Kinase 2 protein (LNP) via strong electrostatic interaction, which not only benefited to stably deliver anticancer drug doxorubicin (DOX) into the tumor cells for pH/NIR-responsive chemotherapy, but also provided strong photoacoustic, photothermal performances and stimulated generation of reactive oxygen species (ROS) for imaging-guided PTT/PDT/CDT combined therapy. This work promised a simple all-in-one multimodal theranostic platform to augment the potential antitumoral therapeutic outcomes.
