Early diagnosis and treatment can significantly enhance the prognosis of patients with non-small cell lung cancer (NSCLC); however, current diagnostic methods remain plagued by inadequate sensitivity or excessive invasiveness. This study describes the innovative construction of a dual-fluorescence, dual-target biosensor based on copper nanoclusters (CuNCs) and hollow mesoporous silica nanospheres (HMSNs) for highly sensitive detection of CD91 and CD151 on the surface of NSCLC exosomes. Specifically, HMSNs regulate the fluorescence signals of red and blue glutathione-CuNCs by loading Ce 3+ and Fe 3+ and conjugating a pH-responsive chitosan coating: Ce 3+ significantly enhances the fluorescence signal of red CuNCs by inducing their aggregation, whereas Fe 3+ quenches the fluorescence signal of blue CuNCs through oxidation. Exosomes are captured using a CD63 aptamer conjugated to streptavidin magnetic beads. HMSN-Ce 3+ -CS-antiCD151 and HMSN-Fe 3+ -CS-antiCD91 probes are then employed to target exosome surface proteins CD151 and CD91, respectively, allowing synchronous detection of dual-fluorescence and dual targets. We then measured and analyzed the change ratio of fluorescence signals, and optimized the detection parameters while evaluating the sensitivity, stability, and linear range of the system. The detection limits of red and blue fluorescence systems were 1058 particles·mL −1 and 239 particles·mL −1 , and ranges of linearity were 4 × 10 3 –4 × 10 8 particles·mL −1 and 7 × 10 2 –7 × 10 6 particles·mL −1 , respectively. Overall, this study introduces a novel strategy for the early screening of NSCLC, with high sensitivity and minimal invasiveness. The complementary design of the dual-fluorescence system extends its detection range and holds potential for clinical application.

Lesion core is the area with the most serious injury and vigorous repair. Existing nanocarriers are difficult to break through the targeted delivery to lesion core and the precise treatment for intracellular and extracellular microenvironment. Herein, a cellular membrane-engineered nanovesicle (CMEV) with hierarchical structure is constructed using double emulsion-extrusion method, by integrating neutrophil membrane, functional antibody and gelled drug-loaded core, as ‘three-stage booster’ to target the lesion core and deliver catestatin (CST), a small therapeutic peptide, for ischemic cardiomyopathy therapy. By coating neutrophil membrane outside the shell, CMEV is endowed with the function of neutrophil-like migration to achieve the first stage of tissue targeting. Based on the specific anchoring to injured myocardium, myosin light chain 3 (MLC3) antibody was embedded to fulfill the second stage of CMEV accumulation in lesion core. The gelled core containing CST-sodium alginate (NaAlg) with pH-responsive shell is prepared by ionic cross-linking to accomplish the third stage of precise CST administration. Triggered by the microenvironment, NaAlg electrostatically adheres to the lesion core for sustained release, enhancing the efficacy of CST in improving cardiomyocyte apoptosis, excessive fibrosis, macrophage polarization and angiogenesis. Thus, the “three-stage booster” nanovesicle significantly ameliorates cardiac function and adverse remodeling to treat ischemic cardiomyopathy. This article is protected by copyright. Using poly(acrylic acid) (abbreviated as PAA) as a shell, bimodal mesoporous silicas (abbreviated as BMMs) as a core, and bipyridine-proline (abbreviated as Z) as an active species, a facile approach for preparation of the [email protected] nanocomposites with core–shell structure was proposed, which was used as a suitable pH-sensitive catalyst for asymmetric aldol reaction between p-nitrobenzaldehyde and cyclohexanone. Their microstructural features and physicochemical properties were extensively characterized by various methods, such as powder X-ray diffraction patterns, N 2 adsorption–desorption isotherms, the scanning (and transmission) electron microscopy images, the dynamic light scattering measurements, the thermogravimetric analysis, the elemental analysis, and the Fourier transform infrared spectroscopy spectra. Particularly, their fractal evolution, the pair distance distribution function, and the Porod plots were demonstrated via the small-angle X-ray scattering patterns. The results elucidated that the PAA was successfully incorporated onto the external amino-modified surfaces of the mesoporous BMMs, and the particles having PAA grafted thereon had a pH-responsive shell in thickness of around 0.97 nm for controlled release of Z species by adjusting pH value of the reaction medium. Besides, the synthesized [email protected] presented the satisfactory catalytic yields (around 80%) in comparison with the homo- and heterogeneous catalytic system using Z or [email protected] as the catalysts.
