Improvement of pale, soft, and exudative (PSE)-like chicken breast protein is urgently needed, to reduce the economic losses induced by its downgraded processing quality. The pH-shifting treatment increases the emulsifying properties of the myofibrillar protein (MP) of PSE-like meat, but the emulsion stability under various environmental conditions has yet to be explored. The creaming index under 60 and 80 °C decreased from 20.1% to 28.9%–16.1% and 26.7%, respective, indicating a higher stability against heating. pH-shifting significantly increased the stability of the emulsion, particularly at pH 7.0 and 7.5. Also, the pH-shifted emulsion could more effectively resist instability brought by freeze-thaw cycles. However, at lower salt concentrations, the stability of the pH-shifted emulsion was poor. Throughout the 48-h storage period at 4 °C, the pH-shifted group reached stability faster than the control, and the turbiscan index values were stabilized at 1.8 and 4.7, respectively. Overall, pH-shifting could improve the emulsion stability of PSE-like MP against heating, pH altering, salt content changing, freezing-thawing, and storage.

Alaska pollock proteins (PPs) are aquatic proteins with a compact structure, which leads to poor solubility and limited application in food industries. This study explored the structural assembly and gelation behavior of the PP-soluble aggregates prepared by the refolding processing of acidic (pH 1.5–7) or alkaline (pH 12–7) pH-shifting treatment. PPs refolded at alkali pHs produced more soluble aggregates than at acid pHs. Lots of myosin heavy lights (MHCs) could interact with actin to form soluble aggregates at high pH rather than low pH. When shifting back to pH 7, the solubility of alkaline-shifted PPs was boosted to over 60% with a more flexible structure, while acidic-shifted PPs showed reduced solubility (29%) with increased rigidity. Molecular characteristics results indicated that alkaline-shifted PPs comprising MHCs and actin exhibited higher surface hydrophobicity and sulfhydryl groups. These structural modifications induced by alkaline pH-shifting contributed to improving the gelling, emulsifying and foaming properties of PPs. Intriguingly, both acidic- and alkaline-shifted PPs showed a retarded digestive behavior. These novel insights may be adaptable to other animal proteins for modulating the protein structure-functionality, expanding their applications in food industries.

In the current study, the interfacial rheological properties of alkali pH-shifted MP under pH 11.0 → 7.0, pH 11.5 → 7.0 and pH 12.0 → 7.0 have been firstly investigated. The molecular weight and microstructure of myofibrillar protein (MP) treated by alkali pH-shifting were investigated by gel-permeation chromatography (GPC) and transmission electron microscopy (TEM). Results indicated a swollen, organized and aggregated structure of refolded MP. Overall, the molecular change in refolded MP led to faster protein adsorption towards the O/W interface, enhanced intermolecular interaction between adsorbed proteins and improved elasticity of interfacial film compared to native MP. After β-mercaptoethanol (β-ME) was added, the advantage of refolded MP in diffusion phase no more existed due to the disruption of aggregation and decreased molecular size, besides the penetration and rearrangement were interfered because the disruption of disulfide bond inhibited the formation of interfacial protein layers. With the protein concentration increasing, MP molecules crowding at O/W (oil in water) surface limited the development of a highly elastic network. As revealed by Tween 20 displacement, MP layers with higher interfacial elasticity were more resistant to displacement. Moreover, after displacement the coalescence stability of emulsion droplet was enhanced but the mechanical strength of interfacial MP layer was reduced. Altogether MP (0.04 or 0.2 mg/mL) treated by pH 11.0 → 7.0 exhibited the best adsorption performance and interfacial properties, which were important in stablizing emulsions. Thus, interfacial behavior regulation by alkali pH-shifting may be a applicable process for developing meat protein-based emulsions in food industry.

Electrospun fibers of hydrophilic polymers meet challenges in a rapid degradation of fiber matrices and discharge of antibiotics to comply with requirements of infection control as a dermal regeneration template. In the current study, a pH conversion process is initially developed to ensure fluent electrospinning, an efficient in situ cross-linking of electrospun gelatin fibers with oxidized alginate and simultaneous loading of gentamicin sulfate (GS) and hydrophobic ciprofloxacin into fibers. The dual drug-loaded fibers indicate a complete release of GS during 6 d and a sustained release of ciprofloxacin for over three weeks, and the antibiotics release indicates significant growth inhibitions on Pseudomonas aeruginosa and Staphylococcus epidermidis . The wound healing efficacy is evaluated on a deep burn model infected with 10 8 CFU of P. aeruginosa . Compared with fibers with loaded individual drugs, the concomitant release of GS and ciprofloxacin significantly reduces the bacteria numbers in wound and livers, at around 2.30 × 10 5 and 1.25 × 10 3 CFU after 3 d, respectively. The wound re-epithelization, blood vessel formation, collagen deposition, and tissue remodeling process are accelerated with a complete healing observed after 21 d. This study provides a feasible strategy to design cross-linked hydrophilic fibers with an extended drug release for biomedical applications.
