GABAB receptor ligand-directed trimethyl chitosan/tripolyphosphate nanoparticles and their pMDI formulation for survivin siRNA pulmonary delivery

Suhui Ni,Yun Liu,Yue Tang,Jing Chen,Shuhan Li,Ji Pu,Lidong Han

The effect of gene silencing by survivin siRNA (siSurvivin) on the proliferation and apoptosis of lung tumor has been attracted more interest. GABA B receptor ligand-directed nanoparticles consisting of baclofen functionalized trimethyl chitosan (Bac-TMC) as polymeric carriers, tripolyphosphate (TPP) as ionic crosslinker, and siSurvivin as therapeutic genes, were designed to enhance the survivin gene silencing. GABA B receptor agonist baclofen (Bac) was initially introduced into TMC as a novel ligand. This Bac-TMC/TPP nanoparticles increased the uptake of survivin siRNA through the interaction with GABA B receptor, further resulted in efficient cell apoptosis and gene silencing. For siRNA-loaded nanoparticles pulmonary delivery, mannitol was utilized for it delivery into pressurized metered dose inhalers (pMDI). The fine particle fractions of this formulation was (45.39   ±   2.99)% indicating the appropriate deep lung deposition. These results revealed that this pMDI formulation containing Bac-TMC/TPP nanoparticles would be a promising siRNA delivery system for lung cancer treatment.

Engineering liquid pMDI into water-processable powder: Manufacture and application as waterborne additive

Qin Chen,Zhen Xu,Zongxing Sun,Yanjun Sun,Jixuan Song,Xiao Zhang,Siqi Huan,Long Bai,Jiyou Gu

Petroleum-derived polyurethane using isocyanates by solvent-based processing causes undesired environmental pollution and health effects. Waterborne polyurethane is an alternative to solve this issue, however, they are inefficient as additives and inconvenient for transportation and usage owing to the large amount of water. Herein, water-processable polymethylene polyphenyl polyisocyanate (PAPI, referred as pMDI) powder modified by biobased castor oil is developed. The preparation of pMDI powder is characterized by its appearance, content of –NCO groups, and storage life. The protective mechanism of –NCO groups is explored by a battery of techniques, including microscopies, FTIR, TGA , DSC and reaction kinetics. The optimal formulation for powdered pMDI uses 0.6 wt% SDS prior to de-hydration, which retains a high level of residual –NCO groups up to two-week storage. The formation of polyurea shells covering pMDI powder upon water-processing minimizes contact and interaction between –NCO and moisture in air, blocking consumption of –NCO groups. The pMDI powder can be used as a crosslinking agent for waterborne wood adhesives, showing great bonding performances, particularly improving wet strength (0.84 MPa) at 18 wt% loading level. This strategy offers a simple, green, and cost-effective route to engineer hazardous, high-risk liquid isocyanates into environmentally-friendly, low-risk, and versatile water-processable materials that are suitable as waterborne additives.

PMDI cross-linked rare earth/liquid metal reinforced ANF/MXene membranes for multifunctional electromagnetic interference shielding

Mengxia Shen,Xinyu Xu,Jiale Qi,Xiaojie Li,Bailiang Xue,Meng Zhu,Zhengliang Zhang,Xunfei Zheng,Bin Li,Zhen Shang

With the miniaturization of electronic devices, the creation of efficient and lightweight electromagnetic interference (EMI) shielding materials is of imminent significance. In this context, we reported an electromagnetic shielding membrane with hydrophobic, antioxidative, and high temperature stability. The chemically cross-linked Aramid nanofibers (ANF)/MXene@Ga/Gd 2 O 3 (CAMG) nanocomposite membranes exhibit a considerable tensile strength and possess exceptional flexibility. The CAMG shielding membranes demonstrate multiple heterogeneous interfaces and a magnetic-dielectric synergistic system, thereby effectively enhancing the absorption shielding efficiency, with the specific shielding effectiveness per unit volume (SSE/t) reaching 6788 dB cm 2 g −1 . Furthermore, the composite membranes demonstrate remarkable electrothermal capabilities, acting as pliable heaters with notable, swiftly replicable, and consistent electrothermal effects at reduced voltages. Additionally, they possess superior heat dissipation efficiency and ensure a uniform heat dispersion. The exceptional electromagnetic shielding performance and multifunctionality of the C-ANF/MXene@Ga/Gd 2 O 3 shielding membranes imply their instructive relevance in next-generation fields such as flexible electronics and aerospace.

Formulation and evaluation of novel reverse microemulsions containing salmon calcitonin in hydrofluoroalkane propellants

Ziyun Shan,Yinhe Tan,Lingzhen Qin,Ge Li,Xin Pan,Zhouhua Wang,Xiu Yu,Qinruo Wang,Chuanbin Wu

To develop reverse microemulsion as a potential strategy for pulmonary delivery of salmon calcitonin (sCT) in HFA134a propellant of pressurized metered dose inhalers (pMDIs), pluronic P85 (P85) was chosen as the most appropriate surfactant to form microemulsions containing sCT. Formulation parameters, including the surfactant and ethanol content, water content, and sCT loading, were optimized to obtain two desired pMDI formulations A and B with clear and transparent appearance, Tyndall effect, good physical stability and aerosolization properties. Aerosolization properties of the optimized pMDIs were assessed by next generation impactor (NGI) and twin-stage impactor (TSI), and the dose of sCT in each stage was assayed by HPLC. The fine particle fraction (FPF) of formulations A and B were both at the range of approximately 28.0–36.0%. Cytotoxicity studies indicated the cell viability determined by MTT assay only slightly dropped when the A549 cells were exposed to the pMDI formulations. Pharmacological study performed on the male Wistar rats showed the intratracheal administration of the microemulsion pMDIs containing sCT exhibited similar but prolonged hypocalcemic activity compared with the intravenous injection of sCT solution. Therefore, such reverse microemulsions are potential for pulmonary delivery of therapeutic peptides using HFA-pMDIs.
