Pogostemonis Herba is a functional food approved in Asian countries. Its major constituent, patchouli alcohol (PA), possesses a gastroprotective effect and is reported to transform into β-patchoulene (β-PAE) under acidic conditions. To investigate whether β-PAE, the metabolite of PA, has a protective effect on the gastrointestinal tract, the formation of β-PAE by gastric juice and the anti-ulcerogenic potential of β-PAE against ethanol-induced gastric injury were evaluated. The Results indicated that PA was converted to β-PAE by rat gastric juice. Additionally, β-PAE was significantly better than PA at reducing the area of gastric ulcer. The overproduction of malondialdehyde, tumour necrosis factor-α, interleukin (IL)-1β, IL-6, Fas, FasL and caspase-3 was markedly inhibited by β-PAE while the underproduction of superoxide dismutase, glutathione and catalase was significantly improved. β-PAE also regulated the NF-κB and ERK1/2 signalling pathways. Our findings suggest that β-PAE has potential therapeutic efficacy for antiulcer treatment.

Ulcerative colitis (UC) is defined as a chronic inflammatory disease with recurrent episodes, and current therapeutic strategies for this gastrointestinal disorder often fail to achieve satisfactory clinical outcomes. This clinical challenge underscores an urgent requirement to develop alternative treatment strategies. Patchouli alcohol (PA), a natural compound exhibiting anti-inflammatory and antioxidant properties, has shown potential; however, its precise mechanisms of action in UC remain to be fully elucidated. Our study shows that PA can effectively alleviate the symptoms of dextran sulfate sodium (DSS)-induced UC in mice. This therapeutic effect is achieved by reducing intestinal barrier impairment and inhibiting inflammatory reactions. In an LPS-induced inflammatory model using RAW264.7 cells, PA significantly downregulated the mRNA expression of il-1β and il-6 . Mechanistically, protein disulfide isomerase (PDI) was identified as a direct target of PA. By inhibiting IRE1 activation via PDI, PA mitigates endoplasmic reticulum stress, thereby reducing intestinal epithelial injury and ultimately alleviating the severity of UC. In this research, it is determined that the ability of PA to protect the intestinal epithelial barrier is brought about by its specific targeting of PDI, which alleviates consequent endoplasmic reticulum stress and prevents cellular damage.

Background Multidrug-resistant Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) are emerging in large numbers, while the rapid development of antimicrobial nanomaterials offers new opportunities to combat bacterial infections and antimicrobial resistance. Objectives We employed the nanomaterial MIL-101(Fe) as a carrier loaded with Patchouli alcohol (PA) to form the complex MIL-101(Fe)-PA. and to investigate the inhibitory activity of MIL-101(Fe)-PA against S. aureus and E. coli . Methods According to the literature,we prepared MIL-101(Fe)-PA by solvent method. Characterization of MIL-101(Fe)-PA was carried out using scanning electron microscopy(SEM), Fourier transform infrared(FT-IR) spectroscopy,X-ray diffractometer (XRD) and Brunauer-Emmett-Teller (BET) adsorption analysis. In the bacteriostatic experiments, its bacteriostatic effect was investigated by Minimal inhibitory concentration (MIC) experiments, cell proliferation, and growth curves of bacteria. Then its cytotoxicity was explored by cytotoxicity experiments. Results The characterization results indicated that we successfully prepared the complex MIL-101(Fe)-PA,which showed significant bacteriostatic effects against S. aureus and E. coli at 2 mg/mL. The inhibitory effect was stronger than that of MIL-101(Fe) as well as PA. Conclusion It shows that MIL-101(Fe)-PA has excellent inhibitory effect on S. aureus and E. coli.

Almost all of the candidate drugs for ischemic stroke failed to be translated from bench to beside. One important reason is that animals used in experimental studies cannot mimic ischemic patients due to lack of comorbidities like hypertension, diabetes and obesity. Therefore, it is better to test candidate drugs not only in normal animals but also in animals with comorbidities. Patchouli alcohol (PA), a natural tricyclic sesquiterpene in the traditional Chinese herb Pogostemonisherba, is well recognized for its anti-inflammation function in various inflammatory diseases. And as inflammation plays a very important role in cerebral ischemia/reperfusion (I/R) injury process and determines the ultimate brain damage, we hypothesized that PA could protect against cerebral I/R injury through its anti-inflammation ability. In this study, the effects of PA on cerebral I/R injury were evaluated in normal mice and obese mice. In normal mice with cerebral I/R injury, PA treatment reduced the infarct volume and neurological deficits in a dose- and time-dependent manner. PA treatment alleviated BBB dysfunction, inhibited mRNA and protein levels of TNF-α and IL-1β and modulated the activation of MAPKs signaling pathways. Moreover, PA also reduced infarct volume, alleviated the BBB dysfunction and inhibited inflammation in ob/ob mice with cerebral I/R injury. In conclusion, we demonstrated for the first time that PA could protect against cerebral I/R injury not only in normal mice but also in obese mice via inhibiting inflammation, suggesting that PA can be a potential drug for clinical treatment of ischemic stroke.

Patchouli alcohol (PA) is a tricyclic sesquiterpene isolated from Pogostemon cablin, which exerts
anti‑inflammatory, anti‑influenza and cognitive‑enhancing bioactivities. The present study aimed to investigate the protective effects of PA on acute lung injury (ALI) induced by intratracheal instillation of lipopolysaccharide (LPS) in mice. Dexamethasone was used as a positive drug for protection against LPS‑induced ALI. The results of the present study demonstrated that pretreatment with PA significantly increased survival rate, attenuated histopathologic damage and lung edema, and decreased the protein content in the bronchoalveolar lavage fluid (BALF) of mice with ALI. Furthermore, PA significantly inhibited the expression levels of proinflammatory cytokines, including tumor necrosis factor (TNF)‑α and interleukin (IL)‑6 in the BALF, downregulated the levels of myeloperoxidase and malondialdehyde, and upregulated the activity levels of superoxide dismutase and glutathione peroxidase in lung tissue. These results indicated that PA may exert potent protective effects against LPS‑induced ALI in mice, the mechanisms of which are possibly associated with the anti‑inflammatory and antioxidative activities of PA.
