Biodegradation characteristics of patulin by Saccharomyces cerevisiae during fermentation

Chao Yang,Bangzhu Peng

Patulin is a mycotoxin that contaminates apple-based products and poses a serious threat to human health. Saccharomyces cerevisiae can efficiently degrade PAT during fermentation. However, the degradation mechanism of PAT remained unclear. This study investigated the degradation of PAT by S. cerevisiae and the formation of degradation products. The results showed that 20 μg/mL of PAT was completely degraded by S. cerevisiae at 5 d of fermentation, and cycloheximide inhibited the degradation process. Both extracellular and intracellular enzymes can degrade PAT, but the latter had a higher degradation capacity. PAT exhibited the highest induction capacity of intracellular enzymes at 2 d, which was a 6.52% higher degradation rate than the uninduced. Additionally, PAT was firstly degraded to Z-ascladiol and then further to E-ascladiol by intracellular enzymes. The findings provide new insights into the biodegradation mechanism of PAT and a theoretical basis for removing methods of PAT contamination in apple-based products.

Mechanistic insights into patulin biodegradation by Geotrichum candidum XG1 in apple juice

Tingwen Wang,Gang Yang,Kewei Chen,Muying Du,Zsolt Zalán,Ferenc Hegyi,Sameh Awad,Nassra Dabou,Amel Ahmed Ibrahim,Jianquan Kan

Patulin (PAT) is a neurotoxic fungal metabolite that commonly contaminates fruits and their derivatives, causing substantial economic loss and serious harm to human health. In this study, we screened a PAT-degrading yeast strain, Geotrichum candidum XG1, among 12 probiotic strains. It efficiently degraded 96.80 % of PAT (5 μg/mL) within 48 h in potato dextrose broth and completely degraded PAT in fresh apple juice. G. candidum XG1 degraded PAT mainly by secreting intracellular enzymes, which were induced by PAT, and the degradation rate increased with increasing induction time. Ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry identified ascladiol as a PAT degradation product that disappeared within 96 h. Notably, both G. candidum XG1 and its intracellular enzymes affected the quality of apple juice, and G. candidum XG1 enhanced the flavor of apple juice. Overall, these results highlight the potential of G. candidum XG1 to remove PAT from apple juice.

Mechanism of patulin biodegradation by a reductase from Saccharomyces cerevisiae and its potential application to apple juice

Chao Yang,Chen Hu,Lingxuan Huang,Bangzhu Peng

Bioenzymatic degradation exhibits great potential for mycotoxins removal. So far, little is known about patulin (PAT) degrading enzymes from Saccharomyces cerevisiae . Here, the degradation mechanism of PAT by a free methionine-R-sulfoxide reductase (FRMSR) from S. cerevisiae was investigated. The results showed that purified FRMSR had high degradability without cofactor and displayed strong substrate specificity. The optimal degradation conditions in aqueous solution were 37 °C and pH 7.0. Isothermal titration calorimetry and molecular docking suggested that the PAT degradation by FRMSR was related to the hydrogen bonds formed between amino acids with PAT. Site-specific mutagenesis indicated that the mutation of Asp151 had the most significant effect on the degradation rate. Furthermore, the addition of FRMSR successfully degraded 88.16 % of PAT in apple juice without affecting its soluble solids content, pH value, titrable acidity and total phenols. These findings could provide valuable insights into the development of PAT-degrading enzymes in apple products and their industrial applications.

Patulin removal from apple juice using a novel cysteine-functionalized metal-organic framework adsorbent

Manshun Liu,Jing Wang,Qingfeng Yang,Na Hu,Wentao Zhang,Wenxin Zhu,Rong Wang,Yourui Suo,Jianlong Wang

Patulin (PAT) is one of the most common toxic contaminants of apple juice, which causes severe food safety issues throughout the apple industry. In order to remove PAT efficiently, a metal-organic framework-based adsorbent (UiO-66(NH 2 )@Au-Cys) was successfully synthesized and used for PAT removal from juice-pH simulation solution and real apple juice. Batch adsorption experiments were systematically performed to study the adsorption behavior for PAT. The results showed that adsorption process could be well described by the Pseudo-second order model and Freundlich isotherm model. The maximum adsorption capacity (4.38 µg/mg) was 10 times higher than the microbe-based biosorbents. Thermodynamic investigation demonstrated that adsorption process was spontaneous and endothermic. Furthermore, no marked cytotoxicity on NIH 3T3 cell lines was observed when the concentration of the adsorbent was lower than 10 μg/mL. Therefore, UiO-66(NH 2 )@Au-Cys is a potential adsorbent for PAT removal from apple juice with little quality changes.

Identification and application of a novel patulin degrading enzyme from Cyberlindnera fabianii

Chenyu Song,Wei Xu,Cuie Guang,Ting Xue,Wanmeng Mu

Patulin (PAT) is a mycotoxin commonly found in fruits and vegetables, prompting the need for effective removal and detoxification methods, which have garnered significant research attention in recent years. Among these methods, the utilization of microbial-derived enzymes stands out due to their mild operating conditions, specificity in targeted functional groups, and the production of non-toxic by-products, making it a preferred degradation approach. In this study, a novel PAT-degrading enzyme derived from Cyberlindnera fabianii ( Cyfa -SDR) was identified, demonstrating its highest catalytic activity at pH 7.0 and 80 °C against PAT. This temperature tolerance level represents the highest reported for PAT-degrading enzymes to date. The enzyme was further characterized as a short-chain dehydrogenase through analysis of its amino acid composition, conserved GXXXGXG motif, and dependency on NADPH. Moreover, the study evaluated the efficiency of PAT degradation by Cyfa -SDR at varying substrate and enzyme concentrations, surpassing the performance of other PAT-degrading enzymes, thus highlighting its substantial potential for the biological control of PAT. In conclusion, the enzymatic treatment using the PAT-degrading enzyme Cyfa -SDR presents a viable and promising solution for enhancing the quality and safety of fruit juice.

Action Mode of the Mycotoxin Patulin as a Novel Natural Photosystem II Inhibitor

Yanjing Guo,Weizhe Liu,He Wang,Xiaoxiong Wang,Sheng Qiang,Hazem M. Kalaji,Reto Jörg Strasser,Shiguo Chen

A biocontrol method plays an important role in weed management. In this study, we aimed to clarify the phytotoxicity of the mycotoxin patulin (PAT) and reveal its mode of action as a new natural photosystem II (PSII) inhibitor. Phytotoxicity test showed that PAT has herbicidal activity and causes significant leaf lesions on Ageratina adenophora. Under a half-inhibition concentration I50 (2.24 μM), the observed significant decrease in oxygen evolution rate and the increase in the J-step of the chlorophyll fluorescence rise OJIP curve indicated that PAT strongly reduces photosynthetic efficiency by blocking electron transport from the primary to secondary plastoquinone acceptors (QA to QB) of PSII. Molecular modeling of PAT docking to the A. adenophora D1 protein suggested that PAT bounds to the QB site by forming hydrogen bonds to histidine 252 in the D1 protein. It is proposed that PAT is a new natural PSII inhibitor and has the potential to be developed into a bioherbicide or used as a template scaffold for discovering novel derivatives with more potent herbicidal activity.

Simultaneous detection of patulin and ochratoxin A based on enhanced dual-color AuNCs modified aptamers in apple juice

Pengfei Ma,Hualin Guo,Ke Li,Yin Zhang,Huiqing Guo,Zhouping Wang

Patulin (PAT) and ochratoxin A (OTA) are the two main mycotoxins present in apples. Herein, a sensitive aptasensor for simultaneous detection of PAT and ochratoxin OTA was developed. Dual-color gold nanoclusters (AuNCs) with enhanced fluorescence properties were synthesized and employed as fluorescence amplifiers. Two separated fluorescence peaks at 650 nm and 530 nm were monitored simultaneously by employing single excitation (405 nm), corresponding to the aptamer probes of Cys@BSA-AuNCs-Apt PAT and Arg@ATT-AuNCs-Apt OTA , respectively. The fluorescent aptasensor demonstrated satisfying specificity, storage ability and accuracy. Under the optimal experimental conditions, the linear detection range for PAT and OTA was 0.10–50 ng/mL, with the limit of detection of 0.09 ng/mL and 0.06 ng/mL, respectively. Most importantly, practicability of the constructed aptasensor were confirmed by conducting the determination of PAT and OTA in apple juice sample, indicating the great potential of the aptasensor in practical detection applications.

High sensitive and selective electrochemical aptasensor for patulin detection based on a novel assembling strategy using hierarchically porous carbon

Cuiwen Jiang,Yanli Wang,Jing Liang,Liping Xie,Zhongdan Liang,Kejing Huang,Huiling Li,Tao Li,Dejiao Ning,Li Tang,Xiaochuan Huang,Lihong Luo,Yu Ya,Feiyan Yan

Patulin (PAT) in food is a menace to human health. Hence, a sensitive and precise approach to PAT detection is significant. In this research, an innovative electrochemical assay was developed for sensitively determining PAT by assembling aptamers on a carboxylation hierarchically porous carbon (HPC-COOH) modified glassy carbon electrode. Diverse analytical methods were used to describe electrode morphology, structure, chemical composition, and electrochemistry properties. The HPC-COOH, with a large surface area, played a crucial role as a unique substrate to effectively immobilize aptamers and enhance target PATs' recognition and capture ability. Simultaneously, the excellent electrical conductivity of HPC-COOH was significant for the signal amplification of sensors. Under optimized conditions, the aptasensor enabled efficient PAT detection in a broad concentration range (0.5–5.0×10 6 ng/L) with a low detection limit (0.25 ng/L). The designed aptasensor showed high sensitivity, selectivity, reproducibility, and stability under optimized conditions. The sensors’ feasibility for practical applications was further demonstrated by analyzing apple juice and haw juice samples.

Effects of Patulin Stress on the Physiology, Fermentation Performance, and Aroma Profile of Saccharomyces cerevisiae During Fermentation

Chao Yang,Lingxuan Huang,Kuo Huang,Stephen Y. K. Seah,Bangzhu Peng

Patulin (PAT) is a mycotoxin commonly found in apples and their derived products, posing a significant threat to human health. Saccharomyces cerevisiae CICC 31084 exhibited excellent ability in removing PAT during fermentation. However, limited studies have addressed the physiological and fermentation-related responses of S . cerevisiae under PAT stress. This study investigated the effect of different concentrations of PAT on S . cerevisiae in simulated apple juice. The results revealed that the PAT removal process affected the growth and metabolism of S . cerevisiae . PAT stress increased the thickness and altered the structure of yeast cell walls while upregulating the expression of genes associated with cell wall biosynthesis. Furthermore, oxidative stress induced by PAT triggered elevated glutathione levels, catalase activity, and ATPase activity, suggesting a robust adaptive response. A total of 1 µg/mL PAT accelerated the changes of fermentation parameters, whereas 10 µg/mL impaired yeast performance. GC-MS analysis revealed that PAT influenced the content of key aroma compounds in the simulated juices. PAT concentration was positively correlated with the production of octanoic acid and ethyl octanoate and negatively correlated with the formation of phenylethyl alcohol, hexanoic acid, and esters. The results could provide a theoretical basis for the application of PAT detoxification by S . cerevisiae in cider, and the identified aroma compounds could serve as potential markers for PAT contamination.

Sensitive determination of Patulin by aptamer functionalized magnetic surface enhanced Raman spectroscopy (SERS) sensor

Zhiming Guo,Lingbo Gao,Shuiquan Jiang,Hesham R. El-Seedi,Islam M. El-Garawani,Xiaobo Zou

Food pollution caused by Patulin (PAT) seriously threatens the safety of human diets and has attracted extensive attention. Early and accurate detections of PAT are essential to prevent further toxin spreading and contamination. A surface enhanced Raman scattering (SERS) aptasensor was fabricated by combining a gold-silver core-shell structure containing signal molecule (ADANRs) and chitosan modified magnetic nanoparticles (CS-Fe 3 O 4 ). The modified ADANRs with the complementary chain of the aptamer (SH-cDNA-ADANRs) which were served as the signal probes and the aptamer modified CS-Fe3O4 (NH 2 -apt-CS-Fe 3 O 4 ) were served as the capture probes. In addition to the ability to recognize the target PAT, the capture probe also showed a strong enrichment ability under the action of external magnetic force. The intraparticle plasma coupling between the inner gold core and the outer silver shell can greatly improve the SERS activity of the signal molecules. SERS aptasensor was used to collect the spectra of actual apple samples spiked with different PAT content. The minimum detection limit of SERS aptasensor for detecting PAT in actual samples was 0.0384 ng/mL and the recovery rate range was from 96.3% to 108%. In conclusion, the sensitive and specific SERS aptasensor detection of PAT based on aptamer functionalized nanoparticles exhibited great potential for practical application in mycotoxin detection and analysis.

Strand displacement amplification combining photothermal-colorimetric dual mode test strip for ultra-sensitive detection of patulin

Yi Zhang,Jing Cao,Jie Zhou,Wen-Long Wang,Zhenghua Xu,Yong-Wei Feng,Xiao-Dong Huang,Feng-De Yu,Xin Yang

Patulin (PAT) is a fungal toxin often found in fruit and juice products. The sensitive and fast detection of PAT can help the control of PAT-induced health risk. However, the lack of efficient and stable antibody for PAT hindered the advancement of its test strips. Here, we constructed a nucleic acid-based lateral flow test strip (NALFTS) and used photothermal-colorimetric dual mode for the detection of PAT. The binding of PAT in samples to the aptamer on magnetic separation probe MNPs-Apt@cDNA induced the release of cDNA competitively. Subsequently, cDNA initiates strand displacement amplification (SDA), generating numerous single-strand DNA (ssDNA). These ssDNA molecules were analyzed by a test strip with photothermal-colorimetric dual signal. The developed method demonstrated a limit of detection (LOD) of 5 pg mL −1 and a quantitative range of 0.01 ∼ 320 ng mL −1 in photothermal mode, and a LOD of 0.1 ng mL −1 and a quantitative range of 0.1 ∼ 40 ng mL −1 in colorimetric mode. This SDA-NALFTS offered dual signals corroborating each other to provide more reliable results for rapid screening or accurate monitoring of PAT in juice.

Operando Photoelectrochemical Surface-Enhanced Raman Spectroscopy: Interfacial Mechanistic Insights and Simultaneous Detection of Patulin

Shuda Liu,Na Dong,Shuyun Meng,Yuye Li,Yuqing Li,Siyuan Wang,Dong Liu,Tianyan You

Comprehending the biosensing mechanism of the biosensor interface is crucial for sensor development, yet accurately reflecting interfacial interactions within actual detection environments remains an unsolved challenge. An operando photoelectrochemical surface-enhanced Raman spectroscopy (PEC-SERS) biosensing platform was developed, capable of simultaneously capturing photocurrent and SERS signals, allowing operando characterization of the interfacial biosensing behavior. Porphyrin-based MOFs (Zr-MOF) served as bifunctional nanotags, providing a photocurrent and stable Raman signal output under 532 nm laser irradiation. Aptamer was used to bridge the Zr-MOF and the silver-encased gold nanodumbbells (AuNDs@AgNPs). The simultaneous in situ acquisition of target-induced PEC and SERS signal responses facilitated the correlation of electron transfer information from the photocurrent with the distance information from the SERS signal. It revealed the biosensing mechanism in which target-induced aptamer conformational bending drove the Zr-MOF to approach the electrode. However, the increase in charge transfer observed through conventional electrochemical methods contradicts the conclusions drawn from the operando PEC-SERS analysis. Comprehensive analysis indicated that redox probes introduced during the non-in-situ measurement process became adsorbed within the MOF pores, potentially affecting the judgment of the biosensing mechanism. In addition, the operando PEC-SERS biosensor simultaneously obtained two independent signals, providing self-verification to improve the accuracy and reliability of patulin detection. The linear ranges were 1 pg mL–1–10 ng mL–1 for the PEC method and 1 pg mL–1–100 ng mL–1 for the SERS method, respectively. This work provides a powerful tool for determining the interface characteristics of biosensors.
