A liquid chromatography with tandem mass spectrometry method was developed and applied for the investigation of penthiopyrad photolysation and hydrolysation in different aqueous solutions, and the potential enantioselectivity of this chiral fungicide was further assessed. Good linearity was obtained (R2 > 0.99) for R-(−)- and S-(+)-penthiopyrad in different matrices and the respective limits of detection and quantitation of both enantiomers were 0.003 μg mL−1 and 0.01 μg mL−1. The most suitable illumination intensity for photolysing penthiopyrad was 400 W. Other factors such as pH, salinity and flavonoid also influenced penthiopyrad photolysation. In an acidic solution, the photolytic degradation rate of penthiopyrad enantiomers decreased due to their chemical structures and physicochemical characteristics. Different salinities affected penthiopyrad photolysation differently; acceleration occurred in solutions with salinities of 0.5% or 3.0% due to effective catalysis by photosensitive chloride radical anions, and when the salinity was 1.0% or 2.0%, the inhibitory effect of inorganic salt decreased the photolysation rate. The addition of different flavonoids could significantly inhibit the photolysation of penthiopyrad by decreasing degradation 5–597 times through photo-quenching effects. Under some conditions, the S-(+)-enantiomer was photolysed faster than its antipode, indicating that the photolysation of penthiopyrad was enantioselective. During the hydrolysation process, penthiopyrad degraded faster in the acidic solution than in the other two aqueous solutions with different pH levels, which was attributed to the hydrogen ion-induced catalytic reaction. In sterile/nonsterile natural water matrices, the hydrolysation rate of penthiopyrad enantiomers was also influenced by pH, in the degradation order of tap water (pH 7.1) > rain water (pH 7.3) > lake water (pH 8.8). Enantiopure enantiomers were hydrolysed more slowly than racemic enantiomers due to their mutual promotion effects. The configuration stability of penthiopyrad enantiomers was preserved during the degradation process. Similar enantioselectivities of the hydrolysation of penthiopyrad and preferential degradation of the S-(+)-enantiomer were observed in some aqueous solutions. The results could provide some data to more accurately demonstrate the enantioselective environmental fate and risk of penthiopyrad in aquatic environments.

Penthiopyrad, a chiral fungicide with a riskier S-enantiomer, exhibited poorly understood stereoselective behaviors in mammals. This study investigated the enantioselective accumulation and metabolism of penthiopyrad enantiomers in mammals using in vivo, in vitro, and in silico approaches. Penthiopyrad accumulated in the liver and kidneys at concentrations over 10-fold higher than those in the blood. S-penthiopyrad showed preferential absorption with 1.6-fold higher bioavailability than the R-enantiomer. Twenty-three metabolites (12 novel) were identified, with CYP-mediated N-demethylation as the dominant pathway. S-penthiopyrad was preferentially metabolized in mouse liver microsomes with a 1.5-fold shorter half-life, while human liver microsomes favored the R-enantiomer. CYP2E1, CYP2C (mouse), and CYP3A4 (human) mediated stereoselective metabolism, with the preferential metabolism of the R-enantiomer by human CYP3A4 attributed to its low binding energy and favorable conformation. This study comprehensively clarified the stereoselective behavior and metabolic pathway of penthiopyrad in mammals, providing valuable metabolic insights for further toxicological evaluation of penthiopyrad.

Penthiopyrad (PEN), a succinate dehydrogenase inhibitor (SDHI) fungicide, is widely used in agriculture; however, its misuse can induce phytotoxicity in cucumber. In our previous study, a dual-functional nano-pesticide (HPC@PEN@MSe) with both detoxification and growth-promoting properties was developed based on mesoporous nano-selenium (MSe) and hydroxypropyl cellulose (HPC). The present study investigated the regulatory effects of this nano-pesticide on cucumber root exudates and photosynthetic parameters under controlled hydroponic conditions. Cucumber plants were exposed to PEN (10 mg/L) or HPC@PEN@MSe (at an equivalent PEN concentration) for 14 days, with three biological replicates per treatment. Changes in cucumber root exudates under PEN and HPC@PEN@MSe stress were analyzed using ion exchange chromatography (IEC), synchronous fluorescence spectroscopy, three-dimensional (3D) fluorescence spectroscopy, Fourier transform infrared spectroscopy (FTIR), and two-dimensional correlation spectroscopy (2D-COS). The results demonstrated that, compared with PEN treatment alone, HPC@PEN@MSe better preserved the chemical profile of root exudates, including amino acid composition, fluorescent components, and functional group characteristics. In parallel, leaf photosynthetic parameters were measured. After 14 days of exposure, the net photosynthetic rate in the HPC@PEN@MSe group was 2.17 times higher than that in the PEN-only group. In summary, these findings suggest that, in addition to its direct physiological protective effects, HPC@PEN@MSe may indirectly promote plant health by maintaining the overall stability of the root exudate chemical profile. This provides new insight into the rhizosphere ecological effects of functionalized nano-pesticides.

Penthiopyrad is a widely used chiral fungicide for controlling rust and Rhizoctonia diseases. Development of optically pure monomers is an important strategy to realize amount reduction and increment effects of penthiopyrad, wherein, fertilizers as the co-exiting nutrient supplement may alter the enantioselective residues of penthiopyrad in soil. In our study, influences of urea, phosphate, potash, NPK compound, organic granular, vermicompost and soya bean cake fertilizers on enantioselective persistence of penthiopyrad were fully evaluated. This study demonstrated that R -(−)-penthiopyrad dissipated faster than S -(+)-penthiopyrad during 120 days. High pH, available nitrogen, invertase activities and reduced available phosphorus, dehydrogenase , urease, catalase activities were situated to benefit removing the concentrations of penthiopyrad and weakening enantioselectivity in soil. With respect to the impact of different fertilizers on soil ecological indicators, vermicompost contributed to enhanced pH. Urea and compound fertilizer played an absolute advantage in promoting available nitrogen. All fertilizers didn't go against available phosphorus. Dehydrogenase responded negatively to phosphate, potash and organic fertilizers . Urea increased invertase, besides, it and compound fertilizer both diminished urease activity. The catalase activity was not activated by organic fertilizer. Based on all the findings, soil application of urea and phosphate fertilizers was recommended and considered as a better option to exhibit high efficiency for the dissipation of penthiopyrad. The combined environmental safety estimation can effectively guide the treatment of fertilization soils in line with the nutrition requirements and pollution regulation from penthiopyrad.

Colletotrichum leaf disease (CLD), mainly caused by the Colletotrichum gloeosporioides species complex (CGSC), is a serious disease of rubber tree in most rubber-growing countries. Few fungicides are available for rubber farmers to manage this disease. To control CLD effectively, it is important to test the baseline sensitivity and efficacy of fungicides against CGSC isolates. Penthiopyrad is a new succinate dehydrogenase inhibitor (SDHI) that has been registered for use in many countries. In this study, baseline sensitivity to penthiopyrad was established by mycelial growth methods using 91 CGSC isolates collected from rubber trees in China. The EC 50 values of penthiopyrad ranged from 0.243 to 3.377 mg/L with a mean EC 50 value of 1.685 mg/L. Penthiopyrad greatly decreased the mycelial biomass and increased cell membrane permeability of CGSC isolates. The mycelia were severely distorted, and the number of top branches increased with penthiopyrad treatment. Penthiopyrad inhibited succinate dehydrogenase activity, causing a decrease in the ATP content of CGSC isolates, and significantly inhibited conidia germination and appressorium formation on rubber tree leaves. The protective and curative efficacy of 100 mg/L penthiopyrad reached up to 98.53% and 66.79% against CLD, respectively. Our results demonstrated the high inhibitory activity of penthiopyrad against CGSC isolates and its potential for CLD management on rubber tree.

Ion interference including copper (Cu + )/calcium (Ca 2+ ) overload activate cell-specific death channels, damage mitochondria and disrupt cellular homeostasis, showing great potential in anti-tumor therapy. However, the complex metabolic environment and the powerful self-protection of tumors cause clinical failure of ion interference. Thus, metabolic disruption is expected an innovative strategy for the enhancement of ion interference. Herein, CuS-α-CHCA&penthiopyrad@CaCO 3 -RGD nanoparticles (CCPCR NPs) are prepared to provide a Cu + /Ca 2+ dual-overload anti-tumor therapy assisted by metabolic-symbiosis-destruction strategy, realizing the collapse of tumor self-protection. Specifically, precise CuS and CaCO 3 delivery triggered irreversible Cu + /Ca 2+ dual-overload and reactive oxygen species (ROS) attack toward 4T1 cells. Meanwhile, α-CHCA and penthiopyrad disturbed the metabolic symbiotic environment by disrupting the TCA cycle and preventing lactate efflux to aggravate intracellular acidosis and promote Fenton-like reaction of Cu + , enhancing the sensitivity of tumor cells to copper death and Ca 2+ overload. Further, aided by the metabolism of symbiosis, the destroyed tumor cells further activate the polarization of M1 macrophages and the maturation and antigen cross-presentation of dendritic cells (DCs), which further eliminate tumor cells. In summary, an amplified anti-tumor dual-ion interference strategy assisted by metabolic symbiotic destruction is established, which is of great significance in improving the clinical effect of ion therapy for tumors.
