Chiral pesticide permethrin promotes the antibiotic resistance genes dissemination by transformation: Different chiral isomers engage in distinct regulatory pathways

Chenxi Li,Linjie Wang,Qi Dai,Yang Chong,Satoshi Utsunomiya,Honggui Wang,Ya Zhang,Jie Han

The global dissemination of antibiotic resistance genes (ARGs) poses an increasingly urgent threat to both environmental and human health. The extensive use of chiral permethrin (PM), the most popular synthetic type I pyrethroid insecticide worldwide, has led to its increased detection in aquatic environments. However, our understanding of PM's role in spreading ARGs is still limited. Here, we systematically assessed the effects of two chiral isomers of 1R-cis-PM (CPM) and 1R-trans-PM (TPM) on the dissemination of ARGs in the aquatic environments by using a natural transformation (NT) model comprising plasmid pWH1274 and Acinetobacter baylyi ADP1. It was found that reactive oxygen species (ROS) was the main factor facilitating the NT of ARGs mediated by CPM and TPM, although their respective production mechanisms exhibited distinct pathways: CPM generates ROS primarily through the primary electron transport chain (ETC), whereas TPM does so via a secondary ETC. Furthermore, CPM enhanced NT by improving bacterial competent state, while TPM promotes it by enhancing recombination.  It was confirmed that both CPM and TPM have the potential to accelerate the spread of ARGs through distinct mechanisms. These findings will help us understand that different chiral isomers may pose risks through distinct mechanisms.

An ultrasensitive solid-state electrochemiluminescence sensor based on Ni-MOF@Ru(bpy)32+ and Au NPs@TiO2 for determination of permethrin

Shuning Yang,Li Tian,Weiwei Fu,Huiling Li,Chao Li,Yujia Song,Ruidan Li,Yanjia Guo,Lun Zhao

The novel TiO 2 and Ni-MOF materials were synthesized and utilized for the detection of permethrin (PET). A highly sensitive solid-state electrochemiluminescence (ECL) sensor was developed based on Ni-MOF@Ru(bpy) 3 2+ and Au NPs@TiO 2 . In this sensing platform, Ru(bpy) 3 2+ -Tripropyl Amine (TPrA) was used as a luminescent signal, Ni-MOF acted as a carrier to carry more luminescent reagents Ru(bpy) 3 2+ . Au NPs acted as promoters facilitated electron transport and TiO 2 could further enhance the luminescence intensity of the system by synergistical interaction with Au NPs. The possible mechanisms of signal amplification were investigated. The ECL intensity decreased significantly with increasing PET concentration, enabling the determination of PET amount through the observation of the change in ECL signal intensity (ΔI). Under optimal experimental conditions, the linear range of PET concentration from 1.0 × 10 −11  mol L −1 to 1.0 × 10 −6  mol L −1 , with a detection limit of 3.3 × 10 −12  mol L −1 (3S/N). This method was successfully applied to determine PET in various vegetable samples.

Ultrasensitive electrochemiluminescence biosensor for permethrin based on iron oxide nanomaterials and Au nanoparticles

Yingjie Zhao,Li Tian,Shiqi Wu,Xin Zhang,Zhuo Sun,Yue Hu,Yue Wang,Ruizhan Chen,Juan Lu

A sensitive electrochemiluminescence (ECL) sensor has been developed for the determination of permethrin based on iron oxide nanomaterials (Fe 2 O 3 ) and Au nanoparticles (Au NPs). The ECL response of luminol/O 2 system is enhanced obviously due to the synergy effect between Fe 2 O 3 and Au NPs. The possible mechanism of ECL phenomenon is studied. Furthermore, the ECL emission increases with adding permethrin, and the determination of permethrin could be achieved by monitoring the ECL intensity. Under the optimal conditions, the novel ECL sensor achieved an efficient detection of permethrin range from 2.6 × 10 −9  mol L −1 to 4.3 × 10 −5  mol L −1 with a detection limit of 8.7 × 10 −10  mol L −1 . The proposed strategy can provide a simple and reliable approach for determining of permethrin, indicating that it has great application prospects in pesticide detection.

Acute and chronic toxicity of deltamethrin, permethrin, and dihaloacetylated heterocyclic pyrethroids in mice

Qiuyan Zhu,Yang Yang,Zhiting Lao,Yingying Zhong,Kun Zhang,Suqing Zhao

BACKGROUND Pyrethroids, a class of insecticides, that act on the nervous system of insects. Frequent consumption of foods with pyrethroid residues increase the risk of developmental and neurological diseases in humans. Assessing the toxicity of novel synthetic pyrethroids to mammals is also critical to the development of agrochemicals. RESULTS Using mice as models, the acute and chronic toxicity of deltamethrin, permethrin, dihaloacetylated pyrethroids to mammals was researched by gavage administration. Acute toxicity assessment displayed that the median lethal dose (LD 50 ) of deltamethrin, permethrin and dihaloacetylated heterocyclic pyrethroids DCA-O, DCA-01, and DCA-11 tested were greater than 500 mg/kg of weight. Furthermore, chronic toxicity assessment demonstrated that deltamethrin, permethrin caused epidermal damage near the genitals, while dihaloacetylated heterocyclic pyrethroids DCA-O, DCA-01, and DCA-11 showed no relevant symptoms. However, both the acute and chronic toxicity assessment suggested that pyrethroids exposure induced mice loss weight. Additionally, the elevated plus maze (EPM) test showed that pyrethroids caused anxiety-like behaviors and no motor defects in Kunming mice. Beside, during the sucrose preference test (SPT), 60-day pyrethroids exposure increased excitatory behaviors in mice. However, the neurochemical studies displayed that pyrethroids exposure increased the total amount of glutamate (Gln), glutamine (Glu) and γ -aminobutyric acid (GABA) in the mice's blood. CONCLUSION Pyrethroids exposure induced weight loss in mice, although the acute oral toxicity of deltamethrin, permethrin and dihaloacetylated heterocyclic pyrethroids DCA-O, DCA-01, and DCA-11 was low. However, regarding chronic toxicity, deltamethrin, permethrin and dihaloacetylated heterocyclic pyrethroids DCA-O, DCA-01, and DCA-11 induced anxiety-like behaviors, excitatory behaviors, Gln-Glu-GABA circulatory dysfunction in blood. Particularly, deltamethrin, though permethrin also had reproductive toxicity. © 2020 Society of Chemical Industry

Photodegradation kinetics, mechanism and aquatic toxicity of deltamethrin, permethrin and dihaloacetylated heterocyclic pyrethroids

Qiuyan Zhu,Yang Yang,Zhiting Lao,Yingying Zhong,Kun Zhang,Suqing Zhao

Photochemical methods attracted much research interests for their high-efficiency and low secondary pollution. Decomposition of synthetic pyrethroids , the fourth major group of insecticides in use worldwide, was also of great significance due to their possible environmental risks. The photodegradation of deltamethrin , permethrin and dihaloacetylated heterocyclic pyrethroids in methanol/acetone = 9/1 (by volume) by a 400 W mercury lamp was examined. The t 1/2 of tested pyrethroids was less than 25 min, except for cis -permethrin with a t 1/2 of up to 50 min. The trans -isomer of permethrin and compound DCA-01 with a smaller t 1/2 might be more susceptible to degradation than their cis -isomer. Besides, the photodegradation of pyrethroids was divided into twelve pathways including isomerization , ester hydrolysis, ester bond cleavage, C O bond cleavage, 3,3-dimethylacrylate formation, double bond break, C1-C3 bond cleavage in cyclopropyl, reductive dehalogenation, decarboxylation , nucleophilic reagents attack on lone pair electrons on oxygen atoms in the phenyl ether, cyano hydrolysis, and halogenated hydrocarbon hydrolysis. The ECOSAR program displayed that pyrethroids and most of their photodegradation products were toxic to fish, daphnid, green algae . Particularly, some photodegradation products were more harmful to aquatic organisms than their parents.

Enhancing electrochemiluminescence by modifying Fe3CuO4 and CdS@ZnS: A novel ECL sensor for highly sensitive detection of permethrin

Qian Wu,Li Tian,Xiangyu Shan,Shuning Yang,Huiling Li,Chao Li,Juan Lu

A novel electrochemiluminescence (ECL) sensor was constructed for the detection of permethrin by modifying Fe 3 CuO 4 –Ru(bpy) 3 2+ and GO-CdS@ZnS on the electrode. Fe 3 CuO 4 was used as a carrier to adsorb more luminous reagent Ru(bpy) 3 2+ to promote luminescence. Meanwhile, it also can be used as a co-reaction promoter to amplify the initial signal of Ru(bpy) 3 2+ -tri-n-propylamine (TPrA) system. When GO-CdS@ZnS was introduced into this system, the ECL signal was further enhanced. The porous nature of graphene oxide (GO) was utilized to load a large amount of CdS@ZnS, in which CdS@ZnS acted as the co-reactant to amplify the ECL signal. The amount of permethrin (PMT) increased and the ECL signal decreased. Under the optimum conditions, the ECL response was linearly related to the logarithm of PMT concentration. The developed ECL sensor allowed for sensitive determination of PMT and exhibited a wide linear range from 1.0 × 10 −11  mol L −1 to 1.0 × 10 −7  mol L −1 . The limit of detection was 3.3 × 10 −12  mol L −1  (S/N = 3). It can be used for the detection of PMT in vegetable samples.

Permethrin exposure impacts zebrafish lipid metabolism via the KRAS-PPAR-GLUT signaling pathway, which is mediated by oxidative stress

Afrasyab Khan,Bibimaryam Khan,Shakeel Hussain,Yuhan Wang,Weijun Mai,Yongzhong Hou

Permethrin (Per) is a widely used and frequently detected pyrethroid pesticide in agricultural products and the environment. It may pose potential toxicity to non-target organisms. Per has been reported to affect lipid homeostasis, although the mechanism is undefined. This study aims to explore the characteristic transcriptomic profiles and clarify the underlying signaling pathways of Per-induced lipid metabolism disorder in zebrafish liver. The results showed that environmental exposure to Per caused changes in the liver index, histopathology, and oxidative stress in zebrafish. Moreover, transcriptome results showed that Per heavily altered the pathways involved in metabolism, the immune system, and the endocrine system. We conducted a more in-depth analysis of the genes associated with lipid metabolism. Our findings revealed that exposure to Per led to a disruption in lipid metabolism by activating the KRAS-PPAR-GLUT signaling pathways through oxidative stress. The disruption of lipid homeostasis caused by exposure to Per may also contribute to obesity, hepatitis, and other diseases. The results may provide new insights for the risk of Permethrin to aquatic organisms and new horizons for the pathogenesis of hepatotoxicity.

FTW SERS probes with Ag NCs-GO composite structure excited by evanescent wave for in situ detection of permethrin

Pan Tao,Yang Luo,Kai Zheng,Jing Cao,Li Zhu,Xunsi Wang,Wei Zhang,Shixun Dai,Qiushun Zou,Peiqing Zhang

Background Permethrin is a pesticide used to kill insects, and once used in excess, it poses a great threat to the environment and human health, therefore, it is necessary to realize the rapid and accurate detection of permethrin. Fiber optic surface enhanced Raman scattering (SERS) probes have the advantages of small volume and can be used for remote monitoring, which have great potential for application in achieving in-situ detection of pesticide residues. Results Fiber taper waist (FTW) SERS probes modified by silver nanocubes-graphene oxide (Ag NCs-GO) composite structures were prepared for in situ detection of permethrin in lake water. Evanescent wave was used to excite SERS signals from Ag NCs-GO composite substrates, which was designed to enhance signals through expanded excitation light-analyte contact area. Ag NCs can provide stronger hot spots, and GO further enhances the sensitivity of SERS through electron transfer and improves the stability of the detection. The detection limit of prepared FTW SERS probe for Rhodamine 6G (R6G) was calculated to be 8.5×10 -9 M and RSD were all below 5% using the drop-casting method. The in-situ detection limit of R6G and permethrin in water was tested to be 10 -6 M and 10 -5 M, respectively. Significance This work utilizes fiber optic evanescent waves to excite SERS signals, and adopts a special FTW structure and SERS substrates to achieve good performance. The synergistic enhancement of electromagnetism and chemistry is realized while increasing the contact area, which improves the sensitivity of in situ detection of the pesticide residue permethrin. It Promotes the application of fiber optic SERS sensors in pesticide residue detection.

Probing the potential mechanism of permethrin exposure on Alzheimer's disease through enantiomer-specific network toxicology, multi-spectroscopic, and docking approaches

Tianzhu Guan,Ning Li,Ya Gao,Mingyuan Gao,Qin Hu,Yajun Gao,Lixia Xiao,Zhenquan Yang,Qiaoquan Liu

Latest observations indicated that exposure of organic environmental neurotoxins may increase the potential risk of Alzheimer's diseases (AD). As a suspected food-derived risk factor, permethrin, composed of cis -isomer and trans -isomer, is widely used as a broad-spectrum pyrethroid insecticide in agricultural crops for the arthropod pests controlling. Thus, evaluating the impact of permethrin exposure is of great importance to human health. In this study, we performed the toxicological network approach to decipher AD-related mechanisms of cis -permethrin and trans -permethrin. Based on the toxicological network construction and central network topological analysis, human serum albumin (HSA) was selected as the core targets in AD-related developing. From the analysis of the steady state and time-resolved fluorescence quenching of HSA in presence of permethrin mixture, it has been inferred that the nature of the quenching mainly originates from the dynamic modes. Experimentally, the thermodynamic parameters revealed hydrophobic interactions and van der Waals forces played a major role during quenching process. Tryptophan synchronous fluorescence spectra were blue shifted whereas the position of tyrosine synchronous spectra was red shifted during the complex formation. Three-dimensional fluorescence together with FT-IR experiment confirmed that permethrin caused the secondary structure changes in HSA. To better understand the binding patterns between HSA and cis/trans -permethrin, theoretical calculation and molecular docking were implemented. According to the electrostatic potential map, the electrophilic attack region corresponds for electron rich oxygen atoms, while the nucleophilic attack regions were mainly located at over the benzene rings and methyl on cyclopropane ring of permethrins. Docking results shown that cis -permethrin and trans -permethrin located in hydrophobic pocket nearby Domain IIA with the different binding affinity (−7.6 and −9.2 kcal/mol), which consistent with the competitive displacement experiment. All these findings generated in the present study facilitated the elucidation of the molecular mechanism details between permethrin mixture and HSA, which provided fresh insights into the links between environmental exposure and AD-related adverse health outcomes.

Pyrazole permethrin-induced liver toxicity in mice involves the PI3K/AKT/NF-κB pathway: Integrated network pharmacology and experimental validation

Jingjing Chen,Bin Liu,Yuanyuan Li,Binwu Zhuang,Dong Su,Yunlong Sang,Zibo Dong,Hao Liu

Pyrazole permethrin (also known as pyraclostrobin, abbreviated as PP) is a widely used fungicide that is chemically stable and fat-soluble; however, due to its accumulation in the environment and bioaccumulation, it poses potential health risks. This study systematically elucidates PP-induced liver injury through a combination of network toxicology and in vitro/in vivo investigations. Network toxicology identified 328 potential targets and 12 core genes associated with PP-induced liver injury. GO and KEGG enrichment analyses revealed pathway enrichment emphasizing biological processes such as inflammation, oxidative stress, apoptosis, and lipid metabolism. In vivo experiments in mice revealed that after 28 days of exposure, histopathological analysis showed minor local inflammatory cell infiltration and extensive hepatocyte necrosis and vacuolation in both low- and high-dose groups. High-dose PP significantly elevated serum transaminase (AST and ALT) activity. Biochemical analysis revealed that PP treatment reduced antioxidant enzyme activity (CAT, T-AOC, GSH-Px) while increasing malondialdehyde (MDA) levels, inducing oxidative stress. Additionally, PP exposure enhanced the expression of inflammatory cytokines (IL-1β, TNF-α). This indicates activation of oxidative stress and inflammatory pathways: oxidative damage occurs via depletion of the antioxidant enzyme system, while inflammatory cascades are mediated through the PI3K/AKT/NF-κB signaling axis. This study reveals key molecular targets and pathways involved in PP-induced hepatotoxicity, suggesting that subacute exposure risks should be fully considered when establishing safety standards for agricultural chemical use.

Title: Co-exposure to polystyrene microplastics and permethrin induces gill injury in Gobiocypris rarus involving oxidative stress and microbiota dysbiosis

Yuyang Ma,Xiaotian Wang,Huan He,Xiangxuan Wang,Xinbin Zhang,Long Zhu,Shiqi Fan,Yufeng Shen,Chengke Zhu

Microplastics and pesticides often coexist in freshwater environments; however, their combined effects on teleost branchial tissues remain unclear. In this study, adult Gobiocypris rarus were exposed to polystyrene microplastics (PS-MPs, 1 mg/L), permethrin (PM, 0.5 μg/L), or their combination for 21 days. Gill toxicity was evaluated using histopathology, scanning electron microscopy, biochemical assays, immunofluorescence staining, transcriptomics, and 16S rRNA sequencing. Our results revealed that both single and co-exposure manifested as sublethal toxicity in gill tissues, with co-exposure generally inducing more pronounced and widespread adverse effects. Histological and ultrastructural observations showed that co-exposure exacerbated lamellar deformation, epithelial vacuolation, microridge disruption, and epithelial surface damage. 8-OHdG staining revealed the most severe oxidative DNA damage in the PM group. Biochemical analyses showed decreased superoxide dismutase (SOD) activity in all exposure groups and increased malondialdehyde (MDA) levels in PM-containing groups. TUNEL analysis showed the most intense apoptotic signal in the co-exposure group. Transcriptomic analysis revealed that co-exposure induced the largest number of differentially expressed genes and mainly affected lipid transport, membrane-related metabolism, programmed cell death, immune regulation, and PPAR signaling. In addition, 16S rRNA sequencing showed that co-exposure disrupted the gill-associated microbiota, with a loss of dominant commensal bacteria and an increase in opportunistic taxa. Combined analysis further showed that microbial changes were closely associated with oxidative injury, apoptosis-related responses, and immune-related indicators. Overall, these findings indicate that PS-MPs and PM co-exposure exacerbates gill toxicity through multi-level injury processes, providing a theoretical basis for evaluating the ecological risks of microplastic–pesticide co-contamination in freshwater fish.
