A fluorescent “turn off–on” pattern for the detection of herring sperm DNA (hsDNA) had been designed through utilizing the interaction between polymyxin B sulfate (PMBS) and hsDNA as an inherent performance and the fluorescent transformation of glutathione (GSH)-capped CdTe quantum dots (QDs) as an external manifestation. Due to the occurrence of the photoinduced electron transfer from the QDs to PMBS, the fluorescence of GSH-capped CdTe QDs could be effectively quenched by PMBS, causing the system into “off” state. With the addition of hsDNA, the quenched fluorescence of GSH-capped CdTe QDs could be restored for the reason that PMBS embedded into hsDNA double helix structure to form new complex and peeled off from the surface of GSH-capped CdTe QDs, leading the system into “on” condition. Corresponding experimental results illustrated that the relative recovered fluorescence intensity of GSH-capped CdTe QDs–PMBS system was near proportional to the concentration of hsDNA within the range of 0.059–15.0 μg mL −1 . This proposed method demonstrated a good linear correlation coefficient of 0.9937 and a detection limit (3 σ / K ) of 0.018 μg mL −1 for hsDNA. This dual-directional fluorescent biosensor overcame the selectivity problem commonly existed in the traditional mono-directional fluorescence detection mode and owned perfect analysis applications in biochemical DNA monitoring.

Disinfection by-products (DBPs) formed in chlorination and chloramination are proved to be cytotoxic and genotoxic and arouse increasing attention. However, previous studies of DBP precursors mainly focused on free amino acids (AAs) and few papers evaluated DBPs' formation potential of combined AAs. This study demonstrated that typical carbonaceous (C-) DBPs, trihalomethanes (THMs) and typical nitrogenous (N-) DBPs, dichloroacetonitrile (DCAN), trichloroacetonitrile (TCAN) and trichloronitromethane (TCNM) could be formed during chlorination and chloramination of polymyxin B sulfate (PBS), a common polypeptide antibiotic working against Gram-negative bacterial infections. The effects of major parameters, including disinfectant dose, contact time, solution pH, temperature, bromide concentration and chloramination mode were evaluated in batch experiments. Different kinds of DBPs exhibited different characteristics as disinfectant dose or contact time increased. Solution pH and temperature affected the formation of DBPs greatly. The formation pathways of different DBPs from chlor(am)ination of PBS were also proposed. Combined AAs, such as PBS, were proved to be important precursors of DBPs during disinfections.

Pulmonary infections caused by multidrug-resistant bacteria have become a significant threat to human health. Bacterial biofilms exacerbate the persistence and recurrence of pulmonary infections, hindering the accessibility and effectiveness of antibiotics. In this study, a dry powder inhalation (DPI) consisting of polymyxin B sulfate (PMBS) inhalable microparticles and high-lectin-affinity (HLA) sugar (i.e., raffinose) carriers was developed for treating pulmonary infections and targeting bacterial lectins essential for biofilm growth. The formulated PMBS-HLA DPIs exhibited particle sizes of approximately 3 μm, and surface roughness varied according to the drug-to-carrier ratio. Formulation F5 (PMBS: raffinose = 10: 90) demonstrated the highest fine particle fraction (FPF) value (64.86%), signifying its substantially enhanced aerosol performance, potentially attributable to moderate roughness and smallest mass median aerodynamic particle size. The efficacy of PMBS-HLA DPIs in inhibiting biofilm formation and eradicating mature biofilms was significantly improved with the addition of raffinose, suggesting the effectiveness of lectin-binding strategy for combating bacterial biofilm-associated infections. In rat models with acute and chronic pulmonary infections, F 5 demonstrated superior bacterial killing and amelioration of inflammatory responses compared to spray-dried PMBS (F 0 ). In conclusion, our HLA carrier-based formulation presents considerable potential for the efficient treatment of multidrug-resistant bacterial biofilm-associated pulmonary infections.

The clinical application of polymyxin B (PMB) is limited by its nephrotoxic effects, making the reduction of PMB-induced nephrotoxicity has become a pressing concern for clinicians. Tetrahydrocurcumin (THC), known for its beneficial characteristics in biological functions, presents an attractive option for intervention therapy to mitigate PMB-induced nephrotoxicity. However, the underlying mechanism of how THC mitigates PMB-induced nephrotoxicity is still poorly understood. Here, we first evaluated the potential of THC intervention therapy to mitigate PMB-induced nephrotoxicity in an in vitro model of PMB-induced cell injury. Moreover, we demonstrated that THC effectively protected HK-2 cells from PMB-induced apoptosis by using cell counting kit-8 and flow cytometry assay. THC could also suppress PMB-induced endoplasmic reticulum (ER) stress via PERK/eIF2α/ATF4/CHOP pathway. In addition, using PERK inhibitor GSK2606414 to inhibit ER stress also alleviated PMB-induced apoptosis. Taken together, these findings provide novel insights that THC possesses the ability to alleviate PMB-induced nephrotoxicity by inhibiting the ER stress-mediated PERK/eIF2α/ATF4/CHOP axis, which sheds light on the benefits of THC as an intervention strategy to reduce PMB-induced nephrotoxicity, thus providing a potential avenue for improved clinical outcomes in patients receiving PMB treatment.

Thioglycolic acid-capped (TGA-capped) CdTe/CdS quantum dots (QDs) were prepared in the water phase. The interaction of TGA-CdTe/CdS QDs with polymyxin B sulfate (POBS) was investigated by ultraviolet–visible (UV–vis) absorption, fluorescence (FL), resonance Rayleigh scattering (RRS) spectroscopy and the measurement of fluorescence lifetime . Under optimum conditions, not only the UV–vis absorption and fluorescence spectra were changed, but also the RRS intensity was greatly enhanced. These phenomena offered useful techniques for the determination of POBS with fluorescence quenching and RRS methods . The linear ranges and detection limits of POBS were 0.09–5.25 μg mL −1 and 27.5 ng mL −1 for the fluorescence quenching technique, 0.02–6.0 μg mL −1 and 6.36 ng mL −1 for the RRS technique. Among them, the RRS approach obtained the higher sensitivity. Accordingly, a novel rapid, convenient, and highly sensitive RRS method for the determination of POBS was proposed and applied to detect POBS in pharmaceutical preparations with satisfactory results. Furthermore, the suitable reaction conditions, affecting factors as well as the influence of coexisting substances were studied. Some possible reaction mechanisms were also discussed.
