Serratia marcescens, a gram-negative bacterium, found in a wide range of ecological niches can produce several high-value products, including prodigiosin, althiomycin, and serratamolide. Among them, prodigiosin has attracted attention due to its immunosuppressive, antimicrobial, and anticancer properties. However, the regulatory mechanisms behind prodigiosin synthesis in Serratia marcescens remains limited. Here, a transposon mutant library was constructed to identify the genes related to prodigiosin synthesis, and BVG90_02415 gene encoding a peptidoglycan synthesizing enzyme D-Ala-D-Ala carboxypeptidase DacA was found to negatively regulates prodigiosin synthesis. Quantitative measurements revealed that disruption of dacA increased prodigiosin production 1.46-fold that of the wild-type strain JNB5-1 in fermentation medium. By comparing differences in cell growth, pigA gene expression level, cell morphology, membrane permeability, and intracellular prodigiosin concentration between wild-type strain JNB5-1 and dacA mutant SK4-72, results revealed that the mechanism for hyper-producing of prodigiosin by the dacA mutant was probably that dacA disruption enhanced prodigiosin leakage, which in turn alleviated feedback inhibition of prodigiosin and increased expression of pig gene cluster. Collectively, this work provides a novel insight into regulatory mechanisms of prodigiosin synthesis and uncovers new roles of DacA protein in regulating cell growth, cell morphology, and membrane permeability in Serratia marcescens. Finally, this study offers a new strategy for improving production of high-value compounds in Serratia marcescens.

The recurrence of drug-resistant and expensive treatment drugs are major causes of the low survival rate of multiple myeloma (MM) patients. Exploring a safe, effective, low-cost and novel drug treatment for MM is a promising strategy to relieve the burden of MM patients. In this study, we found that prodigiosin could inhibit MM cell proliferation and induce MM cell apoptosis, however, it had a lesser cytotoxic effect on normal B cells within the IC 50 range of MM cells. In addition, prodigiosin could inhibit the growth of xenograft MM cells in mice. Transcriptomics and targeted amino acid metabolomics confirmed that prodigiosin could regulate amino acid metabolism, and decrease in amino acid utilization by down-regulated aminoacyl tRNA synthetases expression, resulting in slower growth of MM. In conclusion, prodigiosin exerts anticancer effects on MM cells by interfering with the use of amino acids, indicating its potential novel therapeutic application in MM.

Py-Pro undergoes Type I photobleaching via superoxide anion radical generation. • NBT assays and EPR spectra confirmed light-induced ROS formation in the Py-Pro system. • Replacing pyrrole donors with thiophene or phenyl groups improved photostability. • DFT calculations correlated donor strength with ROS generation and photodegradation. Prodigiosin, a promising microbial pigment for sustainable textile dyeing, suffers from poor lightfastness, the mechanistic origin of which remains poorly understood. Herein, a synthetic analogue (Py-Pro) was employed to elucidate its photodegradation behavior. Under simulated illumination, Py-Pro undergoes self-sensitized photobleaching via a Type I pathway, generating superoxide anion radicals ( O 2 − · ). HPLC-MS analysis identified the major degradation products, supporting oxidative cleavage of the conjugated chromophore. Replacing the electron-rich pyrrole donor with thiophene and phenyl groups yields two analogues (Thio-Pro and Ph-Pro), which exhibit enhanced photostability, reducing degradation from 57.48% (Py-Pro) to 17.57% and 11.41%, respectively. Density functional theory (DFT) calculations reveal that the strong donor character promotes intramolecular charge transfer, reduces the S 1 -T 2 energy gap, and increases spin-orbit coupling, thereby facilitating intersystem crossing and enhancing O 2 − · generation. These findings provide mechanistic insight and a rational strategy for improving the photostability of natural dyes. Healing of infected chronic wounds faces dual challenges: persistent inflammation and impaired angiogenesis. To address these, SN-PB@PG nanocomplexes were prepared by hybridisation of nitroprusside (SNP) with Prussian blue (SN-PB NPs) and loaded with prodigiosin (PG). Under near-infrared (NIR) irradiation, SN-PB NPs generated mild hyperthermia, facilitating the release of nitric oxide (NO) and PG to combat bacterial biofilms and multidrug-resistant pathogens. The in vivo assay using diabetic infected wounds demonstrated that SN-PB@PG NPs with NIR reduced the wound area to 10.6% by the 11th day, which is superior to that of control group (29.6%). In the flap transplantation experiments, the data showed SN-PB@PG NPs with NIR group only have a necrobiosis of 3.8% of flaps on the 8th day, which is superior to 31.3% of the control group. Additionally, the release of NO promoted vascular regeneration by up-regulating vascular endothelial growth factor (VEGF) and platelet endothelial cell adhesion molecule-1 (CD31), and regenerated sarcomeric tissue by down-regulating MMP-9. The results indicated that the combination of SN-PB@PG NPs with gas and photothermal therapy exerted a combined antibacterial and wound healing effect. Compared to traditional clinical methods such as surgical debridement and hyperbaric oxygen therapy, this new strategy efficiently addresses issues of infection and healing, which is convenient for clinical application.

Polylactic acid (PLA), a biodegradable polymer derived from renewable resources, represents a promising candidate for sustainable textiles. Nevertheless, its practical application remains limited by the requirement for high-temperature dyeing, which can induce polymer hydrolysis and lead to the loss of fiber strength. To address this limitation, PLA fabric was treated with an eco-friendly natural deep eutectic solvent (NaDES) composed of glycerol and citric acid. The treatment was found to enhance fiber surface roughness and internal looseness, which facilitated dye diffusion and allowed for a significant reduction in dyeing temperature. When dyed with microbial prodigiosin, the treated PLA fabric achieved a color depth at 70 °C that was equivalent to untreated fabric at 90 °C, while also exhibiting a 93.56% bacteriostatic rate against Staphylococcus aureus due to the inherent antibacterial property of microbial prodigiosin. This work provides a novel and sustainable strategy for the eco-friendly dyeing of PLA textiles.
