Preparation and physicochemical characterization of inclusion complex of pterostilbene-hydroxypropyl-β-cyclodextrin for enhancing the application of pterostilbene in cosmetics

Dan Yang,Hui Liu,Hong Meng,Shujing Li

Pterostilbene (PTS) is a kind of natural plant polyphenol with a high antioxidant capacity. However, it is a challenge to apply PTS in cosmetic formulations due to its lower water solubility and stability. In order to overcome these limitations, a PTS-hydroxypropyl-β-cyclodextrin (PTS-HP-β-CD) inclusion complex with a 1:1 host–guest molar ratio was prepared using the stirred solution method. The inclusion complex of PTS-HP-β-CD was characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), proton nuclear magnetic resonance ( 1 H NMR), two-dimensional rotating frame nuclear Overhauser effect spectroscopy (2D ROESY), thermogravimetric analysis (TG), and differential scanning calorimetry (DSC). Additionally, the photostability and antioxidant activity of PTS and PTS-HP-β-CD were also evaluated. After complexation with HP-β-CD, the water solubility of PTS was found to be 81-fold higher by phase solubility. The results of XRD, SEM, 1 H NMR, and 2D ROESY demonstrated that PTS was successfully encapsulated by HP-β-CD and formed a 1:1 inclusion complex. After the formation of the PTS-HP-β-CD inclusion complex, TG, DSC, UV irradiation, DPPH radical scavenging activity, hydroxyl radical scavenging activity, and superoxide anion radical scavenging activity revealed that the stability and antioxidant activity of PTS were enhanced. Therefore, this work could provide a promising approach to improve the application of PTS or other polyphenolic substances in cosmetic formulations.

Encapsulation of pterostilbene in HP-β-CD via HPH and pH-shift method

Xuanyi Zhang,Xiaojun Lin,Jingwen Wang,Xiaoyu Luo,Baojun Xu

This study focused on enhancing the stability and bioaccessibility of pterostilbene (PTS) by encapsulating it with hydroxypropyl-β-cyclodextrin (HP-β-CD) using high-pressure homogenization (HPH) and pH-shift methods. The encapsulation process was performed at pressures of 50, 100, and 150 MPa along with three different treatments, achieving a maximum encapsulation efficiency of 94.34% and a loading capacity of 14.82% at 150 MPa. The SEM, XRD, and FTIR analyses confirmed encapsulation by revealing significant morphological changes, crystal structure modifications, and host-guest interactions. PTS-HP-β-CD complexes retained 82.22% of PTS at 4°C (28 days) and 76% under UVB (120 min), compared to 56.03% and 21% for free PTS, respectively. Additionally, these complexes exhibited enhanced DPPH scavenging rates (44.95% at 0.02 mg/mL), nearly twice that of free PTS. In the release studies, the complexes’ release behavior aligned with the Higuchi model. Encapsulated PTS revealed lower digestion rates in in-vitro gastric (32.24% vs. 49.88%) and intestinal phases (63.98% vs. 91.01%). Additionally, there was no observable toxicity for PTS-HP-β-CD to human epidermal keratinocytes, supporting its potential use in cosmetics, food, and pharmaceuticals. These results demonstrated that HPH combined with pH-shift methods effectively improved the stability and bioaccessibility of PTS, offering a promising strategy for its formulation.

Transcriptomic and Physiological Analysis of the Effects of Exogenous Phloretin and Pterostilbene on Resistance Responses of Stylosanthes against Anthracnose

Shizi Zhang,Yunfeng Xu,Fang Wang,Liyun Yang,Lijuan Luo,Lingyan Jiang

Anthracnose caused byColletotrichum gloeosporioidesis a destructive disease ofStylosanthes(stylo). Combination treatment of phloretin and pterostilbene (PP) has been previously shown to effectively inhibit the conidial germination and mycelial growth ofC. gloeosporioidesin vitro. In this study, the effects of PP treatment on the growth ofC. gloeosporioidesin vivo and the biocontrol mechanisms were investigated. We found that exogenous PP treatment could limit the growth ofC. gloeosporioidesand alleviate the damage of anthracnose in stylo. Comparative transcriptome analysis revealed that 565 genes were up-regulated and 239 genes were down-regulated upon PP treatment during the infection byC. gloeosporioides. The differentially expressed genes were mainly related to oxidative stress and chloroplast organization. Further physiological analysis revealed that application of PP afterC. gloeosporioidesinoculation significantly reduced the accumulation of O2•−level and increased the accumulation of antioxidants (glutathione, ascorbic acid and flavonoids) as well as the enzyme activity of total antioxidant capacity, superoxide dismutase, catalase, glutathione reductase, peroxidase and ascorbate peroxidase. PP also reduced the decline of chlorophyll a + b and increased the content of carotenoid in response toC. gloeosporioidesinfection. These results suggest that PP treatment alleviates anthracnose by improving antioxidant capacity and reducing the damage of chloroplasts, providing insights into the biocontrol mechanisms of PP on the stylo against anthracnose. Keywords:phloretin;pterostilbene;ROS scavenging;chloroplasts;anthracnose;stylo

Zonghui Tang,Panchen Fang,Yuting Tao,Qiuye Huang,Xuefei Xu,Xiaoyan Cheng,Faraz Hussain,Xueling Li,Jin Liang,Shuming Ye,Yue Sun

To explore the potential applications of pea protein isolate-pectin-pterostilbene complex (PPI-PEC-PT) in the sauce industry, its solubility, antioxidant capacity and oxidative stability were measured. The results indicated that PPI-PEC-PT exhibited a solubility of 77.67 %, more than double that of PPI-PT (31.93 %). The DPPH radical scavenging capacity of PPI-PEC-PT reached up to 78.52 % at a concentration of 50 μg/mL. After accelerated oxidation (60 °C, 7 days), the peroxide value (8.15 mmol/L) and malondialdehyde content (11.50 mmol/L) of PPI-PEC-PT emulsion were significantly lower than those of PPI. Additionally, when applied to walnut butter, PPI-PEC-PT significantly enhanced its gel properties. In conclusion, PPI-PEC-PT could not only serve as a natural antioxidant but also be utilized as a food additive to enhance the gelling properties of products in the food industry.

Pterostilbene-Loaded Soluplus/Poloxamer 188 Mixed Micelles for Protection against Acetaminophen-Induced Acute Liver Injury

Kehong Dong,Mei Zhang,Ying Liu,Xintao Gao,Xiaochen Wu,Dayong Shi,Chuanlong Guo,Jing Wang

Excessive acetaminophen (APAP) induces excess reactive oxygen species (ROS), leading to liver damage. Pterostilbene (PTE) has excellent antioxidant and anti-inflammatory activities, but poor solubility limits its biological activity. In this study, we prepared PTE-loaded Soluplus/poloxamer 188 mixed micelles (PTE-MMs), and the protective mechanism against APAP-induced liver injury was investigated. In vitro results showed that PTE-MMs protected H2O2-induced HepG2 cell proliferation inhibition, ROS accumulation, and mitochondrial membrane potential destruction. Immunofluorescence results indicated that PTE-MMs significantly inhibited H2O2-induced DNA damage and cGAS-STING pathway activation. For in vivo protection studies, PTE-MMs (25 and 50 mg/kg) were administered orally for 5 days, followed by APAP (300 mg/kg). The results showed that APAP significantly induced injury in liver histopathology as well as an increase in serum aspartate aminotransferase and alanine aminotransferase levels. Moreover, the above characteristics of APAP-induced acute liver injury were inhibited by PTE-MMs. In addition, APAP-induced changes in the activities of antioxidant enzymes such as SOD and GSH in liver tissue were also inhibited by PTE-MMs. Immunohistochemical results showed that PTE-MMs inhibited APAP-induced DNA damage and cGAS-STING pathway activation in liver tissues. For in vivo therapeutic effect study, mice were first given APAP (300 mg/kg), followed by oral administration of PTE-MMs (50 mg/kg) for 3 days. The results showed that PTE-MMs exhibited promising therapeutic effects on APAP-induced acute liver injury. In conclusion, our study shows that the Soluplus/poloxamer 188 MM system has the potential to enhance the biological activity of PTE in the protection and therapeutic of liver injury.

The enhancement of surimi gels by utilizing coconut oil pre-emulsion loaded with pterostilbene and tempo modified cellulose

Fang Tian,Wenyuchu Chen,Weiliang Guan,Yujin Li,Zan Ni,Chongsheng Zhang,Xiaona Wang,Luyun Cai

Surimi is widely appreciated for its preferred texture and nutritional profile. However, maintaining its superior gel-forming properties while preventing nutrient loss caused by the washing process remains challenging. In this study, pre-emulsions made from pea protein isolate (PPI), pterostilbene (PTE), TEMPO-oxidized cellulose (TOC), and coconut oil (CO) were used as a fat substitute to improve the gel properties of surimi. These were incorporated into brown croaker ( Miichthys miiuy ) surimi. The results showed that the PPI-CO-TOC-PTE pre-emulsion (P-C-T-P) had a small d 4,3 particle size (283.5 nm). Molecular docking simulations demonstrated that TOC enhanced hydrogen bonding interactions between PPI and PTE. Adding P-C-T-P significantly improved the gel strength (2002.33 ± 48.74 g × mm) and water-holding capacity (91.40 ± 0.37 %) of the surimi gels. Furthermore, the protein network structure was denser in the P-C-T-P group. The surimi with P-C-T-P showed higher storage and loss moduli and the highest levels of hydrogen bonding, hydrophobicity, and β-sheet content. Thus, P-C-T-P addition improved surimi gel properties compared with directly supplying lipids to surimi.

A new antioxidant made from a pterostilbene functionalized graphene nanocomposite as an efficient treatment for dry eye disease

Lin Mimi,Sun Xueqin,Ye Sihao,Chen Youyi,Gao Jing,Yuan Feng,Lin Na,Lawson Tom,Liu Yong,Deng Ruzhi

Dry eye disease is a common condition that affects the eyes. It is caused by problems with the tear film and the tear dynamics. Dry eye can be caused by an increase in the amount of reactive oxygen species (ROS) in the corneal epithelium. The treatment for dry eye typically focuses on relieving the uncomfortable symptoms by using eye drops such as artificial tears, antibiotics, and by using anti-inflammatory/immunosuppressive agents such as cyclosporine, and lifitegrast. However, the recovery of patients with dry eye can take several years particularly if the symptoms are severe. This is because the present treatment approaches for dry eye are not based on its cause, e.g., the oxidative stress arising from the rapid increase in ROS. This work describes a new type of antioxidant made from pterostilbene (PS) and carboxyl-chitosan modified graphene (CG). The use of a hydrophilic two-dimensional CG nanosheet to improve the properties of PS is reported. Superior enhanced properties including better cellular permeability, long sustained release period (over 30 h), and antioxidant properties, were realized by using PS-CG. A hyperosmotic (HS) damaged human corneal epithelial cell (HCEC) model was used for antioxidant tests. This model has an intracellular ROS level 4 times more than that of a control group. The ROS content was declined efficiently to the same amount as normal cells in the PS-CG treated HS group. There was a significant decline in the content of lactate dehydrogenase (LDH) and the apoptosis rate of HCEC in the PS-CG treated HS group when compared to that seen in the HS model. Real-time polymerase chain reaction (PCR) and western blots (WB) were used to understand the antioxidant mechanism of PS-CG. The results showed that the antioxidant was working by activating the Keap1-Nrf2-ARE signalling pathway. In vivo testing testing using a dry eye mouse model suggested that the PS-CG acted as an efficient antioxidant. More tear production and healthier corneal and conjunctival epithelial cells were achieved when PC-CG was applied to this model. The use of PS-CG could be a new strategy for treating dry eye and other ocular diseases caused by ROS.

Pterostilbene attenuates the proliferation and differentiation of TNF‑α‑treated human periodontal ligament stem cells

Min Yi,Guanglei Wang,Jianhua Niu,Minghui Peng,Yi Liu

Periodontitis is a common chronic inflammatory oral disease. The objective of periodontal treatment is to control infection whilst regenerating damaged periodontal tissue. The present study aimed to determine the potential effects of pterostilbene (PTE), a representative stilbene compound, on the proliferation and differentiation of human periodontal ligament stem cells (hPDLSCs). Different concentrations (1, 5, 10 and 20 µM) of PTE were applied to hPDLSCs, after which Cell Counting Kit‑8 and western blotting assays were performed to examine the protein levels of Ki67, PCNA, p‑IκBα, IκBα, Bcl‑2, Bax, cleaved caspase3. The effect of PTE on the release of inflammatory factors, including IL‑1β, IL‑6 and IL‑10 was assessed by RT‑qPCR. The apoptosis of TNF‑α‑induced hPDLSCs was evaluated by TUNEL assay and western blotting. Additionally, the role of PTE in hPDLSC mineralization was evaluated using alizarin red staining. The expression levels of mineralization indices, including RUNX2 and ALP were subsequently determined using western blotting. Subsequently, the target of PTE was predicted using TargetNet database and AutoDock v4.2 software and verified using western blotting. The results of the present study revealed that PTE promoted the proliferation of hPDLSCs in a concentration‑dependent manner. Furthermore, PTE treatment decreased the release of inflammatory factors and alleviated the apoptosis of TNF‑α‑induced hPDLSCs. PTE was also demonstrated to promote the formation of mineral nodules in TNF‑α‑induced hPDLSCs. The Targetnet database, along with molecular docking, indicated that histone deacetylases (HDACs) were the probable targets of PTE upstream of regulating periodontitis. The results of western blotting implied that TNF‑α significantly increased expression levels of HDAC2, 4, 6 and 8, whilst PTE treatment markedly decreased HDAC4, 6 and 8 expression in a concentration‑dependent manner compared with the TNF‑α group, which further confirmed these conclusions. In summary, results of the present study revealed that PTE promoted TNF‑α‑induced hPDLSC proliferation and differentiation, whilst alleviating inflammation and apoptosis. PTE also inhibited the expression of HDACs, which may be involved in the mechanism of periodontitis.

The antibacterial mechanism of pterostilbene derived from xinjiang wine grape: A novel apoptosis inducer in Staphyloccocus aureus and Escherichia coli

Xueyan Ren,Peipei An,Xin Zhai,Shuo Wang,Qingjun Kong

Antibiotic overuse has caused concerns owing to the emergence of antibiotic resistant microorganisms and the accompanying decreased bacteria-inhibiting effectiveness of proffered treatments. Pterostilbene, derived from XinJiang wine grape, is a natural polyphenol, and has attracted considerable attention for its promising antifungal effects. However, its inhibitory activities and mechanisms of action against bacteria are still unknown. In this study, pterostilbene inhibited the growth of Gram-positive Staphyloccocus aureus and Gram-negative Escherichia coli effectively. Electron microscope observations showed that the morphological structures of S. aureus and E. coli were significantly damaged by pterostilbene. Cells became irregular, shriveled, and lysed, while nucleic acids and proteins leaked, and DNA content decreased. Furthermore, confocal laser scanning microscopy experiments revealed that bacteria treated with pterostilbene produced reactive oxygen species, and the cytomembrane potential increased. To further clarify the molecular mechanism, changes in the expression levels of four genes in two bacteria were confirmed by qRT-PCR. Pterostilbene cause the up-regulation of DinF (an oxidative stress gene). An associated down-regulation of PaL, MtgA, and NagA (genes involved in cell wall synthesis) is also attributed to pterostilbene treatment. This is the first time that the mechanism of pterostilbene against S. aureus and E. coli is presented in literature.

Pterostilbene prevents methylglyoxal-induced cytotoxicity in endothelial cells by regulating glyoxalase, oxidative stress and apoptosis

Dan Tang,Wei Xiao,Wen-Ting Gu,Zhi-Tong Zhang,Shu-Hong Xu,Zhi-Quan Chen,You-Hua Xu,Lu-Yong Zhang,Shu-Mei Wang,Hong Nie

Methylglyoxal (MGO), a cytotoxic byproduct of glycolysis in biological systems, can induce endothelial cells dysfunction, implicated in diabetic vascular complications. Pterostilbene (PTS), a naturally occurring resveratrol derivative, is involved in various pharmacological activities . This study aimed to explore the effects of PTS on MGO induced cytotoxicity in human umbilical vein endothelial cells (HUVECs) and the underlying mechanisms for the first time. In the current study, it has been demonstrated that PTS could enhance the level of glyoxalase 1 (GLO-1) and elevate glutathione (GSH) content to active the glyoxalase system, resulting in elimination of the toxic MGO as well as advanced glycation end products (AGEs) in HUVECs. Meanwhile, PTS could also suppress oxidative stress and thus exert cytoprotective effects by elevating Nrf2 nuclear translocation and the corresponding down-stream antioxidant enzymes in MGO induced HUVECs. In addition, PTS could alleviate MGO induced apoptosis in HUVECs via inhibition of oxidative stress and associated downstream mitochondria-dependent signaling apoptotic cascades, as characterized by preventing caspases family activation. Taken together, these findings suggest that PTS could protect against MGO induced endothelial cell cytotoxicity by regulating glyoxalase, oxidative stress and apoptosis, suggesting that PTS could be beneficial in the treatment of diabetic vascular complications.

Pterostilbene Eliminates MRSA Independent of Metabolic State and Effectively Prevents Biofilm Formation in Milk Matrices

Xiaoyong Huang,Huiting Yang,Chenli Wang,Shiqi Yan,Xiaomin Ren,Zilong Sun

The relentless spread of antimicrobial resistance poses a severe threat to global public health, food safety, and environmental security. Natural products with potent antimicrobial activity offer prospective substitutes for traditional antibiotics and chemical preservatives. Here, we demonstrate that pterostilbene (PT), a natural dietary compound, exhibits rapid lytic activity against methicillin-resistantStaphylococcus aureus(MRSA). PT displayed metabolism-independent bactericidal effects, effectively eradicating dormant persister cells within one hour, though its activity was partially attenuated under anaerobic conditions. Mechanistically, PT disrupted membrane integrity by increasing permeability, dissipating membrane potential, depleting cellular ATP, and suppressing reactive oxygen species (ROS) generation. Its efficacy was modulated by membrane phospholipid composition, with phosphatidylglycerol (PG) and cardiolipin (CL) critically influencing antimicrobial potency. Crucially, PT robustly inhibited MRSA biofilm formation in milk. These findings highlight PT’s potential as a structurally stable, natural antimicrobial for controlling resilient MRSA, particularly against biofilm-associated and persister subpopulations in food systems.

Whole-process biomimetic synthesis of melanin-like amino acid surfactants by 2-iodoxybenzoic acid mediated regioselective oxidation of pterostilbene

Hong Zhou,Yinna Wu,Xiao Song,Junwei Zhu,Wenbin Liu

Amino acid surfactants have garnered increasing attention as green and safe alternatives. Bioinspired by the melanogenesis pathway, this study developed a novel melanin-like amino acid surfactant with a melanin mimetic structure by conjugating glycine to o-quinone. Pterostilbene, a versatile natural monophenol, was oxidized to form o-quinone crystals by 2-iodoxybenzoic acid in a manner analogous to tyrosinase. The molecular design achieved whole-process of melanin biomimetics in four aspects: substrate, oxidant, synthetic route, and target product structure. The chemical structure was identified using Fourier-transform infrared spectroscopy, mass spectrometry, and nuclear magnetic resonance spectroscopy. Melanin-like amino acid surfactant exhibited ultralow critical micelle concentration of 70 nM, along with excellent performance in emulsifying, dispersing, foaming, solubilizing, and wetting. It featured exceptional multifunctionalities, including low irritation, biocompatibility, antioxidant activity, and antibacterial activity. This design enables biomimetic synthesis of multifunctional surfactants using any monophenol substrate, expanding reaction accessibility and structural diversity for amino acid surfactant.
